Inclusion Complexes of Colchicine with Acyclic Cucurbit[n]urils to Improve Its Photostability and Retain Anticancer Activity.

Colchicine is known for its significant anti-inflammatory, antigout, and anticancer activities, and recent research has increasingly focused on its potential cardiovascular protective effects. However, its poor photostability restricts its broader applications. To address this limitation, supramolecular host-guest encapsulation has emerged as an effective strategy for enhancing the stability of guest molecules. In this study, two types of acyclic cucurbit[n]urils (ACBs, referred to as M1 and M2) were synthesized as host molecules, while colchicine was utilized as the guest molecule to form inc

Molecular encapsulation of hydroxychloroquine by cucurbit[n]urils: A combined molecular dynamics simulation and quantum chemical study.

The inclusion complexation of hydroxychloroquine (HCQ), a biologically active pharmaceutical compound, with three cucurbit[n]urils, CB[6], CB[7], and CB[8], was investigated using a combination of molecular dynamics simulations and quantum chemical calculations. The results indicate that HCQ can be accommodated within the cavity of CB[6], preferentially inserting its aliphatic side chain into the host interior. For CB[7], two distinct binding modes were observed, in which either the aliphatic chain or the aromatic quinoline ring enters the cavity, reflecting greater conformational flexibility

Acyclic Cucurbit[n]uril-type Receptors: Preparation, Molecular Recognition Properties and Biological Applications.

This article traces the development of acyclic cucurbit[n]uril-type receptors with a focus on work from the Isaacs group. First, we describe the synthesis of methylene bridged glycoluril dimers capped with aromatic sidewalls which allowed us to probe the interconversion of the S- and C-shaped dimers which is a fundamental step in CB[n] formation. The C-shaped compounds were found to undergo discrete self-assembly (dimerization) in both water and organic solvents which lead us to investigate multicomponent self-sorting systems. We supressed the self-association of 8 by electrostatic repulsion i

Macrocycles and Acyclic Cucurbit[n]urils as Pseudo[2]catenane Partners for Long-Acting Neuromuscular Blocks and Rapid Reversal In Vivo.

Long-acting neuromuscular blocks followed by rapid reversal may provide prolonged surgeries with improved conditions by omitting repetitive or continuous administration of the neuromuscular blocking agent (NMBA), eliminating residual neuromuscular block and minimizing postoperative recovery, which, however, is not clinically available. Here, we demonstrate that imidazolium-based macrocycles (IMCs) and acyclic cucurbit[n]urils (ACBs) can form such partners by functioning as long-acting NMBAs and rapid reversal agents through a pseudo[2]catenation mechanism based on stable complexation with Ka v

Inhibited Hydrolysis of 4-(N,N-Dimethylamino)aryl Imines in a Weak Acid by Supramolecular Encapsulation inside Cucurbit[7]uril.

Schiff bases are imines formed by condensing primary amines with aldehydes (aldimines) or ketones (ketimines), and they are common functional groups in nature. This current study prepared 10 different aldimine and ketimine derivatives, based on the 4-(N,N-dimethylamino)aryl imine scaffold, and measured hydrolysis rate constants at pH 6.0, where the mole fraction of protonated imine was very high. The hydrolysis rate constants for the free protonated imines exhibited a logarithmic correlation with the calculated electrophilicity index (ω). Imine hydrolysis was reduced by 15-97-fold by the prese

Acyclic Cucurbit[n]uril-Type Containers as Receptors for Neuromuscular Blocking Agents: Structure-Binding Affinity Relationships.

Acyclic cucurbit[n]uril molecular containers 1 and 2C3 have previously been shown to strongly bind to the neuromuscular blocking agents rocuronium, vecuronium, pancuronium, and cisatracurium in vitro by optical methods and to reverse neuromuscular block in vivo in rats. In this paper we study the in vitro binding of a panel of acyclic CB[n]-type receptors toward the four neuromuscular blocking agents and acetylcholine to develop structure-binding affinity relationships. The selected variants include those with different aromatic sidewalls (e.g. 1Me4 with dimethyl o-xylylene walls; 3 with 1,8-l

Computational insights into fenbendazole encapsulation by Cucurbit[n]urils (n = 7, 8): Implications for drug delivery and nanocarrier design through integrated structural, electronic, thermodynamic, and spectroscopic investigations.

Fenbendazole (fenben), a benzimidazole anthelmintic with emerging anticancer potential, faces significant challenges in clinical applications due to its poor aqueous solubility and potential toxicity at high doses. To address these limitations, this study explores the encapsulation of fenben within cucurbit[7]uril (CB[7]) and cucurbit[8]uril (CB[8]) using computational methods to evaluate host-guest interactions, stability, and electronic properties. Density functional theory (DFT) calculations at the APFD/6-31+G(d) level revealed that CB[7] forms more stable complexes with fenben than CB[8],

Cucurbit[7]uril Inhibits Islet Amyloid Polypeptide Aggregation by Targeting N Terminus Hot Segments and Attenuates Cytotoxicity.

