Salicylic Acid-Induced Elicitation of Nepetalactone and Rosmarinic Acid Biosynthesis in Naked Catmint (Nepeta nuda L.): Metabolomic and Transcriptional Insights.

Salicylic acid (SA) is involved in plant defense responses to environmental stressors by modulating gene expression and specialized metabolites production, enhancing plant adaptive resilience through systemic signaling pathways. This study investigates the impact of exogenous application of SA on the metabolism of iridoids and phenolic compounds-characteristic specialized metabolites of the Nepeta species, associated with diverse biological activities. Nepetalactone (NL) is a characteristic monoterpene iridoid, while rosmarinic acid (RA) represents the most abundant phenolic compound within th

Chemosensory Responses to the Repellent Nepeta Essential Oil and Its Major Component Nepetalactone by Aedes aegypti (Diptera: Culicidae), a Vector of Zika Virus.

Nepeta essential oil (Neo; catnip) and its major component, nepetalactone, have long been known to repel insects including mosquitoes. However, the neural mechanisms through which these repellents are detected by mosquitoes, including the yellow fever mosquito Aedes aegypti (L.), an important vector of Zika virus, were poorly understood. Here we show that Neo volatiles activate olfactory receptor neurons within the basiconic sensilla on the maxillary palps of female Ae. aegypti. A gustatory receptor neuron sensitive to the feeding deterrent quinine and housed within sensilla on the labella of

Enhancing the Phenolic Value of Polyphenol-Rich Extra Virgin Olive Oils Through Optimized Water Extraction, Macroporous Resin Recovery and Spray Drying.

Olive oil polyphenols are well-established for their health-promoting properties. A recently emerged polyphenol-rich extra virgin olive oil (EVOO) was subjected to a water extraction process, optimized using response surface methodology (RSM). The effects of salt content and the water-to-oil ratio on the total phenolics were initially evaluated. Maximal phenolic yield was obtained by a water/EVOO ratio of 9.6:1 (v:v) and 0% added salt, although the addition of 3% salt provided an oil-free extract with a satisfactory phenolic yield compared to the optimized extract. Further, the phenolics of th

Styrene, (+)-trans-(1R,4S,5S)-4-Thujanol and Oxygenated Monoterpenes Related to Host Stress Elicit Strong Electrophysiological Responses in the Bark Beetle Ips typographus.

Bark beetles kill apparently vigorous conifers during epidemics by means of pheromone-mediated aggregation. During non-endemic conditions the beetles are limited to use trees with poor defense, like wind-thrown. To find olfactory cues that help beetles to distinguish between trees with strong or weak defense, we collected volatiles from the bark surface of healthy felled or standing Picea abies trees. Furthermore, living trees were treated with methyl jasmonate in order to induce defense responses. Volatiles were analyzed by combined gas chromatography and electroantennographic detection (GC-E

Evaluating repellence properties of catnip essential oil against the mosquito species Aedes aegypti using a Y-tube olfactometer.

The mosquito species Aedes aegypti (L.) is known to act as a vector in the transmission of various diseases, including dengue fever and yellow fever. The use of insect repellents is one of precautionary measures used to mitigate the risk of these diseases in humans by reducing mosquito biting. Nepetalactone, a potent natural insect repellent primarily found in catnip (Nepeta cataria) essential oil, has emerged as a promising candidate for mosquito repellence. Here, we evaluated the potential of catnip essential oil (> 95% nepetalactone) for use as a mosquito repellent. Using a Y-tube olfactome

Revealing the molecular mechanism of biosynthesis and transcriptional regulation of PAs, caffeine and linalool globally under simulative stress in coffee plants.

Coffee has become one of the most popular beverages worldwide due to the variety of bioactive compounds, which also play crucial roles against biotic and abiotic stresses. However, little is known about how these defensive compounds are produced in coffee. Here, we found that the whole biosynthetic pathways and the production of caffeine and proanthocyanidins (PAs) were promoted under Methyl Jasmonate (MeJA) treatment. Co-expression data showed that some transcription factors were shared by caffeine and PA regulation, and further several candidate caffeine regulators were identified. The biosy

Effect of jasmonic acid on major terpenes and density of glandular trichomes in Lippia graveolens kunth (Verbenaceae).

The effect of exogenous application of jasmonic acid (JA) on the concentration of main terpenes and density of glandular trichomes was investigated in the Mexican oregano, propagated from seeds from 3 localities. JA 1 mM was applied locally and to the whole plant. JA locally applied increased the number of trichomes, with a mean of 20 trichomes more with respect to the controls in plants from Tecomavaca and Zapotitlán Salinas, and significantly increased the thymol concentration by 185% systemically and 255% locally, compared to the control. JA applied to the whole plant decreased the number o

Dynamics of the enhanced emissions of monoterpenes and methyl salicylate, and decreased uptake of formaldehyde, by Quercus ilex leaves after application of jasmonic acid.

