Deltaproteobacteria and Spirochaetes-Like Bacteria Are Abundant Putative Mercury Methylators in Oxygen-Deficient Water and Marine Particles in the Baltic Sea.

Methylmercury (MeHg), a neurotoxic compound biomagnifying in aquatic food webs, can be a threat to human health via fish consumption. However, the composition and distribution of the microbial communities mediating the methylation of mercury (Hg) to MeHg in marine systems remain largely unknown. In order to fill this knowledge gap, we used the Baltic Sea Reference Metagenome (BARM) dataset to study the abundance and distribution of the genes involved in Hg methylation (the hgcAB gene cluster). We determined the relative abundance of the hgcAB genes and their taxonomic identity in 81 brackish m

Associations of gut bacterial classes Clostridia and Deltaproteobacteria with type 2 diabetes and Alzheimer's disease: A two-sample Mendelian randomization study.

Using a 2-sample Mendelian randomization (MR) approach, we evaluated MR evidence for genetically proxied effects of 10 pre-specified gut bacterial classes on type 2 diabetes mellitus (T2DM) and Alzheimer disease (AD), and assessed MR evidence for a direct genetically proxied effect of T2DM on AD. Mediation by T2DM was explored where feasible. We performed 2-sample MR using MiBioGen gut microbiota GWAS summary statistics as exposure, and DIAMANTE (T2DM) and European AD GWAS data as outcomes. Inverse-variance weighted (IVW) MR was the primary analysis, with MR-Egger, weighted median, and weighte

Mercury methylation and hydrogen sulfide production among unexpected strains isolated from periphyton of two macrophytes of the Amazon.

The periphyton of macrophytes had previously been identified as important spots for mercury methylation in the Amazon basin, but the microorganisms that facilitate methylation in such compartment are still to be identified. Here, bacteria were isolated from periphyton associated with Eichhornia crassipes and Polygonum densiflorum in Widdel and Pfennig medium and tested for mercury methylation with a stable isotope tracer technique using (198)HgCl, hydrogen sulfide production and molybdate inhibition. Three Pleomorphomona spp., one unidentified Deltaproteobacteria, two Klebsiella spp., and one

Two Marine Desulfotomaculum spp. of Different Origin are Capable of Utilizing Acetone and Higher Ketones.

Degradation of acetone and higher ketones has been described in detail for aerobic and nitrate-reducing bacteria. Among sulfate-reducing bacteria, degradation of acetone and other ketones is still an uncommon ability and has not been understood completely yet. In the present work, we show that Desulfotomaculum arcticum and Desulfotomaculum geothermicum are able to degrade acetone and butanone. Total proteomics of cell-free extracts of both organisms indicated an involvement of a thiamine diphosphate-dependent enzyme, a B12-dependent mutase, and a specific dehydrogenase during acetone degradati

Performance of Anaerobic Biotrickling Filter and its Microbial Diversity for the Removal of Stripped Disinfection Byproducts.

The objective of this research was to evaluate the biodegradation of chloroform by using biotrickling filter (BTF) and determining the dominant bacteria responsible for the degradation. The research was conducted in three phases under anaerobic condition, namely, in the presence of co-metabolite (Phase I), in the presence of co-metabolite and surfactant (Phase II) and in the presence of surfactant but no co-metabolite (Phase III). The results showed that the presence of ethanol as a co-metabolite provided 49% removal efficiency. The equivalent elimination capacity (EC) was 0.13 g/(m3.hr). The

Bidirectional Mendelian randomization links gut microbiota to primary biliary cholangitis.

Primary biliary cholangitis (PBC) and gut microbiota (GM) are epidemiologically correlated but the causal inter-relationships remain poorly understood. We aim to explore the causal relationships between GM and PBC. Using the MiBioGen consortium, GWAS data for GM at the species level and the largest publicly available PBC GWAS data to date, we performed a bidirectional two-sample Mendelian randomization by the inverse variance weighted, MR-Egger, weighted median, weighted model and MR-PRESSO to elucidate the potential causal role of GM in PBC. To measure the heterogeneity of instrumental variab

Salmonella produces sulfide to compete with Escherichia coli in the gut lumen.

Hydrogen sulfide production is a characteristic that distinguishes Salmonella serovars from closely related species, such as Escherichia coli, but its biological significance remains obscure. Here, we show that PhsABC and AsrABC-mediated hydrogen sulfide production by Salmonella enterica serovar (S.) Typhimurium was linked to an increased abundance of Deltaproteobacteria and inhibition of cytochrome bd oxidase-mediated aerobic respiration of Escherichia coli in the murine large intestine. Functional phsABC and asrABC operons provided a growth benefit to S. Typhimurium in the cecum of Enterobac

Response of mercury methylation to algal bloom decomposition or elevated CO2 in surface sediments from the East China Sea.

