Development of a cell culture-based method for detecting infectious tick-borne encephalitis virus (TBEV) in milk products.

Tick-borne encephalitis outbreaks have been reported in Europe after consumption of raw milk products from infected animals. While molecular methods are commonly used in viral foodborne outbreak investigations due to their sensitivity, specificity and rapidity, there are very few methods to detect infectious tick-borne encephalitis virus (TBEV) in milk products for routine use/analyses. To address this gap, we developed a cell culture-based method to detect infectious TBEV in artificially contaminated raw goat milk and raw goat cheese, and evaluated the sensitivity of TBEV infectivity assays.

Potential roles of nitrate and live yeast culture in suppressing methane emission and influencing ruminal fermentation, digestibility, and milk production in lactating Jersey cows.

Concern over the carbon footprint of the dairy industry has led to various dietary approaches to mitigate enteric CH4 production. One approach is feeding the electron acceptor NO3-, thus outcompeting methanogens for aqueous H2. We hypothesized that a live yeast culture (LYC; Saccharomyces cerevisiae from Yea-Sacc 1026, Alltech Inc., Nicholasville, KY) would stimulate the complete reduction of NO3- to NH3 by selenomonads, thus decreasing the quantity of CH4 emissions per unit of energy-corrected milk production while decreasing blood methemoglobin concentration resulting from the absorbed inter

Unravelling the effect of autochthonous scotta-derived vs. commercial starter cultures in pilot-scale hard sheep milk cheese production: A multi-omics and sensory combined approach.

Autochthonous whey-based (scotta-innesto) starters are increasingly recognized as a valuable resource to preserve microbial biodiversity and sensory identity in hard sheep milk cheeses produced under PDO-type specifications. In this study, two indigenous scotta-innesto cultures collected in Sardinia (Italy) in the 1960s were compared with a widely used commercial starter in pilot-scale hard sheep milk cheese manufacture produced according to Pecorino Romano PDO specifications. To this end, an integrated multidisciplinary approach was employed by combining starter metagenomics, culture-dependen

Nutritional and healthful aspects of cultured and culture-containing dairy foods.

Nutritional and therapeutic qualities of fermented dairy products are reviewed. Partial hydrolysis of milk constituents (proteins, fats, and lactose) in yogurt, cheese, and other cultured dary foods appears to contribute to their increased digestibility. Lactase and other constituent enzymes of various culturing organisms should contribute to assimilation of lactose by lactose intolerant individuals. Several lactic cultures synthesize certain B-vitamins in fermented dairy products. In contrast, directly acidified dairy products do not exhibit such enhancement in B-vitamins. The hypocholestremi

Exploring the metabolic dynamics, storage stability, and functional potential of Lacticaseibacillus rhamnosus Probio-M9-fermented milk.

Lacticaseibacillus rhamnosus Probio-M9 (Probio-M9), a probiotic strain sourced from healthy breast milk, is recognized for its resilience to gastric and bile acids, along with its potential health benefits for infants. Its unique origin may influence its metabolic properties and effectiveness, and it has garnered increasing interest within the scientific community in recent years. However, research on its efficacy as a starter culture for milk fermentation and the associated metabolic shifts remains limited. This study aimed to track the dynamic metabolomic changes of Probio-M9 during the ferm

The potential value of cultured dairy products for child nutrition.

The author suggests that under certain conditions cultured milk, rather than fluid milk, can be used for infant and child nutrition as well as for school milk programs. Some of the major problems with fluid milk, fresh or reconstituted, are discussed. A review of the literature indicates that the assumption of lactose intolerance among many populations is exaggerated. Inappropriate handling of pasteurized milk very often is responsible for a high bacterial count and organoleptic defects. Such quality defects are more pronounced in countries with a warm climate. The use of polluted water in the

Lactose malabsorption from yogurt, pasteurized yogurt, sweet acidophilus milk, and cultured milk in lactase-deficient individuals.

The use of fermented dairy foods is common in areas of the world where lactase deficiency is prevalent. Recently, we have shown that the digestion of lactose from yogurt is enhanced as compared to that from milk. This enhanced digestion is apparently due to inherent B-galactosidase in yogurt which is active in the gastrointestinal tract after consumption of the yogurt. Furthermore, yogurt is well tolerated by lactase-deficient subjects resulting in little or no gastrointestinal distress. Since other fermented and microbial-containing dairy foods are consumed worldwide and may also contain some

Foods for Special Dietary Needs: Non-dairy Plant-based Milk Substitutes and Fermented Dairy-type Products.

A growing number of consumers opt for plant-based milk substitutes for medical reasons or as a lifestyle choice. Medical reasons include lactose intolerance, with a worldwide prevalence of 75%, and cow's milk allergy. Also, in countries where mammal milk is scarce and expensive, plant milk substitutes serve as a more affordable option. However, many of these products have sensory characteristics objectionable to the mainstream western palate. Technologically, plant milk substitutes are suspensions of dissolved and disintegrated plant material in water, resembling cow's milk in appearance. They

Clostridium botulinum in cattle and dairy products.

The use of plastic-wrapped and nonacidified silage as cattle feed has led to an increasing number of botulism outbreaks due to Clostridium botulinum Groups I-III in dairy cattle. The involvement of Groups I and II organisms in cattle botulism has raised concern of human botulism risk associated with the consumption of dairy products. Multiplication of C. botulinum in silage and in the gastrointestinal tract of cattle with botulism has been reported, thus contamination of the farm environment and raw milk, and further transmission through the dairy chain, are possible. The standard milk pasteur

Rediscovering acidophilus milk, its quality characteristics, manufacturing methods, flavor chemistry and nutritional value.

Fermented dairy products represent an excellent source for a wide range of probiotics with several health benefits that add to the positive value of conventional milk. Acidophilus milk is a regular milk enriched with Lactobacillus acidophilus, one of the beneficial bacterial species found in the colon, that renders the product to exhibit an immunostimulant, anti-hypercholesterolemic, anti-inflammatory, or chemopreventive effect especially against colon cancer; in addition, for lactose intolerance treatment. However, most of bacterial species are very demanding and require specific nutritional

Molecular detection and culture of Mycobacterium avium subsp. paratuberculosis in pasteurized and raw retail milk in Kampala, Uganda.

BACKGROUND: The hypothesis that Mycobacterium avium subsp. paratuberculosis (MAP) is a zoonotic pathogen has gained traction with the repeated isolation of MAP from patients with Crohn's disease. As consumption of contaminated milk and dairy products represents a primary suspected route for human exposure to MAP, this study was designed to investigate the presence of MAP in retail milk sold in Kampala, Uganda's Capital and central hub for the nation's milk distribution. METHODS: A total of 772 milk samples were collected and analysed sequentially using two molecular assays targeting the IS900

Matrix-associated spatial structure contributes to compositional stability of a complex multispecies starter culture under microbial perturbation: Insights from kefir grains.

Complex multispecies starter cultures can enrich the metabolic and sensory diversity of fermented foods, but industrial use is limited by compositional drift, overgrowth of dominant taxa, and vulnerability to exogenous microbial contamination. Kefir grains (KG), a matrix-encapsulated complex starter culture with stable serial propagation, provide a model for how spatial structure contributes to starter stability. We compared matrix-associated KG with free kefir-fermented milk (FKFM), representing a grain-free liquid-phase kefir microbial community, under a large-scale microbial challenge. Duri
