Systems Metabolic Engineering of Clostridium tyrobutyricum for 1,3-Propanediol Production From Crude Glycerol.

Clostridium tyrobutyricum has emerged as a non-pathogenic microbial cell factory capable of anaerobic production of various value-added products, such as butyrate, butanol, and butyl butyrate. This study reports the first systematic engineering of C. tyrobutyricum for the heterologous production of 1,3-propanediol (1,3-PDO) from industrial by-product crude glycerol. Initially, the glycerol reductive pathway for 1,3-PDO production was constructed, and the unique glycerol oxidation pathway in C. tyrobutyricum was elucidated. Subsequently, the glycerol metabolism and 1,3-PDO synthesis pathways we

Mechanisms of Clostridium tyrobutyricum removal through natural creaming of milk: A microscopy study.

Clostridium tyrobutyricum is the main spoilage agent of late blowing defect (LBD) in Grana Padano and Parmigiano-Reggiano cheeses; LBD is characterized by openings and holes and is sometimes accompanied by cracks and an undesirable flavor. Even a very few spores remaining in the cheese curd may cause LBD; thus, it is essential to eradicate them during milk natural creaming. By this process, most of the bacteria, somatic cells, and spores rise to the top of the milk, together with the fat globules, and are removed with the cream. Previous studies suggested that milk immunoglobulins mediate the

Development of an in vivo fluorescence based gene expression reporter system for Clostridium tyrobutyricum.

C. tyrobutyricum, an acidogenic Clostridium, has aroused increasing interest due to its potential to produce biofuel efficiently. However, construction of recombinant C. tyrobutyricum for enhanced biofuel production has been impeded by the limited genetic engineering tools. In this study, a flavin mononucleotide (FMN)-dependent fluorescent protein Bs2-based gene expression reporter system was developed to monitor transformation and explore in vivo strength and regulation of various promoters in C. tyrobutyricum and C. acetobutylicum. Unlike green fluorescent protein (GFP) and its variants, Bs2

Integrating Metabolic Modeling and Targeted Supplementation for the Rapid Detection of Clostridium tyrobutyricum in Dairy Products.

Clostridium tyrobutyricum is a major cause of late blowing defects (LBDs) in cheese, resulting in substantial economic losses. Early detection is critical for maintaining product quality. In this study, we developed a rapid detection approach integrating genome-scale metabolic modeling (GEM) with systematic culture optimization. Three media were evaluated, identifying RCM at 38.5 °C as the optimal condition for reducing the lag phase. Flux Balance Analysis (FBA) revealed that targeted supplementation with magnesium, zinc, Vitamin B6, and L-tryptophan significantly enhanced metabolic flux throu

Engineering Clostridium Tyrobutyricum for High Butanol Production through Induction Expression of Exogenous NADPH-Dependent HBD.

Clostridium tyrobutyricum Δcat1::adhE2 is a promising cell factory for butanol production because of its robustness, high butanol tolerance, and minimal butyrate production. However, excessive acetate and ethanol production remains a major bottleneck limiting its butanol yield. Coexpressing an exogenous hbd(Ck) encoding the NADPH-dependent 3-hydroxybutyryl-CoA dehydrogenase (HBD) from Clostridium kluyveri with adhE2 could increase the C4 carbon flux, resulting in increased butanol and decreased acetate and ethanol production. However, constitutively overexpressing hbd(Ck) in Δcat1::adhE2 shows

Non-sterile fermentation of butyl butyrate by Clostridium tyrobutyricum with an anti-microbial contamination system.

Microbial contamination is a major challenge in industrial fermentation, while conventional sterilization is energy-intensive and costly. Here, an anti-contamination system was developed in Clostridium tyrobutyricum for non-sterile production of butyl butyrate (BB). C. tyrobutyricum L319 grow on unconventional nitrogen sources, namely formamide (50 mM) and urea (20 mM), as well as phosphite (50 mM) as the sole phosphorus source. Heterologous expression of phosphite dehydrogenase (ptxD) further enhanced phosphite utilization and raised the intracellular level of reducing equivalents. The engine

A metabolic relay for caproic acid production in Huangshui: Cooperative interaction between Clostridium tyrobutyricum and Caproiciproducens.

