Phytochemical and biological investigation of the roots of the wild Daucus carota ssp. carota afforded three new and four known compounds, including four sesquiterpenes daucane esters (1-3 [new], and 4), one polyacetylene (5), one sesquiterpene coumarin (6), and sitosterol glucoside. The structures of the new compounds were determined by comprehensive NMR studies, including DEPT, COSY, NOESY, HMQC and HMBC analyses. Based on an agar diffusion assay, 1, 2 and 4-6 were screened and found to contain a range of low antibacterial activities against four gram positive (Staphylococcus aureus, Strepto

Antigens derived from various pathogens can readily be synthesized at high levels in plants in their authentic forms. Such antigens administered orally can induce an immune response and, in some cases, result in protection against a subsequent challenge. We here report the expression of rabies virus G protein into carrots. The G gene was subcloned into the pUCpSSrabG vector and then used to transform carrot embryogenic cells by particle bombardment. The carrot cells were selected in liquid medium, a method previously unreported. The presence of the transgene was verified by PCR, and by RT-PCR.

The effect of carrot extract on carbon tetrachloride (CCl4)-induced acute liver damage was evaluated. The increased serum enzyme levels (viz., glutamate oxaloacetate transaminase, glutamate pyruvate transaminase, lactate dehydrogenase, alkaline phosphatase, sorbitol and glutamate dehydrogenase) by CCl4-induction were significantly lowered due to pretreatment with the extract. The extract also decreased the elevated serum bilirubin and urea content due to CCl4 administration. Increased activities of hepatic 5'-nucleotidase, acid phosphatase, acid ribonuclease and decreased levels of succinic de

The development of virus-free, oral vaccines against poliovirus capable of inducing mucosal protective immunity is needed to safely combat this pathogen. In the present study, a carrot cell line expressing the poliovirus VP2 antigen was established at the level of callus and cell suspensions, exploring the effects of culture media (MS and B5), supplementation with urea, phytoregulators (2,4-D : KIN), and light conditions (continuous light, photoperiod, and total darkness). The best callus growth was obtained on B5 medium supplemented with 2 mg/L of 2,4-D + 2 mg/L kinetin and 0.0136 g/L of urea

A dichloromethane (DCM) extract of carrot roots was found to stimulate insulin-dependent glucose uptake (GU) in adipocytes in a dose dependent manner. Bioassay-guided fractionation of the DCM extract resulted in the isolation of the polyacetylenes falcarinol and falcarindiol. Both polyacetylenes were able to significantly stimulate basal and/or insulin-dependent GU in 3T3-L1 adipocytes and porcine myotube cell cultures in a dose-dependent manner. Falcarindiol increased peroxisome proliferator-activated receptor (PPAR)γ-mediated transactivation significantly at concentrations of 3, 10 and 30 μM

Chiral alcohols are essential intermediates in pharmaceuticals, agrochemicals, and advanced materials. Conventional asymmetric reduction of ketones relies on costly metal catalysts with significant environmental impact. Biocatalysis, particularly whole-cell systems, offers a sustainable alternative, providing high regio- and stereoselectivity under mild conditions. Daucus carota (carrot) roots serve as a promising biocatalyst due to their broad substrate compatibility and natural cofactor recycling ability, reducing reliance on toxic reagents and energy-intensive processes, making them both en

OBJECTIVE: To develop a biphasic system for use in plant tissue-mediated biotransformations to overcome the low water solubilities of substrates and inhibitory effects of products.
RESULTS: Commonly-used organic solvents and ionic liquid were assayed using the biphasic system to reduce water-insoluble benzoyl cyanide by Daucus carota (wild carrot) tissue. Dibutyl phthalate was the most suitable solvent. The optimal moisture content of dibutyl phthalate, the ratio of biomass to substrate, and the concentration of benzoyl cyanide were 50 % (v/v), 100 g mM(-1), and 12 mM l(-1), respectively. Und

Postharvest diseases can cause significant losses in carrot (Daucus carota) yield during storage. In this study, the effects of soil quality and microbial diversity in bulk field soil and the rhizosphere on carrot postharvest storability were investigated. Microbial genera in bulk soil and rhizosphere samples were identified by Illumina MiSeq amplicon sequencing of the bacterial 16S rRNA gene and fungal ITS regions. Disease symptoms in the stored carrots were monitored after 6 months of cold storage, and the main pathogens were identified. Field sites with mineral soils, which had higher pH va

The cultivation of Daucus carota L. (carrot) on soils polluted with trace elements (TE) constitutes an innovative phytomanagement approach, combining significant ecological and economic benefits. This plant species exhibits a notable capacity to extract trace elements from the soil while generating valuable biomass, primarily intended for hydrodistillation to produce high-quality essential oils (EO). Carrots exhibit an enhanced capacity to accumulate cadmium in their leaf tissues, with a high bioconcentration factor (BCF = 1.71), as well as significant zinc accumulation. Furthermore, the cult

Carotenoids are pigments with important nutritional value in the human diet. As antioxidant molecules, they act as scavengers of free radicals enhancing immunity and preventing cancer and cardiovascular diseases. Moreover, α-carotene and β-carotene, the main carotenoids of carrots (Daucus carota) are precursors of vitamin A, whose deficiency in the diet can trigger night blindness and macular degeneration. With the aim of increasing the carotenoid content in fruit flesh, three key genes of the carotenoid pathway, phytoene synthase (DcPSY2) and lycopene cyclase (DcLCYB1) from carrots, and carot

The increasing food waste generated along the food chain should be considered as a source of high-value compounds, with the aim of improving the circularity of productions. In this study, carrot pomace, the major by-product of carrot juice processing, was used as a source of carotenoids. For the valorisation of this by-product, different non-conventional extraction methods of carotenoids such as Ultrasound-Assisted Extraction (UAE) and Pressurised Liquid Extraction (PLE) have been developed. For the latter, the main parameters influencing the extraction have been optimised using a multivariate
