Endosomal system of Paramecium: coated pits to early endosomes.

A detailed morphological and tracer study of endocytosis via coated pits in Paramecium multimicronucleatum was undertaken to compare endocytic processes in a free-living protozoon with similar processes in higher organisms. Permanent pits at the cell surface enlarge, become coated and give rise to coated vesicles (188 +/- 41 nm in diameter) that enclose fluid-phase markers such as horseradish peroxidase (HRP). Both the pits and vesicles are labeled by the immunogold technique when a monoclonal antibody (mAb) raised against the plasma membrane of this cell is applied to cryosections. The HRP is

Induction of mutant dynamin specifically blocks endocytic coated vesicle formation.

Dynamin is the mammalian homologue to the Drosophila shibire gene product. Mutations in this 100-kD GTPase cause a pleiotropic defect in endocytosis. To further investigate its role, we generated stable HeLa cell lines expressing either wild-type dynamin or a mutant defective in GTP binding and hydrolysis driven by a tightly controlled, tetracycline-inducible promoter. Overexpression of wild-type dynamin had no effect. In contrast, coated pits failed to become constricted and coated vesicles failed to bud in cells overexpressing mutant dynamin so that endocytosis via both transferrin (Tfn) and

A glycolipid-anchored prion protein is endocytosed via clathrin-coated pits.

The cellular prion protein (PrPc) is a glycolipid-anchored, cell surface protein of unknown function, a posttranslationally modified isoform of which PrPSc is involved in the pathogenesis of Creutzfeldt-Jakob disease, scrapie, and other spongiform encephalopathies. We have shown previously that chPrP, a chicken homologue of mammalian PrPC, constitutively cycles between the cell surface and an endocytic compartment, with a transit time of approximately 60 min in cultured neuroblastoma cells. We now report that endocytosis of chPrP is mediated by clathrin-coated pits. Immunogold labeling of neur

Flat clathrin lattices are dynamic actin-controlled hubs for clathrin-mediated endocytosis and signalling of specific receptors.

Clathrin lattices at the plasma membrane coat both invaginated and flat regions forming clathrin-coated pits and clathrin plaques, respectively. The function and regulation of clathrin-coated pits in endocytosis are well understood but clathrin plaques remain enigmatic nanodomains. Here we use super-resolution microscopy, molecular genetics and cell biology to show that clathrin plaques contain the machinery for clathrin-mediated endocytosis and cell adhesion, and associate with both clathrin-coated pits and filamentous actin. We also find that actin polymerization promoted by N-WASP through t

In situ mapping of late-stage biomechanical coordination during clathrin-mediated endocytosis.

Clathrin-mediated endocytosis (CME) requires precise coordination between membrane deformation and force-generating protein assemblies, yet how these forces are dynamically organized in living cells remains poorly defined. Using multi-dimensional single particle tracking (SPT), we indirectly visualize the mechanical motions of individual clathrin-coated pits during the late stages of CME. We uncover a temporally ordered sequence of rotational behaviors that reflect distinct modes of membrane remodeling preceding vesicle scission. While dynamin-dependent in-plane twisting is a common feature of

Mis-assembly of clathrin lattices on endosomes reveals a regulatory switch for coated pit formation.

The clathrin-coated pit lattice is held onto the plasma membrane by an integral membrane protein that binds the clathrin AP-2 subunit with high affinity. In vitro studies have suggested that this protein controls the assembly of the pit because membrane bound AP-2 is required for lattice assembly. If so, the AP-2 binding site must be a resident protein of the coated pit and recycle with other receptors that enter cells through this pathway. Proper recycling, however, would require the switching off of AP-2 binding to allow the binding site to travel through the endocytic pathway unencumbered.

Bacterial lipopolysaccharide can enter monocytes via two CD14-dependent pathways.

