Lignocellulose (dry plant biomass) is an abundant cheap inedible residue of agriculture and wood industry with great potential as a feedstock for biotechnological processes. Lignocellulosic substrates can serve as valuable resources in fermentation processes, allowing the production of a wide array of chemicals, fuels, and food additives. The main obstacle for cost-effective conversion of lignocellulosic hydrolysates to target products is poor metabolism of the major pentoses, xylose and L-arabinose, which are the second and third most abundant sugars of lignocellulose after glucose. We study

Xylitol, known for its health benefits, is a valuable compound in the food and pharmaceutical industries. However, conventional chemical production methods are often unsustainable for large-scale applications, prompting the need for alternative approaches. This study demonstrates a significant enhancement in xylitol production using microbial cell factories, optimized through metabolic engineering. Two synthetic pathways were combined, and the introduction of a novel NADPH-dependent xylitol dehydrogenase further boosted xylitol yields, achieving 0.14 g xylitol/g glucose-a record-high yield for

Hundreds of ribotypes of the gastrointestinal pathogen Clostridioides difficile have emerged over the last three decades, yet the factors driving their emergence are poorly understood. Recently, there has been an increase in infections caused by PCR ribotype 023 (RT023) strains in Europe. We profiled the growth of seven RT023 strains in 190 unique carbon sources and found they were able to grow in xylitol, a sugar alcohol used as a food additive in humans and animals. Other ribotypes of C. difficile tested (n = 19) displayed little to no growth in 0.5% xylitol and were growth-inhibited in high

L-Xylulose is an intermediate in certain metabolic pathways and is classified as a rare sugar. It shows important physiological effects such as acting as an inhibitor of α-glucosidase and decreasing blood glucose, and it can be employed to produce other significant rare sugars, such as L-ribose and L-xylose which contribute to the production of antiviral drugs. Chemical synthesis of L-xylulose was performed, but it is difficult and low yielding. The biotransformation from xylitol to L-xylulose by xylitol 4-dehydrogenase was studied intensively. This review describes the occurrence of L-xylulos

UNLABELLED: <p>Introduction: d-Xylose and l-arabinose are major components of plant biomass and as such also important carbon sources for most fungi and attractive compounds for biotechnology. Most fungi use the pentose catabolic pathway to convert these sugars through several redox reactions. One of the steps in this pathway is the conversion of l-xylulose to xylitol, catalyzed by l-xylulose reductase (LXR). Genetic studies in Aspergillus niger revealed the involvement of two LXR-encoding genes, lxrA and lxrB.
METHODS: LxrA and LxrB were heterologously pro

BACKGROUND: The principal roots (CHW) and lateral roots (FZ) of Aconitum carmichaelii Debx. are widely used medicinal herbs in Asia. Although the two roots are derived from the same plant, they exhibit differences in anti-inflammatory activity (AIA). In the market, small FZ is frequently passed off as CHW. Therefore, revealing AIA-related markers and biosynthetic regulatory mechanisms between CHW and FZ are critical for the quality control of these medicinal materials.
OBJECTIVE: To identify AIA-related markers and reveal the biosynthetic regulatory mechanisms of these markers.
METHODS: A "m

The tea cultivar 'Xiaoxueya', a temperature-sensitive albino mutant, is a rare tea germplasm because of its highly enriched amino acid content and brisk flavour. In comparison with green leaf tissues of 'Xiaoxueya', albino leaves show significant deficiency in chlorophylls and carotenoids and severely disrupted chloroplasts. Furthermore, the accumulation of quality-related secondary metabolites is altered in 'Xiaoxueya' albino leaf, with significantly increased contents of total amino acids, theanine, and glutamic acid and significantly decreased contents of alkaloids, catechins, and polypheno

Iron (Fe) deficiency is one of the most common micronutrient imbalances limiting plant growth globally, especially in arid and saline alkali regions due to the decreased availability of Fe in alkaline soils. Malus halliana grows well in arid regions and is tolerant of Fe deficiency. Here, a physiological and metabolomic approach was used to analyze the short-term molecular response of M. halliana roots to Fe deficiency. On the one hand, physiological data show that the root activity first increased and then decreased with the prolongation of the stress time, but the change trend of root pH was

A mutant ribitol dehydrogenase (RDH-F) was purified from Klebsiella aerogenes strain F which evolved from the wild-type strain A under selective pressure to improve growth on xylitol, a poor substrate used as sole carbon source. The ratio of activities on xylitol (500 mM) and ribitol (50 mM) was 0.154 for RDH-F compared to 0.033 for the wild-type (RDH-A) enzyme. The complete amino acid sequence of RDH-F showed the mutations. BACKGROUND: MYB transcription factors play critical roles in secondary metabolite biosynthesis in medicinal plants and are crucial for abiotic stress responses. However, no studies have identified BcMYB genes in Bupleurum chinense DC. or analyzed their expression profiling under temperature stress. The role of BcMYB in B. chinense's response to temperature stress and its regulation of saikosaponins synthesis remains unclear.
RESULTS: This study performed a genome-wide identification of the BcMYB gene family in B. chinense using its genomic data, and analyzed the changes in saikosaponins cont

Camphora officinarum Nees ex Wall (C. officinarum), a citral-rich aromatic plant, is recognized as an unrivalled natural source of citral for spice production. Its large-scale cultivation in China via the sustainable coppice-rotation system has markedly revitalized rural economies. Nevertheless, the genetic basis driving its prolific citral accumulation remains elusive. We conducted an integrated transcriptomic and metabolomic analysis comparing three citral type accessions (C1, C2, C3) with a non-citral type control (C0). Metabolomic profiling identified 904 leaf metabolites, with terpenoids

Various internal and external factors negatively affect the homeostatic equilibrium of organisms at the molecular to the whole-body level, inducing the so-called state of stress. Stress affects an organism's welfare status and induces energy-consuming mechanisms to combat the subsequent ill effects; thus, the individual may be immunocompromised, making them vulnerable to pathogens. The information presented here has been extensively reviewed, compiled, and analyzed from authenticated published resources available on Medline, PubMed, PubMed Central, Science Direct, and other scientific database
