Targeting Cryptochromes in Chronic Diseases.

The circadian clock generates 24-h molecular rhythms through transcription-translation negative feedback loops (TTFLs) and regulates daily physiological processes such as sleep-wake cycles, body temperature, hormone secretion, metabolism, and immune function. Cryptochromes (CRY1 and CRY2) are essential components of the mammalian circadian clock as the transcriptional repressors in TTFLs. Disruption of the circadian clock by shiftwork or mutations of clock genes disturbs daily physiological rhythms and poses serious risks to human health. Misregulations of CRY in humans and mice induce chronic

Role of Cryptochrome-1 and Cryptochrome-2 in Aldosterone-Producing Adenomas and Adrenocortical Cells.

Mice lacking the core-clock components, cryptochrome-1 (CRY1) and cryptochrome-2 (CRY2) display a phenotype of hyperaldosteronism, due to the upregulation of type VI 3β-hydroxyl-steroid dehydrogenase (Hsd3b6), the murine counterpart to the human type I 3β-hydroxyl-steroid dehydrogenase (HSD3B1) gene. In the present study, we evaluated the role of CRY1 and CRY2 genes, and their potential interplay with HSD3B isoforms in adrenal pathophysiology in man. Forty-six sporadic aldosterone-producing adenomas (APAs) and 20 paired adrenal samples were included, with the human adrenocortical cells HAC15 u

Atomistic Insights into Cryptochrome Interprotein Interactions.

It is striking that the mechanism by which birds sense geomagnetic fields during the biannual migration seasons is not entirely understood. A protein believed to be responsible for avian magnetoreception is the flavoprotein cryptochrome (CRY), which fulfills many of the criteria for a magnetic field sensor. Some experiments, however, indicate that magnetoreception in birds may be disturbed by extremely weak radio frequency fields, an effect that likely cannot be described by an isolated CRY protein. An explanation can possibly be delivered if CRY binds to another protein inside a cell that wou

Cryptochrome-Related Abiotic Stress Responses in Plants.

It is well known that light is a crucial environmental factor that has a fundamental role in plant growth and development from seed germination to fruiting. For this process, plants contain versatile and multifaceted photoreceptor systems to sense variations in the light spectrum and to acclimate to a range of ambient conditions. Five main groups of photoreceptors have been found in higher plants, cryptochromes, phototropins, UVR8, zeitlupes, and phytochromes, but the last one red/far red wavelengths photoreceptor is the most characterized. Among the many responses modulated by phytochromes, t

Circadian clock cryptochrome proteins regulate autoimmunity.

The circadian system regulates numerous physiological processes including immune responses. Here, we show that mice deficient of the circadian clock genes Cry1 and Cry2 [Cry double knockout (DKO)] develop an autoimmune phenotype including high serum IgG concentrations, serum antinuclear antibodies, and precipitation of IgG, IgM, and complement 3 in glomeruli and massive infiltration of leukocytes into the lungs and kidneys. Flow cytometry of lymphoid organs revealed decreased pre-B cell numbers and a higher percentage of mature recirculating B cells in the bone marrow, as well as increased num

Evolution of Proteins of the DNA Photolyase/Cryptochrome Family.

Proteins of the cryptochrome/DNA photolyase family (CPF) are phylogenetically related and structurally conserved flavoproteins that perform various functions. DNA photolyases repair DNA damage caused by UV-B radiation by exposure to UV-A/blue light simultaneously or subsequently. Cryptochromes are photoreceptor proteins regulating circadian clock, morphogenesis, phototaxis, and other responses to UV and blue light in various organisms. The review describes the structure and functions of CPF proteins, their evolutionary relationship, and possible functions of the CPF ancestor protein.

Functional Characterization of Conserved Positively Charged Residues Surrounding the Ligand-Binding Pocket in Plant Cryptochromes.

Plant cryptochromes are blue light-responsive photoreceptor proteins that regulate various photoresponses throughout plant growth and development. Compared with their homologous CPD I/III photolyases, plant cryptochromes have lost DNA repair activity but evolved signal transduction functions. The structurally conserved ligand-binding pocket, which corresponds to the photolyase active site, exhibits distinctive features in plant cryptochromes, with key binding residues being altered relative to photolyases. Notably, the distribution of charged amino acids around this pocket differs markedly bet

Microscopic dissection of chirality-induced spin selectivity in cryptochrome.