Host recognition and disposal of LPS, an important Gram-negative bacterial signal molecule, may involve intracellular processes. We have therefore analyzed the initial pathways by which LPS, a natural ligand of glycosylphosphatidylinositol (GPI)-anchored CD14 (CD14-GPI), enters CD14-expressing THP-1 cells and normal human monocytes. Exposure of the cells to hypertonic medium obliterated coated pits and blocked 125I-labeled transferrin internalization, but failed to inhibit CD14-mediated internalization of [3H]LPS monomers or aggregates. Immunogold electron microscope analysis found that CD14-b

Rapid endocytosis of interleukin 2 receptors when clathrin-coated pit endocytosis is inhibited.

The cytokine interleukin 2 (IL2) is produced by activated helper T lymphocytes and modulates the growth and activity of cells expressing high-affinity surface IL2 receptors that transduce its signaling. After ligand binding to receptors on the plasma membrane, receptor-ligand complexes are rapidly endocytosed and IL2 is degraded in acidic compartments. The best known receptor-mediated endocytosis pathway involves clathrin-coated pits. Receptors that carry an internalization signal recognized by adaptors on the cytosolic side of the plasma membrane are clustered into the coated pits and enter c

Super-Resolution Microscopy Reveals Alterations in Clathrin Structure in Human Cancer Tissue.

Super-resolution microscopy holds great promise for detailed structural analysis of proteins, yet its application in the investigations of protein structures in situ remains sparse. Clathrin-coated pit-mediated endocytosis (CME) plays a key role in human cancer. This study aimed to discover whether there are structural changes in clathrin pits in cancer. Immunofluorescence combined with super-resolution structured illumination microscopy (SR-SIM) on normal and cancerous prostate tissue was used to reveal novel details of clathrin structure and biology. Clathrin (heavy-chain) plaques and pits,

Exocytosis in the antral gastrin cells of mouse, rat, and guinea pig after stimulation by carbamylcholine.

In order to capture the exocytotic figures of gastrin cells in the pyloric antrum of the stomach, we examined antral cells of the mouse, rat, and guinea pig by electron microscopy following stimulation with the cholinergic secretagogue carbamylcholine. Increased numbers of omega profiles indicative of exocytosis were seen in the basal or lateral cell membrane after stimulation with carbamylcholine. The number of exocytotic figures in stimulated gastrin cells was higher in the guinea pig than in the mouse and rat. Coated and non-coated omega profiles and coated pits in the plasma membrane were

Study of staphylococcal toxic shock syndrome toxin in human epithelial cell culture.

Staphylococcal toxic shock syndrome toxin (TST) inhibited growth of normal human epithelial (Chang) cells in culture, increasing the generation time 28% and 64% at concentrations of 4 X 10(-7)M and 8 X 10(-7)M, respectively. Fluorescence and electron microscopy of the cells treated with TST revealed the location of TST in the coated pits, specialized areas of the cell membrane known to contain high-affinity receptors for other polypeptide ligands. TST was labeled with 125I without detectable damage to the molecule and was shown to bind specifically to epithelial cells. A 100-fold excess of unl

Vasopressin stimulates formation of coated pits in rat kidney collecting ducts.

The water permeability of collecting ducts is greatly increased by the antidiuretic hormone, vasopressin. This permeability increase is associated with the appearance of intramembrane particle (IMP) clusters on the luminal plasma membranes of principal cells of the collecting duct epithelium. IMP aggregates have also been related to an increase in water permeability of two other vasopressin-sensitive epithelia, the amphibian urinary bladder and the amphibian epidermis, and it has been proposed that these specialized membrane domains might represent specific water-permeable membrane patches, in

N-WASP deficiency impairs EGF internalization and actin assembly at clathrin-coated pits.

WASP and WAVE family proteins promote actin polymerization by stimulating Arp2/3-complex-dependent filament nucleation. Unlike WAVE proteins, which are known to drive the formation of protrusions such as lamellipodia and membrane ruffles, vertebrate cell functions of WASP or N-WASP are less well established. Recent work demonstrated that clathrin-coated pit invagination can coincide with assembly of actin filaments and with accumulation of N-WASP and Arp2/3 complex, but the relevance of their recruitment has remained poorly defined. We employed two-colour total internal reflection microscopy t

Monocyte-related functions expressed in cell lines established from human pancreatic adenocarcinoma. I. Comparative analysis of endocytotic activity, lysosomal enzyme secretion, and superoxide anion production.

