Nucleotide sequence, genome organization and phylogenetic analysis of Strawberry pallidosis associated virus, a new member of the genus Crinivirus.

The complete nucleotide sequence of Strawberry pallidosis associated virus (SPaV), a newly identified member of the genus Crinivirus, family Closteroviridae has been determined. RNA 1 is 8067 nucleotides long and encodes at least three open reading frames (ORFs). The first ORF (ORF 1a) specifies a multifunctional protein that has papain-like proteinase, methyltransferase and RNA helicase domains. The RNA-dependent-RNA polymerase is encoded in ORF 1b and is probably expressed by a +1 ribosomal frameshift. The 3' ORF of RNA 1 encodes a small protein with two potential transmembrane helices. Discovery of Novel Chromone Derivatives Containing a Sulfonamide Moiety as Anti-ToCV Agents through the Tomato Chlorosis Virus Coat Protein-Oriented Screening Method.

A number of novel chromone derivatives containing sulfonamide moieties were designed and synthesized, and the activity of compounds against tomato chlorosis virus (ToCV) was assessed using the ToCVCP-oriented screening method. Comparative molecular field analysis (CoMFA) and comparative molecular similarity index analysis (CoMSIA) models were established based on the dissociation constant (Kd) values of the target compounds, and compound 35 was designed and synthesized with the aid of CoMFA and CoMSIA models. The study of affinity interaction indicated that compound 35 exhibited excellent affi

Physalis angulata: A New Natural Host of Tomato chlorosis virus in Brazil.

Tomato chlorosis virus (ToCV) and Tomato infectious chlorosis virus (TICV) are the two Solanaceae-infecting Crinivirus species (family Closteroviridae) of worldwide importance. In Brazil, only ToCV has been detected under natural conditions infecting tomato (Solanum lycopersicum), sweet pepper (Capsicum annuum), and potato (S. tuberosum), causing foliar chlorosis (1, 3). However, there are no formal reports of alternative weed hosts of ToCV. During crop surveys in Capão Bonito, São Paulo State, Brazil (May 2011), a high incidence (above 20%) of plants of the weed, cut leaf ground cherry (Physa

Suppression of Bta11975, an α-glucosidase, by RNA interference reduces transmission of tomato chlorosis virus by Bemisia tabaci.

BACKGROUND: Tomato chlorosis virus (ToCV) is mainly vectored by Bemisia tabaci in China, which has a worldwide distribution, and greatly reduces the yields of tomato and other vegetables. At present, control of ToCV has been focused mainly by the use of insecticides to control whitefly populations. Transcriptome sequencing showed high expression of the B. tabaci Bta11975 gene, an α-glucosidase (AGLU) during ToCV acquisition by whitefly Mediterranean (MED) species. To investigate the role of Bta11975 gene in ToCV acquisition and transmission by B. tabaci MED, we used RNA interference (RNAi) to

Tomato Chlorosis Virus Minor Coat Protein as a Novel Target To Screen Antiviral Drugs.

Minor coat protein (mCP), an important component of tomato chlorosis virus (ToCV), plays a significant role in the process of virus assembly and movement and is directly related to the virus-insect transmission. Therefore, ToCV mCP could be considered as a potent target for anti-ToCV drugs. In this study, ToCV mCP was first cloned, expressed, purified, and a novel target to screen the antiviral agents. The results showed that some antiviral compounds bound to ToCV mCP with strongly affinities in vitro, including quinazoline derivatives 4a and 4b, Ningnanmycin, and Ribavirin. Subsequently, thre

Integrating the tomato chlorosis virus with the tobacco rattle virus facilitates the direct infection of tomato.

Tomato chlorosis virus (ToCV) threatens tomato production, resulting in substantial yield losses. An efficient inoculation system for ToCV can accelerate the isolation and utilization of tomato-resistant genetic resources. However, infectious ToCV clones cannot directly infect tomato plants. In this study, we cloned the full-length ToCV genome, including RNA1 and RNA2 molecules, and integrated their corresponding DNA fragments separately into the less virulent tobacco rattle virus-based pTRV2GW vector. Agrobacterium suspensions carrying pTRV1, pTRV2GW-RNA1, and pTRV2GW-RNA2 plasmids were mixed

Real-time detection of cucurbit chlorotic yellows virus by RT-LAMP.