Chirality-induced spin selectivity (CISS) has been proposed as a mechanism that enhances magnetic field sensitivity of radical pairs in magnetosensitive proteins, such as cryptochrome, through the recruitment of net spin polarization during radical pair formation and recombination. However, existing studies have been largely limited to radical pairs undergoing one-step electron transfer recombination or phenomenological CISS models. Here, we develop a microscopically derived model of CISS acting during both formation and recombination within a radical pair in a multi-step donor-bridge-acceptor

Role of heterozygous and homozygous alleles in cryptochrome-deficient mice.

The circadian rhythms of physiology and behavior are based on molecular systems at the cellular level, which are regulated by clock genes, including cryptochrome genes, Cry1 and Cry2. In mammals, the circadian pacemaker in the suprachiasmatic nucleus (SCN) of the hypothalamus maintains the circadian rhythms throughout the body. Cry1 and Cry2 play distinct roles in regulating the circadian rhythm. However, the different effects of manipulating clock genes in heterozygous and homozygous alleles, Cry1 and Cry2, remain unclear. Therefore, this study aimed to understand the haplosufficiency of cryp

Deregulated expression of cryptochrome genes in human colorectal cancer.

BACKGROUND: Circadian disruption and deranged molecular clockworks are involved in carcinogenesis. The cryptochrome genes (CRY1 and CRY2) encode circadian proteins important for the functioning of biological oscillators. Their expression in human colorectal cancer (CRC) and in colon cancer cell lines has not been evaluated so far. METHODS: We investigated CRY1 and CRY2 expression in fifty CRCs and in the CaCo2, HCT116, HT29, SW480 cell lines. RESULTS: CRY1 (p = 0.01) and CRY2 (p < 0.0001) expression was significantly changed in tumour tissue, as confirmed in a large independent CRC dataset.

Cryptochrome 1b: a possible inducer of visual lateralization in pigeons?

The visual system of adult pigeons shows a lateralization of object discrimination with a left hemispheric dominance on the behavioural, physiological and anatomical levels. The crucial trigger for the establishment of this asymmetry is the position of the embryo inside the egg, which exposes the right eye to light falling through the egg shell. As a result, the right-sided retina is more strongly stimulated with light during embryonic development. However, it is unknown how this embryonic light stimulation is transduced to the brain as rods and cones are not yet functional. A possible solutio

Cryptochrome interaction networks across different tissues in Drosophila melanogaster.

BACKGROUND: Drosophila CRYPTOCHROME (dCRY) is a blue light-sensitive protein involved in various biological processes, including photoreception, circadian rhythm regulation, synaptic plasticity, and magnetoreception. Its circadian function is strictly connected with light: upon light exposure, dCRY undergoes a conformational change, becoming active and binding to various proteins. However, it can also form complexes in the absence of light, with its function varying depending on the cell type in which it is expressed. RESULTS: Here, we use an experimental approach based on co-immunoprecipitati

Magnetoreception: activated cryptochrome 1a concurs with magnetic orientation in birds.

The radical pair model proposes that the avian magnetic compass is based on radical pair processes in the eye, with cryptochrome, a flavoprotein, suggested as receptor molecule. Cryptochrome 1a (Cry1a) is localized at the discs of the outer segments of the UV/violet cones of European robins and chickens. Here, we show the activation characteristics of a bird cryptochrome in vivo under natural conditions. We exposed chickens for 30 min to different light regimes and analysed the amount of Cry1a labelled with an antiserum against an epitope at the C-terminus of this protein. The staining after e

Are stress responses to geomagnetic storms mediated by the cryptochrome compass system?

A controversial body of literature demonstrates associations of geomagnetic storms (GMS) with numerous cardiovascular, psychiatric and behavioural outcomes. Various melatonin hypotheses of GMS have suggested that temporal variation in the geomagnetic field (GMF) may be acting as an additional zeitgeber (a temporal synchronizer) for circadian rhythms, with GMS somehow interfering with the hypothesized system. The cryptochrome genes are known primarily as key components of the circadian pacemaker, ultimately involved in controlling the expression of the hormone melatonin. Cryptochrome is identif

Circadian and photic regulation of cryptochrome mRNAs in the rat pineal gland.