Jasmonic acid (JA) is a signalling compound with a key role in both stress and development in plants, and is reported to elicit the emission of volatile organic compounds (VOCs). Here we studied the dynamics of such emissions and the linkage with photosynthetic rates and stomatal conductance. We sprayed JA on leaves of the Mediterranean tree species Quercus ilex and measured the photosynthetic rates, stomatal conductances, and emissions and uptake of VOCs using proton transfer reaction mass spectrometry and gas chromatography after a dark-light transition. Jasmonic acid treatment delayed the i

Characterization of four terpene synthase genes involved in nerolidol and linalool biosynthesis in Santalum album leaves induced by methyl jasmonate.

Santalum album is highly valued for its fragrant essential oil from heartwood. Volatile terpenoids involved in aroma formation in plants can be emitted in response to a variety of environmental stresses. However, the regulatory mechanisms underpinning the response of S. album to external stresses are not yet known. In this study, the regulatory mechanism of S. album leaves was investigated after the application of methyl jasmonate (MeJA), a stressor. Eight classes of volatile organic compounds were identified in S. album leaves, including terpenes, aldehydes, alcohols, ketones, esters, benzeno

Discovery of novel skeletons of aphid repellents based on the key fragment of nepetalactone and plant volatiles.

BACKGROUND: Aphids are significant agricultural pests, causing extensive crop damage and transmitting plant viruses. Conventional insecticide-based pest control faces challenges such as human and environmental toxicity and resistance. As alternative solutions, pheromone-based repellents have emerged as a promising approach. However, the instability of key compounds like (E)-β-Farnesene (EBF) has limited their effectiveness in field applications. To address these limitations, this study focuses on developing novel aphid repellent compounds based on plant volatiles and nepetalactone key fragment

Differential volatile emissions and salicylic acid levels from tobacco plants in response to different strains of Pseudomonas syringae.

Pathogen-induced plant responses include changes in both volatile and non-volatile secondary metabolites. To characterize the role of bacterial pathogenesis in plant volatile emissions, tobacco plants, Nicotiana tabacum L. K326, were inoculated with virulent, avirulent, and mutant strains of Pseudomonas syringae. Volatile compounds released by pathogen-inoculated tobacco plants were collected, identified, and quantified. Tobacco plants infected with the avirulent strains P. syringae pv. maculicola ES4326 (Psm ES4326) or pv. tomato DC3000 (Pst DC3000), emitted quantitatively different, but qual

Exploring the Immunotherapeutic Potential of Oleocanthal against Murine Cutaneous Leishmaniasis.

Leishmaniasis is a major tropical disease with increasing global incidence. Due to limited therapeutic options with severe drawbacks, the discovery of alternative treatments based on natural bioactive compounds is important. In our previous studies we have pointed out the antileishmanial activities of olive tree-derived molecules. In this study, we aimed to investigate the in vitro and in vivo antileishmanial as well as the in vivo immunomodulatory effects of oleocanthal, a molecule that has recently gained increasing scientific attention. Pure oleocanthal was isolated from extra virgin olive

Evaluating repellence properties of a catnip essential oil-based mosquito repellent using the human landing catch method in Eastern Uganda.

Mosquitoes (Diperta: Culidae) act as vectors for several diseases, including malaria, dengue fever and yellow fever. Mosquito repellents represent one of the primary measures used to reduce the risk of these diseases in humans by reducing mosquito landing and biting events. Nepetalactone, a natural insect repellent primarily found in the essential oil of catnip (Lamiaceae; Nepeta cataria), has established mosquito repellence properties, but has not been widely exploited as a mosquito repellent in malaria-endemic regions such as Sub-Saharan Africa. Here, we evaluated the potential of a locally

Genetics, phosphorus availability, and herbivore-derived induction as sources of phenotypic variation of leaf volatile terpenes in a pine species.

Oleoresin produced and stored in pine tree leaves provides direct resistance to herbivores, while leaf volatile terpenes (LVT) in the resin are also powerful airborne infochemicals. Resin concentration and profile show considerable spatial and temporal phenotypic variation within and among pine populations. LVT biochemistry is known to be under genetic control, and although LVT should be plastic to diverse abiotic and biotic environmental factors such as nutrient availability and herbivore attack, little is known about their relative contributions and interactive effects. The aim of this paper

Structural basis of sex pheromone detection in aphids.

Sex pheromones play a central role in regulating animal behavior and reproduction. In insects, these signals are perceived through specialized odorant receptors (ORs) that mediate species-specific communication and safeguard genetic integrity. However, the structural basis of sex pheromone detection remains largely unresolved. Here, we identified two ORs in the pea aphid Acyrthosiphon pisum, along with the conserved OR co-receptor (Orco), which together mediate recognition of the pheromone components nepetalactone and nepetalactol. Functional assays demonstrated that ApOR21-Orco and ApOR22-Orc

Methyl hexenoate and linalool emitted by hot peppers repel western flower thrips (Frankliniella occidentalis) and attract their predator (Orius laevigatus).

A plant emits diverse volatile compounds and communicates with other organisms at different trophic levels. Hot pepper, Capsicum annuum, leaves produced different compositions of 41 organic compounds upon an infestation by the western flower thrips, Frankliniella occidentalis. These compounds induced a non-preference behavior to the thrips while they attracted a natural enemy, Orius laevigatus. Among the differentially emitted compounds, Z-3-methyl hexenoate (MeH) and linalool repelled the herbivore but attracted the predator. MeH or linalool biosynthesis was suppressed by virus-induced gene s
