Marine Bacterial Dextranases: Fundamentals and Applications.

Dextran, a renewable hydrophilic polysaccharide, is nontoxic, highly stable but intrinsically biodegradable. The α-1, 6 glycosidic bonds in dextran are attacked by dextranase (E.C. 3.2.1.11) which is an inducible enzyme. Dextranase finds many applications such as, in sugar industry, in the production of human plasma substitutes, and for the treatment and prevention of dental plaque. Currently, dextranases are obtained from terrestrial fungi which have longer duration for production but not very tolerant to environmental conditions and have safety concerns. Marine bacteria have been proposed as

Purification and characterization of a dextranase from Sporothrix schenckii.

A dextranase (EC 3.2.1.11) was purified and characterized from the IP-29 strain of Sporothrix schenckii, a dimorphic pathogenic fungus. Growing cells secreted the enzyme into a standard culture medium (20 degrees C) that supports the mycelial phase. Soluble bacterial dextrans substituted for glucose as substrate with a small decrease in cellular yield but a tenfold increase in the production of dextranase. This enzyme is a monomeric protein with a molecular mass of 79 kDa, a pH optimum of 5.0, and an action pattern against a soluble 170-kDa bacterial dextran that leads to a final mixture of gl

Inhibition of rat dental caries by dextranase from a strain of Spicaria violacea.

Dextranase AD17 obtained from a culture liquor of a strain of Spicaria violacea was assessed for its ability to inhibit the development of dental caries in conventional Sprague-Dawley rats which had been infected with one of the Streptococcus mutans strains. MT6R (serotype c), OMZ 176R (d), or MT-703R (e). These experiments showed that caries was significantly inhibited when rats were given cariogenic diets No. 2000 and drinking water containing AD17 at a concentration of 10 units/g, as compared to control rats not given dextranase. The inhibitory effects of AD17 were more prominent in smooth

Biochemical properties of a Flavobacterium johnsoniae dextranase and its biotechnological potential for Streptococcus mutans biofilm degradation.

Cariogenic biofilms have a matrix rich in exopolysaccharides (EPS), mutans and dextrans, that contribute to caries development. Although several physical and chemical treatments can be employed to remove oral biofilms, those are only partly efficient and use of biofilm-degrading enzymes represents an exciting opportunity to improve the performance of oral hygiene products. In the present study, a member of a glycosyl hydrolase family 66 from Flavobacterium johnsoniae (FjGH66) was heterologously expressed and biochemically characterized. The recombinant FjGH66 showed a hydrolytic activity again

Effect of mutanase and dextranase on biofilms of cariogenic bacteria: A systematic review of in vitro studies.

Matrix-degrading enzymes are promising non-biocidal adjuncts to dental biofilm control and caries prevention. By disrupting the biofilm matrix structure, enzymes may prevent biofilm formation or disperse established biofilms without compromising the microbial homeostasis in the mouth. This study reviewed whether treatment with mutanase and/or dextranase inhibits cariogenic biofilm growth and/or removes cariogenic biofilms in vitro. An electronic search was conducted in PubMed, EMBASE, Scopus, Web of Science, Cochrane, and LIVIVO databases. Manual searches were performed to identify additional

In Vivo Digestibility of Carbohydrate Rich in Isomaltomegalosaccharide Produced from Starch by Dextrin Dextranase.

Slowly digestible carbohydrates are needed for nutritional support in diabetic patients with malnutrition. They are a good source of energy and have the advantage that their consumption produces a low postprandial peak in blood glucose levels because they are slowly and completely digested in the small intestine. A high-amount isomaltomegalosaccharide containing carbohydrate (H-IMS), made from starch by dextrin dextranase, is a mixture of glucose polymers which has a continuous linear structure of α-1,6-glucosidic bonds and a small number of α-1,4-glucosidic bonds at the reducing ends. It has

Expression, purification and characterization of a cold-adapted dextranase from marine bacteria and its ability to remove dental plaque.

Dental plaque is a high-incidence health concern, and it is caused by Streptococcus mutans. Dextranase can specifically hydrolyze ɑ-1,6-glycosidic linkages in dextran. It is commonly used in the sugar industry, in the production of plasma substitutes, and the treatment and prevention of dental plaque. In this research work, we successfully cloned and expressed a cold-adapted dextranase from marine bacteria Catenovulum sp. DP03 in Escherichia coli. The recombinant dextranase named Cadex2870 contained a 2511 bp intact open reading frame and encoded 836 amino acids. The expression condition of re

Destruction of single-species biofilms of Escherichia coli or Klebsiella pneumoniae subsp. pneumoniae by dextranase, lactoferrin, and lysozyme.

