Two Marine Desulfotomaculum spp. of Different Origin are Capable of Utilizing Acetone and Higher Ketones.

Degradation of acetone and higher ketones has been described in detail for aerobic and nitrate-reducing bacteria. Among sulfate-reducing bacteria, degradation of acetone and other ketones is still an uncommon ability and has not been understood completely yet. In the present work, we show that Desulfotomaculum arcticum and Desulfotomaculum geothermicum are able to degrade acetone and butanone. Total proteomics of cell-free extracts of both organisms indicated an involvement of a thiamine diphosphate-dependent enzyme, a B12-dependent mutase, and a specific dehydrogenase during acetone degradati

A Novel View on the Taxonomy of Sulfate-Reducing Bacterium 'Desulfotomaculum salinum' and a Description of a New Species Desulfofundulus salinus sp. nov.

Two thermophilic spore-forming sulfate-reducing strains, 435T and 781, were isolated from oil and gas reservoirs in Western Siberia (Russia) about 50 years ago. Both strains were found to be neutrophilic, chemoorganotrophic, anaerobic bacteria, growing at 45-70 °C (optimum, 55-60 °C) and with 0-4.5% (w/v) NaCl (optimum, 0.5-1% NaCl). The major fatty acids were iso-C15:0, iso-C17:0, C16:0, and C18:0. In sulfate-reducing conditions, the strains utilized H2/CO2, formate, lactate, pyruvate, malate, fumarate, succinate, methanol, ethanol, propanol, butanol, butyrate, valerate, and palmitate. In 200

Isolation of sulfate-reducing bacteria from the terrestrial deep subsurface and description of Desulfovibrio cavernae sp. nov.

Deep subsurface sandstones in the area of Berlin (Germany) located 600 to 1060 m below the surface were examined for the presence of viable microorganisms. The in situ temperatures at the sampling sites ranged from 37 to 45 degrees C. Investigations focussed on sulfate-reducing bacteria able to grow on methanol and triethylene glycol, which are added as chemicals to facilitate the long-term underground storage of natural gas. Seven strains were isolated from porewater brines in the porous sandstone. Three of them were obtained with methanol (strains H1M, H3M, and B1M), three strains with triet

[Effects of different vegetation restoration patterns on soil bacterial community in coastal wetlands following Spartina alterniflora removal].

Vegetation restoration following Spartina alterniflora removal is critical for restoring ecological functions in coastal wetlands. However, how different restoration patterns influence soil bacterial communities remains insufficiently understood. In this study, we investigated restored wetlands in the Minjiang River Estuary, southeastern China, including salt marshes dominated by Phragmites australis and Cyperus malaccensis, and Kandelia obovata mangroves of two stand ages (6- and 16-year-old), with unrestored wetlands as control. We used high-throughput sequencing to characterize soil bacteri

Microbial reduction of sulfur dioxide with pretreated sewage sludge and elemental hydrogen as electron donors.

It has been demonstrated tht heat- and alkali-pretreated-sewage sludge may serve as an electron donor and carbon source for SO2 reduction by Desulfovibrio desulfuricans. A continuous D. desulfuricans culture was operated for 6 mo with complete reduction of SO2 to H2S. The culture required only minor amounts of mineral nutrients in addition to pretreated sewage sludge. It has also been shown that the sulfate-reducing bacterium Desulfotomaculum orientis can be grown on H2 as an energy source, CO2 as a carbon source, and SO2 as a terminal electron acceptor. Complete reduction of SO2 to H2S was ob

[Clinical significance of sulfate-reducing bacteria for ulcerative colitis].

Ulcerative colitis(UC) is colon localized disease. Broad epithelial cell damage, crypt abscesses and accumulation of neutrophils are recognized for UC. Although the cause of UC is indistinct at this time, there is a growing consensus that abnormal intestinal microflora would be related with UC. There have been several evidences that excessive production of hydrogen sulfide by bacteria in colon would be associated with UC. Sulfate reducing bacteria are able to utilize sulfate as an electron receptor for dissimilation of organic substrate and hydrogen gas, resulting in generating toxic hydrogen

Anaerobic oxidation of methane coupled to sulfate reduction: Consortium characteristics and application in co-removal of H2S and methane.

