Brain-Wide Synaptic Inputs to Aromatase-Expressing Neurons in the Medial Amygdala Suggest Complex Circuitry for Modulating Social Behavior.

Here, we reveal an unbiased view of the brain regions that provide specific inputs to aromatase-expressing cells in the medial amygdala, neurons that play an outsized role in the production of sex-specific social behaviors, using rabies tracing and light sheet microscopy. While the downstream projections from these cells are known, the specific inputs to the aromatase-expressing cells in the medial amygdala remained unknown. We observed established connections to the medial amygdala (e.g., bed nucleus of the stria terminalis and accessory olfactory bulb) indicating that aromatase neurons are a

The medial amygdala's neural circuitry: Insights into social processing and sex differences.

The amygdala, a critical part of the limbic system, is essential for processing social stimuli and regulating stress responses. Among its various neuronal nuclei, the medial amygdala (MeA) remains one of the least studied in humans. The MeA plays a key role in receiving inputs from the olfactory system through pheromones, as well as from crucial areas such as the hypothalamus, hippocampus, and reward system. This allows the MeA to integrate external stimuli with the organism's internal state, finetuning social interactions, endocrine responses, and innate behaviors. Recent advances in neurosci

The Dorsal Raphe Regulates the Duration of Attack through the Medial Orbitofrontal Cortex and Medial Amygdala.

The dorsal raphe (DR) is an evolutionarily conserved brain structure that is involved in aggressive behavior. It projects onto numerous cortical and limbic areas underlying attack behavior. The specific neurocircuit through which the DR regulates aggression, however, is largely unclear. In this study we show that DR neurons expressing CaMKIIα are activated by attack behavior in mice. These neurons project to the medial aspect of the orbitofrontal cortex (OFC; MeOC) and the medial amygdala (MeA), two key regions within the neural circuit known to control aggressive behavior. Using an in vivo op

Sex differences in afferents and efferents of vasopressin neurons of the bed nucleus of the stria terminalis and medial amygdala in mice.

Steroid-sensitive vasopressin (AVP) neurons in the bed nucleus of the stria terminalis (BNST) and medial amygdala (MeA) have been implicated in the control of social behavior, but the connectional architecture of these cells is not well understood. Here we used a modified rabies virus (RV) approach to identify cells that provide monosynaptic input to BNST and MeA AVP cells, and an adeno-associated viral (AAV) anterograde tracer strategy to map the outputs of these cells. Although the location of in- and outputs of these cells generally overlap, we observed several sex differences with differen

Alarm pheromone activates posterior medial amygdala-to-bed nucleus of the stria terminalis projections in male rats.

Animals communicate using olfactory signals called pheromones. Alarm pheromones are evolutionarily conserved and alert conspecifics to danger. In rats, a mixture of hexanal and 4-methylpentanal was identified as the main component that evokes anxiety responses by activating the anterior bed nucleus of the stria terminalis (aBNST). In addition, the posteroventral medial amygdala (pvMeA) was proposed to transmit the alarm pheromone signal to the aBNST. Here, we assessed whether the binary mixture activates direct pvMeA-to-aBNST projections or indirect projections via other MeA subdivisions in ma

Dominance status modulates activity in medial amygdala cells with projections to the bed nucleus of the stria terminalis.

The medial amygdala (MeA) controls several types of social behavior via its projections to other limbic regions. Cells in the posterior dorsal and posterior ventral medial amygdala (MePD and MePV, respectively) project to the bed nucleus of the stria terminalis (BNST) and these pathways respond to chemosensory cues and regulate aggressive and defensive behavior. Because the BNST is also essential for the display of stress-induced anxiety, a MePD/MePV-BNST pathway may modulate both aggression and responses to stress. In this study we tested the hypothesis that dominant animals would show greate

Defensive behaviors and brain regional activation changes in rats confronting a snake.

In the present study, we examined behavioral and brain regional activation changes of rats). To a nonmammalian predator, a wild rattler snake (Crotalus durissus terrificus). Accordingly, during snake threat, rat subjects showed a striking and highly significant behavioral response of freezing, stretch attend, and, especially, spatial avoidance of this threat. The brain regional activation patterns for these rats were in broad outline similar to those of rats encountering other predator threats, showing Fos activation of sites in the amygdala, hypothalamus, and periaqueductal gray matter. DREADD-Induced Inhibition of the Medial Amygdala and Ventromedial Nucleus of the Hypothalamus Affects Experience-Induced Enhancement of Paced Mating Behavior in Female Rats.

