Anti-neutrophil cytoplasm-associated glomerulonephritis.

Wegener's granulomatosis, microscopic polyangiitis, and renal limited vasculitis are associated with circulating anti-neutrophil cytoplasm antibodies and are an important cause of rapidly progressive glomerulonephritis. This review gives an account of recent advances in the understanding of the pathogenesis underlying these conditions and how these may lead to future treatments. Consideration is given to recent clinical trials in the management of anti-neutrophil cytoplasm antibodies (ANCA)-associated vasculitides.

Antineutrophil cytoplasm autoantibodies and vasculitis.

Antineutrophil cytoplasm autoantibodies are useful diagnostic serologic markers for a variety of well-known primary vasculitic syndromes, including Wegener's granulomatosis, microscopic polyangiitis, and idiopathic necrotizing and crescentic glomerulo-nephritis. More recently antineutrophil cytoplasm autoantibodies have been found in other vasculitic syndromes, such as Churg-Strauss syndrome, Henoch-Schönlein purpura, and some nonvasculitic diseases such as rheumatoid arthritis, inflammatory bowel disease, and autoimmune hepatobiliary diseases. There is now evidence to suggest that infection m

Anti-neutrophil cytoplasm antibodies (ANCA).

Circulating antibodies directed against cytoplasmic constituents of granulocytes and monocytes (anti-neutrophil cytoplasm antibodies, ANCA) are found in about 80% of active cases of Wegener's granulomatosis and microscopic polyarteritis. Antibodies are detected by indirect immunofluorescence on normal peripheral blood leucocytes, or by ELISA or radio-immunoassay using a neutrophil cytoplasm extract. There are 2 patterns of staining seen on indirect immunofluorescent examination of leucocytes and a number of different molecules recognised on Western blots. In Wegener's granulomatosis diffusely

Anti-neutrophil cytoplasm antibodies can recognize vascular endothelial cell-bound anti-neutrophil cytoplasm antibody-associated autoantigens.

Anti-neutrophil cytoplasm antibodies (ANCA) are strongly associated with the development of systemic vasculitis. Myeloperoxidase and proteinase-3 have been identified as targets for P-ANCA and C-ANCA, respectively. Both enzymes are released from neutrophil azurophil granules following neutrophil activation and both are highly cationic. Purified myeloperoxidase is demonstrated to bind non-convalently to endothelial cell membranes, to retain its enzymic function following binding, and to retain its antigenicity for P-ANCA. Endothelial cell-bound myeloperoxidase enhances complement-dependent cyto

Antineutrophil cytoplasm antibody (ANCA) associated vasculitis.

In 1982 we first reported the presence of antineutrophil cytoplasm antibodies (ANCA) in 8 patients with systemic vasculitis and segmental necrotizing glomerulonephritis. The results of long-term follow-up are described. Screening of 7,500 serum samples revealed positive ANCA in 9 additional patients with vasculitis. Eighty-eight other patients with vasculitis were ANCA negative, including 7 with microscopic polyarteritis nodosa (MPAN) and 8 with Wegener's granulomatosis (WG). Conversely, ANCA were never detected in the absence of vasculitis. Fourteen patients presenting with glomerulonephritis

Mesangial cell cytoplasm and glomerular disease.

In this study we have examined mesangial cell cytoplasm as seen by light microscopy of plastic embedded renal biopsy material. An assessment of mesangial cell morphology has been made in each of the 350 renal biopsies, and this has been correlated with the disease process. A surprisingly close correlation with the type of disease present was demonstrated. Thus it appears that an increase in mesangial cell cytoplasm correlates with an active process involving the mesangial area. It is suggested that the index of mesangial cell reactivity may be useful in diagnosis of glomerular disease processe

POLIOVIRUS: GROWTH IN NON-NUCLEATE CYTOPLASM.

Cytoplasmic fragments were produced by micromanipulation of cells from a human amnion cell line cultured on coverslips. The cultures were infected with type 1 (Mahoney) poliovirus, and incubated for 7 hours with tritiated uridine (H3U). Fluorescent antibody to the poliovirus indicated antigenic sites in a number of non-nucleate fragments. By autoradiography the incorporation of H3U was demonstrated at some of the same sites. The occurrence of poliovirus antigen at the same site as induced synthesis of RNA in non-nucleate cytoplasm of mammalian cells indicates that poliovirus infection and grow

[Wegener's granulomatosis and anti-cytoplasm antibodies].

20 patients with biopsy-proven Wegener's granulomatosis (WG) and 95 control patients underwent determination of anticytoplasmic antibodies (ACPA) by the indirect immunofluorescence technique to assess the specificity and sensitivity of ACPA for WG. Of 14 untreated patients with WG, 13 were ACPA-positive. All these patients became ACPA-negative under immunosuppressive treatment. 4 patients treated with immunosuppressive drugs and 2 patients in remission after termination of therapy had a negative ACPA-test. ACPA were detected in 9 patients of the control group (two patients with Henoch Schoenle

Antineutrophil cytoplasm antibodies in Kawasaki disease.

Autoantibodies against components of neutrophil cytoplasm develop during adult vasculitic diseases such as Wegener's granulomatosis and microscopic polyarteritis, and they are predominantly of the IgG class. Similar but distinct antibodies have been described in children with Kawasaki disease and both IgM and IgG class antibodies are represented. This adds another clinically distinct childhood form of vasculitis to the adult forms in which autoantibodies to neutrophil cytoplasmic antigens have been detected.

