Neurostimulation to Improve Cognitive Flexibility.

Cognitive flexibility, the capacity to adapt behaviors in response to changing environments, is impaired across mental illnesses, including depression, anxiety, addiction, and obsessive-compulsive disorder. Cortico-striatal-cortical circuits are integral to cognition and goal-directed behavior and disruptions in these circuits are linked to cognitive inflexibility in mental illnesses. We review evidence that neurostimulation of these circuits can improve cognitive flexibility and ameliorate symptoms, and that this may be a mechanism of action of current clinical therapies. Further, we discuss

Cognitive Flexibility in Primary Dystonia.

Objectives Although primary dystonia is typically characterized as a movement disorder, it is also associated with cognitive alterations in the domain of executive functioning which may arise from changes in cortico-basal ganglia circuits. Specifically, in comparison to healthy controls, patients with dystonia show deficits in neuropsychological tests of cognitive flexibility. However, it is unclear whether cognitive inflexibility is caused by the pathomechanisms underlying primary dystonia or by confounding factors such as depression or symptom-related distraction. Methods The present study ai

Impaired cognitive flexibility across psychiatric disorders.

OBJECTIVE: Problems with cognitive flexibility have been associated with multiple psychiatric disorders, but there has been little understanding of how cognitive flexibility compares across these disorders. This study examined problems of cognitive flexibility in young adults across a range of psychiatric disorders using a validated computerized trans-diagnostic flexibility paradigm. We hypothesized that obsessive-compulsive spectrum disorders (eg, obsessive-compulsive disorder, trichotillomania, and skin-picking disorder) would be associated with pronounced flexibility problems as they are mo

Specific cognitive flexibility rehabilitation in schizophrenia.

A Cognitive Shift neurocognitive training module was developed in the attempt to ameliorate cognitive flexibility deficits in chronic schizophrenic patients. A procedural training approach hypothesized the exercise of specific neural network processes, identified from theories of frontal and prefrontal lobe functioning. Three male patients who underwent the intensive program demonstrated significant gains in Wisconsin Card Sorting Test performance, gains that were maintained at a six month reassessment. Expanded Brief Psychiatric Rating Scale (a measure of symptomatology) and Life Skills Profi

Cognitive bias modification of inferential flexibility.

This research examines the effects of a cognitive bias modification procedure for facilitating inferential flexibility, on inferences, mood, and state rumination. Participants were presented with training scenarios, followed by two consecutive inferences for each scenario. In the training condition, participants repeatedly practiced shifting from a negative inference to a positive one. But in the control condition, the two inferences were of the same valence and no shifting occurred. The training successfully promoted the intended inferential shift on new scenarios (d = 0.72). Moreover, traine

Cognitive Control and Flexibility in the Context of Stress and Depressive Symptoms: The Cognitive Control and Flexibility Questionnaire.

Cognitive control and (cognitive) flexibility play an important role in an individual's ability to adapt to continuously changing environments. In addition to facilitating goal-directed behaviors, cognitive control and flexibility have been implicated in emotion regulation, and disturbances of these abilities are present in mood and anxiety disorders. In the context of stressful experiences, the reported studies examined processes related to cognitive control and flexibility, emotional regulation and depressive symptoms. To this end, a brief (18-item) self-report measure - the Cognitive Contro

C1ql3 promotes cognitive flexibility behavior in mice.

Cognitive flexibility, the ability to adapt behavioral strategies based on changing circumstances, is deficient in patients with certain neuropsychiatric disorders, such as autism spectrum disorder and schizophrenia. Cognitive flexibility can be measured in humans and animal models using attentional set-shifting tasks, which require activity of the prefrontal cortex (PFC) and other key brain regions. We demonstrate that C1QL3, a protein involved in regulating excitatory synapse density in the PFC and other brain circuits, is important for attentional set-shifting, using a mouse model of C1ql3

Acute psilocybin enhances cognitive flexibility in rats.

Psilocybin has been shown to improve symptoms of depression and anxiety when combined with psychotherapy or other clinician-guided interventions. To understand the neural basis for this pattern of clinical efficacy, experimental and conceptual approaches that are different than traditional laboratory models of anxiety and depression are needed. A potential novel mechanism is that acute psilocybin improves cognitive flexibility, which then enhances the impact of clinician-assisted interventions. Consistent with this idea, we find that acute psilocybin robustly improves cognitive flexibility in

Temperament and cognitive flexibility across lemur species.

Temperament can shape patterns of individual differences in animal cognition and behaviour. An influential hypothesis proposes that bolder and more exploratory individuals are less cognitively flexible than shy individuals. However, there is also evidence that the relationship between temperament and cognition may vary depending on the particular social and ecological challenges an individual faces. Here, we test the link between temperament and cognitive flexibility by examining seven lemur species (N = 65) with targeted variation in diet and social organization: Coquerel's sifakas, Propithec

Impaired cognitive flexibility and disrupted cognitive cerebellum in degenerative cerebellar ataxias.

