Engineering Cupriavidus necator for L-threonine production.

L-threonine plays pivotal roles in the medicine, food, and feed industries. In this work, C. necator H16 was engineered for heterotrophic and autotrophic production of L-threonine. Blocking poly-hydroxybutyrate biosynthesis redirected the carbon flux toward L-threonine, resulting in a 6.8-fold increase in L-threonine titer relative to the wild type (from 5.56 mg/L to 38.03 mg/L). Deletion of genes involved in L-threonine catabolism further minimized L-threonine loss. Moreover, strategies including sufficient precursor supply, screening of key enzymes, relief of feedback inhibition, and efficie

Isopropanol production with engineered Cupriavidus necator as bioproduction platform.

Alleviating our society's dependence on petroleum-based chemicals has been highly emphasized due to fossil fuel shortages and increasing greenhouse gas emissions. Isopropanol is a molecule of high potential to replace some petroleum-based chemicals, which can be produced through biological platforms from renewable waste carbon streams such as carbohydrates, fatty acids, or CO2. In this study, for the first time, the heterologous expression of engineered isopropanol pathways were evaluated in a Cupriavidus necator strain Re2133, which was incapable of producing poly-3-hydroxybutyrate [P(3HB)].

Cupriavidus necator as a model organism for CO2-based biotechnology.

Cupriavidus necator H16 is a chemolithoautotrophic model organism that can grow in a simple mineral medium supplemented with the gases H2, O2 and CO2. This unique metabolic capability makes it a promising organism for sustainable biotechnology applications such as the production of bioplastics, single cell proteins and bulk chemicals from renewable resources. However, gas fermentation, particularly with explosive gas mixtures like H2 and O2, poses considerable technical and safety challenges. Effective control of gas mixtures, pressurized environments, mass transfer from gas to liquid and gas

Engineering Mixotrophy in the Chemolithoautotrophic Cupriavidus necator through Hydrogenase Induction.

Mixotrophy offers a promising strategy for biosynthesis by simultaneously utilizing organic carbon and CO2; however, mixotrophic microorganisms are rarely isolated outside of photoautotrophic microalgae. In this study, the chemoautotroph Cupriavidus necator H16 was found to preferentially consume fructose when coexisting with CO2 and H2, switching to utilization of only CO2 and H2 after fructose depletion. Transcriptomic analysis revealed significant differences in genes involved in energy metabolism, electron generation, and the respiratory chain. The molecular mechanism underlying the inabil

Optimizing Cupriavidus necator H16 as a host for aerobic C1 conversion.

Biological systems capable of converting CO2 or CO2-derived, single-carbon (C1) compounds can be used to reduce or reverse carbon emissions while establishing a circular bioeconomy to provide sustainable sources of the fuels, foods, and materials humanity relies on. A robust bioeconomy will rely upon a variety of microorganisms capable of assimilating C1 compounds and converting them to valuable products at industrial scale. While anaerobic microbes are ideal hosts for production of short-chain acids and alcohols, microbes capable of aerobic respiration are well suited for biosynthesis of high

Oxygen-dependent redox control enables growth-decoupled biotransformations in Cupriavidus necator.

Genetic instability and environmental heterogeneity present persistent challenges to the stable microbial production of reduced, value-added chemicals. This work shows that limiting oxygen availability redirects electrons from respiration toward targeted reductive pathways in Cupriavidus necator, steering growth-decoupled biotransformations - exemplified by the conversion of acetoin to 2,3-butanediol and of glycerol to 1,3-propanediol. In cell suspension assays under a hydrogen-containing atmosphere, anoxic conditions stabilised near-stoichiometric acetoin-to-2,3-butanediol conversion (93% mol

Physiology-informed use of Cupriavidus necator in biomanufacturing: a review of advances and challenges.

Biomanufacturing offers a potentially sustainable alternative to deriving chemicals from fossil fuels. However, traditional biomanufacturing, which uses sugars as feedstocks, competes with food production and yields unfavourable land use changes, so more sustainable options are necessary. Cupriavidus necator is a chemolithoautotrophic bacterium capable of consuming carbon dioxide and hydrogen as sole carbon and energy sources, or formate as the source of both. This autotrophic metabolism potentially makes chemical production using C. necator sustainable and attractive for biomanufacturing. Add

Persistent low PHB accumulation during pilot-scale gas fermentation in Cupriavidus necator.