The acquisition of sexual experience leads female rats to return to the male more quickly after intromissions, receive intromissions at a faster rate, spend more time with the male during mating, and exhibit more proceptive behaviors during paced mating tests, suggesting heightened sexual motivation. Brain areas critical for sexual experience-enhanced paced mating behavior are unknown. Here, designer receptors exclusively activated by designer drugs (DREADDs) were used to test the hypothesis that silencing the medial amygdala (MeA) or ventromedial nucleus of the hypothalamus (VMH) across four

Retinal organoids: a window into human retinal development.

Retinal development and maturation are orchestrated by a series of interacting signalling networks that drive the morphogenetic transformation of the anterior developing brain. Studies in model organisms continue to elucidate these complex series of events. However, the human retina shows many differences from that of other organisms and the investigation of human eye development now benefits from stem cell-derived organoids. Retinal differentiation methods have progressed from simple 2D adherent cultures to self-organising micro-physiological systems. As models of development, these have coll

c-Fos protein expression in the anterior amygdaloid area and nc. accumbens in the hypoxic rat brain.

INTRODUCTION: By examining the production of c-Fos protein, we analyzed the response to the ischemic attack in different brain tissue, two of which are regions of the limbic system: the anterior amygdaloid area and nc. accumbens. MATERIAL AND METHODS: We used the model of rat brain ischemia--four-vessel occlusion, and Pulsinelli's method. The rats were treated in two ways, according to which they were divided into two groups: a total ischemia (ligation of four blood vessels, i.e. electrocauterization of the vertebral artery with bilateral ligation of the carotid artery--the so-called R-group

Neonatal maternal separation opposes the facilitatory effect of castration on the respiratory response to hypercapnia of the adult male rat: Evidence for the involvement of the medial amygdala.

Respiratory manifestations of panic disorder (PD) include a greater respiratory instability and enhanced responsiveness to CO2 compared to normal individuals. Although the prevalence of PD is approximately three times greater in women compared to men, the origins of this sexual dimorphism remain poorly understood. Similar to PD patients, adult female rats previously subjected to neonatal maternal separation (NMS) show an increase in their ventilatory response to CO2 . Because this effect of NMS is not observed in males, we hypothesised that testosterone prevents NMS-induced hyper-responsivenes

Estrogen signaling in the medial amygdala decreases emotional stress responses and obesity in ovariectomized rats.

Declining estradiol (E2), as occurs during menopause, increases risk for obesity and psychopathology (i.e., depression, anxiety). E2 modulates mood and energy homeostasis via binding to estrogen receptors (ER) in the brain. The often comorbid and bidirectional relationship between mood and metabolic disorders suggests shared hormonal and/or brain networks. The medial amygdala (MeA) is abundant in ERs and regulates mood, endocrine, and metabolic stress responses; therefore we tested the hypothesis that E2 in the MeA mitigates emotional and metabolic dysfunction in a rodent model of surgical men

The Dissociative Subtype of Posttraumatic Stress Disorder: Unique Resting-State Functional Connectivity of Basolateral and Centromedial Amygdala Complexes.

Previous studies point towards differential connectivity patterns among basolateral (BLA) and centromedial (CMA) amygdala regions in patients with posttraumatic stress disorder (PTSD) as compared with controls. Here we describe the first study to compare directly connectivity patterns of the BLA and CMA complexes between PTSD patients with and without the dissociative subtype (PTSD+DS and PTSD-DS, respectively). Amygdala connectivity to regulatory prefrontal regions and parietal regions involved in consciousness and proprioception were expected to differ between these two groups based on diffe

Angiotensinergic receptors in the medial amygdaloid nucleus differently modulate behavioral responses in the elevated plus-maze and forced swimming test in rats.

