Kinetic analysis of aggregation data.

Aggregation of repeat-containing proteins is associated with neurodegenerative disorders; a specific example is the established link between expansion of the polyglutamine domain in huntingtin and the appearance of nuclear inclusions in Huntington's disease. This connection between aggregation and pathology has motivated numerous investigations into the kinetics of aggregation. Quantitative analysis of kinetic data is needed both for comparative purposes (e.g., to compare the effect of different compounds on aggregation kinetics) and for mechanistic insight. Here we describe some analytical eq

Modeling Protein Aggregation Kinetics from NMR Data.

We provide an overview of the practical aspects of using NMR spectroscopy to follow the time course of protein fibril formation (aggregation) and quantitatively model the kinetics of aggregation processes. Following a brief survey of the theoretical foundations of the kinetics of protein aggregation and its inhibition, the modeling of aggregation kinetics, from data acquired by a series of fast two-dimensional 1H-15N correlation NMR spectra, is described. Examples are drawn from our recent NMR-based studies of (1) the aggregation kinetics of a pathogenic huntingtin exon-1 protein whose fibrill

Aggregated Indexing of Biomedical Time Series Data.

Remote and wearable medical sensing has the potential to create very large and high dimensional datasets. Medical time series databases must be able to efficiently store, index, and mine these datasets to enable medical professionals to effectively analyze data collected from their patients. Conventional high dimensional indexing methods are a two stage process. First, a superset of the true matches is efficiently extracted from the database. Second, supersets are pruned by comparing each of their objects to the query object and rejecting any objects falling outside a predetermined radius. Thi

Phase-independent wireless data aggregation via optical in-sensor computing.

The data-intensive nature of the Internet of Things (IoT) significantly challenges conventional communication systems. While over-the-air computation integrates communication and computation to reduce data aggregation burdens, it requires strict synchronization of transmitted signals, increasing system overhead. Here, we report an optical in-sensor wireless data aggregation paradigm that exploits photocarrier trapping in defect-dominated persistent photoconductance sensors to reduce dependence on synchronization constraints. Within a single communication window, the system reliably aggregates

Actigraph data are reliable, with functional reliability increasing with aggregation.

Motion sensor devices such as actigraphs are increasingly used in studies that seek to obtain an objective assessment of activity level. They have many advantages, and are useful additions to research in fields such as sleep assessment, drug efficacy, behavior genetics, and obesity. However, questions still remain over the reliability of data collected using actigraphic assessment. We aimed to apply generalizability theory to actigraph data collected on a large, general-population sample in middle childhood, during 8 cognitive tasks across two body loci, and to examine reliability coefficients

The interpretation of intensity and aggregation data for infections of Schistosoma haematobium.

The relationships between mean intensities of infection, the aggregation of infection among hosts, and host age are analysed using data from 2 large (> 3000 individuals) field studies of Schistosoma haematobium infection. The data show a convex relationship between mean intensity and age, a convex relationship between an inverse index of aggregation and age, and an age-dependent relationship between the mean and aggregation of infection intensity when levels of infection are high. These patterns are qualitatively compared with the output of mathematical models based on an immigration-death pro

Individual participant data meta-analyses compared with meta-analyses based on aggregate data.

BackgroundMeta-analyses based on individual participant data (IPD-MAs) allow more powerful and uniformly consistent analyses as well as better characterisation of subgroups and outcomes, compared to those which are based on aggregate data (AD-MAs) extracted from published trial reports. However, IPD-MAs are a larger undertaking requiring greater resources than AD-MAs. Researchers have compared results from IPD-MA against results obtained from AD-MA and reported conflicting findings. We present a methodology review to summarise this empirical evidence .ObjectivesTo review systematically empiric

Molecular Autopsy for Sudden Death in the Young: Is Data Aggregation the Key?

The Scripps molecular autopsy study seeks to incorporate genetic testing into the postmortem examination of cases of sudden death in the young (<45 years old). Here, we describe the results from the first 2 years of the study, which consisted of whole exome sequencing (WES) of a cohort of 50 cases predominantly from San Diego County. Apart from the individual description of cases, we analyzed the data at the cohort-level, which brought new perspectives on the genetic causes of sudden death. We investigated the advantages and disadvantages of using WES compared to a gene panel for cardiac disea

Aggregate data modelling: A fast implementation for fitting pharmacometrics models to summary-level data in R.

Pharmacometric modelling is traditionally performed using individual level data. Recently a new method was developed to fit pharmacometric models to summary level - or aggregate - data. This methodology allows for jointly modelling different data sources, once transformed into aggregate data. As such, the method can be applied to a combination of individual data, pharmacometric models, and aggregate data. In this study we aimed to (1) implement this methodological framework into an accessible R package (admr) and (2) develop a novel algorithm with enhanced computational efficiency. The develop

Familial Aggregation of Endemic Congenital Hypothyroidism Syndrome in Congo (DR): Historical Data.

