Characteristics, Antioxidant Activity Stability, and Anti-Fatigue Activity of Hydrolysates from Cucumaria frondosa Tentacles.

This study aimed to assess the impact of alcalase, trypsin, flavourzyme, and neutrase on the characteristics, antioxidant activity stability, and anti-fatigue activity of hydrolysates derived from Cucumaria frondosa tentacles (CFTHs). The results demonstrate that favourzyme hydrolysates exhibited the highest degree of hydrolysis (DH). Zeta potential and particle size measurements indicated that hydrolyzed peptides treated with favourzyme appeared aggregated and exhibited larger particle sizes. The antioxidant properties of CFTHs demonstrated good thermal stability, pH stability, and enhanced s

Antioxidant and ACE-Inhibitory Activity of Protein Hydrolysates Produced from Atlantic Sea Cucumber (Cucumaria frondosa).

Atlantic sea cucumber is a benthic marine echinoderm found in Northwest Atlantic waters and is harvested mainly for its body wall. The body wall, along with internal organs and aquaphyrangeal bulb/flower, is a rich source of proteins, where the latter parts are often considered as processing discards. The objective of this research was to produce protein hydrolysates from sea cucumber tissues (body wall, flower, and internal organs) with bioactive properties associated with antioxidants, DNA and LDL cholesterol oxidation inhibition, and angiotensin-I-converting enzyme (ACE) inhibitory effects.

Structure and Anti-Inflammatory Activity of a New Unusual Fucosylated Chondroitin Sulfate from Cucumaria djakonovi.

Fucosylated chondroitin sulfate CD was isolated from the sea cucumber Cucumaria djakonovi collected from the Avachinsky Gulf of the eastern coast of Kamchatka. Structural characterization of CD was performed using a series of non-destructive NMR spectroscopic procedures. The polysaccharide was shown to contain a chondroitin core [→3)-β-d-GalNAc-(1→4)-β-d-GlcA-(1→]n where about 60% of GlcA residues were 3-O-fucosylated, while another part of GlcA units did not contain any substituents. The presence of unsubstituted both at O-2 and O-3 glucuronic acid residues in a structure of holothurian chond

Intermediate molecular weight-fucosylated chondroitin sulfate from sea cucumber Cucumaria frondosa is a promising anticoagulant targeting intrinsic factor IXa.

Thromboembolic diseases pose a serious risk to human health worldwide. Fucosylated chondroitin sulfate (FCS) is reported to have good anticoagulant activity with a low bleeding risk. Molecular weight plays a significant role in the anticoagulant activity of FCS, and FCS smaller than octasaccharide in size has no anticoagulant activity. Therefore, identifying the best candidate for developing novel anticoagulant FCS drugs is crucial. Herein, native FCS was isolated from sea cucumber Cucumaria frondosa (FCScf) and depolymerized into a series of lower molecular weights (FCScfs). A comprehensive a

Sequential Extraction of Bioactive Saponins from Cucumaria frondosa Viscera: Supercritical CO2-Ethanol Synergy for Enhanced Yields and Antioxidant Performance.

This study investigates the sequential extraction of lipids and saponins from C. frondosa viscera. Lipids were extracted using supercritical carbon dioxide (scCO2) in the presence of ethanol (EtOH) as a co-solvent. Subsequently, the lipid-extracted viscera underwent three saponin extraction approaches, scCO2-scCO2, scCO2-EtOH, and scCO2-hot water, resulting in saponin-rich extracts. Process parameter investigation for saponin extraction from scCO2-defatted viscera revealed minimal effects of temperature, pressure, extraction time, static extraction, and EtOH concentration on saponin yields, al

Antithrombotic activities of fucosylated chondroitin sulfates and their depolymerized fragments from two sea cucumbers.

Fucosylated chondroitin sulfate (FCS), a glycosaminoglycan extracted from the body wall of sea cucumber, is a promising antithrombotic agent. The chemical structures of FCSc isolated from sea cucumber Cucumaria frondosa and its depolymerized fragment (dFCSc) were characterized for the first time. Additionally, anticoagulant and antithrombotic activities were evaluated in vitro and in vivo. The results demonstrated that dFCSc exhibited better antithrombotic-hemorrhagic ratio than native FCSc on the electrical induced arterial thrombosis model in rats. Compared to FCSt obtained from Thelenota an

Dynamic mechanical characterization of a mutable collagenous tissue: response of sea cucumber dermis to cell lysis and dermal extracts.

The dermis of the holothurian Cucumaria frondosa is a mutable collagenous tissue (MCT). In this study, the inner and outer regions of the dermis were separated and used to make two different tissue extracts. These extracts were applied to intact pieces of dermis, one invoking a stiff mechanical state and the other invoking a compliant state. The extracts were effective on tissues incubated in artificial sea water (ASW) and in those incubated in Ca(2+)-chelated ASW. Furthermore, the extracts were effective on both fresh tissues and tissues in which the cells had been lysed by freeze-thawing, in

Saponins of North Atlantic Sea Cucumber: Chemistry, Health Benefits, and Future Prospectives.

Frondosides are the major saponins (triterpene glycosides) of the North Atlantic sea cucumber (Cucumaria frondosa). Frondosides possess amphiphilic characteristics due to the presence of various hydrophilic sugar moieties and hydrophobic genin (sapogenin). Saponins are abundant in holothurians, including in sea cucumbers that are widely distributed across the northern part of the Atlantic Ocean. Over 300 triterpene glycosides have been isolated, identified, and categorized from many species of sea cucumbers. Furthermore, specific saponins from sea cucumbers are broadly classified on the basis

Eicosapentaenoic acid-enriched phospholipids suppressed lipid accumulation by specific inhibition of lipid droplet-associated protein FSP27 in mice.

BACKGROUND: Sea cucumber is a rich source of eicosapentaenoic acid in the form of eicosapentaenoic acid-enriched phospholipids (EPA-PL). It is known to be efficacious in preventing obesity. However, few studies have focused on the role of EPA-PL in inhibiting lipid accumulation by lipid droplets (LDs). This study first investigated the effect of EPA-PL from sea cucumber on the formation of LDs and the underlying mechanism in C57BL/6J mice. The mice were randomly divided into two groups and treated for 8 weeks or 3, 7, and 14 days with either (i) a high-sucrose diet (model group), (ii) a high-s

Tensilin-like stiffening protein from Holothuria leucospilota does not induce the stiffest state of catch connective tissue.

The dermis of sea cucumbers is a catch connective tissue or mutable connective tissue that exhibits large changes in mechanical properties. A stiffening protein, tensilin, has been isolated from the sea cucumber Cucumaria frondosa. We purified a similar protein, H-tensilin, from Holothuria leucospilota, which belongs to a different family to C. frondosa. H-tensilin appeared as a single band with an apparent molecular mass of 34 kDa on SDS-PAGE. No sugar chain was detected. Tryptic fragments of the protein had homology to known tensilin. H-tensilin aggregated isolated collagen fibrils in vitro
