Engineering Cupriavidus necator for L-threonine production.

L-threonine plays pivotal roles in the medicine, food, and feed industries. In this work, C. necator H16 was engineered for heterotrophic and autotrophic production of L-threonine. Blocking poly-hydroxybutyrate biosynthesis redirected the carbon flux toward L-threonine, resulting in a 6.8-fold increase in L-threonine titer relative to the wild type (from 5.56 mg/L to 38.03 mg/L). Deletion of genes involved in L-threonine catabolism further minimized L-threonine loss. Moreover, strategies including sufficient precursor supply, screening of key enzymes, relief of feedback inhibition, and efficie

Multiresistent Cupriavidus pauculus infection in an immunocompromised elderly patient.

Cupriavidus pauculus is a gram-negative bacillus aerobic bacteria widely distributed in nature that can cause, in rare cases, serious infections both in immunocompromised and immunocompetent patients. We describe a case of an elderly patient admitted in emergency room with septic shock and diagnosed with a urinary tract infection. During his hospital stay, his clinical and analytical conditions have deteriorated. Blood cultures were positive for C. pauculus only sensitive to minocycline. Despite every effort, due to multiple comorbidities and a nosocomial pneumonia, the patient ends up dying.

Isopropanol production with engineered Cupriavidus necator as bioproduction platform.

Alleviating our society's dependence on petroleum-based chemicals has been highly emphasized due to fossil fuel shortages and increasing greenhouse gas emissions. Isopropanol is a molecule of high potential to replace some petroleum-based chemicals, which can be produced through biological platforms from renewable waste carbon streams such as carbohydrates, fatty acids, or CO2. In this study, for the first time, the heterologous expression of engineered isopropanol pathways were evaluated in a Cupriavidus necator strain Re2133, which was incapable of producing poly-3-hydroxybutyrate [P(3HB)].

Cupriavidus necator as a model organism for CO2-based biotechnology.

Cupriavidus necator H16 is a chemolithoautotrophic model organism that can grow in a simple mineral medium supplemented with the gases H2, O2 and CO2. This unique metabolic capability makes it a promising organism for sustainable biotechnology applications such as the production of bioplastics, single cell proteins and bulk chemicals from renewable resources. However, gas fermentation, particularly with explosive gas mixtures like H2 and O2, poses considerable technical and safety challenges. Effective control of gas mixtures, pressurized environments, mass transfer from gas to liquid and gas

Unveiling the biotransformation mechanism of indole in a Cupriavidus sp. strain.

Indole, an important signaling molecule as well as a typical N-heterocyclic aromatic pollutant, is widespread in nature. However, the biotransformation mechanisms of indole are still poorly studied. Here, we sought to unlock the genetic determinants of indole biotransformation in strain Cupriavidus sp. SHE based on genomics, proteomics and functional studies. A total of 177 proteins were notably altered (118 up- and 59 downregulated) in cells grown in indole mineral salt medium when compared with that in sodium citrate medium. RT-qPCR and gene knockout assays demonstrated that an indole oxygen

Cupriavidus pauculus bacteremia in a child on extracorporeal membrane oxygenation.

We report a case of bacteremia secondary to Cupriavidus pauculus in a 15-month-old boy on extracorporeal membrane oxygenation (ECMO). The source of the organism was water in the thermoregulator reservoir. The child responded well to cefepime and ciprofloxacin, a delayed oxygenator change out and replacement of the thermoregulator reservoir with a unit that was cleaned and decontaminated with sodium hypochlorite. Isolation of Cupriavidus pauculus from a patient on ECMO support should raise suspicion of the reservoir as a source.

[A case of ventilator-associated pneumonia caused by Cupriavidus pauculus].

Cupriavidus pauculus (formerly CDC Group IVc-2) is a non-fermentative, motile, gram-negative bacillus, rarely associated with human infections. It has been isolated from water, water from ultrafiltration systems and bottled mineral water. To date, 19 cases of bacteremia, two cases of peritonitis and one case of tenosynovitis associated with C. pauculus have been reported in English literature. In this paper, we report the first case of C. pauculus ventilator-associated pneumonia (VAP) in Turkey. A 47 years-old female with breast cancer was performed total mastectomy six years ago and received

Engineering Mixotrophy in the Chemolithoautotrophic Cupriavidus necator through Hydrogenase Induction.

Mixotrophy offers a promising strategy for biosynthesis by simultaneously utilizing organic carbon and CO2; however, mixotrophic microorganisms are rarely isolated outside of photoautotrophic microalgae. In this study, the chemoautotroph Cupriavidus necator H16 was found to preferentially consume fructose when coexisting with CO2 and H2, switching to utilization of only CO2 and H2 after fructose depletion. Transcriptomic analysis revealed significant differences in genes involved in energy metabolism, electron generation, and the respiratory chain. The molecular mechanism underlying the inabil

Challenge of Cupriavidus gilardii infection in an immunocompromised child: a case report.