Two "hot segments" within an islet amyloid polypeptide are responsible for its self-assembly, which in turn is linked to the decline of β-cells in type 2 diabetes (T2D). A readily available water-soluble, macrocyclic host, cucurbit[7]uril (CB[7]), effectively inhibits islet amyloid polypeptide (IAPP) aggregation through ion-dipole and hydrophobic interactions with different residues of the monomeric peptide in its random-coil conformation. A HSQC NMR study shows that CB[7] likely modulates IAPP self-assembly by interacting with and masking major residues present in the "hot segments" at the N

Recent Advances in the Construction and Application of Anion-Induced Cucurbit[n]uril-Based Supramolecular Assemblies via Outer-Surface Interactions.

Cucurbit[n]urils (abbreviated as Q[n]s or CB[n]s) are synthesized through the condensation of formaldehyde and glycoluril in a hydrochloric acid environment. These molecules exhibit three distinctive properties concerning their charge distribution: a neutral internal cavity, a positively charged outer surface, and a negatively charged portal carbonyl oxygen atoms. These unique characteristics facilitate the formation of supramolecular frameworks with a range of anions and metal polyanions. The primary focus of this review is on the creation of anion-induced Q[n]-based supramolecular self-assem

Structure-Activity Relationship Studies Leading to the Discovery of Highly Water-Soluble and Biocompatible Acyclic Cucurbit[n]uril FY-3451 as a Universal Antagonist That Rapidly Reverses Neuromuscular Blocking Agents In Vivo.

The development of a reversal agent that can rapidly reverse clinically used nondepolarizing neuromuscular blocking agents (NMBAs) has long been a challenge. Here, we report the synthesis of a series of highly water-soluble acyclic cucurbit[n]urils (acCBs). Systematic structure-activity relationship studies reveal that introducing two propylidene units on the peripheral benzene rings not only remarkably improves the activity of the corresponding derivative acCB6 (FY 3451) in reversing the neuromuscular block of rocuronium, cisatracurium, vecuronium, and pancuronium, the four clinically used NM

Arene-Cucurbiturils: Benzene-Containing Cucurbituril[2,4] and Benzene-Containing Cucurbituril[3,6].

Benzene-containing cucurbituril[2,4] and benzene-containing cucurbituril[3,6], the structures of cucurbit[n]urils with some glycoluril units replaced by benzene rings, are constructed through condensation of specific benzene-containing motifs. These novel macrocycles inherit cucurbit[n]urils' concave cavity and provide chemical modification potentiality and chromophores. Benzene-containing cucurbituril[2,4] shows a C2-symmetric macrocycle, while benzene-containing cucurbituril[3,6] exhibits a distinctive two-cavity structure (one big and one small) that resembles a gourd and could potentially

Organic supramolecular assemblage-confined photoluminescence.

Organic supramolecular hosts can selectively bond to small functional molecules with chromophores, inhibiting molecular motion and isolating quenching factors through spatial confinement, thereby enhancing fluorescence and phosphorescence emission and expanding their applications in chemistry, biology, and materials science. This review mainly focuses on supramolecular luminescent systems constructed from cucurbit[n]urils, cyclodextrins, and other macromolecules. It is very important that cucurbit[n]urils and cyclodextrins both possess rigid hydrophobic cavities, in which cucurbit[n]urils can

Cucurbit[7]uril: an emerging candidate for pharmaceutical excipients.

Cucurbit[7]uril (CB[7]), belonging to the cucurbit[n]uril family (CB[n], n = 5-8, 10, or 13-15), may form host-guest complexes with a variety of small molecules of biomedical interest. The physical and chemical properties of the complexed drugs are often improved as a result of this complexation, suggesting the potential application of CB[7] as a pharmaceutical excipient. This review has summarized the most recent research progress reported between 2011 and early 2017 regarding the biocompatibility of CB[7] and the influence of CB[7] on the stability, solubility, biouptake, and biological acti

A supramolecular cucurbit[8]uril-based rotaxane chemosensor for the optical tryptophan detection in human serum and urine.

Sensing small biomolecules in biofluids remains challenging for many optical chemosensors based on supramolecular host-guest interactions due to adverse interplays with salts, proteins, and other biofluid components. Instead of following the established strategy of developing alternative synthetic binders with improved affinities and selectivity, we report a molecular engineering approach that addresses this biofluid challenge. Here we introduce a cucurbit[8]uril-based rotaxane chemosensor feasible for sensing the health-relevant biomarker tryptophan at physiologically relevant concentrations,

Differential Modulation of the Aggregation of N-Terminal Truncated Aβ using Cucurbiturils.

Modulating the aggregation of Aβ has long been considered to be one of the potential methods to treat Alzheimer's disease (AD). It has been found that different Aβ species, including N-terminal truncated or/and modified Aβ, co-exist in the brain of AD patients. Yet, there is currently little detailed work about the specific modulation of these Aβ species which hinders us to understand their roles in patients' brain. Using thioflavin T (ThT) kinetics and transmission electron microscope, here we showed that cucurbit[7]uril and cucurbit[8]uril could inhibit the aggregation of both Aβ4-40 and Aβ1
