Delay of iris flower senescence by cytokinins and jasmonates.

It is not known whether tepal senescence in Iris flowers is regulated by hormones. We applied hormones and hormone inhibitors to cut flowers and isolated tepals of Iris × hollandica cv. Blue Magic. Treatments with ethylene or ethylene antagonists indicated lack of ethylene involvement. Auxins or auxin inhibitors also did not change the time to senescence. Abscisic acid (ABA) hastened senescence, but an inhibitor of ABA synthesis (norflurazon) had no effect. Gibberellic acid (GA3 ) slightly delayed senescence in some experiments, but in other experiments it was without effect, and gibberellin i

Cytokinins and plant immunity: old foes or new friends?

Cytokinins are plant growth promoting hormones involved in the specification of embryonic cells, maintenance of meristematic cells, shoot formation and development of vasculature. Cytokinins have also emerged as a major factor in plant-microbe interactions during nodule organogenesis and pathogenesis. Microbe-originated cytokinins confer abnormal hypersensitivity of cytokinins to plants, augmenting the sink activity of infected regions. However, recent findings have shed light on a distinct role of cytokinins in plant immune responses. Plant-borne cytokinins systemically induce resistance agai

Chemically Induced Resistance to Pathogen Infection in Arabidopsis by Cytokinin (Trans-Zeatin) and an Aromatic Cytokinin Arabinoside.

This study compares the ability of the cytokinin (CK) trans-zeatin (tZ) and the CK sugar conjugate 6-(3-methoxybenzylamino)purine-9-arabinoside (BAPA) to induce resistance against the bacterial pathogen Pseudomonas syringae in Arabidopsis thaliana. Treatment with either tZ or BAPA significantly reduced bacterial growth after a later infection. This chemically induced resistance (IR) required the CK receptor AHK3, highlighting its critical role in mediating resistance by tZ and BAPA. This is remarkable as these compounds show either high or no affinity for this CK receptor, respectively. Surpri

Cytokinin-enhanced accumulation of indole alkaloids in Catharanthus roseus cell cultures - The factors affecting the cytokinin response.

Cytokinins were found to stimulate the alkaloid synthesis induced by removing auxin from the medium of a cell line of Catharanthus roseus. Diluting the mineral salts of the culture medium decreased the alkaloid production but increased the "sensitivity" of the cells. Addition of high levels of Ca(2+), Mg(2+) or Sr(2+) to B5 media in which the mineral salts were diluted to 5-40%, increased the alkaloid production. The latter effect is related only partially to enhanced osmotic potential.

Rapid profiling of cytokinins using supercritical fluid chromatography coupled with tandem mass spectrometry.

BACKGROUND: The determination of plant hormones is still a very challenging analytical discipline, mainly due to their low concentration in complex plant matrices. Therefore, the involvement of very sensitive high-throughput techniques is required. Cytokinins (CKs) are semi-polar basic plant hormones regulating plant growth and development. Modern methods for CK determination are currently based on ultra-high performance liquid chromatography-tandem mass spectrometry (UHPLC-MS/MS), which enables the separation of CK isomeric forms occurring endogenously in plants. Here, ultra-high performance

Plant hormone cytokinins control cell cycle progression and plastid replication in apicomplexan parasites.

Cytokinins are plant hormones that are involved in regulation of cell proliferation, cell cycle progression, and cell and plastid development. Here, we show that the apicomplexan parasites Toxoplasma gondii and Plasmodium berghei, an opportunistic human pathogen and a rodent malaria agent, respectively, produce cytokinins via a biosynthetic pathway similar to that in plants. Cytokinins regulate the growth and cell cycle progression of T. gondii by mediating expression of the cyclin gene TgCYC4. A natural form of cytokinin, trans-zeatin (t-zeatin), upregulated expression of this cyclin, while a

Extraction and Analysis of Cytokinins from Mammalian Tissues by HPLC-HRMS.

Cytokinins (CKs) are adenine-derived plant hormones regulating almost every aspect of plant growth and development. But CK-like compounds may also occur in microbial as well as mammalian systems. Detecting CKs in animal matrices is challenging due to low abundance and complex backgrounds. We describe a high-performance liquid chromatography-high-resolution mass spectrometry (HPLC-HRMS) protocol for CK extraction, purification, and quantification. Samples are frozen or lyophilized, homogenized, and extracted with methanol: water (80:20, v/v) containing stable isotope labeled CKs as internal sta

[Immunotherapy of cancer using cytokinins. Use and perspectives in lung oncology].

Progress in genetics now enables the synthesis of molecules acting on the regulation of the immune system which are called cytokines. Currently there are several cytokines, Interferon (IFN), Interleukin 2 (IL2), tumour necrosis factor (TNF) as well as haematopoietic growth factors and these are the object of study in clinical trials. Interferon has already been used in the therapy of hairy cell leukaemia, Kaposi sarcoma associated with AIDS(SIDA) and metastasis of malignant melanoma. Interleukin 2 allows for an increase in the cytotoxic activity of NK cells in producing LAK cells, the lymphocy

Pattern of auxin and cytokinin responses for shoot meristem induction results from the regulation of cytokinin biosynthesis by AUXIN RESPONSE FACTOR3.

De novo organ regeneration is an excellent biological system for the study of fundamental questions regarding stem cell initiation, cell fate determination, and hormone signaling. Despite the general belief that auxin and cytokinin responses interact to regulate de novo organ regeneration, the molecular mechanisms underlying such a cross talk are little understood. Here, we show that spatiotemporal biosynthesis and polar transport resulted in local auxin distribution in Arabidopsis (Arabidopsis thaliana), which in turn determined the cytokinin response during de novo shoot regeneration. Altered cytokinin metabolism affects cytokinin, auxin, and abscisic acid contents in leaves and chloroplasts, and chloroplast ultrastructure in transgenic tobacco.

