Human embryo culture media comparisons.

Every program of assisted reproduction strives to maximize pregnancy outcomes from in vitro fertilization and selecting an embryo culture medium, or medium pair, consistent with high success rates is key to this process. The common approach is to replace an existing medium with a new one of interest in the overall culture system and then perform enough cycles of IVF to see if a difference is noted both in laboratory measures of embryo quality and in pregnancy. This approach may allow a laboratory to select one medium over another but the outcomes are only relevant to that program, given that t

Serum-free media for lymphoid culture.

A serum-free medium is described that we have been using since almost ten years when growing a number of lymphoid cell lines. Serum-free media offer several advantages before media containing even well standardized animal sera. As an example of studies where serum-free media offer advantages we present data demonstrating the use of a defined medium when monitoring glycosidase enzyme levels in lymphoid cells.

Production of culture filtrates of Sporothrix schenckii in diverse culture media.

Mycelial-form Sporothrix schenckii was studied to determine if growth in complex (Sabouraud-dextrose) or defined culture media (minimal medium, Gibco Medium 199 with and without Hepes buffer) with differing initial pH levels would affect expression of antigen components. Cultures were evaluated by continuous monitoring and serial sampling for various parameters. Great variation was seen in the protein and antigenic patterns induced by the different media. The expression of a 55 kDa component accompanied by significant acidification of the culture medium at the beginning of the stationary growt

The action of light on culture media.

Failure of growth of staphylococci on nutrient agar plates was shown to be due to previous exposure to sunlight in an exceptionally well lit laboratory. Such plates inhibited the growth of Staph. aureus, Staph. albus, some Proteus species and Providence, but not that of various other bacteria. The affected species are found to be more susceptible to the action of hydrogen eroxide added to the medium. The effect was produced in plates of five different nutrient agars, but not in two others which contained casein. Plates awaiting inoculation should be protected from light; a white fabric blind w

Focus on Bioburden Culture Media and Medical Devices.

Medical devices are a vital part of the global health care system that can have a far-reaching impact on patient treatment. Therefore, they must be sterile to ensure patient safety. The prevalent microorganism's type on a medical device, also known as "bioburden", is a useful indicator of a potential contamination source. Indeed, bioburden is a potential risk to the patient not only because the sterilization process might not be completely effective, but also post-processing because of the possible presence of residual materials. Although bioburden may be confidently killed by destructive ster

Improved Neisseria gonorrhoeae culture media without atmospheric CO2.

Bacterial culture on solid media is the crucial step in diagnosing Neisseria gonorrhoeae infections and is the gold standard for determining their antimicrobial resistance profile. However, culture of Neisseria spp. can be challenging in resource poor areas, relying on specialist incubators or other methods of supplying 5% CO2 for growth of the bacteria. Even when such incubators are available, the CO2 to run them may be scarce; there were CO2 shortages during the COVID-19 pandemic, for example. Although culture jars with gas packs or candles can be used, these are inefficient in terms of use

[Pediatric bacteremia strains grow in blood culture media].

Bacteremia strains may influence the clinical therapy, the isolation of bacteremia strains is important for patients. In order to understand the distribution and the growth in the blood culture media of bacteremia strains, from January 1982 through December 1988 we studied the blood culture sent to us from the pediatric ward. These were three hundred and forty one bacterial strains (3.1%) isolated from patients. In the meantime we also studied the bacteremia strains growth in the blood culture media. In these positive cultures, there were 83 gram-positive, and 258 gram-negative strains. The 83

Growth of Trichomonas vaginalis in commercial culture media.

There are only two commercially available, ready-to-use culture media which are approved by the Food and Drug Administration for clinical diagnosis of vaginal trichomoniasis: Kupferberg's STS and Diamond's medium (modified). Diamond's medium (Klaas modification), recommended by the Centers for Disease Control for the isolation of Trichomonas vaginalis, was compared in vitro to Kupferberg's (STS) medium. Growth studies using six fresh clinical isolates, all from different patients, showed that while generation time was about 6 h in both STS and Diamond's, the period of exponential growth was lo

[Culture media for antibiotic sensitivity testing (author's transl)].

The medium is undoubtedly one of the most important factors influencing the sensitivity testing techniques. The possible role of pH and different components of media, as such as the concentration of peptone, tymidine or tymine, riboflavin, cations (MG++, Ca++, Fe++), mineral salts, agar and phosphate (added eventually with buffer solutions) is discussed. The conclusions point to the need of synthetic media which are suitable for growing all common pathogens and to not interfere with the antibacterial activity of drugs.

Vitamins in cell culture media: Stability and stabilization strategies.

Nowadays, chemically defined cell culture media (CCM) have replaced serum- and hydrolysate-based media that rely on complex ingredients, such as yeast extracts or peptones. Benefits include a significantly lower lot-to-lot variability, more efficient manufacturing by reduction to essential components, and the ability to exclude components that may negatively influence growth, viability, or productivity. Even though current chemically defined CCMs provide an excellent basis for various mammalian biotechnological processes, vitamin instabilities are known to be a key factor contributing to the v

Laboratory diagnosis of Haemophilus ducreyi: sensitivity of culture media.

Laboratory confirmation of the clinical diagnosis of chancroid requires the isolation of Haemophilus ducreyi. Enriched gonococcal (GcHbS) and Mueller-Hinton agar (MHHb) both support the growth of most strains of H. ducreyi. In this study we compared the isolation rate of H. ducreyi on GcHbs and MHHb media in 111 men with genital ulcer disease. A second culture was obtained in 84 men at 48 hr in order to determine the reproducibility of H. ducreyi culture. The sensitivity of a single and a sequential pair of cultures on GcHbS was 67% and 74%, respectively, on men with presumed chancroid. The se

Draining ears in acute otitis media: reliability of culture.

The purulent exudates in the ear canal of 69 children who presented with acute otorrhea were cultured directly onto standard media. No effort was made to either suction out the exudate lying in the canal or to use antiseptic agents on the canal wall. Potential pathogens such as H. influenzae, S. pneumoniae and S. pyogenous were recovered from two-thirds of the patients who suffered spontaneous otorrhea. Reliable cultures can be obtained from such patients provided the cultures are plated expeditiously on solid agar.

BIPHASIC PARTICULATE MEDIA FOR THE CULTURE OF FILTER-FEEDERS.

1. Over 200 parthenogenetic generations of the freshwater Cladocera Moina macrocopa were obtained aseptically in three artificial media. 2. The media have two phases: a liquid phase supplying mineral salts, water soluble vitamins, nucleic acids, and a liver extract and a fine particulate phase made from coagulated proteins, starch and lipid factors. 3. The particulate phase supplies the bulk nutrients very efficiently; hence, this type of media may be useful for growing other filter-feeders.

Culture media influences Candida parapsilosis growth, susceptibility, and virulence.

INTRODUCTION: Candida parapsilosis, a pathogenic yeast associated with systemic infections, exhibits metabolic adaptability in response to nutrient availability. METHODS: We investigated the impact of RPMI glucose supplemented (RPMId), TSB, BHI and YPD media on C. parapsilosis growth, morphology, susceptibility (caspofungin and amphotericin B), and in vivo virulence (Galleria mellonella) in planktonic and biofilm states. RESULTS: High-glucose media favors growth but hinders metabolic activity and filamentation. Media promoting carbohydrate production reduces biofilm susceptibility. Virulence
