Photoantimicrobial anthraquinones in Australian fungi of the genus Cortinarius.

Dermocyboid Cortinarius species of both hemispheres are usually intensely colored and known to contain anthraquinones. Australian dermocyboid fungi have distinct evolutionary histories which are clearly different from Northern Hemisphere species of similar appearance. Allopatric speciation often results in different metabolic adaptations. We were especially interested in the diversity of anthraquinonic pigments and in their photoantimicrobial potential. In this study 33 dried samples from eighteen Australian dermocyboid Cortinarius taxa were extracted with methanol and analyzed via HPLC-DAD-MS

Resurrection of Cortinarius coalescens: taxonomy, chemistry, and ecology.

Cortinarius coalescens Kärcher & Seibt is a rare European species of the subgenus Phlegmacium, section Phlegmacioides, neglected in recent molecular studies. New primers (CortF and CortR) designed for species in the section Phlegmacioides allowed to obtain ITS rDNA sequence data from the holotype collection of C. coalescens; according to the results, this epithet has priority over C. crassorum Rob. Henry ex Rob. Henry, C. pardinus Reumaux, and C. parargutus Bidaud, Moënne-Locc. & Reumaux. Morphological and ecological observations on recent collections of C. coalescens from the Czech Republic i

[Cortinarius orellanus: a poisonous mushroom causing late acute renal insufficiency].

Three cases of intoxication due to ingestion of Cortinarius orellanus are presented. The hallmark of the clinical picture is acute tubulo-interstitial nephritis appearing about 10 days after ingestion of the mushrooms. This time lag is specific for intoxication by Cortinarius orellanus. In all the three cases, acute gastroenteritic disorders and hepatocellular damage were observed initially and disappeared spontaneously. Histological examination reveals acute tubulointerstitial nephritis. Experimental data are discussed in regard to the clinical observations.

[Acute renal failure as a sequela of mushroom poisoning with Cortinarius speciocissimus].

BACKGROUND: Poisoning with the mushroom Cortinarius speciocissimus is a rare event. Within 2-17 days after ingestion unspecific symptoms appear. Characteristically, this state is soon followed by irreversible kidney damage which finally requires chronic hemodialysis. CASE REPORT: A 46-year-old man was admitted with nausea, vomiting and acute anuria 9 days after he had eaten Cortinarius speciocissimus. Hemodialysis had to be started. Kidney biopsy revealed an interstitial nephritis. This finding corresponds to earlier case reports about intoxication with orellanine, the poison of Cortinarius s

Mushroom poisoning due to Cortinarius speciosissimus: electron microscope study in rats.

The effects of a poisonous mushroom, Cortinarius speciosissimus, on rat kidney were studied by transmission electron microscopy. Suspension of dried mushroom (500 mg/kg body weight) was administered as a single dose directly into the esophagus. The kidneys were fixed by perfusion, and both cortex and medulla were sampled. C. speciosissimus toxin acted primarily on the epithelial cells of the proximal tubules. No changes were seen in the glomeruli. First ultrastructural changes were observed at 2 days in the renal cortex. The most prominent damage occurred at 5 days, when most of the proximal t

Flavin-dependent halogenases from Cortinarius mushrooms for the regioselective chlorination of anthraquinone pigments.

Flavin-dependent halogenases (FDHs) are valuable catalysts for the late-stage diversification of small molecule scaffolds, modulating their biological activities and/or introducing orthogonal functionalities for regiospecific derivatization. This study broadens the repertoire of FDHs by discovering two such enzymes (CkHAL1 and CsHAL2) from Cortinarius mushrooms that chlorinate anthraquinone skeletons from the emodin family when expressed in Saccharomyces cerevisiae. As opposed to the phenolic FDH CcRadH from the ascomycete Chaetomium chiversii that can modify only neutral anthraquinones such a

Synthesis and characterization of cortinarins - cryptic cyclic peptides from mushrooms of the genus Cortinarius.

