Global transcriptomic response of Escherichia coli to p-coumaric acid.

The aromatic compound p-coumaric acid (p-CA) is a secondary metabolite produced by plants. This aromatic acid and derived compounds have positive effects on human health, so there is interest in producing them in biotechnological processes with recombinant Escherichia coli strains. To determine the physiologic response of E. coli W3110 to p-CA, dynamic expression analysis of selected genes fused to a fluorescent protein reporter as well as RNA-seq and RT-qPCR were performed. The observed transcriptional profile revealed the induction of genes involved in functions related to p-CA active export

Photocatalytic degradation of p-coumaric acid over TiO2 suspensions.

The photocatalytic degradation of p-coumaric acid, a biorecalcitrant phenolic compound typically found in olive processing and wine distillery waste waters, has been investigated in aqueous heterogeneous solutions containing semiconductor powders as photocatalysts. The disappearance of the organic molecule follows approximately a pseudo-first kinetic order according to the Langmuir-Hinselwood model. The most important reaction intermediates were determined by GS-MS and HPLC measurements, suggesting a reaction pathway for the photodegradation. Various commercial photocatalysts were compared wit

Biocatalysis with In-Situ Product Removal Improves p-Coumaric Acid Production.

Natural and pure p-coumaric acid has valuable applications, and it can be produced via bioprocessing. However, fermentation processes have so far been unable to provide sufficient production metrics, while a biocatalytic process decoupling growth and production historically showed much promise. This biocatalytic process is revisited in order to tackle product inhibition of the key enzyme tyrosine ammonia lyase. In situ product removal is proposed as a possible solution, and a polymer/salt aqueous two-phase system is identified as a suitable system for extraction of p-coumaric acid from an alka

[Heterologous biosynthesis and production optimization of p-coumaric acid in Streptomyces coelicolor].

p-coumaric acid is an important bioactive natural product and a crucial precursor for the synthesis of downstream phenylpropanoids. Compared with plant extraction and chemical synthesis methods, microbial production of p-coumaric acid offers unique advantages, including environmental friendliness, short production cycles, and high conversion efficiency. Streptomyces species possess a complete shikimate pathway and an extensive aromatic amino acid metabolic network, being promising chassis for the production of p-coumaric acid. This study aims to achieve efficient heterologous biosynthesis of p

Methanol-based biosynthesis of p-coumaric acid by engineered Pichia pastoris.

p-Coumaric acid (p-CA) is a key aromatic precursor for the biosynthesis of flavonoids, stilbenoids, and other high-value phenylpropanoids. While microbial production of p-CA typically relies on sugar-based substrates, methanol offers a sustainable and cost-effective alternative, though its use for aromatic biosynthesis remains unexplored. Here, we report the first de novo production of p-CA from methanol using engineered methylotrophic yeast Pichia pastoris. Through heterologous expression of a tyrosine ammonia-lyase and implementing a balanced push-pull strategy in the shikimate pathway using

Influence of p-coumaric acid on doxorubicin-induced oxidative stress in rat's heart.

The therapeutic value of doxorubicin (DOX) as anticancer antibiotic is limited by its cardiotoxicity. The implication of natural phenolic acids in the prevention of many pathologic diseases has been reported. Herein, the ability of p-coumaric (PC) acid, a member of phenolic acids, to protect rat's heart against DOX-induced oxidative stress was investigated. Three main groups of albino rats were used; DOX, PC, and PC plus DOX-receiving animals. Corresponding control animals were also used. DOX was administered i.p. in a single dose of 15mgkg(-1). PC alone, in a dose of 100mgkg(-1), was orally a

Metabolic engineering of Bacillus subtilis for enhanced p-Coumaric acid production and antimicrobial applications.

