Serum-free media for lymphoid culture.

A serum-free medium is described that we have been using since almost ten years when growing a number of lymphoid cell lines. Serum-free media offer several advantages before media containing even well standardized animal sera. As an example of studies where serum-free media offer advantages we present data demonstrating the use of a defined medium when monitoring glycosidase enzyme levels in lymphoid cells.

Two novel serum-free media for the culture of Trichomonas vaginalis.

Definitive diagnosis of Trichomonas vaginalis, one of the common causes of sexually transmitted diseases in Turkey, relies on the microscopic examination of both fresh preparations and culture material of genital secretions and urine. The aim of this study was to compare the diagnostic efficacies of two culture media, serum-free TB1 and TB2, including iron and vitamin B12, respectively, with the well-known medium, TYM. Growth rate reached peak levels 48 h in TYM and 72 h in both TB1 and TB2 after inoculation. The highest amount of viable trophozoites has been obtained from TB1, almost equal to

Comparison of culture media supplements identifies serum components in self-reported serum-free preparations.

BACKGROUND: Cell culture media are essential for cell expansion and many cells still depend on blood-derived supplements for optimal growth. From a regulatory perspective, these materials are ideally xeno-free, serum-free or even chemically defined. However, differences in composition and in performance are seldom clear from the terminology used in this field. METHODS: Therefore, we set out to investigate the presence of serum in seven serum-free media (SFM) and link that information to the performance and cost. We used fetal bovine serum and five human platelet lysate preparations (hPL) for c

Effects of serum-free culture media on human apical papilla cells properties.

OBJECTIVE: Aiming at more effective and safer cell therapies, the objective of this study was to evaluate the biological properties of human apical papilla cells cultured in the absence of serum supplementation in comparison to cells cultured with fetal bovine serum (FBS). DESIGN: Two apical papilla cell populations were isolated from third molars with incomplete rhizogenesis, and cultured in four different media: minimum essential Eagle medium - alpha modification (alpha-MEM); alpha-MEM supplemented with FBS (alpha-MEM + FBS); Dulbecco's modified Eagle medium/nutrient mixture F-12 (DMEM/F12)

Low-cost food-grade alternatives for serum albumins in FBS-free cell culture media.

Cultivated meat may be an ethical, environmentally friendly, antibiotic-free meat alternative of the future. As of now, one of the main limiting factors for bringing cultivated meat to the market is the high cost of the cell culture media and their great dependency on serum albumins, production of which is predicted to become a major bottleneck of this industry. Here, using bovine muscle stem cells, we optimized serum free B8/B9 medium. We identified several food grade, low-price medium stabilizers, exhibiting comparable or even superior stabilization of the B8 medium in short- and long-term c

Serum-free, xeno-free culture media maintain the proliferation rate and multipotentiality of adipose stem cells in vitro.

BACKGROUND AIMS: Human adipose stem cells (ASC) are an abundant, readily available population of multipotent progenitor cells that reside in adipose tissue. ASC have been shown to have therapeutic applicability in pre-clinical studies, but a standardized expansion method for clinical cell therapy has yet to be established. Isolated ASC are typically expanded in medium containing fetal bovine serum (FBS); however, sera and other culturing reagents of animal origin in clinical therapy pose numerous safety issues, including possible infections and severe immune reactions. METHODS: To identify op

Culture of transgenic Drosophila melanogaster Schneider 2 cells in serum-free media based on TC100 basal medium.

Requirements of eliminating animal proteins from cell culture have intensified in recent years, with the pressure of regulatory agencies related to biopharmaceuticals production. In this work, the substitution of fetal bovine serum by yeastolate and a soy hydrolysate (Hy Soy) for the culture of Drosophila melanogaster Schneider 2 cells transfected for the production of rabies virus G glycoprotein was evaluated. TC100 supplemented with glucose, glutamine, lipid emulsion and Pluronic F68 was employed as basal medium. Results show that yeastolate was more efficient on cell growth stimulation than

New perspectives on serum-free cell culture: Harnessing microbial resources for innovative media development in food biomanufacturing.

Cell culture is a fundamental technique in food biomanufacturing. However, the inherent limitations of traditional serum-containing culture systems hinder the scalable application of cell culture in the food industry, thus driving the development of serum-free cell culture. This review summarizes the current status and technical challenges of serum-free media (SFM) for cells relevant to food biomanufacturing. In this context, we systematically compile recent advances in the use of microbial resources as potential supplements for SFM, given that their functional ingredients are capable of meeti

A comparison of drug-induced cardiotoxicity in rat embryos cultured in human serum or protein free media.

