The photochemical behavior of 5-S-cysteinyldopa (5-S-CD), a colorless product of melanocyte metabolism, was investigated in neutral phosphate buffer with biologically relevant UV radiation. Exposure of 5-S-CD to pyrex-filtered UV light (wavelengths > 320 nm) was found to induce an oxygen-dependent reaction, leading to, besides abundant polymeric materials, the benzothiazine derivatives I and II (two diastereoisomers). Melanoma is one of the most lethal and malignant cancers and its incidence is increasing worldwide, and Japan is not an exception. Although there are numerous therapeutic options for melanoma, the prognosis is still poor once it has metastasized. 5-S-cysteinyldopa concentrations were determined by high-pressure liquid chromatography and electrochemical detection in plasma from normally pigmented patients and patients with oculocutaneous albinism, both tyrosinase-positive and tyrosinase-negative. The plasma 5-S-cysteinyldopa concentrations were similar in all three groups, suggesting that 5-S-cysteinyldopa can be produced by mechanisms which do not involve tyrosinase.

Seventy-six Caucasians, 30 men and 46 women, were investigated for the 24-hour excretion of 5-S-cysteinyl dopa in the urine during the months of September to November, 1973. No subject had had strong sun exposure for at least 4 weeks prior. A preliminary finding of a variation of 5-S-cysteinyldopa with season necessitated this precaution. The excretion varied between 9.0 and 242 mug/24 hours. The mean value in men was 100 mug/24 hours and in women 77.8 mug/24 hours. Malignant melanoma originating outside the skin is very rare, whereas primary malignant melanoma of the lung is extremely rare. 5-S-Cysteinyldopa (5-S-CD), a melanin metabolite, has been reported to be a prognostic marker for cutaneous malignant melanoma. This is the first report in the English language literature dealing with primary malignant melanoma of the lung using serum 5-S-Cysteinyldopa levels to monitor the effects of surgery and chemotherapy.

5-S-cysteinyl-dopa (cysdopa) and 5-S-cysteinyl-dopamine (cysdopamine) are oxidized in vitro by soybean lipoxygenase (LOX) in the presence of hydrogen peroxide giving rise to the corresponding pheomelanins. Urinary excretion of the phaeomelanin precursor 5-S-cysteinyldopa (5-S-CD) and the eumelanin metabolite 6-hydroxy-5-methoxyindole-2-carboxylic acid (6H5MI-2-C) was studied in black and albino mice. The urinary concentration of 5-S-CD was 31.7 ng/ml in black and 16.1 ng/ml in albino mice. The concentration of 6H5MI-2-C was 21.0 ng/ml in the urine of black mice. A 35-year-old male presented with a variant of neurocutaneous melanosis with leptomeningeal malignant melanoma. He had three pigmented nevi from birth. He suffered diplopia followed by headache. T1-weighted magnetic resonance (MR) imaging revealed hydrocephalus and a small linear hyperintense lesion in the right frontal cortex. Several parts of the cortical sulci and the brain surface were slightly enhanced by gadolinium. Ventriculoperitoneal shunting was performed and extensive pigmented leptomeninges were recognized. Open biopsy established the diagnosis of leptomeningeal malignant melanoma.
