Susceptibility of Chickens to Porcine Deltacoronavirus Infection.

Porcine deltacoronavirus (PDCoV) is a novel swine enteropathogenic coronavirus with worldwide distribution. PDCoV belongs to the Deltacoronavirus (DCoV) genus, which mainly includes avian coronaviruses (CoVs). PDCoV has the potential to infect human and chicken cells in vitro, and also has limited infectivity in calves. However, the origin of PDCoV in pigs, the host range, and cross-species infection of PDCoV still remain unclear. To determine whether PDCoV really has the ability to infect chickens in vivo, the three lines of chicken embryos and specific pathogen free (SPF) chickens were inocu

Computational design of an ultrapotent deltacoronavirus miniprotein inhibitor.

Multiple spillovers of porcine deltacoronavirus (PDCoV) into humans in Haiti highlight its zoonotic potential and the need for targeted interventions. No approved vaccines or therapeutics are available for use in humans against any DCoVs. Here, we report the de novo design of PDCoV miniprotein inhibitors (aka minibinders, MBs) and show that one of them, MB11, binds with picomolar affinity to the PDCoV receptor-binding domain (RBD). MB11 potently inhibits PDCoV, outcompeting monoclonal antibodies, and cross-reacts with and broadly neutralizes a panel of distantly related DCoVs. We determined a

Characterization and evolution of porcine deltacoronavirus in the United States.

Porcine deltacoronavirus (PDCoV) was identified in multiple states across the United States (US) in 2014. In this study, we investigate the presence of PDCoV in diagnostic samples, which were further categorized by case identification (ID), and the association between occurrence, age, specimen and location between March and September 2014. Approximately, 7% of the case IDs submitted from the US were positive for PDCoV. Specimens were categorized into eight groups, and the univariate analysis indicated that oral fluids had 1.89 times higher odds of detecting PDCoV compared to feces. While the 4

CRISPR/Cas13a-based rapid detection method for porcine deltacoronavirus.

BACKGROUND: Porcine deltacoronavirus (PDCoV) is a newly discovered porcine intestinal pathogenic coronavirus with a single-stranded positive-sense RNA genome and an envelope. PDCoV infects pigs of different ages and causes acute diarrhea and vomiting in newborn piglets. In severe cases, infection leads to dehydration, exhaustion, and death in sick piglets, entailing great economic losses on pig farms. The clinical symptoms of PDCoV infection are very similar to those of other porcine enteroviruses. Although it is difficult to distinguish these viral infections without testing, monitoring PDCoV

[Expression and immunogenicity of porcine deltacoronavirus S-trimer protein].

This study aims to express the S-trimer protein of porcine deltacoronavirus (PDCoV) and study its immunogenicity. Targeting the extracellular domain (D20-N1 077) of PDCoV S protein, we introduced the signal peptide sequence (secretoglobin family 1D member 1, SCGB1D1) at the N-terminus and the T4 phage Foldon motif at the C-terminus, and cloned the recombinant sequence into pcDNA3.1(+) vector. The S-trimer fusion protein was expressed in the ExpiCHO expression system and purified, and the molecular weight of the S-trimer fusion protein was analyzed by non-denaturing polyacrylamide gel electroph

Genomic Epidemiology, Evolution, and Transmission Dynamics of Porcine Deltacoronavirus.

The emergence of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) has shown once again that coronavirus (CoV) in animals are potential sources for epidemics in humans. Porcine deltacoronavirus (PDCoV) is an emerging enteropathogen of swine with a worldwide distribution. Here, we implemented and described an approach to analyze the epidemiology of PDCoV following its emergence in the pig population. We performed an integrated analysis of full genome sequence data from 21 newly sequenced viruses, along with comprehensive epidemiological surveillance data collected globally over the l

Outbreak of porcine deltacoronavirus in pigs in central China, 2024.

Porcine deltacoronavirus (PDCoV) is an emerging enteropathogenic coronavirus responsible for acute diarrhea, vomiting, and dehydration in piglets, thereby posing a substantial threat to the global swine industry. In December 2024, a severe diarrheal disease outbreak occurred on an intensive pig farm in Hubei Province, located in central China. Here, we describe the isolation and genomic characterization of a novel PDCoV strain, designated SP2403-2, obtained from the intestinal contents of an affected piglet. The virus was successfully propagated in cell culture and confirmed by immunofluoresce

Porcine deltacoronavirus infection is inhibited by Griffithsin in cell culture.

Porcine deltacoronavirus (PDCoV) is an emerging porcine enteric coronavirus that causes severe diarrhea in piglets and results in serious economic losses. There are no effective vaccines and antiviral drugs to prevent and treat PDCoV infection currently. Griffithsin (GRFT) is a lectin with potent antiviral activity against enveloped viruses because of its ability to specifically bind N-linked high-mannose oligosaccharides. GRFT has been reported to possess antiviral activity against severe acute respiratory syndrome coronavirus (SARS-CoV), Middle East respiratory syndrome coronavirus (MERS-CoV

First report and phylogenetic analysis of porcine deltacoronavirus in Mexico.

