Differential response of lens crystallins and corneal crystallins in degenerative corneas.

Corneal degenerations, occurring either spontaneously or as a complication to other diseases, cause vision problems by endangering corneal transparency. Our past cornea research projects involving mice revealed that some recruited mice presented corneal problems similar to human corneal degeneration. The present study examines the histology of diseased mice corneas, including ultrastructure. Genome-wide microarray and proteomic methods were utilized to screen for molecular changes in the diseased corneas. It was found that abnormalities affected mainly anterior layers of the corneas. The most

Ontogeny of human lens crystallins.

The soluble proteins from prenatal and neonatal human lenses were fractionated by gel filtration into four distinct size classes viz. high molecular weight alpha-crystallin (HM-alpha), alpha-crystallin, intermediate molecular weight (IMW) proteins and low molecular weight (LMW) proteins. Extinction coefficients of the isolated proteins were determined and used to calculate the proportions of each fraction on a weight basis. The distributions of polypeptides within each of these fractions were analyzed by SDS gel electrophoresis and isoelectric focussing, followed by densitometric scanning of t

Role of crystallins in diabetic complications.

BACKGROUND: Crystallins are the major structural proteins of vertebrate eye lens responsible for maintaining the refractive index of the lens. However, recent studies suggest that they also have a functional significance in non-lenticular tissues. Prolonged uncontrolled diabetes results in the development of macro and microvascular complications that are the leading causes of morbidity and mortality in diabetic patients all over the world. SCOPE OF REVIEW: Recent studies have shown that crystallins play an instrumental role in diabetes and its complications. Therefore, this review highlights

Gamma crystallins of the human eye lens.

BACKGROUND: Protein crystallins co me in three types (α, β and γ) and are found predominantly in the eye, and particularly in the lens, where they are packed into a compact, plastic, elastic, and transparent globule of proper refractive power range that aids in focusing incoming light on to the retina. Of these, the γ-crystallins are found largely in the nuclear region of the lens at very high concentrations (>400 mg/ml). The connection between their structure and inter-molecular interactions and lens transparency is an issue of particular interest. SCOPE OF REVIEW: We review the origin and p

Effect of lens crystallins aggregation on cataract formation.

Cataracts represent one of the leading causes of blindness globally. The World Health Organization's 2019World Report on Vision indicates that approximately 65.2 million individuals worldwide experience varying degrees of visual impairment or blindness attributable to cataracts. The prevalence of this condition is significantly increasing, largely due to the accelerated aging of the global population. The lens of the eye is primarily composed of crystallins, which are categorized into three families: α-, β-, and γ-crystallins. The highly ordered structure and interactions among these crystalli

The genetic landscape of crystallins in congenital cataract.

BACKGROUND: The crystalline lens is mainly composed of a large family of soluble proteins called the crystallins, which are responsible for its development, growth, transparency and refractive index. Disease-causing sequence variants in the crystallins are responsible for nearly 50% of all non-syndromic inherited congenital cataracts, as well as causing cataract associated with other diseases, including myopathies. To date, more than 300 crystallin sequence variants causing cataract have been identified. METHODS: Here we aimed to identify the genetic basis of disease in five multi-generation

Protein-protein association properties of human βB2-crystallins.

Protein-protein association events are involved in many physiological and pathological processes. Cataract disease is a pathology that manifests protein aggregation of crystallins. β-Crystallins are present in a high proportion in the eye lens. Therefore, the structural study of the dimerization properties of crystallins can shed light on the first stages of protein aggregation. In the present work, we examine the protein-protein association profiles of the human βB2-crystallin by employing extensive coarse-grained molecular dynamics (CG-MD) and the Markov state analysis. Interestingly, our re

Function and Aggregation in Structural Eye Lens Crystallins.

Crystallins are transparent, refractive proteins that contribute to the focusing power of the vertebrate eye lens. These proteins are extremely soluble and resist aggregation for decades, even under crowded conditions. Crystallins have evolved to avoid strong interprotein interactions and have unusual hydration properties. Crystallin aggregation resulting from mutation, damage, or aging can lead to cataract, a disease state characterized by opacity of the lens.Different aggregation mechanisms can occur, following multiple pathways and leading to aggregates with varied morphologies. Interactions of chlorpromazine with alpha-, beta- and gamma-crystallins.

The binding parameters (binding affinity constant, K and number of binding sites, p) has been determined spectrofluorometrically for chlorpromazine (CPZ) binding to the lens proteins--alphaL-crystallin, betaL-crystallin and gamma-crystallin. The binding affinity constants for CPZ binding to alphaL- and gamma-crystallins are higher than the binding affinity constants for 3betaL-crystallin, although the number of CPZ binding sites for betaL-crystallin is comparatively higher than the number for the other two lens proteins. CPZ causes local conformational changes around the tryptophan moieties of

S-methylated cysteines in human lens gamma S-crystallins.

