Expanding the diversity of Desulfovibrio from the human gut: Desulfovibrio intestinivivus sp. nov., Desulfovibrio desulfopostgatei sp. nov. and Desulfovibrio faecalis sp. nov.

Three strictly anaerobic, Gram-negative, curved rod-shaped bacteria, designated strains QI0430T, QI0434T and QI0442T, were isolated from faecal samples of healthy adult donors in the UK. Phylogenomic analysis based on 31 single-copy marker genes showed that all three isolates formed distinct lineages. Whole-genome average nucleotide identity and digital DNA-DNA hybridization values between each isolate and its nearest phylogenetic neighbours were <95.6 and <63%, respectively, supporting their status as separate species. Genome sizes ranged from 3.41 to 3.59 Mb, with DNA G+C contents of 56.91-5

Bacteremia caused by Desulfovibrio fairfieldensis.

Desulfovibrio species are anaerobic gram-negative, pleomorphic bacilli rarely causing infection in humans. In the present report, we describe a case of bacteremia caused by Desulfovibrio fairfieldensis. The patient, for whom biapenem was administered, rapidly improved without any sequelae. As far as we know, this is the first case report of infection by Desulfovibrio species in Japan.

Desulfovibrio in the Gut: The Enemy within?

Desulfovibrio (DSV) are sulfate-reducing bacteria (SRB) that are ubiquitously present in the environment and as resident commensal bacteria within the human gastrointestinal tract. Though they are minor residents of the healthy gut, DSV are opportunistic pathobionts that may overgrow in the setting of various intestinal and extra-intestinal diseases. An increasing number of studies have demonstrated a positive correlation between DSV overgrowth (bloom) and various human diseases. While the relationship between DSV bloom and disease pathology has not been clearly established, mounting evidence

Comparative proteomics reveals hydrogenase-centered steel-dependent growth and corrosion in Desulfovibrio vulgaris and Desulfovibrio ferrophilus.

Comparative proteomic analyses including ecologically distinct sulfate-reducing strains and different cellular fractions are scarce, limiting our understanding of corrosion-related mechanisms under anoxic conditions and the identification of protein signatures relevant to microbiologically influenced corrosion (MIC). To capture both freshwater and marine adaptations, we investigated the well-established SRB model strains Desulfovibrio vulgaris and Desulfovibrio ferrophilus. We compared energy metabolism, electron transport pathways, and growth patterns in both strains cultivated with lactate,

Desulfovibrio Bacteria Are Associated With Parkinson's Disease.

Parkinson's disease (PD) is the most prevalent movement disorder known and predominantly affects the elderly. It is a progressive neurodegenerative disease wherein α-synuclein, a neuronal protein, aggregates to form toxic structures in nerve cells. The cause of Parkinson's disease (PD) remains unknown. Intestinal dysfunction and changes in the gut microbiota, common symptoms of PD, are evidently linked to the pathogenesis of PD. Although a multitude of studies have investigated microbial etiologies of PD, the microbial role in disease progression remains unclear. Here, we show that Gram-negati

The first case of bacteremia caused by Desulfovibrio legallii.

We report the first documented case of bacteremia caused by Desulfovibrio legallii. A 55-year-old man with multiple system atrophy was admitted to our hospital for rehabilitation. During hospitalization, he developed a fever and rigors. Blood cultures were obtained, and the anaerobic blood culture yielded spiral-shaped, Gram-negative rods. The isolated strain could not be identified by matrix-assisted laser desorption/ionization time-of-flight mass spectrometry but was subsequently identified as D. legallii by 16S rRNA gene sequencing analysis.

Desulfovibrio species are potentially important in regressive autism.

Autism is a complex disorder with no specific diagnostic test so the disease is defined by its characteristics including cognitive defects, social, communication and behavioral problems, repetitive behaviors, unusual sensitivity to stimuli such as noise, restricted interests, and self stimulation. The incidence of this disease has increased remarkably in recent years and was 110/10,000 children (∼1%) in multiple areas of the US in 2007. The financial burden on families and communities is enormous. In terms of predisposing factors, heredity plays a role in some subjects, but it is clear that en

Bile Acid Sequestration Attenuates Desulfovibrio-Induced Hepatic Injury.

Desulfovibrio (DSV), sulfate-reducing gut bacteria that generate hydrogen sulfide (H2S), can impact host health through diverse mechanisms including bile acid (BA) metabolism. Although intestinal overgrowth of DSV expands the BA pool and promotes liver injury, its causal role in hepatic pathophysiology remains incompletely defined. Here, by employing complementary interventions of cholic acid (CA) supplementation and the BA sequestrant cholestyramine in mouse models, we show that DSV-driven liver injury is mediated by aberrant BA metabolism coupled with gut microbial remodeling. CA alone induc

Cobalamin-mediated mercury methylation by Desulfovibrio desulfuricans LS.

The prominence of sulfate reducers in mercury biomethylation prompted the examination of the methyl carrier and mercury methylation activity of Desulfovibrio desulfuricans LS. There was a low degree of mercury tolerance and a high degree of methylation during fermentative growth; the opposite was true during sulfate reduction. During 2 days of fermentative growth, up to 37% of HgCl2 was methylated at 0.1 micrograms/ml, but only 1.5% was methylated at 10.0 micrograms/ml. Less than 1% of the added HgCl2 was methylated under sulfate-reducing conditions. D. desulfuricans LS radioimmunoassay result

Experimental evolution reveals nitrate tolerance mechanisms in Desulfovibrio vulgaris.