Cucurbit chlorotic yellows virus (CCYV) of the genus Crinivirus and family Closteroviridae, is an emerging infectious agent transmitted by whiteflies that mainly infects cucumber, melon and watermelon plants. Here, we developed a procedure based on reverse transcription (RT) followed by loop-mediated isothermal amplification (LAMP) to detect CCYV in real-time by using a fluorescent dye and incubating at 65 °C. This method detected CCYV in RNA extracts at about 10 min and in nylon membrane-filtered crude extracts between 55 and 95 min. The detection was sensitive, ten times higher than RT follo

Silencing of the Prophenoloxidase Gene BtPPO1 Increased the Ability of Acquisition and Retention of Tomato chlorosis virus by Bemisia tabaci.

Tomato chlorosis virus (ToCV) has seriously impacted tomato production around the world. ToCV is semi-persistently transmitted by the whitefly, Bemisia tabaci, which is a serious agricultural pest in the world. However, the interaction mechanism between ToCV and its whitefly vector is still poorly understood. Our previous transcriptome analysis demonstrated that the expression level of an immune-related gene, prophenoloxidase (PPO), in B. tabaci increased after ToCV acquisition, which indicates that the PPO may be involved in the interaction mechanism between the ToCV and its vector. To determ

Field evaluation of Cucurbit yellow stunting disorder virus transmission by Bemisia tabaci.

Cucurbit yellow stunting disorder virus (CYSDV) is a whitefly-transmitted Crinivirus (Closteroviridae) that impacts melon production in many parts of the world including the USA. It has been responsible for melon crop loss in the southwestern U.S. since 2006 when it was first identified. Control strategies have revolved mainly around chemical control, but research to identify suitable products and approaches to implementing them have lagged. The current study investigated the performance of four systemic insecticides in the field while concurrently tracking CYSDV disease progression after cont

Multiplex RT-PCR for simultaneous detection of seven cucurbit viruses found in Florida cucurbits.

Virus detection of phytopathogens has benefited greatly from advancements in diagnostic technologies; however, not all laboratories have access to high-cost platforms. To address this limitation, a single-tube multiplex reverse transcription-polymerase chain reaction (m50RT-PCR) assay was developed for the simultaneous detection of six RNA viruses - cucurbit chlorotic yellows virus (CCYV), squash vein yellowing virus (SqVYV), papaya ringspot virus-W (PRSV-W), watermelon crinkle leaf-associated virus 1 (WCLaV-1) and WCLaV-2, cucurbit yellow stunting disorder virus (CYSDV) and one DNA virus, cuc

Crude sap based colorimetric duplex RT-RPA assay for rapid detection of citrus yellow vein clearing virus and citrus yellow mottle-associated virus in citrus.

Citrus yellow vein clearing virus (CYVCV) and citrus yellow mottle-associated virus (CiYMaV) frequently co-infect citrus, posing challenges for reliable diagnosis and certification systems. In this study, a duplex reverse transcription recombinase polymerase amplification (RT-RPA) assay was developed for the simultaneous detection of both viruses using crude leaf sap of citrus. The assay operates under isothermal conditions (40 °C, 25 min) and incorporates an endogenous internal control (Elongation factor 1α) to support assay reliability and reduce the likelihood of false-negative results. The

In Vitro Identification and In Vivo Confirmation of Inhibitors for Sweet Potato Chlorotic Stunt Virus RNA Silencing Suppressor, a Viral RNase III.

Sweet potato virus disease (SPVD), caused by synergistic infection of Sweet potato chlorotic stunt virus (SPCSV) and Sweet potato feathery mottle virus (SPFMV), is responsible for substantial yield losses all over the world. However, there are currently no approved treatments for this severe disease. The crucial role played by RNase III of SPCSV (CSR3) as an RNA silencing suppressor during the viruses' synergistic interaction in sweetpotato makes it an ideal drug target for developing antiviral treatment. In this study, high-throughput screening (HTS) of small molecular libraries targeting CSR
