Identification of specific corrinoids reveals corrinoid modification in dechlorinating microbial communities.

Cobalamin and other corrinoids are essential cofactors for many organisms. The majority of microbes with corrinoid-dependent enzymes do not produce corrinoids de novo, and instead must acquire corrinoids produced by other organisms in their environment. However, the profile of corrinoids produced in corrinoid-dependent microbial communities, as well as the exchange and modification of corrinoids among community members have not been well studied. In this study, we applied a newly developed liquid chromatography tandem mass spectrometry-based corrinoid detection method to examine relationships

Versatility in corrinoid salvaging and remodeling pathways supports corrinoid-dependent metabolism in Dehalococcoides mccartyi.

Corrinoids are cobalt-containing molecules that function as enzyme cofactors in a wide variety of organisms but are produced solely by a subset of prokaryotes. Specific corrinoids are identified by the structure of their axial ligands. The lower axial ligand of a corrinoid can be a benzimidazole, purine, or phenolic compound. Though it is known that many organisms obtain corrinoids from the environment, the variety of corrinoids that can serve as cofactors for any one organism is largely unstudied. Here, we examine the range of corrinoids that function as cofactors for corrinoid-dependent meta

Complex samples cyanide detection with immobilized corrinoids.

Colorimetric solid phase with spatially separated extraction and detection zones as a rapid, effective and economic method for the optical detection of cyanide in complex samples is described. The system is seven times more sensitive for the optical detection of cyanide than the same class of chemical sensors used under homogeneous conditions. The application of the method in the detection of (i) endogenous cyanide in colored plant samples and of (ii) hydrogen cyanide in tobacco smoke is shown. The optical detection of multiple anions within a single sample has been demonstrated in principle f

The chemistry of the cobalt corrinoids - Recent advances and emerging themes. Part 1: The chemistry and industrial production of cobalt corrinoids.

This is the first part of a three-part review of advances in cobalt corrinoid (B12) research published between 2020 and 2025, focusing on the chemistry and industrial production of cobalt corrinoids. Although vitamin B12 is widely recognised for its biological and clinical importance, its derivatives also constitute a distinctive class of organometallic compounds whose behaviour emerges from the interplay between cobalt redox chemistry, corrin macrocycle structure, and axial ligation. Recent work increasingly treats corrinoids not as structurally conserved cofactors with narrowly defined funct

Investigating weak axial ligation in corrinoids by X-ray absorption spectroscopy: Implications for corrinoid iron-sulfur protein.

Corrinoid iron‑sulfur protein (CFeSP) is the methyltransferase in the Wood-Ljungdahl pathway, yet the geometric and electronic structure that enables its higher catalytic efficiency relative to small-molecule analogs remains a subject of debate. While axial ligation is known to tune the reactivity of B12-dependent enzymes, weak interactions in "base-off" species like methylated CFeSP (Me-CFeSP) have been difficult to define using standard structural probes, despite that solution-phase spectroscopy has suggested their presence. Here we utilize Co K-edge X-ray absorption spectroscopy (XAS) and t

Quenching corrinoid-based interactions in a model bacterial coculture.

Microbial community structure is driven, in part, by the metabolic interdependencies of resident microbes. Thus, manipulating specific metabolic interactions represents an attractive way to both understand how microbial communities perform complex functions and alter them for therapeutic or environmental effects. However, it is not yet possible to control the availability of those metabolites produced by some members of the community that are required by others. Here, we report the development of a metabolite "quenching" strategy that disrupts a specific metabolic interaction involving corrino

[Effect of nucleotide containing corrinoids on vitamin B 12 synthesis in Propionibacterium shermanii].

The influence of B12-CN, B12-OH, coenzyme B12, factor III and factor B on the synthesis of vitamin B12 and porphyrins by different strains of P. shermanii was investigated. Neither compound inhibited the development of propionic bacteria or suppressed porphyrin formation. All nucleotide containing analogues of vitamin B12 produced a strong repressive effect on the synthesis of corrinoid compounds regardless of the modifications in the upper and lower cobalt ligands. Factor B containing no nucleotide moiety did not show this effect. It is suggested that the nucleotide moiety of the vitamin B12

Cyanide Scavenging by a Cobalt Schiff-Base Macrocycle: A Cost-Effective Alternative to Corrinoids.

The complex of cobalt(II) with the ligand 2,12-dimethyl-3,7,11,17-tetraazabicyclo-[11.3.1]heptadeca-1(17)2,11,13,15-pentaene (CoN4[11.3.1]) has been shown to bind two molecules of cyanide in a cooperative fashion with an association constant of 2.7 (±0.2) × 10(5). In vivo, irrespective of whether it is initially administered as the Co(II) or Co(III) cation, EPR spectroscopic measurements on blood samples show that at physiological levels of reductant (principally ascorbate) CoN4[11.3.1] becomes quantitatively reduced to the Co(II) form. However, following addition of sodium cyanide, a dicyano

Transcriptomic microarray analysis of corrinoid responsive genes in Dehalococcoides ethenogenes strain 195.

Dehalococcoides ethenogenes strain 195 utilizes corrinoid-containing reductive dehalogenases to reduce the environmental pollutants tetrachloroethene and trichloroethene to ethene. Although corrinoids are essential for dehalogenation activity, strain 195 cannot biosynthesize corrinoids de novo. To improve our understanding of corrinoid physiology in this bacterium, whole-genome microarrays were applied to characterize the transcriptome during growth with excess and limiting concentrations of the corrinoid cyanocobalamin. Additional studies examined the effects of exposure to spent medium from

Side chains of cobalt corrinoids control the sensitivity and selectivity in the colorimetric detection of cyanide.