Methylmercury (MeHg), a neurotoxin bioaccumulated in aquatic food webs, poses risks to ecosystems and human health, with marine products mainly contributing to global human MeHg exposure. The East China Sea (ECS), influenced by anthropogenic Hg inputs and environmental changes like algal boom and elevated CO2, is critical for understanding MeHg dynamics. This study investigated how algal decomposition and elevated CO2 (1000 ppm) affect Hg methylation in the ECS sediments. Surface sediments showed average MeHg concentrations of 0.291 ± 0.069 ng g-1 and a MeHg/THg ratio of 0.67 %. Input of algal

Magnetotaxis in an anaerobic ciliate via tripartite syntrophy.

Magnetotaxis has evolved independently numerous times in bacteria, whereby genetically controlled biomineralization of nano-crystalline magnets results in swimming along Earth's magnetic field lines. Compared to magnetotactic bacteria (MTB), evolutionary mechanisms of magnetotaxis as a trait in eukaryotes remain poorly understood. Here, we report a magnetotactic ciliate, Tropidoatractus magnetotacticus sp. nov., that acquires magnetotaxis via syntrophy. T. magnetotacticus exhibits magnetotaxis due to the magnetic moment of internal ferrimagnetic magnetite (Fe3O4) nanoparticles forming ellipsoi

Biomolecular and isotopic evidence reveals key cellular controls on methylmercury fate during bacterial mercury methylation.

Microbial mercury (Hg) methylation drives the formation of methylmercury (MeHg) hotspots in natural environments, but the cellular pathways that determine MeHg fate and isotopic signatures remain largely unresolved. In this study, we cultured the model sulfate-reducing bacterium Pseudodesulfovibrio hydrargyri BerOc1 anaerobically under fumarate respiration for 30 h and quantified Hg speciation and fraction- and species-specific Hg isotopic compositions. The results showed that intracellular MeHg was rapidly exported and dominated the extracellular pool by 30 h (> 80%). Exported MeHg bound to s

Visualization of Syntrophic Benzene-Fermenting Desulfobacterota ORM2 in a Methanogenic Enrichment Culture Using Fluorescence In Situ Hybridization.

Benzene degradation under anoxic conditions was first reported more than 25 years ago; however, the activation mechanism in the absence of oxygen remains elusive. Progress has been hindered by the difficulty in cultivating anaerobic benzene-degrading enrichment cultures. Our laboratory has sustained a methanogenic enrichment culture harboring Desulfobacterota ORM2, a benzene fermenter distinct from any known genus but related to other known or predicted benzene degraders. ORM2's slow doubling time (∼30 days) and extended lag phase after inoculation complicate its study. We developed a fluoresc

Isolation and cultivation of a novel sulfate-reducing magnetotactic bacterium belonging to the genus Desulfovibrio.

Magnetotactic bacteria (MTB) synthesize magnetosomes composed of membrane-enveloped magnetite (Fe3O4) and/or greigite (Fe3S4) nanoparticles in the cells. It is known that the magnetotactic Deltaproteobacteria are ubiquitous and inhabit worldwide in the sediments of freshwater and marine environments. Mostly known MTB belonging to the Deltaproteobacteria are dissimilatory sulfate-reducing bacteria that biomineralize bullet-shaped magnetite nanoparticles, but only a few axenic cultures have been obtained so far. Here, we report the isolation, cultivation and characterization of a dissimilatory s

Simultaneous determination of mercury methylation and demethylation capacities of various sulfate-reducing bacteria using species-specific isotopic tracers.

The use of species-specific isotopic tracers for inorganic and methyl mercury has allowed the simultaneous determination of the methylation and demethylation potentials of pure culture of isolated sulfate-reducing (SR) bacterial strains using low Hg species concentration levels (7 µg/L (199)Hg(II), 1 µg/L Me(201)Hg). A major advantage of the method reported here is that it can be used to follow simultaneously both the degradation of the species added but also the formation of their degradation products and thus the determination during the same incubation of the specific methylation/demethylat

Diverse Communities of hgcAB+ Microorganisms Methylate Mercury in Freshwater Sediments Subjected to Experimental Sulfate Loading.