Anaerobic sludge from a sewage treatment plant was used to acclimatize microbial colonies capable of anaerobic oxidation of methane (AOM) coupled to sulfate reduction. Clone libraries and fluorescence in situ hybridization were used to investigate the microbial population. Sulfate-reducing bacteria (SRB) (e.g., Desulfotomaculum arcticum and Desulfobulbus propionicus) and anaerobic methanotrophic archaea (ANME) (e.g., Methanosaeta sp. and Methanolinea sp.) coexisted in the enrichment. The archaeal and bacterial cells were randomly or evenly distributed throughout the consortia. Accompanied by s

Control of bacterial spores.

Bacterial spores are much more resistant than their vegetative counterparts. The most dangerous spore-former is Clostridium botulinum which produces a potent neurotoxin that can prove fatal. The most common food poisoning from a spore-former is caused by C. perfringens. Other food poisoning spore-formers include Bacillus cereus, B. subtilis and B. licheniformis. There are a number of non-pathogenic spore-formers including butyric and thermophilic anaerobes that cause significant economic losses to food producers. Some unusual spoilage complaints have been reported, for example, B. sporothermod

A conceptual method to simultaneously inhibit methane and hydrogen sulfide production in sewers: The carbon metabolic pathway and microbial community shift.

In this study, the impact of COD/SO42- ratio in sewage on methane and hydrogen sulfide production in sewer biofilms was investigated by using three identical lab-scale gravity sewer systems. The results showed that the COD/SO42- played a key role in the competition between methanogenic archaea (MA) and sulfate reducing bacteria (SRB). Both the lowest methane and hydrogen sulfide production were obtained at COD/SO42- ratio of 6. The carbon transformation revealed that the activity of both MA and SRB was inhibited at this COD/SO42- ratio. Methanosarcina and Methanobacterium were the two dominant

Selective conversion of carbon monoxide to hydrogen by anaerobic mixed culture.

A new method for the conversion of CO to H2 was developed by anaerobic mixed culture in the current study. Higher CO consumption rate was obtained by anaerobic granular sludge (AGS) compared to waste activated sludge (WAS) at 55 °C and pH 7.5. However, H2 was the intermediate and CH4 was the final product. Fermentation at pH 5.5 by AGS inhibited CH4 production, while the lower CO consumption rate (50% of that at pH 7.5) and the production of acetate were found. Fermentation at pH 7.5 with the addition of chloroform achieved efficient and selective conversion of CO to H2. Stable and efficient H

[Sulfate-Reducing Bacterial Communities in the Water Column of the Gdansk Deep (Baltic Sea)].

Biodiversity of sulfate-reducing bacterial communities in the water column of the Gdansk Deep, Baltic Sea, where H2S had been detected in near-bottom layers, was analyzed by PCR with primers for the 16S rRNA genes of six major phylogenetic subgroups of sulfate-reducing bacteria (SRB). Using denaturing gradient gel electrophoresis followed by sequencing, the nucleotide sequences of reamplified dsrB gene fragments from investigated water samples were determined. For the first time the presence of nucleotide sequences of the dsrB gene was detected by PCR in the water samples from all hydrochemica

Tolerance of a sulfidogenic sludge to trichloroethylene at microcosms level as a basis for a long-term operation of reactors designed for its biodegradation.

Trichloroethylene (TCE) is known as a toxic organic compound found as a pollutant in water streams around the world. The ultimate goal of the present work was to determine the TCE concentration that would be feasible to biodegrade on a long-term basis by a sulfidogenic sludge while maintaining sulfate reducing activity (SRA). Microcosms were prepared with sulfidogenic sludge obtained from a stabilized sulfidogenic UASB and amended with different TCE concentrations (100-300 µM) and two different proportions of volatile fatty acids (VFA) acetate, propionate and butyrate at COD of 2.5:1:1 and 1:1

Microbial community mediates hydroxyl radical production in soil slurries by iron redox transformation.

The generation of reactive oxygen species (ROS) mediated by minerals and/or microorganisms plays a vital but underappreciated role in affecting carbon and nutrient cycles at soil-water interfaces. It is currently unknown which interactions between microbial communities and iron (Fe) minerals produce hydroxyl radical (HO•), which is the strongest oxidant among ROS. Using a series of well-controlled anoxic incubations of soil slurries, we demonstrated that interactions between microbial communities and Fe minerals synergistically drove HO• production (up to ∼100 nM after 21-day incubation). Reductive dechlorination of tetrachloroethene to cis-1, 2-dichloroethene by a thermophilic anaerobic enrichment culture.