Porcine deltacoronavirus has caused great economic losses in the swine industry worldwide. In this study, we carried out the first detection, sequencing and characterization of this virus in Mexico. We analysed 885 rectal samples by multiplex RT-PCR to determine coinfections. In addition, the Spike gene was amplified, sequenced and analysed phylogenetically. We found 85 positive samples for porcine deltacoronavirus, representing 9.6% of the total samples, and we determined that the most frequent coinfection was with porcine epidemic diarrhoea virus (54.1%). Four sequences of Mexican isolates w

Deletion of NS6 attenuates Porcine Deltacoronavirus pathogenicity without compromising immunogenicity.

Porcine deltacoronavirus (PDCoV) is a major enteric pathogen that inflicts lethal diarrhea in sucking piglets. In this study, we constructed an infectious cDNA clone of PDCoV strain JS2021-LX using a bacterial artificial chromosome (BAC) system and generated a recombinant virus (rPDCoV-ΔNS6-EGFP) by replacing NS6 gene with an EGFP reporter via CRISPR/Cas9. In vitro analyses revealed that rPDCoV-ΔNS6-EGFP exhibited attenuated replication kinetics and formed smaller plaque compared to both the parental PDCoV and rescued PDCoV (rPDCoV). Furthermore, we observed that infection with rPDCoV-ΔNS6‑EGF

Identification of a Monoclonal Antibody against Porcine Deltacoronavirus Membrane Protein.

Porcine deltacoronavirus (PDCoV) is an emerging virus that poses a significant threat to the global swine industry. Its membrane (M) protein is crucial for virion assembly and virus-host interactions. We selected the hydrophilic region of M protein for prokaryotic expression, purification, and recombinant protein production. Utilizing hybridoma technology, we prepared the monoclonal antibody (mAb) 24-A6 against M protein. The mAb 24-A6 was shown to be suitable for use in immunofluorescence assays, western blotting, and immunoprecipitation, with specificity for PDCoV and no cross-reactivity wit

Porcine Deltacoronavirus nsp5 Cleaves DCP1A To Decrease Its Antiviral Activity.

Porcine deltacoronavirus (PDCoV) is an emerging swine enteropathogenic coronavirus. The nonstructural protein nsp5, also called 3C-like protease, is responsible for processing viral polyprotein precursors in coronavirus (CoV) replication. Previous studies have shown that PDCoV nsp5 cleaves the NF-κB essential modulator and the signal transducer and activator of transcription 2 to disrupt interferon (IFN) production and signaling, respectively. Whether PDCoV nsp5 also cleaves IFN-stimulated genes (ISGs), IFN-induced antiviral effector molecules, remains unclear. In this study, we screened 14 cl

Deltacoronavirus Modulates circRNA cGLIS3 Metabolism to Evade Host Antiviral Response.

Circular RNAs (circRNAs) are dynamically remodeled during infection, yet how viruses exploit circRNA-RNA binding protein (RBP) circuits remains poorly understood. Here, we report a cGLIS3-IGF2BP2-linked TNF-α regulatory axis triggered by deltacoronavirus infection to inhibit antiviral innate immunity. The host m6A reader insulin-like growth factor 2 mRNA-binding protein 2 (IGF2BP2) and viral nucleocapsid (N) protein promote biogenesis of the m6A-modified cGLIS3 by strengthening the GLIS3 exon 3 circulation. Interaction assay reveals that K-Homology domain of IGF2BP2 or the linker region of vir

Susceptibility of Cell Lines from Different Species to Porcine Deltacoronavirus.

Porcine deltacoronavirus (PDCoV) is an emerging swine enteric coronavirus that causes severe diarrhea in piglets, leading to extensive economic losses in the global swine industry. Previous studies have reported that PDCoV can experimentally infect multiple animal species. However, the replication capability of PDCoV in non-natural host cell lines remains largely unknown. Herein, we investigated the effects of exogenous trypsin on PDCoV replication and systematically evaluated the susceptibility of various cell lines derived from human, murine, and feline to PDCoV. The results showed that high

Porcine Deltacoronavirus Infection Cleaves HDAC2 to Attenuate Its Antiviral Activity.

Protein acetylation plays an important role during virus infection. Thus, it is not surprising that viruses always evolve elaborate mechanisms to regulate the functions of histone deacetylases (HDACs), the essential transcriptional and epigenetic regulators for deacetylation. Porcine deltacoronavirus (PDCoV), an emerging enteropathogenic coronavirus, causes severe diarrhea in suckling piglets and has the potential to infect humans. In this study, we found that PDCoV infection inhibited cellular HDAC activity. By screening the expressions of different HDAC subfamilies after PDCoV infection, we

GRP78 interacts with the Nsp3 of porcine deltacoronavirus and inhibits viral replication.

Porcine deltacoronavirus (PDCoV) is an important intestinal coronavirus, which has caused large economic losses to the global pig industry. Endoplasmic reticulum chaperone protein (GRP78) is involved in viral infection replication and regulates the homeostasis of host cells during pathogen infection. However, the role of GRP78 in PDCoV replication remains unclear. In this study, overexpression or knockdown of GRP78 was used to explore effects on PDCoV replication. The interaction between GRP78 and viral non-structural proteins was determined with co-IP and laser confocal microscopy experiments