Elevated nitrate in the environment inhibits sulfate reduction by important microorganisms of sulfate-reducing bacteria (SRB). Several SRB may respire nitrate to survive under elevated nitrate, but how SRB that lack nitrate reductase survive to elevated nitrate remains elusive. To understand nitrate adaptation mechanisms, we evolved 12 populations of a model SRB (i.e., Desulfovibrio vulgaris Hildenborough, DvH) under elevated NaNO3 for 1000 generations, analyzed growth and acquired mutations, and linked their genotypes with phenotypes. Nitrate-evolved (EN) populations significantly (p < 0.05)

Mercury Uptake by Desulfovibrio desulfuricans ND132: Passive or Active?

Recent studies have identified HgcAB proteins as being responsible for mercury [Hg(II)] methylation by certain anaerobic microorganisms. However, it remains controversial whether microbes take up Hg(II) passively or actively. Here, we examine the dynamics of concurrent Hg(II) adsorption, uptake, and methylation by both viable and inactivated cells (heat-killed or starved) or spheroplasts of the sulfate-reducing bacterium Desulfovibrio desulfuricans ND132 in laboratory incubations. We show that, without addition of thiols, >60% of the added Hg(II) (25 nM) was taken up passively in 48 h by live

Desulfovibrio desulfuricans bacteremia: A case report and literature review.

Desulfovibrio spp. are sulfate-reducing, anaerobic bacteria that are ubiquitously found in the environment. These organisms infrequently cause human infections, and the clinical characteristics of infection with Desulfovibrio spp. remain unclear. Here, we describe a case of Desulfovibrio desulfuricans bacteremia in an 88-year-old Japanese man with a past medical history of thoracic endovascular aortic repair (TEVAR). His chief complaint was hemoptysis for 2 weeks. A chest contrast-enhanced computed tomography demonstrated an enlarged thoracic aortic aneurysm surrounded by a ring-enhanced lesio

[Description of a case of abdominal sepsis due to Desulfovibrio desulfuricans].

Desulfovibrio spp. are strict anaerobes that are ubiquitous in nature. They can reside in the human or animal gastrointestinal tract and, as they are also environmental bacteria, may be present in soil and water. They can persist asymptomatically in the intestine or behave as opportunistic pathogens associated with primary bacteremia and intraabdominal infections. Several Desulfovibrio spp. infections may be underestimated due to their slow growth rate and because many laboratories do not routinely perform anaerobic cultures. Simple tests such as motility detection on a fresh subculture, Gram

[A case of Desulfovibrio desulfuricans cultured from blood in Japan].

We report a case of Desulfovibrio desulfuricans bacteremia in a 60-year-old-man. In our case, anaerobic blood culture bottle turned out positive after five days' incubation. Gram stain showed the presence of slightly-curved Gram negative rod. Suspecting Campylobacter and Helicobacter, we added microaerobic culture while tentatively reporting Campylobacter to the physician. We then added anaerobic culturing with Brucella HK (RS) Agar because microaerobic culture proved the absence of microaerophile. We found small colonies on the third day, then we started anaerobic culture and eventually ident

First isolation of Desulfovibrio from the human vaginal flora.

Four Desulfovibrio species, including 2 subtypes of 1 species, namely, Desulfovibrio piger, Desulfovibrio desulfuricans MB subtype and Essex 6 subtype, Desulfovibrio fairfieldensis, and Desulfovibrio vulgaris, have been isolated from the human oral and intestinal flora, but not previously from the vaginal flora. They are opportunistic pathogens and have been considered as possible environmental and etiologic agents involved in ulcerative colitis and chronic periodontitis. We isolated Desulfovibrio intestinalis from vaginal specimens of four Japanese women; a species which has not been previous

Bacteremia caused by a recently described novel Desulfovibrio species.

An obligately anaerobic, fastidious, slowly growing, spiral, gram-negative bacterium was isolated from the blood of a 75-year-old man with acute onset of pyrexia. The patient responded rapidly to appropriate antibiotic therapy. Extensive investigation failed to detect a focus for the infection. Phenotypically, the organism was consistent with Desulfovibrio species. Microscopic investigation revealed an organism with a vibrioid or spirillioid morphology with rapidly progressive motility by means of a single polar flagellum. Biochemically, the organism produced large amounts of H2S and contained

Enzymatic catalysis of mercury methylation by Desulfovibrio desulfuricans LS.

The recently defined role of methylcobalamin in Hg2+ methylation by Desulfovibrio desulfuricans LS enabled us to reexamine the question of whether the principal source of methylmercury is spontaneous transmethylation or an enzymatically catalyzed process. In cell extracts of D. desulfuricans LS, over 95% of the 57Co label was associated with macromolecules rather than with free cobalamin. Both gel filtration and electrophoresis of cell extracts identified a single corrinoid protein of 40 kDa in size. This finding, in combination with the previously reported light-reversible propyl iodide inhib

Characterization of periplasmic hydrogenase from Desulfovibrio vulgaris Miyazaki K.

Periplasmic hydrogenase [hydrogen:ferricytochrome c3 oxidoreductase, EC 1.12.2.1] from Desulfovibrio vulgaris Miyazaki K (MK) was purified to homogeneity. Its chemical and immunological properties were examined and compared with those of other Desulfovibrio hydrogenases. The pure enzyme showed a specific activity of 1,000 mumol H2 evolution min-1 (mg protein)-1. The enzyme had a molecular weight of 50,000 as estimated by gel filtration and consisted of a single polypeptide chain. The absorption spectrum of the enzyme was characteristic of an iron-sulfur protein and the extinction coefficients