Climate change and food security are tightly linked global challenges. Gas fermentation offers a sustainable route to convert CO₂ into microbial single-cell protein (SCP), with hydrogen-oxidizing bacteria (HOB) emerging as efficient biomass producers via the Calvin-Benson-Bassham cycle. While SCP can replace protein-rich feed components, intracellular polyhydroxybutyrate (PHB) accumulation redirects carbon and reducing power away from biomass and protein synthesis, reducing process efficiency. With this aim, a low amount of PHB is needed to focus on protein production rather than PHB accumulat

Isopropanol production from carbon dioxide in Cupriavidus necator in a pressurized bioreactor.

A bioreactor was designed to provide high gas mass transfer to reach cell and product titres in the g L-1 level from CO2 for realistic, laboratory scale, engineered autotrophic strain evaluation. The design was based on independent CO2, H2 and air inputs and the ability to operate at high pressures. The bioreactor configuration and cultivation strategy enabled growth of Cupriavidus necator strains for long periods, to reach over 3 g L-1 dry cell weight. No negative impact of the high pressure was observed on viability of the strains up to more than 4 bar overpressure. The cultivation was then

Over expression of GroESL in Cupriavidus necator for heterotrophic and autotrophic isopropanol production.

We previously reported a metabolic engineering strategy to develop an isopropanol producing strain of Cupriavidus necator leading to production of 3.4gL-1 isopropanol. In order to reach higher titers, isopropanol toxicity to the cells has to be considered. A toxic effect of isopropanol on the growth of C. necator has been indeed observed above a critical value of 15gL-1. GroESL chaperones were first searched and identified in the genome of C. necator. Native groEL and groES genes from C. necator were over-expressed in a strain deleted for PHA synthesis. We demonstrated that over-expressing gro

Rieske Oxygenase-Catalyzed Biotransformations in Recombinant Cupriavidus necator Fueled by Formate Oxidation.

The use of single carbon (C1) molecules, such as carbon dioxide or formate, is crucial in the transition from a linear, petroleum-based economy to a circular bioeconomy. Formate can serve as both a carbon and energy source, further enhancing its attractiveness as a feedstock. Cupriavidus necator, a lithoautotrophic microbial chassis strain, provides an opportunity to leverage formate for the synthesis of valuable products. However, its ability to grow on formate and the subsequent coupling of that process to recombinantly produced redox enzymes for the efficient production of high-value-added

Metabolomics Reveals Acetate-Formate compatibility for Dual-Carbon PHB biosynthesis in Cupriavidus necator.

The production of polyhydroxybutyrate (PHB) from organic substrates is often limited by energetic imbalance when single-carbon sources are used. Formate, while easily generated via electrochemical CO2 reduction, fails to sustain robust biomass and polymer yields when utilized as a primary carbon source. Similarly, acetate provides the necessary skeletal structures for PHB biosynthesis, but lacks the intrinsic energy to drive high-yield accumulation. To bridge this gap, we implemented a dual-carbon framework in wild-type Cupriavidus necator H16 that decouples energy supply from carbon assimilat

Comparative Genomic Assessment of the Cupriavidus necator Species for One-Carbon Based Biomanufacturing.

The transition from a petroleum-based manufacturing to biomanufacturing is an important step towards a sustainable bio-economy. In particular, biotechnological processes which use one carbon (C1) compounds as feedstock represent an interesting avenue. Many bacterial species evolved naturally to thrive on such compounds, among them Cupriavidus necator, which has been studied in the past due to its range of metabolic capabilities in utilisation and production of compounds of interest. Cupriavidus necator strain H16 is the reference laboratory strain for this species and by far the most extensive

Multilevel Metabolic Engineering of Cupriavidus necator for CO2-Derived Single-Cell Protein Production.

