Proliferative enteropathy involving Lawsonia intracellularis infection in rabbits (Oryctlagus cuniculus).

Five rabbits suffering from diarrhea were diagnosed with proliferative enteropathy (PE). Histopathology revealed a thickened mucosa consisting of hyperplastic intestinal epithelium and infiltration of inflammatory cells mainly consisted of macrophages. In the affected epithelial cytoplasm, numerous curved bacillus-like organisms were observed in the Warthin-Starry silver stain and electron microscopy observation. In polymerase chain reactions, Lawsonia intracellularis-specific DNA fragment were amplified from affected ileal tissue extracted DNA in each case and present 5 cases were confirmed t

The Lawsonia Paradox: The Hidden Burden of Silent Disease.

Lawsonia intracellularis, the etiologic agent of porcine proliferative enteropathy (PPE), represents a growing challenge for modern swine production systems. The infection often occurs in a subclinical form and, together with limited awareness of its true production impact, may delay diagnosis and the timely adoption of control measures. In our opinion, this under-recognition of subclinical infection represents the greatest barrier to successful PPE control. Disease management is further complicated by frequent co-infections, limitations of currently available diagnostic tools, and the progres

Babeisa duncani infection alters gut microbiota profile in hamsters.

The genus Babesia includes parasites that can induce human and animal babesiosis, which are common in tropical and subtropical regions of the world. The gut microbiota has not been examined in hamsters infected by Babesia duncani. Red blood cells infected with B. duncani were injected into hamsters through intraperitoneal route. To evaluate the changes in gut microbiota, DNAs were extracted from small intestinal contents, acquired from hamsters during disease development. Then, the V4 region of the 16S rRNA gene of bacteria was sequenced using the Illumina sequencing platform. Gut microbiota a

Isolation, Passage, and Pathogenicity of a Newly Isolated Lawsonia intracellularis Strain From Hubei, China.

Lawsonia intracellularis is the causative agent of porcine proliferative enteropathy (PPE)-a disease of great economic impact to the global pig industry, but the isolation and continuous passage culture of this bacterial species is very difficult which limits the development of inactivated or live vaccines. While China is the largest pig rearing country in the world, only one study has reported the isolation of L. intracellularis. In this study, we examined 1574 ileal samples collected from 10 slaughterhouses in Hubei Province, China, and obtained 104 samples tested positive for L. intracellul

Cytologic diagnosis of Lawsonia intracellularis proliferative ileitis in a Japanese snow macaque (Macaca fuscata).

Diffuse ileal thickening and ileocecocolic lymphadenomegaly were observed during exploratory laparotomy in a 2-year-old male Japanese snow macaque (Macaca fuscata) that had flu-like signs and diarrhea. Cytologic examination of ileal biopsy imprints revealed many mature, mildly karyolytic neutrophils and fewer well-differentiated lymphocytes, eosinophils, macrophages, and plasma cells in a background containing amorphous, necrotic material. Tightly cohesive sheets of moderately pleomorphic epithelial cells also were seen. The cytologic diagnosis was chronic, active, mixed inflammation with atyp

Hydrogen as an energy source for the human pathogen Bilophila wadsworthia.

The gram-negative anaerobic gut bacterium Bilophila wadsworthia is the third most common isolate in perforated and gangrenous appendicitis, being also found in a variety of other infections. This organism performs a unique kind of anaerobic respiration in which taurine, a major organic solute in mammals, is used as a source of sulphite that serves as terminal acceptor for the electron transport chain. We show here that molecular hydrogen, one of the major products of fermentative bacteria in the colon, is an excellent growth substrate for B. wadsworthia. We have quantified the enzymatic activi

Characterisation of autophagy disruption in the ileum of pigs infected with Lawsonia intracellularis.

Lawsonia intracellularis is the aetiological agent of proliferative enteropathy, an enteric disease endemic in swine. Survival in its intracellular niche of the ileum epithelial lining requires the capacity to subvert, repress or exploit the host immune response to create an environment conducive to bacterial propagation. To better understand how L. intracellularis survives in its intracellular niche, we have performed an investigation into the dynamic relationship between infection and the host autophagy response by immunohistochemistry in experimentally infected porcine ileum samples.Beclin1

Pathogenesis of proliferative enteropathy caused by Lawsoniaintracellularis.