p-Coumaric acid (p-CA) is widely utilized in the food, pharmaceutical and other industries, and has traditionally been derived from plant extraction or chemical synthesis. However, p-CA synthesized by the safe B. subtilis remains poorly explored. In this study, we first engineered a recombinant B. subtilis strain (PBK) capable of synthesizing p-Coumaric acid, achieving an initial yield of 3.81 mg L-1. A high-yielding strain PBnprE was then developed through promoter substitution, with a yield reaching 60.92 mg L-1, and the yield of PBnprE was further increased to 304.04 mg L-1 by optimizing fe

Novel Effect of p-Coumaric Acid on Hepatic Lipolysis: Inhibition of Hepatic Lipid-Droplets.

p-coumaric acid (p-CA), a common plant phenolic acid with multiple bioactivities, has a lipid-lowering effect. As a dietary polyphenol, its low toxicity, with the advantages of prophylactic and long-term administration, makes it a potential drug for prophylaxis and the treatment of nonalcoholic fatty liver disease (NAFLD). However, the mechanism by which it regulates lipid metabolism is still unclear. In this study, we studied the effect of p-CA on the down-regulation of accumulated lipids in vivo and in vitro. p-CA increased a number of lipase expressions, including hormone-sensitive lipase (

Surface Enhanced Raman Scattering for Quantification of p-Coumaric Acid Produced by Escherichia coli.

The number of newly developed genetic variants of microbial cell factories for production of biochemicals has been rapidly growing in recent years, leading to an increased need for new screening techniques. We developed a method based on surface-enhanced Raman scattering (SERS) coupled with liquid-liquid extraction (LLE) for quantification of p-coumaric acid (pHCA) in the supernatant of genetically engineered Escherichia coli (E. coli) cultures. pHCA was measured in a dynamic range from 1 μM up to 50 μM on highly uniform SERS substrates based on leaning gold-capped nanopillars, which showed an

Inhibitory effect of p-coumaric acid by Rhodiola sachalinensis on melanin synthesis in B16F10 cells.

Rhodiola has been widely used in traditional Asian medicine. In this study, we tested the hypopigmentation effects of R. sachalinensis and its active compounds including catechin, chlorogenic acid, p-coumaric acid, and p-tyrosol. Results have shown that only p-coumaric acid inhibits melanin synthesis in B16F10 cells. However, p-coumaric acid did not inhibit tyrosinase activity when L-DOPA was used as a substrate. Instead, p-coumaric acid inhibited tyrosinase activity when L-tyrosine was used as a substrate. We further analyzed the changes of cAMP responsive element binding protein (CREB) phosp

Development of an Efficient Bifunctional Endoxylanase/Feruloyl Esterase Chimera for Coproduction of Ferulic Acid, p-Coumaric Acid, and Xylooligosaccharides.

Bifunctional endoxylanases/feruloyl esterases that can efficiently produce hydroxycinnamic acids and xylooligosaccharides from agricultural residues are highly desirable but underexplored. Here, we report the development of a bifunctional chimera, XynB-SnT-FaeB, which shares low sequence identity (<23%) with its homologues and contains a novel linker peptide. It can hydrolyze model hydroxycinnamate esters and xylans. Due to interdomain synergism, its feruloyl esterase and endoxylanase catalytic efficiencies (kcat/Km) toward feruloylated arabinoxylan were 2.95 and 1.60 times those of FaeB and X

Catabolism of coniferyl aldehyde, ferulic acid and p-coumaric acid by Saccharomyces cerevisiae yields less toxic products.

BACKGROUND: Lignocellulosic substrates and pulping process streams are of increasing relevance to biorefineries for second generation biofuels and biochemical production. They are known to be rich in sugars and inhibitors such as phenolic compounds, organic acids and furaldehydes. Phenolic compounds are a group of aromatic compounds known to be inhibitory to fermentative organisms. It is known that inhibition of Sacchromyces cerevisiae varies among phenolic compounds and the yeast is capable of in situ catabolic conversion and metabolism of some phenolic compounds.
