Fetal Origins of Mental Health: The Developmental Origins of Health and Disease Hypothesis.

The quality of fetal growth and development predicts the risk for a range of noncommunicable, chronic illnesses. These observations form the basis of the "developmental origins of health and disease" hypothesis, which suggests that the intrauterine signals that compromise fetal growth also act to "program" tissue differentiation in a manner that predisposes to later illness. Fetal growth also predicts the risk for later psychopathology. These findings parallel studies showing that antenatal maternal emotional well-being likewise predicts the risk for later psychopathology. Taken together, thes

Developmental origins of health and disease: environmental exposures.

The developmental origins of health and disease (DOHaD) approach has evolved over the past 20 years, and the current hypothesis proposes that fetal adaptations to intrauterine and maternal conditions during development shape structure and function of organs. Here we present a review of some environmental exposures that may trigger fetal maladaptations in these processes, including three examples: exposures to tobacco smoke, antidepressant medication, and folic acid deficits in the food supply. We provide a selected review of current research on the effects of each of these exposures on fetal d

COVID-19 and developmental origins of health and disease.

From the moment of the identification of SARS-CoV-2 as an etiological agent of the severe clinical pictures of pneumonia that were being slowly observed all over the world, numerous studies have been conducted to increase the knowledge about what was an unknown virus until then. The efforts were mainly aimed to acquire epidemiological, microbiological, pathogenetic, clinical, diagnostic, therapeutic and preventive information in order to increase the available weapons to fight an infection which was rapidly taking on the characteristics of the pandemic. Given the topicality of the problem, not

Tryptophan Metabolism in Developmental Origins of Health and Disease.

Tryptophan, an essential amino acid in mammals that is obtained from the diet, has impacts on early life and development. This amino acid is being studied under the Developmental Origins of Health and Disease (DOHaD) concept, which has led to findings of factors from conception to early childhood that affect health and susceptibility to disease. Tryptophan is metabolized mainly through 2 pathways, serotonin (5-HT) and kynurenine. The kynurenine pathway, active in the brain, gut, liver, and placenta, breaks down over 95% of tryptophan and plays roles in inflammation, neurotransmission, immune r

Epigenetic responses and the developmental origins of health and disease.

Maternal and paternal factors influence offspring development and program its genome for successful postnatal life. Based on the stressors during gestation, the pregnant female prepares the fetus for the outside environment. This preparation is achieved by changing the epigenome of the fetus and is referred to as 'developmental programming'. For instance, nutritional insufficiency in utero will lead to programming events that prepare the fetus to cope up with nutrient scarcity following birth; however, offspring may not face nutrient scarcity following birth. This discrepancy between predicted

Environmental monitoring and the developmental origins of health and disease.

Early-life chronic exposure to environmental contaminants, such as bisphenol-A, particulate matter air pollution, organophosphorus pesticides, and pharmaceutical drugs, among others, may affect central tissues, such as the hypothalamus, and peripheral tissues, such as the endocrine pancreas, causing inflammation and apoptosis with severe implications to the metabolism. The Developmental Origins of Health and Disease (DOHaD) concept articulates events in developmental phases of life, such as intrauterine, lactation, and adolescence, to later-life metabolism and health. These developmental phase

Expanding the Developmental Origins of Health and Disease beyond maternal behaviors.

Developmental Origins of Health and Disease (DOHaD) research has long emphasized maternal pregnancy behaviors as key determinants of child health. New evidence recently published in PLOS Medicine suggests the greatest opportunities to improve child health may lie in tackling socioeconomic disadvantage rather than maternal behavior alone.

Programming Bugs: Microbiota and the Developmental Origins of Brain Health and Disease.

It has been nearly 30 years since Dr. David Barker first highlighted the importance of prenatal factors in contributing to the developmental origins of adult disease. This concept was later broadened to include postnatal events. It is clear that the interaction between genetic predisposition and early life environmental exposures is key in this regard. However, recent research has also identified another important factor in the microbiota-the trillions of microorganisms that inhabit key body niches, including the vagina and gastrointestinal tract. Because the composition of these maternal micr

[Developmental origins of health and disease in adults: role of maternal environment].

Many epidemiological studies indicate that environmental conditions during embryonic and fetal development can have an impact on health at adulthood. Animal studies clearly demonstrate that maternal, and even paternal undernutrition or nutritional excess durably modify some epigenetic marks in their offspring, affecting gene expression and physiological adaptations to the environment. It is crucial to better define the effects of early environment on adult phenotype and epigenetic marks in humans and to develop, with the help of animal models, new preventive strategies and treatments.

Diabetes in pregnancy and lung health in offspring: developmental origins of respiratory disease.

Diabetes is an increasingly common complication of pregnancy. In parallel with this trend, a rise in chronic lung disease in children has been observed in recent decades. While several adverse health outcomes associated with exposure to diabetes in utero have been documented in epidemiological and experimental studies, few have examined the impact of diabetes in pregnancy on offspring lung health and respiratory disease. We provide a comprehensive overview of current literature on this topic, finding suggestive evidence that exposure to diabetes in utero may have adverse effects on lung develo

Perinatal biomarkers implying 'Developmental Origins of Health and Disease' consequences in intrauterine growth restriction.