Microbial chain elongation for medium-chain fatty acid production has gained significant attention. This study elucidated how initial pH and carbon sources regulate microbial caproic acid production from organic waste. By successively subculturing pit mud with Huangshui, lactic acid, or ethanol at different pH values (4.0-6.0), we obtained two enriched consortia: NatCom H (from Huangshui) and NatCom E (from ethanol), both of which produce 10 g/L caproic acid at pH 6.0. Key functional divergence was observed: NatCom H maintained production from pH 5.0 to 6.0, whereas NatCom E was functional onl

Integrated co-cultivation and subsequent esterification: Harnessing Saccharomyces cerevisiae and Clostridium tyrobutyricum for streamlined ester production.

The rising demand for natural products is accelerating research into sustainable methods for producing bio-based flavourings like ethyl butyrate. In this study, ethyl butyrate was successfully produced through the enzymatic esterification of butyric acid and ethanol, which were derived from the co-cultivation of Clostridium tyrobutyricum and Saccharomyces cerevisiae. Initial monoculture experiments with both strains were performed to investigate compromised fermentation conditions for co-cultivation. Based on these findings, anaerobic co-cultivation conditions were established at 37 °C and 150

Clostridium tyrobutyricum alleviates Staphylococcus aureus-induced endometritis in mice by inhibiting endometrial barrier disruption and inflammatory response.

Endometritis is an inflammatory disease of the uterus caused by bacterial infection, and it affects both human and animal health. This study aims to investigate the protective effects and molecular mechanisms of probiotics such as Clostridium tyrobutyricum (C. tyrobutyricum) on Staphylococcus aureus (S. aureus)-induced endometritis. The results showed that S. aureus infection significantly induced the pathological damage of the uterus, increased the production of pro-inflammatory cytokines, such as TNF-α and IL-1β, and attenuated the expression of tight junction proteins of uterine tissues. Adaptive laboratory evolution of Saccharomyces cerevisiae enhances butyric acid tolerance and enables co-cultivation with Clostridium tyrobutyricum.

Adaptive laboratory evolution (ALE) was successfully applied to improve the tolerance of Saccharomyces cerevisiae toward butyric acid, enabling its use in co-culture with Clostridium tyrobutyricum for the simultaneous production of ethanol and butyric acid as ester precursors. S. cerevisiae was adapted through serial transfer at progressively increasing butyric acid concentrations up to 20 g L⁻1. The evolved yeast population exhibited significantly enhanced butyric acid tolerance and maintained ethanol production under acid-stress conditions. Interestingly, the evolved population also displaye

Metabolic Engineering of Clostridium tyrobutyricum for High-Yield n-Butanol Production by Increasing Intracellular Reducing Equivalent with NADPH-Dependent 3-Hydroxybutyryl-CoA Dehydrogenase.

Clostridium tyrobutyricum was engineered to overexpress adhE2 encoding the aldehyde/alcohol dehydrogenase and an exogenous NADPH-dependent 3-hydroxybutyryl-CoA dehydrogenase from Clostridium kluyveri (Ckhbd) for n-butanol production. In general, large amounts of butyrate, acetate, and ethanol are also produced from glucose when butanol biosynthesis is hindered by limited intracellular NADH pools. In silico flux balance analysis showed that coupling NADP+/NADPH turnover with butanol production increased the reducing equivalent supply and butanol selectivity over ethanol and acids, thus increasi

A Potential Probiotic for Diarrhea: Clostridium tyrobutyricum Protects Against LPS-Induced Epithelial Dysfunction via IL-22 Produced By Th17 Cells in the Ileum.

Probiotics are clinically used for diarrhea and inflammatory bowel diseases in both humans and animals. Previous studies have shown that Clostridium tyrobutyricum (Ct) protects against intestinal dysfunction, while its regulatory function in the gut needs further investigation and the related mechanisms are still not fully elucidated. This study aims to further verify the protective function of Ct and reveal its underlying mechanisms in alleviating diarrhea and intestinal inflammation. Ct inhibited LPS-induced diarrhea and intestinal inflammation in the ileum. IL-22 was identified and the prot

A Potential Prophylactic Probiotic for Inflammatory Bowel Disease: The Overall Investigation of Clostridium tyrobutyricum ATCC25755 Attenuates LPS-Induced Inflammation via Regulating Intestinal Immune Cells.