Cytokinins (CKs) are involved in the regulation of plant development including plastid differentiation and function. Partial location of CK biosynthetic pathways in plastids suggests the importance of CKs for chloroplast development. The impact of genetically modified CK metabolism on endogenous CK, indole-3-acetic acid, and abscisic acid contents in leaves and isolated intact chloroplasts of Nicotiana tabacum was determined by liquid chromatography/mass spectrometry and two-dimensional high-performance liquid chromatography, and alterations in chloroplast ultrastructure by electron microscopy

Cytokinin and growth of excised roots of Bryophyllum calycinum.

Excised roots of Bryophyllum calycinum require for growth both auxin and cytokinin. This is demonstrated by the poor growth of 2-mm root tips in a basal medium of mineral salts, sucrose, and vitamins supplemented with either an auxin or a cytokinin, and much better growth when the basal medium is supplemented with both auxin and cytokinin. However, both substances are synthesized by the root, as is demonstrated by the growth of large inocula (dry wt 6-7 mg) through many successive passages in a medium limited to mineral salts, sugar, and vitamins.

Cytokinin derivatives as tools to enhance plant stress resilience.

Cytokinins (CKs) are key regulators of plant growth and development. Recently, CKs have been increasingly recognized to have additional roles as modulators of plant adaptation to abiotic and biotic stresses. A large part of CK action in stress adaptation can be attributed to the promotion of antioxidative defense mechanisms. However, the contributions of CKs to stress tolerance can be complex and sometimes contradictory, as both increased and decreased CK signaling can enhance stress resilience depending on the context, a phenomenon referred to as the "cytokinin paradox". In addition, some eff

Cytokinin availability and chloroplast functional lifetime in senescing leaves.

Leaf senescence is often characterized by visible yellowing, reflecting the degradation of photosynthetic pigments and the functional deterioration of photosynthetic complexes, marking a shift from photosynthetic maintenance to nutrient remobilization. Cytokinins are key modulators in this process. This review examines cytokinin action in mature Arabidopsis leaves from the perspective of chloroplast functional lifetime, focusing on how long-distance supply, local metabolism and candidate transport routes may shape cytokinin availability and action during leaf senescence. Cytokinin signaling is

Plant Hormone Cytokinin as Aggregation Modulator of Gelsolin Amyloidosis.

Amyloidosis, a self-assembly of proteins or peptides, is associated with numerous degenerative diseases, such as gelsolin amyloidosis, which remain without a cure. Gelsolin protein is an actin-binding protein, but when aggregated in a diseased state, it is a potential drug target. Specifically, gelsolin mutations, N184K and D187Y, have been linked to renal amyloidosis and systemic progressive deposition of amyloids, respectively. Understanding how such mutations mitigate gelsolin aggregation and how this process can be prevented through small molecule inhibitors is of interest. Herein, we expl

Characterization of a cytokinin-binding protein locus in Mycobacterium tuberculosis.

Cytokinins are adenine-based hormones that have been well-characterized in plants but are also made by bacteria, including the human-exclusive pathogen Mycobacterium tuberculosis. Like plants, M. tuberculosis uses cytokinins to regulate gene expression. We previously established that cytokinin overaccumulation in M. tuberculosis results in a buildup of aldehydes produced during cytokinin breakdown. In plants, dedicated enzymes called cytokinin oxidases convert cytokinins into adenine and various aldehydes. Proteasome degradation-deficient M. tuberculosis, which cannot degrade the cytokinin-pro

Spatiotemporal regulation of lateral root organogenesis in Arabidopsis by cytokinin.

The architecture of a plant's root system, established postembryonically, results from both coordinated root growth and lateral root branching. The plant hormones auxin and cytokinin are central endogenous signaling molecules that regulate lateral root organogenesis positively and negatively, respectively. Tight control and mutual balance of their antagonistic activities are particularly important during the early phases of lateral root organogenesis to ensure continuous lateral root initiation (LRI) and proper development of lateral root primordia (LRP). Here, we show that the early phases of

Two-component circuitry in Arabidopsis cytokinin signal transduction.

Cytokinins are essential plant hormones that are involved in shoot meristem and leaf formation, cell division, chloroplast biogenesis and senescence. Although hybrid histidine protein kinases have been implicated in cytokinin perception in Arabidopsis, the action of histidine protein kinase receptors and the downstream signalling pathway has not been elucidated to date. Here we identify a eukaryotic two-component signalling circuit that initiates cytokinin signalling through distinct hybrid histidine protein kinase activities at the plasma membrane. Histidine phosphotransmitters act as signall

Cytokinin perception in potato: new features of canonical players.

Potato is the most economically important non-cereal food crop. Tuber formation in potato is regulated by phytohormones, cytokinins (CKs) in particular. The present work studied CK signal perception in potato. The sequenced potato genome of doubled monoploid Phureja was used for bioinformatic analysis and as a tool for identification of putative CK receptors from autotetraploid potato cv. Désirée. All basic elements of multistep phosphorelay required for CK signal transduction were identified in the Phureja genome, including three genes orthologous to three CK receptor genes (AHK 2-4) of Arabi

Two classes of compounds which bind to calmodulin in a calcium-dependent manner (neuroleptic drugs and local anaesthetics) were used to investigate the possible involvement of a calcium-dependent regulator protein in the action of the plant hormones cytokinins. The cytokinin-induced synthesis of betacyanin in Amaranthus tricolor seedlings was used as one test system.