Proliferative enteropathy is a common infectious disease affecting pigs, horses and other animals; it is caused by Lawsonia intracellularis, an obligately intracellular bacterium. Lawsonia infection causes a distinctive pathological change, characterized by a rugose thickening of the intestinal mucosa, due to the adenomatous proliferation of immature intestinal crypt epithelial cells, with local depletion of goblet and enteroendocrine cells. The disease has a low infectious dose; transmission occurs readily between young animals via oral contact with infected faecal material derived from other

Evaluation of immune efficacy of Omp2 protein against Lawsonia intracellularis in mice.

Porcine proliferative enteropathy (PPE) is caused by the obligate intracellular bacterium Lawsonia intracellularis. Infection results in an enteric disease characterised by decreased growth performance of pigs, and presents a major economic burden for swine industries worldwide. Since vaccination is an effective technique for controlling PPE, novel effective vaccine platforms are need to be developed. In this study, five proteins of L. intracellularis were screened through animal experiments and the highly immunoprotective Omp2 protein was identified. Then, the immune efficacy of Omp2 was furt

Evaluation of swine enteroids as in vitro models for Lawsonia intracellularis infection1,2.

The enteric pathogen Lawsonia intracellularis is one of the main causes of diarrhea and compromised weight gain in pigs worldwide. Traditional cell-line cultures have been used to study L. intracellularis pathogenesis. However, these systems fail to reproduce the epithelial changes observed in the intestines of L. intracellularis-infected pigs, specifically, the changes in intestinal cell constitution and gene expression. A more physiologically accurate and state-of-the-art model is provided by swine enteroids derived from stem cell-containing crypts from healthy pigs. The objective of this st

Lawsonia intracellularis infection in the large intestines of pigs.

In this study we examined the proliferative enteropathy, caused by the obligate intracellular bacterium Lawsonia intracellularis, in colon of naturally infected pigs, using immunohistochemistry, in situ hybridisation and scanning confocal laser microscopy. When 396 pigs submitted for routine laboratory examination were investigated, large intestinal gross lesions were seen in 93, including 74 cases of L. intracellularis colitis (proliferative enteropathy). Fifty-one pigs without recorded colonic gross lesions revealed L. intracellularis colitis microscopically. In four cases, L. intracellulari

Equine Proliferative Enteropathy in Weanling Foals on A German Breeding Farm: Clinical Course, Treatment and Long-Term Outcome.

The goal of the current report was to describe the clinical signs, therapy and outcome of foals with suspected equine proliferative enteropathy (EPE) due to an infection with Lawsonia intracellularis. Forty foals, born on the same breeding farm, were diagnosed with suspected clinical EPE between September 2019 and January 2020. Data of these cases were analyzed retrospectively regarding the course of the disease, treatment, outcome and long-term prognosis. All horses, including randomly selected control horses, were reassessed about nine months after the suspicion of EPE. The horses affected w

Preparation and characterization of polyclonal and monoclonal antibodies against Lawsonia intracellularis.

Proliferative enteropathy is an intestinal infectious disease caused by the obligate intracellular bacterium Lawsonia intracellularis. Immunohistochemistry staining has superior sensitivity over hematoxylin and eosin and silver staining for detecting L. intracellularis in histological sections. A L. intracellularis-specific monoclonal antibody (MAb) produced in the UK (IG4 MAb) has been described in the literature. However, no monoclonal or polyclonal antibodies are commercially available. Therefore, the objective of this study was to produce and characterize new polyclonal and monoclonal anti

Lawsonia intracellularis associated equine proliferative enteropathy in Danish weanling foals.

