Studies by Garg et al. and Ashworth et al. investigated the functional relevance of a key regulatory protein, cofilin-1, for podocyte actin dynamics (Ashworth et al. in PLos One 5:e12626, 2010; Garg et al. in J Biol Chem 285:22676-22688, 2010). Using different model organisms (zebrafish or transgenic mice), both groups observed a collapse of the glomerular filtration barrier upon inactivation of cofilin-1. In elegant biochemical studies, Garg et al. established that cofilin-1 activity is regulated by nephrin, which is part of the slit diaphragm complex. Two feedback loops stabilize cofilin-1 i

Oxidation of human cofilin-1 (CFL-1) protein has been linked to dysregulation and may promote aggregation into amyloid-like structures implicated in neurodegenerative disorders. Herein, CFL-1 was intentionally oxidized upon incubation with N-chlorotaurine to monitor aggregation through direct electrochemistry. Solution-phase characterization by size-exclusion chromatography (SEC), dynamic light scattering (DLS), fluorescence, and circular dichroism spectroscopy confirmed successful oxidation and showed dimers to high-molecular-weight aggregates exclusively under oxidizing conditions. Actin dynamics determines podocyte morphology during development and in response to podocyte injury and might be necessary for maintaining normal podocyte morphology. Because podocyte intercellular junction receptor Nephrin plays a role in regulating actin dynamics, and given the described role of cofilin in actin filament polymerization and severing, we hypothesized that cofilin-1 activity is regulated by Nephrin and is necessary in normal podocyte actin dynamics. Nephrin activation induced cofilin dephosphorylation via intermediaries that include phosphatidylinositol 3-kinase, SSH1, 14-3-3,

Parkinson's disease (PD) is marked by α-synuclein aggregation, mitochondrial dysfunction, and synaptic degeneration, yet the molecular mediators linking these pathological events remain poorly defined. Full-length cofilin (cofilin FL), an actin-binding protein, can be cleaved by asparagine endopeptidase (AEP) to generate the truncated fragment cofilin 1-138, which is known to exert enhanced neurotoxicity in neurodegenerative disorders. However, its role in PD pathogenesis has not been established. In this study, we demonstrate that cofilin 1-138 is elevated in the striatum of PD patients and α

Elevated homocysteine (Hcy) levels contribute to the onset and progression of childhood epilepsy. However, Hcy-lowering therapies provide limited benefits, suggesting additional pathogenic mechanisms. Here, we identified N-homocysteinylated Cofilin-1 (N-Hcy-CFL1) as a molecular link between elevated Hcy levels and childhood epilepsy. We detected N-Hcy-CFL1 in patients with epilepsy and in a mouse model of hyperhomocysteinemia. Mass spectrometry identified Hcy-dependent lysine modifications in CFL1, and functional assays, including actin depolymerization, the F/G-actin ratio, and axonal elongat

Accreta and gestational trophoblastic disease (ie, choriocarcinoma) are placental pathologies characterized by hyperproliferative and invasive trophoblasts. Cellular proliferation, migration, and invasion are heavily controlled by actin-binding protein (ABP)-mediated actin dynamics. The ABP vasodilator-stimulated phosphoprotein (VASP) carries key regulatory role. Profilin-1, cofilin-1, and VASP phosphorylated at Ser157 (pVASP-S157) and Ser239 (pVASP-S239) are ABPs that regulate actin polymerization and stabilization and facilitate cell metastases. Cofilin-1 (CFL1) is an actin-regulating protein encoded by the CFL1 gene and plays a central role in cytoskeletal dynamics, cell motility, and invasion-processes critical for tumor progression and metastasis. Aberrant CFL1 expression has been linked to poor prognosis, therapeutic resistance, and increased metastatic potential across several human cancers. This review consolidates clinical evidence from studies involving human samples over the past 15 years, highlighting CFL1 as a candidate prognostic and predictive biomarker. We analyze tumor-specific expression patterns, detection methodologi

NOD1 is an intracellular pathogen recognition receptor that contributes to anti-bacterial innate immune responses, adaptive immunity and tissue homeostasis. NOD1-induced signaling relies on actin remodeling, however, the details of the connection of NOD1 and the actin cytoskeleton remained elusive. Here, we identified in a druggable-genome wide siRNA screen the cofilin phosphatase SSH1 as a specific and essential component of the NOD1 pathway. We show that depletion of SSH1 impaired pathogen induced NOD1 signaling evident from diminished NF-κB activation and cytokine release. Chemical inhibiti

OBJECTIVE: Parkinson's disease (PD) lacks reliable, minimally invasive biomarkers for diagnosis. This study aimed to identify and validate PD-specific plasma extracellular vesicle (EV)-associated proteins.
METHODS: Plasma EVs were isolated by ultracentrifugation and characterized. Proteomic profiling of total plasma EVs from PD patients and healthy controls was performed, and a candidate protein identified after analysis was validated by enzyme-linked immunosorbent assay. Diagnostic accuracy was evaluated using receiver operating characteristic and precision-recall curve analysis. Disease spe

Podocyte foot process (FP) is dysregulated in nephrotic syndrome. The effacement of podocyte FPs typically arises following perturbations in the actin cytoskeleton. Recent data suggest that the effects of calcineurin (CaN) inhibitor cyclosporine A (CsA) are independent of its effects on T-cells, and CsA has been identified as stabilizing the actin cytoskeleton through stabilizing synaptopodin in podocytes, and thereby directly reducing proteinuria. Other studies showed that CsA could regulate cofilin-1 directly within tubular epithelial cells. However, whether synaptopodin is the only target o

