Gut intraepithelial lymphocytes and immunity to Coccidia.

Little is known about the physiological functions of the intra-epithelial lymphocytes (IELs) of mucosal tissues, including their role in immunity to infection. Most IELs are T cells, but their surface phenotype and activation characteristics are different from those of peripheral T cells. Here, Vincent McDonald outlines the distinctive features of IELs and presents evidence, particularly from studies of Coccidia, that these cells have an important role in the control of intracellular infection at the mucosal surface.

Infections by Intestinal Coccidia and Giardia duodenalis.

The coccidians Cryptosporidium spp, Cyclospora cayetanensis, and Cystoisospora belli and the flagellate Giardia duodenalis are pathogenic protozoa associated with gastrointestinal manifestations. Diagnosis relies heavily on microscopy, and although ova-and-parasite examinations can detect Giardia and Cystoisospora, Cryptosporidium and Cyclospora often require specific diagnostic requests. Approved non-microscopy methods are available for Giardia and Cryptosporidium, although negative results are frequently followed by microscopic assays. Polymerase chain reaction-based methods are not frequent

Gut Coccidia--Isospora, Cryptosporidium, Cyclospora and Sarcocystis.

The gut Coccidia are members of a large, varied, and exclusively intracellular group of protozoan parasites, four species of which (Isospora, Cryptosporidium, Cyclospora, and Sarcocystis) are human pathogens. The first three, but particularly Cryptosporidium parvum, have moved from medical curiosities to major problems with the coming of the acquired immunodeficiency syndrome (AIDS) epidemic, but are now known to also cause disease in the immunocompetent patient. They are easy to acquire and difficult to remove from the environment and, in the case of cryptosporidiosis, impossible to treat pro

The Coccidia (Apicomplexa) of the Archosauria (Crocodylia: Eusuchia) of the World.

The order Crocodylia (suborder Eusuchia) includes 27 species of alligators, caimans, crocodiles, and gharials that are cosmopolitan in distribution, inhabiting subtropical and tropical locations. Numerous surveys (many of them trivial, with small sample sizes) have reported a wide variety of blood and intestinal apicomplexans from 17/27 (63%) crocodilian hosts, but neither a summation for the group nor a revisionary systematic approach to species evaluation has ever been provided. Herein, we summarize information on the 16 species of apicomplexans that we consider to be valid, including 8 Eime

Do All Coccidia Follow the Same Trafficking Rules?

The Coccidia are a subclass of the Apicomplexa and include several genera of protozoan parasites that cause important diseases in humans and animals, with Toxoplasma gondii becoming the 'model organism' for research into the coccidian molecular and cellular processes. The amenability to the cultivation of T. gondii tachyzoites and the wide availability of molecular tools for this parasite have revealed many mechanisms related to their cellular trafficking and roles of parasite secretory organelles, which are critical in parasite-host interaction. Nevertheless, the extrapolation of the T. gondi

Intestinal coccidia in Cuban pediatric patients with diarrhea.

From May to August 1999, we evaluated 401 patients from a pediatric hospital of Havana City. One group was composed of 113 patients with diarrhea admitted to the Gastroenterology ward and a second consisted of 288 patients without diarrhea, admitted for other reasons, and hospitalized within the same time period. Three stool samples were collected from each child and were examined using three parasitological techniques. When we compared the frequency of parasite species between both groups, we found Cryptosporidium spp. and Cyclospora cayetanensis, only in the group of children with diarrhea (

Survival of coccidia in poultry litter and reservoirs of infection.

The survival of coccidia was studied in poultry litter, dust, soil, and invertebrate animals. The populations of coccidia in litter were recorded during broiler growout in 16 broiler houses and in floor-pen trials involving anticoccidial drugs. The viability of oocysts declined rapidly in poultry litter regardless of the species; it was retained best in 40% moisture at 4 C. Sporocysts from broken oocytes did not survive even short exposure to poultry litter. Survival of oocysts was poorest at temperatures higher than 4 C, regardless of the carrier. In four floor-pen experiments designed to stu

Antigen-specific T cell proliferation following coccidia infection.

Coccidia antigen-specific T lymphocyte proliferation induced by Eimeria acervulina was measured in chickens congenic at the major histocompatibility complex and in two unrelated lines. Sporozoites and merozoites induced high proliferation of lymphocytes following primary infection, with similar changes seen for splenic or peripheral blood lymphocytes. Highest antigen-specific proliferation after both primary and secondary infection was seen in Line 15I5, which contributed the background genome to the congenic lines. In general, merozoites elicited higher proliferation responses than sporozoite

Coccidia species in endemic and native New Zealand passerines.

