Mitochondrial genome of Protobothrops jerdonii (Squamata: Viperidae: Crotalinae).

Protobothrops jerdonii is a common venomous snake that is widely distributed in southwestern China and other adjacent countries of Asia. In this study, the complete mitochondrial genome of P. jerdonii was determined. The circle genome with the 17,239 bp total length contained 13 protein-coding genes, 22 transfer RNA genes, 2 ribosomal RNA genes, and 2 control regions. Overall base composition of the complete mtDNA was 33.13% A, 25.07% T, 29.31% C, and 12.50% G. All the genes in P. jerdonii were distributed on the H-strand, except for the ND6 subunit gene and eight tRNA genes which were encoded

Accelerated evolution of crotalinae snake venom gland serine proteases.

Eight cDNAs encoding serine proteases isolated from Trimeresurus flavoviridis (habu snake) and T. gramineus (green habu snake) venom gland cDNA libraries showed that nonsynonymous nucleotide substitutions have accumulated in the mature protein-coding regions to cause amino acid changes. Southern blot analysis of T. flavoviridis genomic DNAs using two proper probes indicated that venom gland serine protease genes form a multigene family in the genome. These observations suggest that venom gland serine proteases have diversified their amino acid sequences in an accelerating manner. Since a simil

Myocardial infarction during treatment of Crotalinae envenomation: A case report.

While myocardial infarction is a rare, but known, potential side effect of snakebite envenomation, snake antivenom has thus far not been associated with any cardiovascular adverse events. We report the case of a 71-year-old man who developed an anterolateral MI during administration of Crotalidae immune F(ab')2 (equine) (ANAVIP), given as treatment for Crotalidae envenomation. The patient required cardiac catheterization with stenting of the left anterior descending artery and was discharged two days later on long-term clopidogrel and aspirin. Treatment of MI in the setting of envenomation sho

Toxicity of venom from the mamushi, Gloydius blomhoffii, (Squamata, Crotalinae) to centipedes.

Vipers include approximately 300 species and usually feed on vertebrates, but over 30 species of them occasionally eat centipedes. Centipedes have been also known to occur in stomach contents of a Japanese pit viper, mamushi, Gloydius blomhoffii. Toxicity of the venom of mamushi to small mammals has been well studied, but there is no information concerning its toxicity to arthropods. Here, we studied the toxicity of the raw venom to the red-headed centipede, Scolopendra subspinipes mutilans, by comparing with the toxicity to two other common prey animals, a house mouse, Mus musclus, and a pond

Neutralising effects of small molecule toxin inhibitors on nanofractionated coagulopathic Crotalinae snake venoms.

Repurposing small molecule drugs and drug candidates is considered as a promising approach to revolutionise the treatment of snakebite envenoming. In this study, we investigated the inhibiting effects of the small molecules varespladib (nonspecific phospholipase A2 inhibitor), marimastat (broad spectrum matrix metalloprotease inhibitor) and dimercaprol (metal ion chelator) against coagulopathic toxins found in Crotalinae (pit vipers) snake venoms. Venoms from Bothrops asper, Bothrops jararaca, Calloselasma rhodostoma and Deinagkistrodon acutus were separated by liquid chromatography, followed

Electrophysiological modulation of cholinergic neurotransmission by biologically active peptides from Bothrops bilineatus (Viperidae: Crotalinae) venom.

The venom of Bothrops bilineatus, an Amazonian arboreal viper, induces neurotoxicity in mammalian nerve-muscle preparations that is characterized by initial neuromuscular facilitation followed by irreversible blockade. Up until now, the toxins responsible for the neuromuscular excitatory action of this venom have remained unidentified. In this study, we characterized two presynaptically active peptides from B. bilineatus venom using mass spectrometry and electrophysiological analysis at the neuromuscular junction. Fractionation by size-exclusion chromatography yielded eight fractions, with fra

Phospholipase A2 inhibitors (βPLIs) are encoded in the venom glands of Lachesis muta (Crotalinae, Viperidae) snakes.

Phospholipase A(2) inhibitors (PLIs) are glycoproteins secreted by snake liver into the circulating blood aiming the self-protection against toxic venom phospholipases A(2). In the present study, we describe the first complete nucleotide sequence of a βPLI from venom glands of a New World snake, Lachesis muta. The deduced primary structure was compared to other known βPLIs and recent literature findings of other possible roles of PLIs in snakes are discussed.

Structural, biochemical and immunochemical characterization of an acidic phospholipase A2 from Lachesis acrochorda (Viperidae: Crotalinae) venom.

Viperids of the genus Lachesis, also known as bushmasters, are capable of injecting great amounts of venom that cause severe envenomation incidents. Since phospholipases type A2 are mainly involved in edema and myonecrosis within the snakebite sites, in this work, the isolation, amino acid sequence and biochemical characterization of the first phospholipase type A2 from the venom of Lachesis acrochorda, named Lacro_PLA2, is described. Lacro_PLA2 is an acidic aspartic 49 calcium-dependent phospholipase A2 with 93% similarity to the L. stenophrys phospholipase. Lacro_PLA2 has a molecular mass of

Comparative in-vivo toxicity of venoms from South Asian hump-nosed pit vipers (Viperidae: Crotalinae: Hypnale).