BACKGROUND: It is known that retinoid receptor function is attenuated during T cell activation, a phenomenon that involves actin remodeling, suggesting that actin modification may play a role in such inhibition. Here we have investigated the role of actin dynamics and the effect of actin cytoskeleton modifying agents on retinoid receptor-mediated transactivation.
RESULTS: Agents that disturb the F-actin assembly or disassembly attenuated receptor-mediated transcription indicating that actin cytoskeletal homeostasis is important for retinoid receptor function. Overexpression or siRNA-induced k

Alzheimer’s disease (AD) is characterized by the accumulation of neurofibrillary tangles (NFTs) composed of misfolded tau and by early mitochondrial dysfunction. Although tau aggregation has been closely linked to mitochondrial impairment and neuronal toxicity, the molecular factors that amplify tau-induced mitochondrial damage remain incompletely understood. Previous studies have shown that cofilin-1, an actin-binding protein, facilitates tau aggregation and propagation under pathological conditions. In the present study, using tauopathy-based cellular models, we investigated whether cofilin-

Skeletal muscle differentiation requires coordinated cytoskeletal remodeling and transcriptional reprogramming. Members of the actin-depolymerizing factor/Cofilin family regulate actin filament turnover, yet the isoform-specific functions of Cofilin-1 and Cofilin-2 during myogenesis remain incompletely understood. Murine C2C12 myoblasts were used to investigate the expression and function of Cofilin isoforms during differentiation. CRISPR/Cas9-mediated knockout, shRNA-mediated knockdown, immunofluorescence, gene expression analyses, MRTF reporter assays, and pharmacological inhibition of LIM k

Cofilin is an actin-depolymerizing factor that regulates actin dynamics critical for T cell migration and T cell activation. In unstimulated resting CD4 T cells, cofilin exists largely as a phosphorylated inactive form. Previously, we demonstrated that during HIV-1 infection of resting CD4 T cells, the viral envelope-CXCR4 signaling activates cofilin to overcome the static cortical actin restriction. In this pilot study, we have extended this in vitro observation and examined cofilin phosphorylation in resting CD4 T cells purified from the peripheral blood of HIV-1-infected patients. Here, we

Rac1 can affect the migration of neural crest cells by regulating the polymerization of actin and the membrane formation process. But the role of the Rac1 signaling pathway in the pathogenesis of Hirschsprung's disease (HSCR) remains unclear. In order to investigate the mechanism of the abnormal protein phosphorylation of Rac1, Lim-kinase 1 (Limk1) and Cofilin involved in the pathogenesis of HSCR. The protein phosphorylation levels of these proteins were detected by Western blot in 30 samples of HSCR narrow segment, 30 samples of transitional segment tissues, and 14 samples of normal intestina

Alzheimer's disease (AD) is a degenerative neurological disease characterized by various biological signatures, including synaptic dysfunction, β-amyloid plaques, hyperphosphorylated Tau, cofilin-actin rods, and Hirano bodies, all of which are linked to the actin cytoskeleton and its regulators. Additionally, the presence of herpes simplex virus type 1 (HSV-1) in the brains of AD patients has long been suggested as a contributing factor for AD. However, mechanisms by which HSV-1 accelerates AD pathogenesis remain poorly understood. Here we report that HSV-1 infection induces hyperphosphorylati

OBJECTIVE: In recent years, the roles of Neuregulin-1 (NRG-1) in optic nerve injury and retinal cells have been investigated. However, the molecular mechanism by which NRG-1 affects optic nerve injury remains elusive and merits deeper exploration. Hence, this study examined the specific function of NRG-1 in the RhoA/cofilin/F-actin axis in optic nerve injury.
METHODS: Retinal cells were isolated and identified for subsequent experimental uses. Reverse transcription quantitative polymerase chain reaction and Western blot assays were performed to measure NRG-1 expression in retinal cells which

Liver fibrosis is a progressive clinical condition characterised by an abnormal accumulation of extracellular matrix (ECM) components, which are predominantly generated by activated hepatic stellate cells (HSCs) and other fibrogenic cells. This process leads to liver stiffness, which can progress to cirrhosis, an irreversible condition, and ultimately result in liver failure. Obtusifolin (OBT), an anthraquinone derivative isolated from the seeds of Senna obtusifolia, is known for its potent anti-inflammatory and antioxidant properties. The present study evaluated the anti-fibrotic efficacy of

Oxidative stress on cysteine (Cys)-containing proteins has been associated with physiological disorders, as suggested for the human cofilin-1 (CFL-1) protein, in which the oxidized residues are likely implicated in the aggregation process of α-synuclein, leading to severe neuronal injuries. Considering the relevance of the oxidation state of cysteine, quantification of thiols may offer a guide for the development of effective therapies. This work presents, for the very first time, thiol quantification within CFL-1 in solution and on the surface following classic and adapted versions of Ellman'

The cofilin-1 protein, encoded by CFL1, is an actin-binding protein that regulates F-actin depolymerization and nucleation activity through phosphorylation and dephosphorylation. CFL1 has been implicated in the development of neurodegenerative diseases (Alzheimer's disease and Huntington's disease), neuronal migration disorders (lissencephaly, epilepsy, and schizophrenia), and neural tube closure defects. Mutations in CFL1 have been associated with impaired neural crest cell migration and neural tube closure defects. In our study, various computational approaches were utilized to explore singl