Substitution of Co(III)-bound water by cyanide allows the rapid colorimetric detection of micromolar amounts of cyanide with cobalt corrinoids. Negatively charged side chains at the periphery of these metal-based sensors increase the selectivity of cyanide versus thiocyanate, the most disturbing anion, by a factor of 30 through additional supramolecular interactions in water.

The chemistry of the cobalt corrinoids - Recent advances and emerging themes. Part 3. Cobalamins and health.

Vitamin B12 (cobalamin) is an essential micronutrient whose biological importance extends beyond its traditional classification as a haematinic vitamin. This third and final part of a review covering work published between 2020 and 2025 synthesises selected illustrative studies that have advanced understanding of B12 physiology, nutrition, deficiency, delivery, and systems-level biology. At the molecular level, B12 functions as a cofactor in one‑carbon metabolism and mitochondrial pathways, influencing DNA synthesis, methylation capacity, and energy metabolism. These biochemical roles translat

Abiotic reductive deiodination of iodinated organic compounds and X-ray contrast media catalyzed by free corrinoids.

Iodinated X-ray contrast media are known for their stability concerning deiodination in the aquatic environment under aerobic conditions. In this study, we demonstrate the abiotic reductive deiodination of the iodinated contrast media iopromide, iopamidol and diatrizoate in the presence of corrinoids. In addition, triiodinated benzoic acid derivatives with iodine atoms bound at different positions were investigated. Corrinoids like cyanocobalamin (vitamin B12) and dicyanocobinamide served as electron shuttles and as catalysts between the reducing agent (e.g., titanium (III) citrate) and the el

Reductive formation of carbon monoxide from CCl4 and FREONs 11, 12, and 13 catalyzed by corrinoids.

In an earlier publication, we reported that corrinoids catalyze the sequential reduction of CCl4 to CHCl3, CH2Cl2, CH3Cl, and CH4 with titanium(III) citrate as electron donor [Krone, U. E., Thauer, R. K., & Hogenkamp, H. P. C. (1989) Biochemistry 28, 4908-4914]. However, the recovery of these products was less than 50%, indicating that other products were formed. We now report that, under the same experimental conditions, CCl4 is also converted to carbon monoxide. These studies were extended to include FREONs 11, 12, 13, and 14. Corrinoids were found to catalyze the reduction of CFCl3, CF2Cl2,

Refined experimental annotation reveals conserved corrinoid autotrophy in chloroform-respiring Dehalobacter isolates.

Two novel chlorinated alkane-respiring Dehalobacter restrictus strains CF and DCA were isolated from the same enrichment culture, ACT-3, and characterized. The closed genomes of these highly similar sister strains were previously assembled from metagenomic sequence data and annotated. The isolation of the strains enabled experimental verification of predicted annotations, particularly focusing on irregularities or predicted gaps in central metabolic pathways and cofactor biosynthesis. Similar to D. restrictus strain PER-K23, strains CF and DCA require arginine, histidine and threonine for grow

Cobalamin production by Lactobacillus coryniformis: biochemical identification of the synthetized corrinoid and genomic analysis of the biosynthetic cluster.

BACKGROUND: Despite the fact that most vitamins are present in a variety of foods, malnutrition, unbalanced diets or insufficient intake of foods are still the cause of vitamin deficiencies in humans in some countries. Vitamin B12 (Cobalamin) is a complex compound that is only naturally produced by bacteria and archea. It has been reported that certain strains belonging to lactic acid bacteria group are capable of synthesized water-soluble vitamins such as those included in the B-group, as vitamin B12. In this context, the goal of the present paper was to evaluate and characterize the producti

Tetrachloroethene reductive dehalogenase of Dehalospirillum multivorans: substrate specificity of the native enzyme and its corrinoid cofactor.

The substrate specificity of the tetrachloroethene reductive dehalogenase of Dehalospirillum multivoransand its corrinoid cofactor were studied. Besides reduced methyl viologen, titanium(III) citrate could serve as electron donor for reductive dehalogenation of tetrachloroethene (PCE) and trichloroethene to cis-1,2-dichloroethene. In addition to chlorinated ethenes, chlorinated propenes were reductively dechlorinated solely by the native enzyme. trans-1,3-Dichloropropene, 1,1,3-trichloropropene and 2,3-dichloropropene were reduced to a mixture of mono-chloropropenes, 1,1-dichloropropene, and 2

Redox potential changes during ATP-dependent corrinoid reduction determined by redox titrations with europium(II)-DTPA.

Corrinoids are essential cofactors of enzymes involved in the C1 metabolism of anaerobes. The active, super-reduced [CoI ] state of the corrinoid cofactor is highly sensitive to autoxidation. In O-demethylases, the oxidation to inactive [CoII ] is reversed by an ATP-dependent electron transfer catalyzed by the activating enzyme (AE). The redox potential changes of the corrinoid cofactor, which occur during this reaction, were studied by potentiometric titration coupled to UV/visible spectroscopy. By applying europium(II)-diethylenetriaminepentaacetic acid (DTPA) as a reductant, we were able to