INTRODUCTION: Although much reproductive toxicology research is performed in live animals there is increasing use of in vitro techniques primarily to identify potential hazards with human exposure. As many in vitro studies are undertaken using protein free media, the standard protocol is to compare the effect concentration determined in vitro with the predicted therapeutic free plasma concentration in humans. The aim of the present study was to test this rationale by comparing the effect of a small number of therapeutic drugs on heart rate of rodent embryos cultured in human sera or protein fr

Synthesis and composition of glycosaminoglycans by explant cultures of human ciliary body and ciliary processes in serum-containing and serum-free defined media.

Freshly isolated ciliary body explants and tips of ciliary processes derived from human eyes were cultured in serum-containing or serum-free defined medium. These cultures synthesized tissue-bound and medium-released ("excreted") glycosaminoglycans (GAGs), as evidenced by the incorporation of 14C-glucosamine and enzymatic characterization of these labelled GAGs (hyaluronic acid, heparan sulphate, chondroitin sulphate, dermatan sulphate). The GAG synthesis and excretion rate was enhanced by serum. It is suggested that ciliary epithelium performs this function of GAG synthesis also under in vivo

Cell culture of human gingival fibroblasts, oral cancer cells and mesothelioma cells with serum-free media, STK1 and STK2.

The majority of cells are cultured with Dulbecco's modified Eagle's medium (DMEM) or RPMI supplemented with fetal bovine serum (FBS), which contains numerous factors, including cytokines, nutrients and unknown growth factors. These factors may affect cell growth, apoptosis and differentiation. The serum-free medium, STK2, has been previously reported as suitable for the cell culture of human mesenchymal stem cells. However, how STK1 or STK2 affect the cell proliferation of normal and cancer cells remains unknown. The present study examined the growth of the human gingival fibroblast (HGF-1) ce

Differential expression of voltage-gated Ca2+ conductances in human neuroblastoma NB69 cells cultured in defined serum-free and astrocyte-conditioned media.

Voltage-gated Ca2+ conductances were investigated with the whole-cell patch-clamp technique-either using Ca2+ or Ba2+ as charge carriers-in NB69 human neuroblastoma cells plated in "defined" serum-free (DM) and in "astroglial-conditioned" media (CM). Cells expressed the microtubule associated protein 1A when plated in both media, indicating neuronlike differentiation. Cells of similar sizes and shapes were selected for recordings. Different sets of voltage-gated Ca2+ current types were usually expressed in DM- and CM-plated cells. DM-plated cells exhibited a high-voltage-activated current (HVA

Serum-Free Media Formulation Using Marine Microalgae Extracts and Growth Factor Cocktails for Madin-Darby Canine Kidney and Vero Cell Cultures.

The development of serum-free media (SFM) is critical to advance cell culture techniques used in viral vaccine production and address the ethical concerns and contamination risks associated with fetal bovine serum (FBS). This study evaluated the effects of marine microalgal extracts and growth factor cocktails on the activity of Madin-Darby canine kidney (MDCK) and Vero cells. Five marine microalgal species were used: Spirulina platensis (SP), Dunaliella salina (DS), Haematococcus pluvialis (HP), Nannochloropsis salina (NS), and Tetraselmis sp. (TS). DS and SP extracts significantly increased

In vitro culture of human acute lymphoblastic leukemia (ALL) cells in serum-free media; a comparison of native ALL blasts, ALL cell lines and virus-transformed B cell lines.

The aim of this study was to standardize in vitro culture conditions for human acute lymphoblastic leukemia (ALL) cells. The cells were cultured in medium containing 10% fetal calf serum (FCS) and in the four serum-free media X-vivo 10, X-vivo 15, X-vivo 20 and Stem Span. Native ALL blasts could proliferate in all four serum-free media, but the strongest responses were usually observed with Stem Span. Native leukemia blasts were also cultured in the presence of various single cytokines or cytokine combinations. The highest proliferation was usually observed in the presence of Flt3-Ligand (Flt3

A human-serum-free medium can induce more infectious P. falciparum gametocytes than a conventional human-serum-containing medium.

Malaria remains a global health problem, and the standard membrane feeding assay (SMFA) is a key functional assay for development of new interventions to stop malaria transmission from human to mosquito. For SMFA, media with ~ 10% of human serum has been used for infectious gametocyte cultures, however, there are multiple challenges to obtain a suitable human serum. Here we show a human-serum-free culture medium (HSF), which was a mixture of two stem cell culture media and AlbuMAX, supported infectious gametocyte growth. Moreover, the HSF-induced gametocytes elicited significantly higher numbe

Identification of compounds for improved growth of Leptospira in culture and isolation.

Leptospirosis is a recurring global disease of severe illness involving pulmonary and renal involvement in cattle and humans that needs attention to cure. The major challenge in treating leptospirosis disease is the diagnosis of the disease. The culturing of an organism is a gold standard for confirmation of the disease. The growth and optimistic identification of an organism require at least 8 to 14 days because of its slow growth characteristics. We have investigated various media conditions that are prepared based on the wealth of information obtained from 'omic' studies and report a sustai
