Infectious hypodermal and hematopoietic necrosis virus (IHHNV) is a widely distributed and highly pathogenic virus affecting shrimp. Helice tientsinensis, a key benthic crab commonly found in shrimp ponds in northern China, has unknown susceptibility to IHHNV and a potential role in viral transmission. In this study, healthy H. tientsinensis individuals were experimentally infected with IHHNV through non-invasive oral way to mimic natural exposure pathways in shrimp aquaculture systems. By day 15 post-infection, some individuals developed small black lesions on the carapace, which mostly resol

Infectious hypodermal and hematopoietic necrosis virus (IHHNV) is classified as a reportable crustacean disease by the World Organisation for Animal Health (WOAH), which causes poor growth in Penaeus vannamei. According to genome sequence alignment analysis, enzymatic recombinase amplification (ERA) primers and probe were designed based on the ORF1 region of IHHNV, and a real-time ERA assay for IHHNV detection (IHHNV-ERA) was established. The experimental results show that IHHNV-F2/IHHNV-R2 and IHHNV-Probe can effectively amplify the target gene, and the sensitivity is 1.4 × 101 copies/μL with

Densovirus is classified as invertebrate virus belonging to the subfamily Densovirinae of Parvoviridae family. This group of viruses infects only insects and several densoviruses have been isolated from indigenous mosquitoes and mosquito cell lines. A number of mosquitoes, especially Aedes aegypti and Ae. albopictus are important vectors of viruses, which are the major causes of dengue hemorrhagic fever and yellow fever in humans. As densoviruses do not cause any pathology in humans, these viruses have been proposed to be a potential vector for use in biological control of mosquitoes and insec

BACKGROUND: Dengue virus (DENV) is a major public health threat, with Aedes albopictus being the confirmed vector responsible for dengue epidemics in Guangzhou, China. Mosquito densoviruses (MDVs) are pathogenic mosquito-specific viruses, and a novel MDV was previously isolated from Ae. albopictus in Guangzhou. This study aims to determine the prevalence of MDVs in wild Ae. albopictus populations and investigate their potential interactions with DENV and impact on vector susceptibility for DENV.
METHODS: The prevalence of MDV in wild mosquitoes in China was investigated using open access sequ

Loop-mediated isothermal amplification (LAMP) assay is a novel method of gene amplification with high specificity, sensitivity and rapidity, which can be applied for disease diagnosis in shrimp aquaculture. The method is performed under isothermal conditions with a set of four specially designed primers that recognize six distinct sequences of the target. In the present study, according to the conservative regions of non-structural protein gene NS1, a set of four specific primers were designed, and a rapid detection of IHHNV was established by LAMP assay. Bombyx mori densovirus (BmDNV-1), on the basis of the previously reported genome sequence, constitutes by itself a separate genus (Iteravirus) within the Densovirinae subfamily of parvoviruses. Inconsistencies in the genome organization, however, necessitated its reassessment. The genome sequence of new clones was determined and resulted in a completely different genome organization. The corrected sequence also contained conserved sequence motifs found in other parvoviruses. Some amino acids in the highly conserved domain in the unique region of VP1 were shared by critical amino acids in the c

Ecytonucleospora (=Enterocytozoon) hepatopenaei (EHP) is an intracellular parasitic microsporidian, Infectious hypodermal and haematopoietic necrosis virus (IHHNV) is a linear single-stranded DNA virus. Both of them are widespread pathogens in shrimp farming, causing slow growth in shrimp and leading to significant economic losses. Litopenaeus vannamei (L. vannamei), the dominant species in shrimp farming, can be co-infected by EHP and IHHNV. According to our prior research, EHP and IHHNV exhibit synergistic interactions during infection. However, the underlying mechanisms of their co-infectio

Infectious hypodermal and haematopoietic necrosis virus (IHHNV) is the smallest known virus in shrimp, which causes runt-deformity syndrome (RDS) and leads to huge economic loss every year in penaeid shrimp farming. Previous studies have shown that the juvenile Penaeus vannamei is more susceptible to IHHNV infection than the adults, but the mechanism is still unclear. In order to investigate the mechanism of pathogenic differences in IHHNV infection of P. vannamei at different developmental stages, the juvenile and adult P. vannamei were studied by transcriptome high-throughput sequencing to a

White spot syndrome virus (WSSV) and Infectious hypodermal and hematopoietic necrosis virus (IHHNV) are two infectious agents associated to economic losses in shrimp aquaculture. As virus spread occurs through vectors and hosts, this study sought to verify the presence of WSSV and IHHNV in Neohelice granulata crab from Lagoa dos Patos estuary in Brazil and nearby shrimp farms. DNA extractions were performed with phenol/chloroform protocol. Molecular diagnosis was carried out by nested PCR for WSSV and one-step PCR for IHHNV. Results showed the presence of WSSV on crabs of both Lagoa dos Patos