BACKGROUND: Envenoming by south Asian hump-nosed pit vipers (Genus: Hypnale) is a significant health issue in Sri Lanka and in peninsular India. Bites by these snakes frequently lead to local envenoming, coagulopathy and acute renal failure even resulting in death. Recently the genus was revised and the existence of three species viz H. hypnale, H. nepa and H. zara were recognized. There is, however, a paucity of information on the toxicity of the venoms of these species. Hence, we compared the toxic effects of the three Hypnale venoms using BALB/c mice. FINDINGS: Intraperitoneal median letha

Ammodytoxin C gene helps to elucidate the irregular structure of Crotalinae group II phospholipase A2 genes.

Ammodytoxin C is a presynaptically neurotoxic phospholipase A2 (PLA2) expressed in the venom glands of Vipera ammodytes (subfamily Viperinae). The gene spans more than 4 kb and consists of five exons and four introns characteristic of group II phospholipase A2 genes. The first exon encodes the 5' untranslated region, the second exon encodes most of the signal peptide, while exons 3-5 encode three parts of the mature protein. Comparison of the Crotalinae and Viperinae PLA2 genes has shown that Crotalinae PLA2 retain the first intron in their mRNAs. The apparent cause of this retention is a dele

Cross-recognition of a pit viper (Crotalinae) polyspecific antivenom explored through high-density peptide microarray epitope mapping.

Snakebite antivenom is a 120 years old invention based on polyclonal mixtures of antibodies purified from the blood of hyper-immunized animals. Knowledge on antibody recognition sites (epitopes) on snake venom proteins is limited, but may be used to provide molecular level explanations for antivenom cross-reactivity. In turn, this may help guide antivenom development by elucidating immunological biases in existing antivenoms. In this study, we have identified and characterized linear elements of B-cell epitopes from 870 pit viper venom protein sequences by employing a high-throughput methodolo

Identification and functional analysis of a novel bradykinin inhibitory peptide in the venoms of New World Crotalinae pit vipers.

A novel undecapeptide has been isolated and structurally characterized from the venoms of three species of New World pit vipers from the subfamily, Crotalinae. These include the Mexican moccasin (Agkistrodon bilineatus), the prairie rattlesnake (Crotalus viridis viridis), and the South American bushmaster (Lachesis muta). The peptide was purified from all three venoms using a combination of gel permeation chromatography and reverse-phase HPLC. Automated Edman degradation sequencing and MALDI-TOF mass spectrometry established its peptide primary structure as: Thr-Pro-Pro-Ala-Gly-Pro-Asp-Val-Gly

Venom composition, toxicity and cross-neutralization by PoliVal-ICP antivenom, of Mesoamerican jumping pitvipers genus Metlapilcoatlus (Viperidae: Crotalinae).

BACKGROUND: The genus Metlapilcoatlus was recently erected to include six species of stout venomous snakes, known as the jumping pitvipers, which inhabit mountainous areas of Mesoamerica. This group maintains affinity with Atropoides picadoi, another jumping pitviper with restricted distribution in Costa Rica and Panama. Although the venom of A. picadoi and a couple of Metlapilcoatlus species has previously been characterized, little is known about the interspecific and intraspecific variation of the other species that comprise the genus. In this work, we characterize the venoms of five out of

The Protective Action of Coutarea hexandra (Rubiaceae) on the Neuromuscular Blockade Induced by Lachesis muta muta (Viperidae: Crotalinae) Venom.

Envenomations by snakes represent a neglected health problem in tropical and subtropical countries. In South America, Lachesis muta occasionally causes severe human envenomation, with treatment being conditioned to an unspecific antivenom. In this work, we examined the neutralizing ability of Coutarea hexandra stem bark hydroalcoholic extract (Ch-E), including the commercial phytochemicals coumarin and quinine, on the neuromuscular blockade induced by L. m. muta venom in mouse phrenic nerve-diaphragm preparation. Inside the head of Crotalus durissus LINNAEUS, 1758 (Serpentes, Viperidae, Crotalinae): Macroscopic description of the brain with ontogenetic insights.

Neuroanatomy studies in vertebrates have garnered significant attention in recent years, particularly driven by advancements in computerized tomography imaging techniques. Nonetheless, these advancements remain largely constrained to specific vertebrate groups, notably mammals, birds, and fish, leaving studies in reptiles at an incipient stage. In this work, we aim to describe in detail the macroscopic morphology of the brain of Crotalus durissus based on a sample of four young and four adult individuals-three male and five female specimens, providing the first detailed description of the brai

Interisland mutation of a novel phospholipase A2 from Trimeresurus flavoviridis venom and evolution of Crotalinae group II phospholipases A2.

Trimeresurus flavoviridis (Crotalinae) snakes inhabit the southwestern islands of Japan: Amami-Oshima, Tokunoshima, and Okinawa. Affinity and conventional chromatographies of Amami-Oshima T. flavoviridis venom led to isolation of a novel phospholipase A2 (PLA2). This protein was highly homologous (91%) in sequence to trimucrotoxin, a neurotoxic PLA2, which had been isolated from T. mucrosquamatus (Taiwan) venom, and exhibited weak neurotoxicity. This protein was named PLA-N. Its LD50 for mice was 1.34 microg/g, which is comparable to that of trimucrotoxin. The cDNA encoding PLA-N was isolated