BACKGROUND: Cupriavidus gilardii is a multidrug-resistant (MDR) pathogen found in soil and water. Human infection is extremely rare, with only two pediatric cases reported to date, and its clinical features and the antimicrobial strategies remain unclear. The pathogen's MDR nature often poses a therapeutic challenge. This case provides valuable clinical evidence regarding the management of C. gilardii infection in immunocompromised pediatric patients. CASE DESCRIPTION: A 4-year-old boy with acute lymphoblastic leukemia developed severe immunosuppression after reinduction cyclophosphamide, cyt

A Cupriavidus Pauculus Infection in a Patient with a Deep Brain Stimulation Implant.

While deep brain stimulation (DBS) is now standard therapy for the treatment of Parkinson's disease, essential tremor, and dystonia, infections remain one of the most common perioperative complications. In this report, we describe a 58-year-old female with a history of medically refractory Parkinson's disease, who underwent magnetic resonance (MR)-guided bilateral subthalamic DBS. While the initial surgery and programming were successful, she returned in follow-up with signs of a generator pocket infection. She was taken to surgery for hardware explantation, and cultures revealed multispecies

Optimizing Cupriavidus necator H16 as a host for aerobic C1 conversion.

Biological systems capable of converting CO2 or CO2-derived, single-carbon (C1) compounds can be used to reduce or reverse carbon emissions while establishing a circular bioeconomy to provide sustainable sources of the fuels, foods, and materials humanity relies on. A robust bioeconomy will rely upon a variety of microorganisms capable of assimilating C1 compounds and converting them to valuable products at industrial scale. While anaerobic microbes are ideal hosts for production of short-chain acids and alcohols, microbes capable of aerobic respiration are well suited for biosynthesis of high

First case of bloodstream infection caused by Cupriavidus metallidurans in Japan.

We report the first case in Japan of a bloodstream infection caused by Cupriavidus metallidurans. The patient was a 77 years old man hospitalized for treatment of heart failure and diabetes. On day 83 of hospitalization, he developed a fever of 38.0°C, followed three days later by an elevation in C-reactive protein. His condition improved with the administration of cefmetazole, cefepime, and levofloxacin, and antibiotic therapy was discontinued on the 17th day after the onset of fever. Blood cultures collected at the onset of fever revealed growth in the aerobic bottle after 21 h, and Gram sta

Muscular abscess caused by Cupriavidus gilardii in a renal transplant recipient.

Cupriavidus gilardii is a rare cause of human infection. We report a muscular abscess on the right thigh caused by this organism in a renal transplant recipient, who had suffered a septic shock associated with an extensive cellulitis caused by Streptococcus pyogenes. The patient was successfully treated with surgical drainage and intravenous ciprofloxacin for 13 days. This is the first time that C. gilardii is isolated from a human abscess. C. gilardii should be considered as a cause of human infection, especially in immunocompromised patients. Infection caused by this organism may be underdia

Oxygen-dependent redox control enables growth-decoupled biotransformations in Cupriavidus necator.

Genetic instability and environmental heterogeneity present persistent challenges to the stable microbial production of reduced, value-added chemicals. This work shows that limiting oxygen availability redirects electrons from respiration toward targeted reductive pathways in Cupriavidus necator, steering growth-decoupled biotransformations - exemplified by the conversion of acetoin to 2,3-butanediol and of glycerol to 1,3-propanediol. In cell suspension assays under a hydrogen-containing atmosphere, anoxic conditions stabilised near-stoichiometric acetoin-to-2,3-butanediol conversion (93% mol

Isopropanol production from carbon dioxide in Cupriavidus necator in a pressurized bioreactor.

A bioreactor was designed to provide high gas mass transfer to reach cell and product titres in the g L-1 level from CO2 for realistic, laboratory scale, engineered autotrophic strain evaluation. The design was based on independent CO2, H2 and air inputs and the ability to operate at high pressures. The bioreactor configuration and cultivation strategy enabled growth of Cupriavidus necator strains for long periods, to reach over 3 g L-1 dry cell weight. No negative impact of the high pressure was observed on viability of the strains up to more than 4 bar overpressure. The cultivation was then

Over expression of GroESL in Cupriavidus necator for heterotrophic and autotrophic isopropanol production.

We previously reported a metabolic engineering strategy to develop an isopropanol producing strain of Cupriavidus necator leading to production of 3.4gL-1 isopropanol. In order to reach higher titers, isopropanol toxicity to the cells has to be considered. A toxic effect of isopropanol on the growth of C. necator has been indeed observed above a critical value of 15gL-1. GroESL chaperones were first searched and identified in the genome of C. necator. Native groEL and groES genes from C. necator were over-expressed in a strain deleted for PHA synthesis. We demonstrated that over-expressing gro