There is a clinically unmet need for a neuropsychological tool that reflects the pathophysiology of cognitive dysfunction in cerebellar degeneration. We investigated cognitive flexibility in degenerative cerebellar ataxia patients and aim to identify the pathophysiological correlates of cognitive dysfunction in relation to cerebellar cognitive circuits. We prospectively enrolled degenerative cerebellar ataxia patients with age-matched healthy controls who underwent 3 T 3D and resting-state functional MRI. All 56 participants were evaluated with the Scale for Assessment and Rating of Ataxia and

Vascular cognitive disorders: memory, mental speed and cognitive flexibility after stroke.

This study investigated the occurrence of cognitive disorders 1 and 6 months after stroke in a cohort of patients with a first-ever stroke. In addition, it was investigated whether age, sex and level of education are risk factors for vascular cognitive disorders. Memory, simple speed, cognitive flexibility and overall cognitive functioning were examined in 139 patients at 1 and 6 months post-stroke. Inclusion criteria on admission were first cerebral stroke, age>/=40, no other neurological or psychiatric disorders and ability to communicate. Mean age was 69.3 years (S.D.=12.3). Patients were c

Neurocomputational mechanisms underlying cognitive flexibility: a translational perspective.

Cognitive flexibility allows us to adjust behavior when environmental rules change and to recombine prior knowledge in novel situations. However, the computations underlying rule discovery remain unclear: how do rule representations emerge as we interact with the environment? While it is often considered a hallmark of human behavior, converging evidence highlights comparable forms of abstract learning in rodents. Across species, prefrontal activity patterns encode structured representations that support generalization and monitor for rule switches. In this review, we synthesize recent behavior

Cognitive flexibility and depression remission: A narrative review.

INTRODUCTION: In depression, cognitive flexibility - the ability to adjust cognitive strategies in response to a changing environment in order to adapt one's behavior appropriately - is frequently impaired. This impairment is thought to contribute to difficulties in modifying maladaptive cognitive patterns and may influence responsiveness to antidepressant treatments. Treatment resistance remains a major clinical challenge, with approximately half of patients failing to respond to first-line antidepressant therapies. Moreover, residual symptoms characterized by rigid and maladaptive cognitive

Aging Affects Dopaminergic Neural Mechanisms of Cognitive Flexibility.

UNLABELLED: Aging is accompanied by profound changes in the brain's dopamine system that affect cognitive function. Evidence of powerful individual differences in cognitive aging has sharpened focus on identifying biological factors underlying relative preservation versus vulnerability to decline. Dopamine represents a key target in these efforts. Alterations of dopamine receptors and dopamine synthesis are seen in aging, with receptors generally showing reduction and synthesis demonstrating increases. Using the PET tracer 6-[18F]fluoro-l-m-tyrosine, we found strong support for upregulated str

Improved cognitive flexibility after aortic valve replacement surgery.

OBJECTIVES: Aortic valve replacement (AVR) surgery is associated with potential risk to cerebral injury. On the other hand, improved cardiovascular functioning after the surgery may have positive impact on brain health. The aim of this preliminary study was to investigate the impact of AVR surgery on cognition, specifically on higher cognitive control functions, i.e. executive functions, that are typically implicated in cognitive decline due to vascular origin. METHODS: Patients (n = 16) undergoing elective AVR surgery due to aortic stenosis were recruited and their cognitive performance was

Beta-adrenergic modulation of cognitive flexibility during stress.

Stress-induced activation of the locus ceruleus-norepinephrine (LC-NE) system produces significant cognitive and behavioral effects, including enhanced arousal and attention. Improvements in discrimination task performance and memory have been attributed to this stress response. In contrast, for other cognitive functions that require cognitive flexibility, increased activity of the LC-NE system may produce deleterious effects. The aim of the present study was to determine the effect of pharmacological modulation of the LC-NE system on stress-induced impairments in cognitive flexibility perform

Cognitive flexibility in phenotypes of pediatric bipolar disorder.

OBJECTIVE: Clinicians and researchers debate whether children with chronic, nonepisodic irritability should receive the diagnosis of bipolar disorder (BD). To address this debate, we evaluated cognitive flexibility, or the ability to adapt to changing contingencies, in three groups of children: narrow-phenotype BD (NP-BD; full-duration manic episodes of elevated/expansive mood; N = 50; 13.1 +/- 2.9 years), severe mood dysregulation (SMD; chronic, nonepisodic irritability; N = 44; 12.2 +/- 2.1 years), and healthy controls (N = 43; 13.6 +/- 2.4 years). Cognitive flexibility is relevant to sympto

Cognitive flexibility in anorexia nervosa and bulimia nervosa.

The aim of this study was to determine if there are differences in cognitive flexibility in anorexia nervosa and bulimia nervosa. Fifty-three patients with an eating disorder (34 with anorexia nervosa and 19 with bulimia nervosa) and 35 healthy controls participated in the study. A battery of neuropsychological tests for cognitive flexibility was used, including Trail Making B, the Brixton Test, Verbal Fluency, the Haptic Illusion Test, a cognitive shifting task (CatBat) and a picture set test. Using exploratory factor analysis, four factors were obtained: 1: Simple Alternation; 2: Mental Flex