Methylmercury (MeHg) is a bioaccumulative neurotoxin produced by certain sulfate-reducing bacteria and other anaerobic microorganisms. Because microorganisms differ in their capacity to methylate mercury, the abundance and distribution of methylating populations may determine MeHg production in the environment. We compared rates of MeHg production and the distribution of hgcAB genes in epilimnetic sediments from a freshwater lake that were experimentally amended with sulfate levels from 7 to 300 mg L-1. The most abundant hgcAB sequences were associated with clades of Methanomicrobia, sulfate-r

Mercury methylation by dissimilatory iron-reducing bacteria.

The Hg-methylating ability of dissimilatory iron-reducing bacteria in the genera Geobacter, Desulfuromonas, and Shewanella was examined. All of the Geobacter and Desulfuromonas strains tested methylated mercury while reducing Fe(III), nitrate, or fumarate. In contrast, none of the Shewanella strains produced methylmercury at higher levels than abiotic controls under similar culture conditions. Geobacter and Desulfuromonas are closely related to known Hg-methylating sulfate-reducing bacteria within the Deltaproteobacteria.

Genomic analysis of Desulfobulbaceae strain B35, a new hydrothermal vent species of mesophilic bacterium that disproportionates sulfur and respires Fe(III).

Desulfobulbaceae sp. nov. strain B35 is a novel mesophilic, anaerobic, chemolithoautotrophic bacterium isolated from the Lucky Strike deep-sea hydrothermal vent on the Mid-Atlantic Ridge, growing autotrophically by disproportionation of elemental sulfur (S0) and thiosulfate (S₂O₃2-), as well as by respiration of Fe(III) using H₂ as an electron donor. Its genome, assembled into 6 contigs totaling 4,140,770 base pairs, has a G + C content of 60.95% and a completeness of 99.4%. It encodes complete metabolic pathways, including the Wood-Ljungdahl pathway for CO₂ fixation, the tricarboxylic acid cy

Syntrophic pathways for microbial mercury methylation.

Exposure to dietary sources of methylmercury (MeHg) is the focus of public health concerns with environmental mercury (Hg) contamination. MeHg is formed in anoxic environments by anaerobic microorganisms. This process has been studied mostly with single-species culture incubations, although the relevance of such studies to Hg(II)-methylation in situ is limited because microbial activities in the environment are critically modulated by interactions among microbial functional groups. Here we describe experiments in which Hg(II)-methylation was examined within the context of various microbial syn

Gut bacterial microbiota in patients with myasthenia gravis: results from the MYBIOM study.

BACKGROUND: Myasthenia gravis (MG) is an autoimmune neuromuscular disease, with gut microbiota considered to be a pathogenetic factor. Previous pilot studies have found differences in the gut microbiota of patients with MG and healthy individuals. To determine whether gut microbiota has a pathogenetic role in MG, we compared the gut microbiota of patients with MG with that of patients with non-inflammatory and inflammatory neurological disorders of the peripheral nervous system (primary endpoint) and healthy volunteers (secondary endpoint). METHODS: Faecal samples were collected from patients

Determining the Reliability of Measuring Mercury Cycling Gene Abundance with Correlations with Mercury and Methylmercury Concentrations.

Methylmercury (MeHg) is a bioaccumulative toxic contaminant in many ecosystems, but factors governing its production are poorly understood. Recent work has shown that the anaerobic microbial conversion of mercury (Hg) to MeHg requires the Hg-methylation genes hgcAB and that these genes can be used as biomarkers in PCR-based estimators of Hg-methylator abundance. In an effort to determine reliable methods for assessing hgcA abundance and diversity and linking them to MeHg concentrations, multiple approaches were compared including metagenomic shotgun sequencing, 16S rRNA gene pyrosequencing and

Metabolic trade-offs in Desulfosporosus meridiei: dose-dependent FeS nanoparticles modulate extracellular electron transfer and gene expression for mineral transformation.

Biogenic ferrous sulfide nanoparticles (FeS NPs) regulate sulfate (SO42-)-reducing bacteria (SRB)-driven iron/sulfur cycling in SO42--rich anaerobic environments, yet their dose-dependent impacts on SRB metabolism remain unclear. This study revealed how FeS NPs dose modulates Desulfosporosus meridiei (a model SRB) in reducing schwertmannite (Sch). SRB preferentially reduced Fe(III) over SO42- in Sch via FeS NPs-mediated extracellular electron transfer (EET). At low FeS doses (0-6 mM), the dsr gene expression (sulfur metabolism) associated with mineral transformation increased despite a decline