The intrauterine-growth-restricted (IUGR) state, particularly the asymmetric one, has been associated with 'Developmental Origins of Health and Disease' (DOHaD) consequences later in life. Several environmental factors, acting during the phase of foetal developmental plasticity interact with genotypic variation, 'programme' tissue function and change the capacity of the organism to cope with its environment. They may be responsible for chronic illness risk in adulthood. Detection of possible future DOHaD consequences at a very early age, by applying relevant biomarkers, is of utmost importance

Developmental Origins of Health and Disease (DOHaD): Implications for health and nutritional issues among rural children in China.

In China, with fast economic growth, health and nutrition status among the rural population has shown significant improvement in the past decades. On the other hand, burden of non-communicable diseases and prevalence of related risk factors such as overweight and obesity has also increased. Among rural children, the double burden of malnutrition and emerging overweight and obesity has been neglected so far. According to the theory of Developmental Origin of Health and Diseases (DOHaD), malnutrition, including both undernutrition (stunting and wasting) and over-nutrition (overweight and obesity

Uncertainty in Developmental Origins of Health and Disease research: commentary on Nomura et al. (2023).

Nomura et al. (Journal of Child Psychology and Psychiatry, 2023) reported that children whose mothers were pregnant when Superstorm Sandy struck the Eastern Seaboard of the United States had elevated risk of psychopathology. Their study leverages data from a unique cohort of children established prior to Sandy that enabled researchers to investigate children's mental health depending on their prenatal exposure to the storm. Their findings add to mounting evidence that various types of prenatal stressors instigate stress responses that are transmitted to the developing fetus and impart enduring

Developmental Origins of Health and Disease: The History of the Barker Hypothesis and Assisted Reproductive Technology.

Since Barker's publication of "The fetal and infant origins of adult disease" in 1990, significant emphasis has been placed on the intrauterine environment and its effect on adult disease. Historical events such as the Dutch Famine and the 1918 Flu Pandemic have provided organic data about the epigenetic changes that can result from famine, infection, and stress. Mechanisms that allow for intrauterine survival may predispose to adult disease states when the fetus enters a world of abundance. As the field of in vitro fertilization (IVF) has developed and evolved, little attention has been paid

How can the Developmental Origins of Health and Disease (DOHaD) hypothesis contribute to improving health in developing countries?

The relevance of nutrition during pregnancy and early infancy in defining short-term health and survival has been well established. However, the Developmental Origins of Health and Disease (DOHaD) paradigm provides a framework to assess the effect of early nutrition and growth on long-term health. This body of literature shows that early nutrition has significant consequences on later health and well-being. In this article, we briefly present the main consequences of malnutrition that affect human growth and development and consider how the DOHaD paradigm, with its evolutionary implications, m

Developmental Origins of Health and Disease: The role of Clostridioides difficile Colonization of Gut Microbiota During Infancy.

The DOHaD theory was originally proposed to explain the link between fetal development and risk for non-communicable disease - NCD. With advancements in human microbiome research, this theory has been extended to consider postnatal development of the infant gut microbiome and how this contributes to NCD trajectories. C. difficile transiently colonizes the gut microbiota of up to 50% of infants without causing symptoms, but has been associated with disease in the longterm. We reviewed and summarized the current literature to identify prenatal, birth and postnatal factors contributing to C. diff

Developmental origins of adult health and disease: The metabolic role of BDNF from early life to adulthood.

Accumulating evidence suggests that the origins of adult disease may occur during fetal life. Thus, the concept of "developmental programming" has been introduced and supported by epidemiological and experimental data. This concept supports the idea that the nutritional and hormonal status during pregnancy could interfere in metabolism control. The mechanisms responsible for this "developmental programming" remain poorly documented. Current research indicates that neurotrophins and particularly brain-derived neurotrophic factor (BDNF) may play a crucial role in this process. Although mainly ex

Response to uncertainty in developmental origins of health and disease research: commentary on Nomura et al. (2022).

We appreciate the comments of Gilman et al. (2023) on our paper and their acknowledgement of its importance in highlighting the significance of this area of research. Further, their acknowledgment that the primary results of our study are in a range that is similar to those from other published studies of children exposed to highly stressful environmental events emphasizes the validity of our findings and the important extension of our results to children experiencing these events in utero. They, however, raised concerns about some of the results regarding specific types of psychiatric disorde

Developmental origins of health and adult disease: what should neonatologists/paediatricians be considering about the long-term health of their patients?

The developmental origins of health and disease hypothesis is now strongly supported by both animal and human evidence, and as a consequence, obstetricians, neonatologists and paediatricians need to consider the impact that the in utero and early post-natal environment can have on later renal, cardiovascular and metabolic health. Four common clinical scenarios were provided along with animal and human evidence identifying long-term health implications. Suggestions as to how we should translate this growing body of evidence into practice are provided.