Enhancing nitrogen assimilation and coordinating carbon-energy metabolism in the autotrophic chassis Cupriavidus necator remain key challenges for maximizing single-cell protein (SCP) production from CO2. In this study, multilevel engineering was performed on C. necator. First, glnA was overexpressed to enhance nitrogen assimilation, which increased crude protein content to 72.75% in the best-performing strain, GS13. Preliminary physiological characterization suggested that this increase correlated with enhanced heme level. Second, glcR was disrupted to alter carbon allocation toward amino aci

Reducing Cooling Costs for Sustainable Food Biomanufacturing With Thermotolerant Cupriavidus necator H16 Mutant.

Autotrophic CO2 conversion to Single Cell Protein (SCP) using H2 and O2 is gaining commercial traction for food and feed production. Cupriavidus necator H16 is a key chassis organism for this "knallgas" fermentation, but industrial operation is typically constrained to ~30°C due to poor autotrophic growth at higher temperatures. Autotrophic growth at 37°C would enable the use of low-cost cooling water instead of chilled water, offering substantial reduction in cooling expenses. Here, we characterised a spontaneous C. necator mutant that conferred 37°C-thermotolerance when grown autotrophically

The state of metabolic engineering in the versatile bacterial cell factory Cupriavidus necator H16.

Cupriavidus necator strain H16 has emerged as a versatile microbial chassis for sustainable bioproduction due to its metabolic flexibility, enabling growth on a wide range of substrates, including H2/CO2, formate, and organic carbon (waste) sources such as volatile fatty acids. This review first provides a comprehensive overview of recent advances in systems-level understanding and metabolic engineering of C. necator. We next discuss recent advances in omics analyses and genome-scale metabolic modeling that are increasingly used to understand the large genome and wide metabolic portfolio of C.

Comparative characterisation of autotrophic and heterotrophic isopropanol formation by Cupriavidus necator in shake flasks.

Autotrophic cultivation offers a path to carbon-neutral bioproduction, which is increasingly valuable in the context of climate change mitigation. In this study, the production of isopropanol by Cupriavidus necator is used as an example for CO2 valorisation, and a simple shake bottle system is introduced to facilitate the development of aerobic autotrophic cultivation processes and strain screening. Applying 1.5 bar overpressure in the bottle's headspace enhances gas transfer while pressure decrease was shown to be correlated to biomass and product formation, allowing to follow metabolic activ

Genomics and transcriptomics insights into luteolin effects on the beta-rhizobial strain Cupriavidus necator UYPR2.512.

Cupriavidus necator UYPR2.512 is a rhizobial strain that belongs to the Beta-subclass of proteobacteria, able to establish successful symbiosis with Mimosoid legumes. The initial steps of rhizobium-legumes symbioses involve the reciprocal recognition by chemical signals, being luteolin one of the molecules involved. However, there is a lack of information on the effect of luteolin in beta-rhizobia. In this work, we used long-read sequencing to complete the genome of UYPR2.512 providing evidence for the existence of four closed circular replicons. We used an RNA-Seq approach to analyse the resp

An efficient method for the application of PHA-poor solvents to extract polyhydroxybutyrate from Cupriavidus necator.

There are many published studies presenting ethanol and acetone as PHAs-poor solvents, where these two solvents are shown to dissolve <2% (w/v) of PHAs at low temperatures. In this study, the suitability of ethanol and acetone for the recovery of PHB at different temperatures (from room temperature to near boiling point) in Cupriavidus necator was investigated. Experiments were performed using response surface methodology to examine the effects of different temperatures and heating incubation times on recovery percentage using the two solvents. The highest recovery percentage (92.3%) and produ

Unlocking the potential of Cupriavidus necator H16 as a platform for bioproducts production from carbon dioxide.

The rapid global increase in fossil fuel and energy consumption has resulted in the accumulation of greenhouse gases, especially carbon dioxide (CO2), thus contributing to climate change. Therefore, transforming CO2 into valuable products could yield beneficial outcomes. In this review, the capabilities of Cupriavidus necator H16, a light-independent chemoautotrophic bacterium, as a host platform for the transformation of CO2 into diverse products are explored. We begin by examining the progress in synthetic biology toolkits, gas fermentation technologies, and engineering approaches, consideri