Thermophilic anaerobic biodegradation of tetrachloroethene (PCE) was investigated with various inocula from geothermal and nongeothermal areas. Only polluted harbor sediment resulted in a stable enrichment culture that converted PCE via trichloroethene to cis-1, 2-dichloroethene at the optimum temperature of 60 to 65 degrees C. After several transfers, methanogens were eliminated from the culture. Dechlorination was supported by lactate, pyruvate, fructose, fumarate, and malate as electron donor but not by H2, formate, or acetate. Fumarate and L-malate led to the highest dechlorination rate. High genetic and physiological diversity of sulfate-reducing bacteria isolated from an oligotrophic lake sediment.

The community structure of sulfate-reducing bacteria in littoral and profundal sediments of the oligotrophic Lake Stechlin (Germany) was investigated. A collection of 32 strains was isolated from the highest positive dilutions of most-probable-number series, and their partial 16S rRNA gene sequences and genomic fingerprints based on ERIC (enterobacterial repetitive intergenic consensus)-PCR were analyzed. The strains fell into eight distinct phylogenetic lineages, and the majority (70%) showed a close affiliation to the genus Desulfovibrio. Most of the remaining strains (22%) were related to t

Effect of sulfate addition on carbon flow and microbial community composition during thermophilic digestion of cellulose.

Substrates with high sulfate levels pose problems for biogas production as they allow sulfate reducing bacteria to compete with syntrophic and methanogenic members of the community. In addition, the end product of sulfate reduction, hydrogen sulfide, is toxic and corrosive. Here we show how sulfate addition affects physiological processes in a thermophilic methanogenic system by analyzing the carbon flow and the microbial community with quantitative PCR and amplicon sequencing of the 16s rRNA gene. A sulfate addition of 0.5 to 3 g/L caused a decline in methane production by 73-92%, while highe

Anaerobic phenanthrene biodegradation by a newly isolated sulfate-reducer, strain PheS1, and exploration of the biotransformation pathway.

Phenanthrene is a widespread and harmful polycyclic aromatic hydrocarbon that is difficult to anaerobically biodegrade. Current challenges in anaerobic phenanthrene bioremediation are a lack of degrading cultures and limited knowledge of biotransformation pathways. Under sulfate-reducing conditions, pure-cultures and biotransformation processes for anaerobic phenanthrene biodegradation are poorly understood. In this study, strain PheS1, which is phylogenetically closely related to Desulfotomaculum, was found to be a sulfate-reducing phenanthrene-degrading bacterium. Anaerobic phenanthrene biod

Effect of Thermophilic Nitrate Reduction on Sulfide Production in High Temperature Oil Reservoir Samples.

Oil fields can experience souring, the reduction of sulfate to sulfide by sulfate-reducing microorganisms. At the Terra Nova oil field near Canada's east coast, with a reservoir temperature of 95°C, souring was indicated by increased hydrogen sulfide in produced waters (PW). Microbial community analysis by 16S rRNA gene sequencing showed the hyperthermophilic sulfate-reducing archaeon Archaeoglobus in Terra Nova PWs. Growth enrichments in sulfate-containing media at 55-70°C with lactate or volatile fatty acids yielded the thermophilic sulfate-reducing bacterium (SRB) Desulfotomaculum. Enrichme

Seismic shifts in the geochemical and microbial composition of a Yellowstone aquifer.

Seismic energy, like that released by earthquakes, can fracture rock and thereby alter subsurface fluid flow paths, release substrates from inclusions, and expose fresh mineral surfaces capable of reacting with water. However, it is unclear how such seismic-induced changes influence microbial communities. Volcanically active areas experience frequent seismic activity and thus represent ideal locations to examine the influence of seismic-induced geochemical change on subsurface microbial communities. Here, we demonstrate that energy released in an earthquake swarm in 2021 correlated with extens

Analysis of biofilms formed by static culture of bacterial communities in the Inba floodway (Lake Inbanuma-Tokyo Bay).

Microbiologically influenced corrosion (MIC) is a major threat to marine infrastructure, yet the bacterial communities on metal surfaces before corrosion remain poorly understood. In this study, environmental water collected across a salinity gradient (freshwater-brackish-seawater) in the Inba floodway (Lake Inbanuma to Tokyo Bay, Japan) was treated with the extremely low-nutrient R2A medium, after which stainless steel was immersed in the treated water. After 1 week of static incubation, biofilms were formed on stainless steel immersed in the water samples. The early-stage biofilm-forming bac