SCOPE: This study aims to roundly investigate whether Clostridium tyrobutyricum (Ct) alleviates inflammation via regulating immune cells in the small intestines. METHODS AND RESULTS: Mice are pre-treated with different concentrations of Ct follow by LPS stimulation. Ct maintains the mice body weight under inflammation. In response to LPS, 107 CFU mL-1 Ct decreases the mRNA expression of inflammatory cytokines in the duodenum, while 108 CFU mL-1 Ct reduces inflammatory cytokines expression in both duodenum and ileum and protected intestinal morphology. The small intestinal immune cells are ana

Protective effect of Clostridium tyrobutyricum in acute dextran sodium sulphate-induced colitis: differential regulation of tumour necrosis factor-α and interleukin-18 in BALB/c and severe combined immunodeficiency mice.

One of the promising approaches in the therapy of ulcerative colitis is administration of butyrate, an energy source for colonocytes, into the lumen of the colon. This study investigates the effect of butyrate producing bacterium Clostridium tyrobutyricum on dextran sodium sulphate (DSS)-induced colitis in mice. Immunocompetent BALB/c and immunodeficient severe combined immunodeficiency (SCID) mice reared in specific-pathogen-free (SPF) conditions were treated intrarectally with C. tyrobutyricum 1 week prior to the induction of DSS colitis and during oral DSS treatment. Administration of DSS w

Production of isopropyl and butyl esters by Clostridium mono-culture and co-culture.

Production of esters from the acetone-butanol-ethanol (ABE) fermentation by Clostridium often focuses on butyl butyrate, leaving acetone as an undesired product. Addition of butyrate is also often needed because ABE fermentation does not produce enough butyrate. Here we addressed the problems using Clostridium beijerinckii BGS1 that preferred to produce isopropanol instead of acetone, and co-culturing it with Clostridium tyrobutyricum ATCC 25,755 that produced butyrate. Unlike acetone, isopropanol could be converted into ester using lipase and acids. C. tyrobutyricum ATCC 25,755 produced acids

Integrating genomics, phenotypes, and volatile profiling with cheese slurry trials reveals strain-dependent clostridial spoilage originating from silage.

Silage is a primary source of Clostridium spores entering the dairy chain; however, the contributions of spoilage beyond Clostridium tyrobutyricum remain undercharacterized. Here, we integrated whole-genome sequencing with fermentation phenotypes and controlled cheese slurry trials, including volatile profiling measured directly in inoculated slurries, to define strain- and species-specific spoilage modes originating from silage. 15 isolates recovered from silage were confirmed by 16S rRNA sequencing and whole-genome sequencing and analyzed alongside 10 ATCC reference strains for phylogenomic

Enhanced isopropanol and n-butanol production by supplying exogenous acetic acid via co-culturing two clostridium strains from cassava bagasse hydrolysate.

The focus of this study was to produce isopropanol and butanol (IB) from dilute sulfuric acid treated cassava bagasse hydrolysate (SACBH), and improve IB production by co-culturing Clostridium beijerinckii (C. beijerinckii) with Clostridium tyrobutyricum (C. tyrobutyricum) in an immobilized-cell fermentation system. Concentrated SACBH could be converted to solvents efficiently by immobilized pure culture of C. beijerinckii. Considerable solvent concentrations of 6.19 g/L isopropanol and 12.32 g/L butanol were obtained from batch fermentation, and the total solvent yield and volumetric producti

In-Silico Analysis and Engineering of an Aldehyde/Alcohol Dehydrogenase for Alternative Cofactor Utilization and Selective Butanol Production.

Biobutanol production by solventogenic Clostridia is limited by a low butanol titer and yield. To overcome this limitation, Clostridium tyrobutyricum was engineered to overexpress the adhE2 gene encoding a bifunctional aldehyde/alcohol dehydrogenase (AAD) for converting acetyl-CoA/butyryl-CoA to acetaldehyde/butyraldehyde and then to ethanol/butanol. In this study, we aimed to increase butanol biosynthesis in C. tyrobutyricum by engineering AAD targeting on amino acid residues in the enzyme catalytic center that could increase butanol:ethanol ratios and alter cofactor specificity. High-Quality Genomic DNA Extraction Protocol for Bacillus and Clostridium Species.

High-quality DNA with sufficient yield is the goal of DNA extraction protocols. We present an optimized, cost-effective method for extracting next-generation sequencing (NGS)-quality genomic DNA from Bacillus and Clostridium species using the chloroform-isoamyl approach. The protocol involves two main procedures: cultivation of the bacteria under appropriate conditions, followed by DNA extraction through cell lysis, phase separation, DNA precipitation, and cleanup. This method has been successfully applied to several hundred strains of Bacillus and Clostridium species in our laboratory, includ