New Zealand native passerines are hosts to a large variety of gastrointestinal parasites, including coccidia. Coccidian parasites are generally host-specific, obligate intracellular protozoan parasites. In passerine birds, members of the genus Isospora are most common. Under natural conditions, these parasites seldom pose a threat, but stressors such as quarantine for translocation, overcrowding, or habitat changes may cause an infection outbreak that can severely affect wild populations. Although coccidia are important pathogens and have caused mortalities in kiwi (Apteryx spp.) and hihi (Not

A Survey of Coccidia and Nematodes in Pastured Poultry in the State of Georgia.

The aim of this study was to evaluate and quantify the parasitological challenge in pastured poultry production in the state of Georgia. Over the course of 1 yr, fecal samples from six turkey flocks, 10 broiler flocks, and 13 layer flocks were collected on a pastured farm in 2-wk intervals to determine counts of Eimeria oocysts and nematode eggs. Average coccidia counts were 10,198 oocysts per gram of feces (OPG) in broiler flocks, 1470 OPG in layer flocks, and 695 OPG in turkey flocks. The means in broiler and turkey flocks were higher at their first week on pasture. Counts in broilers and la

The relevance of laboratory diagnosis of human cryptosporidiosis and other coccidia.

Human infection by Cryptosporidium spp and other coccidia are due to opportunist non-host specific microorganisms. In HIV seropositive patients, the gastrointestinal symptoms accompanying such infections may be serious and prolonged and may include nausea, low-grade fever, abdominal cramps, anorexia and watery diarrhoea. We studied 188 stool samples from 111 patients (84 men and 27 women) with diarrhoea. A modified Ziehl-Nielsen technique for the detection of Cryptosporidium spp and Isospora belli was employed. The mean age of the patients was 31 years. Cryptosporidium spp was seen in 18% (n =

[Peculiarities of the distribution of Cryptosporidia (Coccidia: Cryptosporydiidae) in monkeys in an apery].

A total of 520 monkeys belonging to 6 species (Macaca mullata, M. fascicularis, M. nemestrina, Cercopithecus aethiops, Papio anubis, and P. hamadrias) were investigated. Total frequency of occurrence of the protozoan Cryptosporidium in the Adler apery constituted 13.8%. The majority of parasites were found in animals with intestinal disorders such as diarrhea. The lowest frequency of cryptosporidias occurrence was revealed in clinically healthy monkeys. Among sick monkeys, the invasion was most common in infants under one year of age. Cryptosporidiosis is rarely found just as it is, and, as a

Effects of components of Artemisia annua on coccidia infections in chickens.

Four experiments were run to test the anticoccidial activity of dried Artemisia annua leaves and several of their chemical constituents for possible use as prophylactic feed additives. When fed over a period of 3 wk at a level of 5%, a dried leaf supplement of A. annua provided significant protection against lesions due to Eimeria tenella but not Eimeria acervulina or Eimeria maxima. When fed over a period of 5 wk at a level of 1% to chicks undergoing immunization with a live vaccine, it provided significant protection in partially immunized chicks against E. acervulina and E. tenella lesions

Coccidia: a review of recent advances on immunity and vaccine development.

The genus Eimeria contains a number of obligate intracellular protozoan parasites with a complicated life-cycle involving both asexual and sexual stages of development. Coccidiosis is caused by Eimeria infecting primarily the intestine of the susceptible host, thereby seriously impairing the growth and feed utilization of poultry and other livestock. The desire to develop a vaccine against Eimeria has promoted active research to elucidate the mechanisms of protective immunity and identification of candidate vaccine antigens. Protozoa are unique in their modes of transmission and nature of dise

In five large calf-houses and seven farms, 943 calves at the age from one to 198 days were examined coprologically; the feces of 224 animals (23.7%) contained coccidium oocysts of the genus Cryptosporidium. The cryptosporidium oocysts were counted in the Bürker chamber by means of the original method, and their mean number in 1 g of feces was the highest in the animals of the age from 11 to 20 days (3.0 +/- 2.4 million in the calves suffering from scours and 4.2 +/- 4.1 million in the calves without scours). Statistical comparison of the mean weights of heifers was performed at the time of the

The cathepsin-D expression in the cyto-invasive stages of coccidia in intestinal mucosa of chickens, and the relevance of this proteinase for intracellular penetration and spreading of coccidia in host tissues.

The Cathepsin-D proteinase was demonstrated immunohistochemically in cases of intestinal coccidiosis in chickens; its expression concerned almost the cyto-invasive stages of reproductive cycle of several strains of coccidia (merozoites of the II-nd generation and also microgametes). The authors consider Cathepsin-D as a potentional "penetration/spreading factor", being significant for intracellular invasion and spreading of coccidia into host cells. In diagnostic practice, an immunohistochemical demonstration of Cathepsin-D is a relatively simple and sensitive method, which brings information