The brain renin-angiotensin system (RAS) has been implicated in anxiety and depression disorders, but the specific brain sites involved are poorly understood. The medial amygdaloid nucleus (MeA) is involved in expression of behavioral responses. However, despite evidence of the presence of all angiotensinergic receptors in this amygdaloid nucleus, regulation of anxiety- and depressive-like behaviors by angiotensinergic neurotransmissions within the MeA has never been reported. Thus, the present study aimed to investigate the role angiotensin II (AT1 and AT2 receptors) and angiotensin-(1-7) (Ma

Abnormalities of fixation, saccade and pursuit in posterior cortical atrophy.

The clinico-neuroradiological syndrome posterior cortical atrophy is the cardinal 'visual dementia' and most common atypical Alzheimer's disease phenotype, offering insights into mechanisms underlying clinical heterogeneity, pathological propagation and basic visual phenomena (e.g. visual crowding). Given the extensive attention paid to patients' (higher order) perceptual function, it is surprising that there have been no systematic analyses of basic oculomotor function in this population. Here 20 patients with posterior cortical atrophy, 17 patients with typical Alzheimer's disease and 22 hea

Sex-related gene expression in the posterodorsal medial amygdala of cycling female rats along with prolactin modulation of lordosis behavior.

The rat posterodorsal medial amygdala (MePD) is sexually dimorphic, has a high concentration of receptors for gonadal hormones and prolactin (PRL), and modulates reproduction. To unravel genetic and functional data for this relevant node of the social behavior network, we studied the expression of ERα, ERβ, GPER1, Kiss1, Kiss1R, PRGR, PRL, PRLR, EGR1, JAK2, STAT5A, and STAT5B in the MePD of males and females along the estrous cycle using the RT-qPCR technique. We also investigated whether PRL in the MePD would affect the sexual behavior display of proestrus females by microinjecting saline, th

PVN AVP neurons projecting to MeA modulate social recognition and anxiety-like behavior in female mice.

Arginine vasopressin (AVP) is well established in regulating social cognition and emotional responses in males. However, its roles in these processes in females and the underlying circuit mechanisms, remain poorly understood. In this study, female mice spent significantly more time sniffing the anogenital region of unfamiliar conspecifics compared to familiar individuals. Fiber photometry recordings revealed that the calcium activity in paraventricular nucleus of the hypothalamus (PVN) AVP neurons was elevated when sniffing unfamiliar mice relative to familiar ones. No significant differences

Estrogen and oxytocin receptors interplay in the medial amygdala to rapidly facilitate social recognition.

The estrogen, 17β-estradiol (E2), rapidly facilitates social recognition in various regions of the social brain network. The medial amygdala (MeA) is heavily involved in the processing of social cues and expresses the three main estrogen receptors (ER): ERα, ERβ, and G Protein-Coupled ER (GPER). All ERs, as well as oxytocin and its receptor (OTR) in the MeA are crucial for social recognition in female mice, suggesting an interplay between ERs and OTR in the rapid facilitation of this social behavior. Here, we demonstrate an interplay between E2 and the three ERs (ERα, ERβ, GPER) with OTR in th

Moderate prenatal alcohol exposure alters GABAergic transmission and the actions of acute alcohol in the medial central amygdala of adolescent rats.

Individuals with prenatal alcohol exposure (PAE) are at a higher risk for developing alcohol use disorder (AUD). Using a rat model of moderate PAE (mPAE) on gestational day 12 (G12; ∼2nd trimesters in humans), a critical period for amygdala development, we have shown disruptions in medial central amygdala (CeM) function, an important brain region associated with the development of AUD. In addition to this, acute ethanol (EtOH) increases GABA transmission in the CeM of rodents in a sex-dependent manner, a mechanism that potentially contributes to alcohol misuse. How mPAE alters acute alcohol's

Modulation of tonic immobility by GABAA and GABAB receptors of the medial amygdala.

Tonic immobility (TI) is a temporary state of profound motor inhibition associated with great danger as the attack of a predator. Previous studies carried out in our laboratory evidenced high Fos-IR in the posteroventral region of the medial nucleus of the amygdala (MEA) after induction of the TI response. Here, we investigated the effects of GABAA and GABAB of the MEA on TI duration. Intra-MEA injections of the GABAA agonist muscimol and GABAB agonist baclofen reduced TI response, while intra-MEA injections of the GABAA antagonist bicuculline and GABAB antagonist phaclofen increased the TI re