The proteins of the eye lens, which do not turn over throughout life, undergo many modifications, some of which lead to senile cataract. We describe a modification, S-methylation of cysteine, that may serve to protect the lens from detrimental modifications. The modification was detected as a +14 Da peak in electrospray ionization mass spectra of human lens gammaS-crystallins. Derivatization of gammaS-crystallin with iodoacetamide showed reaction at only six of the seven cysteines, indicating the modification blocked reaction at one cysteine. Further analysis of the modified gammaS-crystallin

Biochemical comparison of lens crystallins from three reptilian species.

Lens crystallins were isolated from the homogenates of reptilian eye lenses derived from three different species by gel-permeation chromatography and characterized by gel electrophoresis, amino-acid analysis, N-terminal sequence analysis and circular dichroism. Four fractions corresponding to alpha-, delta/epsilon/beta-, beta- and gamma-crystallins were obtained for the crystallins from caiman lenses, whereas delta- and gamma-crystallin fraction were present in lesser amounts or missing in the turtle and snake lenses, respectively. The native molecular masses for these purified fractions and t

The possible role of alpha-crystallins in human senile cataractogenesis.

alpha-Crystallins possess molecular chaperone properties and are one of the most abundant of the lenticular proteins. Posttranslational modifications of these proteins have been implicated as a possible etiology of human cataracts. This article will review current knowledge concerning the effects of known posttranslational modifications upon the molecular chaperone properties and aggregation behavior of alpha-A and alpha-B crystallin. Based upon these effects, experimental approaches will be discussed that may be useful in the development of reagents that may selectively inhibit the cataractog

Study on the crystallinity of PEG on the crystalline size of flavonoids in a crystalline dispersion system.

Poor water solubility and low bioavailability of flavonoids present significant barriers to their development and application. To address these challenges, this study explores the use of crystalline solid dispersions (CSDs) to reduce drug crystalline size and enhance in vivo bioavailability. The CSDs were prepared using a spray-drying technique with chrysin (CHY) and quercetin (QUR) as model drugs and various molecular weights of polyethylene glycol (PEG) as carriers. The authors systematically investigated the factors influencing the interaction between flavonoids and PEG in CSDs. These facto

Novel roles for α-crystallins in retinal function and disease.

α-Crystallins are key members of the superfamily of small heat shock proteins that have been studied in detail in the ocular lens. Recently, novel functions for α-crystallins have been identified in the retina and in the retinal pigmented epithelium (RPE). αB-Crystallin has been localized to multiple compartments and organelles including mitochondria, golgi apparatus, endoplasmic reticulum and nucleus. α-Crystallins are regulated by oxidative and endoplasmic reticulum stress, and inhibit apoptosis-induced cell death. α-Crystallins interact with a large number of proteins that include other cry

Light-induced phosphorylation of crystallins in the retinal pigment epithelium.

Protein phosphorylations have essential regulatory roles in visual signaling. Previously, we found that phosphorylation of several proteins in the retina and retinal pigment epithelium (RPE) is involved in anti-apoptotic signaling under oxidative stress conditions, including light exposure. In this study, we used a phosphoprotein enrichment strategy to evaluate the light-induced phosphoproteome of primary bovine RPE cells. Phosphoprotein-enriched extracts from bovine RPE cells exposed to light or dark conditions for 1h were separated by 2D SDS-PAGE. Serine and tyrosine phosphorylations were vi

In vitro glycation and acetylation (by aspirin) of rat crystallins.

In vitro studies with rat lens crystallins were conducted to explore the mechanism by which aspirin (ASA-acetylsalicylic acid) could inhibit cataractogenesis. The purpose of the present study is to show whether gamma-crystallin is the primary target for glycation by glucose and acetylation by ASA. Lens soluble fractions from one and seven month old Sprague-Dawley rats were incubated with 5 mM [14C]glucose with and without 10 mM ASA. alpha, beta, and gamma-crystallins were separated by molecular sieve HPLC and specific activities of each crystallin determined. In vitro acetylation was also stud

Glycation of crystallins in lenses from aging and diabetic individuals.

Water-soluble crystallins were obtained from clear human lenses of different age (4-81-year-olds) and lenses of individuals showing senile or diabetic cataracts. Levels of early glycation products were high in the high molecular weight material (HM) and the alpha-crystallin fractions, compared with beta- and gamma-crystallins. This difference becomes more prominent upon aging. The content of total early glycation products in HM and alpha-crystallin increases clearly with age, whereas levels remain relatively constant in the beta- and gamma-crystallins. There is an elevation of early products i

[Antibodies against alpha, beta, and gamma crystallins in human serum].

Serum antibodies against alpha, beta, and gamma crystallins in 79 normal subjects and 38 patients with senile cataract were determined by ELISA, with the result that the presence of the antibodies was related to age, not to sex, and the rate of presence in patients with senile cataract did not differ from that of controls. The relationship between the presence of the antibodies and senile cataract was discussed.
