Enterovirus 71 (EV71), a major causative agent of hand, foot, and mouth disease (HFMD), is often associated with severe clinical outcomes. In this study, three rare cyano-containing alkaloids, cyanodaphcalycines A-C (1-3), featuring an unusual 5/6/6/7/5/5-hexacyclic scaffold, and an undescribed daphmanidin A-type alkaloid, daphmanidin G (4), were isolated from the leaves and stems of the medicinal plant Daphniphyllum calycinum Benth., along with three known Daphniphyllum alkaloids (5-7). The previously undescribed structures and their absolute configurations were determined using a combination

Daphniphyllum calycinum (DC), a main component of Chinese patent drug Fengliao-Changweikang, is effectively used to cure bowel disease in the clinic. It was recorded that DC possessed slight toxicity, which was caused by the alkaloids existing in its extract. Unfortunately, to date the toxicity level and toxic constituents are still unclear. The present study is designed to illustrate the acute toxicity and induced organ damages of the total alkaloids as well as to determine the toxic constituents. Based on the above studies, not only was the acute toxicity determined but also hepatic toxicity

Six undescribed Daphniphyllum alkaloids, daphcalycines E-J (1-6), possessing structurally diverse skeletal frameworks, were isolated from the seeds of Daphniphyllum calycinum. Their structures and relative configurations were elucidated by comprehensive spectroscopic analysis, including extensive 2D NMR techniques and single-crystal X-ray diffraction. Compounds 1-6 were evaluated in vitro for cytotoxicity and for their effects on oxygen-glucose deprivation/reoxygenation (OGD/R)-induced injury in H9c2 cells. None of the compounds showed detectable cytotoxicity at 10 μM. Under the OGD/R conditio

Native to the Asia-Pacific region and widely applied in traditional Chinese medicine, the genus Daphniphyllum has produced over 330 known Daphniphyllum alkaloids. Investigations into these alkaloids have shown an exceptional range of interesting bioactivities. Challenging and caged polycyclic architectures and the promising biological profiles make Daphniphyllum alkaloids intriguing synthetic targets. Based on their backbones, these alkaloids can be categorized into 13-35 structurally distinct subfamilies. In addition to our work, almost 30 impressive total syntheses of Daphniphyllum alkaloids

Daphniphyllum neilgherrense (Daphniphyllaceae), an ethnomedicinal plant, is used for skin diseases and various ailments, for which the studies on its anatomical, phytochemical and pharmacological properties are unexplored. Hence, the present study has been aimed to explore anatomical, phytochemical, antimicrobial, antioxidant, and anticancer activities. The anatomy of different parts of the plant is elaborated for the first time. Antibacterial activity observed (inhibition zone) against Gram-positive bacteria (Staphylococcus aureus) and Gram-negative bacteria (Escherichia coli) was 18.10 ± 0.2

ETHNOPHARMACOLOGICAL RELEVANCE: Gastric mucosal injury, commonly induced by ethanol and psychological stress, represents a significant clinical challenge with limited natural therapeutic options. The Li ethnic herbal pair consisting of Daphniphyllum calycinum Benth. and Polygonum hydropiper L. (DCPH), commercially available as Fengliao Changweikang Granules, is traditionally used by the Li people of Hainan, China, for gastrointestinal disorders. However, its gastroprotective effects and underlying mechanisms remain to be systematically characterized.

Three new carbon-skeleton triterpenoids, named daphniphyanoids A-C (1-3), featuring an uncommon A-ring rearranged 5/6/6/6/5-pentacyclic nucleus, along with two biogenetically related known lupane-type triterpenoids (4 and 5), have been isolated from Daphniphyllum calycinum Benth. The determination of their structures was based on the extensive spectroscopic analysis, X-ray single crystal diffraction analysis, and chemical conversion. A plausible biosynthetic pathway for compounds 1-5 was proposed, involving a pinacol rearrangement, Wagner-Meerwein rearrangement, oxidation, and esterification.

Gastric mucosal injury refers to the pathological changes characterized by congestion, edema, bleeding and erosion of the gastric mucosa caused by various factors. This condition is a significant feature of gastritis in which the inflammatory response plays a critical role in its pathogenesis. The herbal pair Daphniphyllum calycinum-Polygonum hydropiper (DCPH), known for its anti-inflammatory and antioxidant properties, has been shown to alleviate gastric mucosal injury. This study aimed to elucidate the mechanism by which DCPH alleviates gastric mucosal injury by integrating network pharmacol

A concise enantioselective total synthesis of (-)-daphenylline, a hexacyclic Daphniphyllum alkaloid with a unique benzene ring, was achieved in 14 steps. The synthesis commences with two chiral stereocenters, C2 and C18, readily installed via Carreira's Ir/amine dual-catalyzed allylation. The allylic bridgehead amine 6 was rapidly prepared through Wickens' photoredox-catalyzed hydrocarboxylation of olefin and CuBr2-catalyzed α-amination of ketone. The tetracycle 4 was formed via Pd-catalyzed reductive Heck reaction or, more concisely, by Krische's Rh-catalyzed reductive 1,6-enyne cyclization.

Covering: up to July 2026The vinylogous Mannich reaction (VMR) enables γ-selective carbon-carbon bond formation between vinylogous nucleophiles and imines or iminium ions, yielding δ-aminocarbonyl frameworks. Compared with the classical Mannich reaction, VMR shows distinct advantages in remote functionalization and convergent bond formation, facilitating the rapid assembly of nitrogen-containing heterocycles, multiple stereogenic centers, and densely functionalized intermediates. As a result, this reaction has become an increasingly valuable strategy for generating complexity in the total synt

The severe side effects of chemosynthetic anti-diarrhea drugs have created an interest in low-toxic alternative plant-derived compounds. FengLiao consists of Polygonum hydropiper Linn. and Daphniphyllum calycinum Bench., and is widely used in China to treat diarrhea due to low levels of toxicity. In this study, the effects of FengLiao were analyzed in a castor oil-induced diarrhea model, using the anti-diarrhea drug, loperamide, as the positive control. The effects were evaluated using stool characteristics and the expression levels of various diarrhea-related factors in the jejunum and liver,