Antineutrophil cytoplasm antibody in crescentic glomerulonephritis.

Antineutrophil cytoplasm antibody (ANCA) has been reported in the sera of adults with Wegener's granulomatosis and microscopic polyarteritis, but this phenomenon has not so far been described in children. We report three children with crescentic glomerulonephritis in whom ANCA concentrations were raised at presentation. Two cases were idiopathic, and the other later developed features of Wegener's granulomatosis. In all three, plasma exchange and immunosuppression removed ANCA, but in only one case was there clinical improvement. This child later developed classical nasal lesions of Wegener's

Macromolecular Crowding in Cytoplasm and Cellular Organelles.

A high concentration of diverse macromolecules causes a densely packed environment, influencing both function and structural change of the cell organelles, cytoplasm, and molecules within. The concept of molecular crowding was for the first time introduced in the 70 s but was only accepted at the end of the 90 s. Crowding demonstrates how the high concentration of macromolecules, such as proteins, nucleic acids, and polysaccharides, influences the behavior and interactions of these molecules. This systematic review explored recent literature that impacted the understanding of molecular crowdin

Myelodysplasia, vasculitis and anti-neutrophil cytoplasm antibodies.

A cutaneous or systemic vasculitis occurs in myelodysplasia as well as in myeloproliferative and lymphoproliferative disorders. The most common lesion is a leucocytoclastic vasculitis, with neurological or joint involvement occurring less often. The vasculitis may appear contemporaneously with or precede the clinical onset of the blood dyscrasia. Occasionally the lesions respond dramatically to the use of steroids but in general, patients with vasculitis have a worse prognosis than those with uncomplicated myelodysplasia. Vasculitis and myelodysplasia appear together too often for the associat

[Anti neutrophil cytoplasm antibody in Wegener's granulomatosis].

Autoantibodies against human neutrophil cytoplasma (ACPA) were assayed in 50 serum samples from patients with systemic vasculitic syndrome by indirect immunofluorescence method. ACPA titer was higher than 32x in 5 patients with active Wegener's granulomatosis (WG) although ACPA of low titer was found in 2 patients with AGA, 1 patient with temporal arteritis. It was confirmed that serial changes of ACPA titer in 5 patients with WG proved to be the selective marker of disease activity and the severity of renal injury in WG. It was concluded that the detection of ACPA is useful both for the early

Anisotropic Subdiffusion in the Cytoplasm of Living Cells.

The cytoplasm of eukaryotic cells is a complex fluid that harbors a variety of membrane-enveloped organelles with distinct spatial distributions and duties. Peroxisomes, for example, are vesicular entities with radii in the sub-micron range, dispersed all over the cytoplasm to provide oxidative reaction vessels across the cell. Here, we have tracked peroxisomes as biologically relevant tracer particles to explore the local structure of the cytoplasm. Focusing on the vast majority of (sub)diffusively moving peroxisomes, we show that these display a significant motion anisotropy even in the abse

Multispecific anti neutrophil cytoplasm antibodies: a case report.

AIM: To report the presence af ANCA with an unusual polyreactivity. CASE REPORT: 50 year-old woman with pulmonary fibrosis whose immunological investigations showed the presence of ANCA with an unusual polyreactivity against several neutrophil proteins (PR3,MPO, BPI, lysozyme, elastase and cathepsine G) which could be related to a polyclonal hypergammaglobulinemia occurring in this patient. CONCLUSION: The international consensus on the testing of ANCA recommends seeking major specificities like MPO and PR3 which are good markers of ANCA-associated vasculitides. The use of multiantigenic ELI

Nucleus-cytoplasm cross-talk in the aging brain.

Aging is a primary risk factor for fatal neurodegenerative disorders, yet the mechanisms underlying physiological healthy aging and pathological aging, and how these mechanisms can divert one scenario to the other, are not completely understood. In recent years, reports indicate that alterations in nucleocytoplasmic transport may be a hallmark of both healthy and pathological aging. In this review, I summarize recent evidence supporting this information, specifically focusing on the association between the nucleocytoplasmic transport and aging of the brain, indicating both common and case-spec

Inverted urothelial papillomas with foamy or vacuolated cytoplasm.

Inverted papillomas of the bladder are uncommon benign neoplasms characterized by endophytic growth of urothelial cells as anastomosing cords, displaying minimal cytologic atypia. Reports of inverted papilloma associated with urothelial carcinoma or urothelial carcinoma arising within inverted papilloma highlight the difficulties in evaluating urothelial lesions with inverted growth patterns. Within the spectrum of findings in inverted papilloma, vacuolization and foamy (xanthomatous-appearing) cytoplasmic changes have not been previously reported. In the current study, we present 5 novel case

[Antineutrophil cytoplasm antibodies (ANCA): description and immunopathological role].

These antibodies are specific for antigens in the cytoplasm of neutrophils. The main antigenic targets are proteinase 3(PR3) and myeloperoxydase (MPO) but other targets have been described without determinant conclusions for clinical practice. Staining patterns can be distinguished by an indirect immunofluorescence test (IFI), in which ethanol fixed neutrophils from healthy donors are incubated with patient's sera. Two patterns are distinguished: cytoplasmic pattern (c-ANCA) and perinuclear pattern (p-ANCA). When ANCA are detected by IFI, from a practical point of view, anti -MPO and anti-PR3