Fine structural analyses of human pancreatic carcinoma have demonstrated their composition of one major cell type, a mucin secreting cell. In lower grades of differentiation the tumor cells change polarity and reduce mucin production and secretion. Instead numerous small vesicles in the cytoplasm and a high frequency of coated pits and coated vesicles beneath the plasma membrane suggest endocytotic activity. One monoclonal antibody (BW 227/19) produced against pancreatic tumor cells of low differentiation, besides binding to primary pancreatic carcinoma in cytofluorometric studies, also reacte

Exocytosis in human salivary glands visualized by high-resolution scanning electron microscopy.

The luminal membrane of salivary acinar cells creates a specialized cell surface area that accepts exocytosis and undergoes dynamic changes during secretion. These changes were visualized three-dimensionally from both the inside and outside of the cell in human parotid and submandibular glands, by application of in vitro secretory stimulation and then of OsO4 maceration to remove cytoplasmic organelles by varying degrees. In control glands treated without secretagogues, the luminal surface of serous acinar cells bore well-developed microvilli with only an occasional incidence of exocytotic pro

Cytoskeletal regulation of human salivary secretion studied by high resolution electron microscopy and confocal laser microscopy.

To study the cell regulation mechanisms of human salivary secretion, surgical specimens of human parotid and submandibular glands were treated in vitro with isoproterenol (beta-agonist), carbachol (muscarinic agonist), and cytochalasin D (microfilament disruptive agent), and morphological changes occurring in serous acinar cells were observed. Control acinar cells treated without secretagogues exhibited only occasional examples of exocytosis. Rabies Internalizes into Primary Peripheral Neurons via Clathrin Coated Pits and Requires Fusion at the Cell Body.

The single glycoprotein (G) of rabies virus (RABV) dictates all viral entry steps from receptor engagement to membrane fusion. To study the uptake of RABV into primary neuronal cells in culture, we generated a recombinant vesicular stomatitis virus in which the G protein was replaced with that of the neurotropic RABV CVS-11 strain (rVSV CVS G). Using microfluidic compartmentalized culture, we examined the uptake of single virions into the termini of primary neurons of the dorsal root ganglion and ventral spinal cord. By pharmacologically disrupting endocytosis at the distal neurites, we demons

VAMP8 function reveals tight linkage between endocytic recycling and endocytosis.

Clathrin-mediated endocytosis (CME) is a multistage process that involves the initiation and stabilization of clathrin-coated pits (CCPs) that invaginate and finally detach from the plasma membrane to form clathrin-coated vesicles (CCVs). Given that Soluble N-ethylmaleimide-sensitive factor attachment receptor (SNARE) proteins are essential for downstream vesicle targeting and fusion events, their recruitment into nascent CCVs has been suggested to be a prerequisite for CME progression. However, which and how SNARE proteins regulate CME remains to be explored. Here, we showed that siRNA-mediat

The ins and outs of membrane movement in biliary epithelia.

We have developed a novel technique for the isolation from normal rat liver of morphologically polar, intrahepatic bile duct epithelial cells which exhibit clathrin-coated pits. Using electron microscopic cytochemistry, we demonstrated receptor-mediated endocytosis of EGF by cultured IBDEC. Also, using freshly isolated polar couplets of IBDEC, we demonstrated that these cells participate in fluid-phase endocytosis. Finally, using a novel fluorescence unquenching assay and our isolated bile duct epithelial cell model, we showed that secretin stimulates exocytosis in IBDEC, a finding compatible

100-kD coated vesicle proteins: molecular heterogeneity and intracellular distribution studied with monoclonal antibodies.

Proteins with molecular weights of around 100,000 (designated 100K) are found in all coated vesicles. Five monoclonal antibodies have been raised against the major 100K proteins of bovine brain coated vesicles, which migrate on SDS gels as three closely spaced bands. One antibody stains the middle band (band B), two stain both upper and lower bands (bands A and C), and two stain the lower band (band C) only. Thus, the polypeptides in bands A and C are related (but not identical), a result confirmed by NH2-terminal sequencing. Other tissues were found to express proteins corresponding to, and c