BACKGROUND: Lawsonia intracellularis, an obligate intracellular bacterium, causes equine proliferative enteropathy, mainly in horses around weaning. This disease is rarely reported in the Scandinavian countries. RESULTS: Five cases of equine proliferative enteropathy were diagnosed between 2008-2016 at the University of Copenhagen Large Animal Teaching Hospital. Cases were Danish Warmbloods and a Friesian horse, aged 6-7 months, presenting with typical clinical signs of lethargy, poor body condition, pyrexia and diarrhea. Clinical pathology was consistent with previous reports of severe hypoa

Desulfovibrio desulfuricans bacteremia in an immunocompromised host with a liver graft and ulcerative colitis.

Desulfovibrio spp. are anaerobic, sulfate-reducing, nonfermenting, Gram-negative bacteria found in the digestive tract of humans. Identification of these species with conventional methods is difficult. The reported case of a Desulfovibrio desulfuricans bacteremia occurring in an immunocompromised host with ulcerative colitis confirms that this organism may be a possible opportunistic human pathogen.

Uncommon isolation of Desulfovibrio vulgaris from a depressed fracture wound on the forehead.

Desulfovibrio spp. are gram negative, obligate anaerobes capable of reducing sulfate. They have caused infections in humans, but very rarely. They are slow growers and difficult to identify. Hence, they are often overlooked and their actual presence goes unnoticed. Here, we describe a case of a 15- year old boy who was involved in a road traffic accident and he presented with seropurulent discharge from a depressed fracture wound on the forehead. Desulfovibrio vulgaris (D.vulgaris), was isolated from the pus discharge, the first to be reported. The characteristic desulfoviridin pigment product

The first case of bacteremia caused by Desulfovibrio legallii.

We report the first documented case of bacteremia caused by Desulfovibrio legallii. A 55-year-old man with multiple system atrophy was admitted to our hospital for rehabilitation. During hospitalization, he developed a fever and rigors. Blood cultures were obtained, and the anaerobic blood culture yielded spiral-shaped, Gram-negative rods. The isolated strain could not be identified by matrix-assisted laser desorption/ionization time-of-flight mass spectrometry but was subsequently identified as D. legallii by 16S rRNA gene sequencing analysis.

Desulfovibrio desulfuricans bacteremia: A case report and literature review.

Desulfovibrio spp. are sulfate-reducing, anaerobic bacteria that are ubiquitously found in the environment. These organisms infrequently cause human infections, and the clinical characteristics of infection with Desulfovibrio spp. remain unclear. Here, we describe a case of Desulfovibrio desulfuricans bacteremia in an 88-year-old Japanese man with a past medical history of thoracic endovascular aortic repair (TEVAR). His chief complaint was hemoptysis for 2 weeks. A chest contrast-enhanced computed tomography demonstrated an enlarged thoracic aortic aneurysm surrounded by a ring-enhanced lesio

Lawsonia intracellularis infected enterocytes lack sucrase-isomaltase which contributes to reduced pig digestive capacity.

Lawsonia intracellularis is endemic to swine herds worldwide, however much is still unknown regarding its impact on intestinal function. Thus, this study aimed to characterize the impact of L. intracellularis on digestive function, and how vaccination mitigates these impacts. Thirty-six L. intracellularis negative barrows were assigned to treatment groups (n  =  12/trt): (1) nonvaccinated, L. intracellularis negative (NC); (2) nonvaccinated, L intracellularis challenged (PC); and (3) L. intracellularis challenged, vaccinated (Enterisol® Ileitis, Boehringer Ingelheim) 7 weeks pre-challenge (VAC

Lawsonia intracellularis and Porcine Circovirus type-2 infection in Estonia.

The present study describes the reasons of post-weaning distress in Estonian pig herds. Here we examined the natural cases of Lawsonia intracellularis and porcine circovirus 2 (PCV2) infection and co-infections. The presence of L. intracellularis in swine herds were tested by PCR and by histopathological methods, whereas PCV2 was detected by real-time-PCR and immunohistochemical stainings. Seven of the 11 investigated herds with signs of post-weaning wasting were infected with L. intracellularis and all 11 herds with PCV2. From the analysed samples 22.2% were infected with L. intracellularis a