Familial aggregation of endemic congenital hypothyroidism (CH) in an iodine-deficient population from northern Congo (Democratic Republic (DR)) was analysed on data collected four decades ago (1979-1980). During a systematic survey of 62 families, 46 endemic CH subjects (44 myxedematous and 2 neurological) were identified based on clinical evidence within a village cohort of 468 subjects. A distribution analysis showed that two families presented significant excess of cases versus a random background distribution. Both families were characterised by two healthy parents having all of their five

Data on correlation between Aβ42 structural aggregation propensity and toxicity in bacteria.

Protein aggregation and amyloid formation is a hallmark of an increasing number of human disorders. Because protein aggregation is deleterious for the cell physiology and results in a decrease in overall cell fitness, it is thought that natural selection acts to purify aggregating proteins during evolution. This data article contains complementary figures and results related to the research article entitled "Selection against toxic aggregation-prone protein sequences in bacteria" (Navarro et al., 2014) [1]. Here, we used the AGGRESCAN3D (A3D) server, a novel in house predictor that forecasts p

The Cosmos Collaborative: A Vendor-Facilitated Electronic Health Record Data Aggregation Platform.

OBJECTIVE: Learning healthcare systems use routinely collected data to generate new evidence that informs future practice. While implementing an electronic health record (EHR) system can facilitate this goal for individual institutions, meaningfully aggregating data from multiple institutions can be more empowering. Cosmos is a cross-institution, single EHR vendor-facilitated data aggregation tool. This work aims to describe the initiative and illustrate its potential utility through several use cases. METHODS: Cosmos is designed to scale rapidly by leveraging preexisting agreements, clinical

A systematic review aggregated data and individual participant data meta-analysis of percutaneous endovascular arteriovenous fistula.

OBJECTIVE: To assess the short-term and mid-term safety and efficacy of percutaneous endovascular arteriovenous fistula (pAVF) creation. METHODS: A systematic search was implemented corresponding to the PRISMA 2020 and the PRISMA for individual participant data (IPD) systematic reviews 2015. Aggregated data from the included studies were obtained and meta-analyzed regarding both the overall pAVF efficacy and the comparison of pAVF with surgical AVF (sAVF). We performed a two-stage IPD meta-analysis for studies comparing pAVF and sAVF regarding primary and secondary patency. Primary end points

Improved Scheme for Data Aggregation of Distributed Oracle for Intelligent Internet of Things.

Oracle is a data supply mechanism that provides real-world data for blockchain. It serves as a bridge between blockchain and the IoT world, playing a crucial role in solving problems such as data sharing and device management in the IoT field. The main challenge at this stage is determining how to achieve data privacy protection in distributed Oracle machines to safeguard the value hidden in data on the blockchain. In this paper, we propose an improved scheme for distributed Oracle data aggregation based on Paillier encryption algorithm, which achieves end-to-end data privacy protection from d

Bayesian Random-Effects Meta-Analysis of Aggregate Data on Clinical Events.

To investigate intervention effects on rare events, meta-analysis techniques are commonly applied in order to assess the accumulated evidence. When it comes to adverse effects in clinical trials, these are often most adequately handled using survival methods. A common-effect model that is able to process data in commonly quoted formats in terms of hazard ratios has previously been proposed for this purpose. In order to accommodate potential heterogeneity between studies, we have extended the model by Holzhauer to a random-effects approach. The Bayesian model is described in detail, and applica

Integration of aggregate data in causally interpretable meta-analysis by inverse weighting.

Obtaining causally interpretable meta-analysis results is challenging when there are differences in the distribution of effect modifiers between eligible trials. To overcome this, recent work on transportability methods has considered standardizing results of individual studies over the case-mix of a target population, prior to pooling them as in a classical random-effect meta-analysis. One practical challenge, however, is that case-mix standardization often requires individual participant data (IPD) on outcome, treatments and case-mix characteristics to be fully accessible in every eligible s

An aggregate accident model based on pooled, regional time-series data.

The determinants of personal injury road accidents and their severity are studied by means of generalized Poisson regression models estimated on the basis of combined cross-section/time-series data. Monthly data have been assembled for 18 Norwegian counties (every county but one), covering the period from January 1974 until December 1986. A rather wide range of potential explanatory factors are taken into account, including road use (exposure), weather, daylight, traffic density, road investment and maintenance expenditure, accident reporting routines, vehicle inspection, law enforcement, seat

The Measurement Properties of Aggregated Relational Data and NSUM-Estimated Network Size.

Aggregated relational data (ARD), derived from questions of the form "How many people do you know who [belong to subpopulation X]?" are widely used to estimate the size and composition of social networks, often adopting the network scale-up method (NSUM). However, their measurement properties are insufficiently studied. The authors address this gap by assessing (1) the test-retest reliability of a large set of ARD questions and NSUM-estimated network sizes and (2) the convergent validity of these network size estimates. This mixed-methods study involved a heterogeneous quota sample of 50 citiz