Expressions of Cry1 and Cry2 mRNA in the rat pineal gland were examined by Northern blot. The levels of Cry1 and Cry2 transcript had a marked circadian rhythm with peaks at circadian time (CT) 20 in constant darkness. But the amplitude of the Cry1 rhythm was higher than that of the Cry2 rhythm. Furthermore, a significant increase in Cry1 mRNA levels was caused by light pulse given at CT 16 but not at CT 4, but the expression of Cry2 was not significantly induced by light pulses given at either CT 4 or CT 16. These results suggest that Cry1 in the pineal gland is regulated by photic and circadi

Cryptochrome 2 stabilization alleviates psoriasis by inhibiting keratinocyte hyperproliferation and inflammation.

Psoriasis, a chronic inflammatory skin disorder, is characterized by aberrant keratinocyte proliferation and immune dysregulation. Although cryptochrome 2 (CRY2), a circadian rhythm regulator, has been implicated in inflammatory conditions, its role in the psoriasis pathogenesis remains elusive. Here, we report a marked downregulation of CRY2 expression in psoriasis, which was reversed upon biological therapy, suggesting its pivotal involvement in disease progression. Using cellular, murine, and in vitro human tissue models, we revealed that CRY2 deficiency exacerbates inflammatory and hyperpr

Chickens' Cry2: molecular analysis of an avian cryptochrome in retinal and pineal photoreceptors.

We have identified and characterized an ortholog of the putative mammalian clock gene cryptochrome 2 (Cry2) in the chicken, Gallus domesticus. Northern blot analysis of gCry2 mRNA indicates widespread distribution in central nervous and peripheral tissues, with very high expression in pineal and retina. In situ hybridization of chick brain and retina reveals expression in photoreceptors and in visual and circadian system structures. Expression is rhythmic; mRNA levels predominate in late subjective night. The present data suggests that gCry2 is a candidate avian clock gene and/or photopigment

The roles of Cryptochrome-1: the circadian clock as a control point in cancer therapy.

In this review, we carefully describe the composition and operation of cryptochrome-1 (CRY1). We introduce four related signaling pathways, namely cAMP/PKA/CREB signaling way, HIFα signaling way, NF-κB signaling way, STAT3 signaling way, FOXO signaling pathway and Hedgehog (HH) signaling pathway. We also emphasize the current understanding of the complex roles CRY1 plays in the progression of various solid tumors. We propose that its function represents a novel therapeutic target that could pave the way for new cancer treatments. Additionally, we summarize three emerging therapeutic strategies

Cryptochrome deficiency enhances transcription but reduces protein levels of pineal Aanat.

Cryptochrome (Cry) 1 and 2 are essential for circadian rhythm generation, not only in the suprachiasmatic nucleus, the site of the mammalian master circadian clock, but also in peripheral organs throughout the body. CRY is also known as a repressor of arylalkylamine-N-acetyltransferase (Aanat) transcription; therefore, Cry deficiency is expected to induce constantly high pineal melatonin content. Nevertheless, we previously found that the content was consistently low in melatonin-proficient Cry1 and Cry2 double-deficient mice (Cry1−/−/Cry2−/−) on C3H background. This study aims to clarify the

Neuroglobin correlates with cryptochrome-1 in obstructive sleep apnea with primary aldosteronism.

BACKGROUND: Neuroglobin (Ngb) is highly expressed in the suprachiasmatic nucleus, and can regulate Per1 gene expression. It is still not known whether Ngb also influences Cryptochrome (Cry). Cry is implicated in hypertension and primary aldosteronism (PA) in mice. There is a strong correlation between Obstructive Sleep Apnea (OSA) and PA. We propose to prove that Ngb and Cry play a role in OSA with PA. METHODS: Subjects were recruited consecutively from residents of Jakarta, Indonesia; subjects aged 30-65 years with moderate to severe OSA and hypertension were included in the study. OSA was d