Cortinarins have been described as cyclic peptides from mushrooms of the genus Cortinarius, and brought into context with mushroom intoxication, particularly causing kidney damage. Herein we report the first total synthesis of the bicyclic peptides cortinarin A and B using iodine-mediated Trp-Cys cyclization to establish their characteristic tryptathionine bridge. Peptide atropisomers, termed ansamers, were observed and characterized by NMR spectroscopy and molecular dynamics simulations. Conformational analysis showed that cortinarins adopt β-turn structures. Furthermore, cortinarins induce s

Intoxication by Cortinarius orellanus: detection and assay of orellanine in biological fluids and renal biopsies.

A woman suffering from acute tubulo-interstitial nephritis was admitted to the hospital ten days after deliberate intoxication by ingestion of Cortinarius orellanus. Orellanine, the main toxin responsible for orellanine poisoning, was detected in biological fluids and renal biopsies. It was assayed by direct spectrofluorimetry on two-dimensional thin-layer chromatograms after specific photodecomposition into orelline. The orellanine concentration was 6.12 mg/l in the plasma (10 days after ingestion). Orellanine levels in renal biopsies were 7 micrograms per 25 mm3 of the first biopsy (13 days

The effect of cyclophosphamide on the experimental inflammation induced by the toxic mushroom Cortinarius speciosissimus in the rat kidney.

A single intraperitoneal dose of cyclophosphamide (150 mg/kg) given at the same time as an oral dose of Cortinarius speciosissimus prevented the renal inflammation induced by this toxic mushroom in the male rat. Furthermore, a scar formation around dilated collecting ducts was clearly reduced by cyclophosphamide treatment. In general the only lesions observed in the cyclophosphamide treated animals were dilated collecting ducts in the outer medullary zone, the epithelia of which were either in regenerative mitosis or were atrophic. Apparently the primary sites of action of Cortinarius toxins i

Two new species of Cortinarius subgenus Telamonia section Firmiores (Agaricales, Basidiomycota) from Europe and North America.

In this study, we describe two new species of Cortinarius subgen. Telamonia sect. Firmiores: Cortinarius curiosus and C. ferrugineochraceus. Both species are well supported by phylogenetic analyses and are characterized by distinct morphological features. Cortinarius curiosus is resolved within the broader /Alboviolacei clade, forming a strongly supported lineage together with the three European taxa and C. obliquus from USA. Morphologically, C. curiosus can be distinguished from the European C. acutispissipes, C. alboviolaceus, and C. paralbocyaneus by first its silvery-whitish pileus and the

[Acute kidney failure following Cortinarius splendens (Fries) or "yellow clubbed foot" mushroom poisoning (so-called orellanus syndrome)].

Intoxications by cortinarii have only been recognized in recent years. Compared to other mushroom poisoning they are characterized by the long incubation time of up to ten days or more, and predominantly hepatorenal symptoms. In this paper the first two Swiss cases of intoxication with Cortinarius phlegmacium splendens are reported. The two patients ate two meals of this mushroom one week apart. Three days after the second meal the patients became acutely ill with severe lumbar pain, polyuria, polydipsia and constipation. Laboratory examinations revealed signs of hepato-renal toxicity with inc

Bioconcentration potential of 210Po and 210Pb in the gypsy mushroom (Cortinarius caperatus): Dietary intake and health risk assessment.

The Gypsy mushroom (Cortinarius caperatus) is prized as an edible fungus in northern climates. This species has a strong predilection for accumulating radiocaesium (134/137Cs), but the potential to accumulate other toxic nuclides is unknown. 210Po and 210Pb activity concentrations were determined in the fruiting bodies of this species and the associated forest soil (0-10 cm layer) collected at 20 sites across Northern Poland. Concentrations of 210Po and 210Pb in mushrooms ranged from 0.92 to 10.0 and 0.36 to 13.1 Bq∙kg-1 db and from 10.5 to 46.6 and 11.0 to 46.2 Bq∙kg-1 db in soils, respective