Densovirus (DNV) is a small single-stranded DNA, non-enveloped virus belonging to the subfamily Densovirinae of the Parvoviridae family. This group of invertebrate viruses infects exclusively insects. Two of the major densoviruses, Aedes aegypti (AaeDNV) and Ae. albopictus (AalDNV), infect mosquitoes that carry viruses responsible for two important public health diseases, namely, dengue hemorrhagic fever and yellow fever. The present study describes cloning, sequencing and phylogenetic analysis of a new densovirus, AalDNV-4, from infected Ae. albopictus C6/36 cell line. The total nucleotide se

Some insects use endogenous reverse transcriptase (RT) to make variable viral copy DNA (vcDNA) fragments from viral RNA in linear (lvcDNA) and circular (cvcDNA) forms. The latter form is easy to extract selectively. The vcDNA produces small interfering RNA (siRNA) variants that inhibit viral replication via the RNA interference (RNAi) pathway. The vcDNA is also autonomously inserted into the host genome as endogenous viral elements (EVE) that can also result in RNAi. We hypothesized that similar mechanisms occurred in shrimp. We used the insect methods to extract circular viral copy DNA (cvcDN

The infectious hypodermal and haematopoietic necrosis virus (IHHNV) represents a significant viral threat to global shrimp aquaculture, leading to considerable economic losses. In this study, we have developed a one-step, one-pot isothermal assay for the detection of IHHNV, employing recombinase-aided amplification in conjunction with clustered regularly interspaced short palindromic repeats-Cas12a (RAA-CRISPR/Cas12a). The assay is performed at a constant temperature of 37°C, achieving a detection limit of 10 copies per reaction for the fluorescence assay and 1 copy per reaction for the latera

Bats are recognized as key reservoirs for diverse viruses, including members of the Parvoviridae family. Vietnam hosts an exceptionally wide variety of bat species, yet the diversity of parvoviruses in these populations remains largely unexplored. This study provides the first survey of parvoviruses in bats from Gia Lai and Dong Nai provinces, screening 150 samples from 26 bat species and detecting parvoviruses in 38 (25%). A total of 45 viral operational taxonomic units (OTUs) were identified, encompassing four parvoviral subfamilies: Parvovirinae, Densovirinae, Penbrevirinae, and Hamavirinae

The mealworm (Tenebrio molitor) is one of the most commonly mass-reared insects for food and feed. Monitoring the health status of commercially reared mealworm populations is of great importance for the early detection of entomopathogens and for preventing pathogen outbreaks. Metagenomic screening is a suitable and commonly used method for detecting entomopathogens. The approach used here previously enabled the discovery of the Tenebrio molitor densovirus (TmDV) (family Parvoviridae, subfamily Densovirinae) in symptomatic larvae. In the present study, the search for TmDV was extended to larvae

Infectious hypodermal and hematopoietic necrosis virus (IHHNV) is widespread in cultured Penaeus monodon and P. vannamei in Thailand. It causes runt-deformity syndrome that is characterized by physical abnormalities and stunted growth in P. vannamei, but causes no apparent disease in P. monodon. In both species, the virus may produce Cowdry Type A inclusions in tissues of ectodermal and mesodermal origin, but these are common in P. vannamei and rare in P. monodon. The virus can be more easily detected in both species by IHHNV-specific PCR primers. By in situ hybridization (ISH) using specific

An outbreak of fever in a village in southern India was reported on 1st September, 2001. The first reported case presented with epistaxis and a platelet count of 27000h(1)/mm3. Clinical, laboratory and entomological evidence supported a diagnosis of dengue fever. One third of the village was affected and 3.7 % of the population presented with haemorrhagic symptoms; none were fatal. Five acute cases tested for dengue specific IgM showed that two were positive. The larvae of Aedes aegypti were discovered from domestic water collections in the village. Spatial analysis done with the help of Geogr

BACKGROUND: Plant based repellent against mosquito borne diseases are used recently because synthetic repellents cause side effects like breathing problem, eye irritation, head ache, cough, etc. The use of natural products for dengue control would protect the environment, reduce dependence on expensive synthetic repellents and also generate local employment.
MATERIAL AND METHODS: Essential oil was isolated by steam distillation which was used against the bites of Aedes aegypti and duration of protection period was assessed. Skin-irritant potential test was also conducted on 25 healthy volunte