The aim of this work was to determine the destructive activity of dextranase, lactoferrin, and lysozyme, against single species biofilms composed of either Klebsiella pneumoniae subsp. pneumoniae or Escherichia coli using the MBEC Assay. Luminescence measurements based on quantitation of the ATP present were used to determine the amount of biofilm elimination and correlated with quantity of live bacteria present in the sample. The data were analyzed employing a two-way ANOVA and Bonferroni post-test. Treatments resulted in percentage reductions of E. coli biofilms ranging from 73 to 98%. Lacto

First implementation of a polymer-polymer aqueous two-phase system to improve lateral-flow immunoassay sensitivity via dextranase integration on paper.

Lateral-flow immunoassays (LFAs) are commonly used to detect proteins in bodily fluids, providing rapid and accessible patient sample testing at the point of care (POC). Although commercially successful, LFAs suffer from low sensitivity. To improve their sensitivity, our group was the first to implement polymer-salt and micellar aqueous two-phase systems (ATPSs) to preconcentrate various targets into a smaller volume prior to LFA application. For small hydrophilic protein targets that distribute fairly evenly in polymer-salt and micellar ATPSs, polymer-polymer ATPSs may be used for preconcentr

A novel GH66 family glycoside hydrolase pseudo-dextranase from Streptococcus thermophilus 17140 in catalyzing multi-step deglycosylation of ginsenoside Rb1.

Rare ginsenosides exhibit enhanced bioavailability and strong pharmacological activities such as anti-aging and immunomodulatory effects, making them valuable in health care food and medicine. We previously identified a Streptococcus thermophilus strain 17140 (St17140) capable of converting rare ginsenosides. In this study, we cloned and expressed a novel GH66 family glycoside hydrolase gene, pseudo-Dextranase from St17140 (PStDex). Despite lacking canonical catalytic residues for β-glucan hydrolysis, PStDex specifically cleaves the glycosidic bonds of ginsenoside Rb1 to produce rare ginsenosi

Colon specific enzyme responsive oligoester crosslinked dextran nanoparticles for controlled release of 5-fluorouracil.

Targeting bioactives selectively to diseased sites is one of the most challenging aspects of cancer therapy. Herein, fabrication of colonic enzyme-responsive dextran based oligoester crosslinked nanoparticles is reported for the controlled release of 5-fluorouracil (5-FU) - an anticancer drug. The 5-FU drug loaded nanoparticles (DNPs, size ~237 ± 25 nm, ζ-potential -17.0 ± 3 mV) were developed by the in-situ crosslinking of dextran with a bifunctional telechelic oligoester followed by the physical drug encapsulation via nanoprecipitation. Conjugation of metronidazole with dextran: a potential pharmaceutical strategy to control colonic distribution of the anti-amebic drug susceptible to metabolism by colonic microbes.

Metronidazole (MTDZ), the drug of choice for the treatment of protozoal infections such as luminal amebiasis, is highly susceptible to colonic metabolism, which may hinder its conversion from a colon-specific prodrug to an effective anti-amebic agent targeting the entire large intestine. Thus, in an attempt to control the colonic distribution of the drug, a polymeric colon-specific prodrug, MTDZ conjugated to dextran via a succinate linker (Dex-SA-MTDZ), was designed. Upon treatment with dextranase for 8 h, the degree of Dex-SA-MTDZ depolymerization (%) with a degree of substitution (mg of MTD

Revisiting the endo and exo mode of action of dextran hydrolyzing enzymes, and their significance for Streptococcus mutans biofilm eradication.

Streptococcus mutans is a major microorganism causing dental caries. The mechanism by which caries arises involve the production of extracellular polysaccharides by S. mutans, which further assemble into a biofilm. Several strategies, including enzymatic deconstruction of the carbohydrate portion of the biofilm, can potentially be used to combat the formation of oral biofilms. We set out to heterologously produce two different S. mutans dextranases: DexA and DexB, from glycoside hydrolase families 66 (GH66) and 13 (GH13), respectively, and to study their capacity to degrade S. mutans biofilms.

Next Generation Aqueous Two-Phase System for Gentle, Effective, and Timely Extracellular Vesicle Isolation and Transcriptomic Analysis.

The isolation of extracellular vesicles (EVs) using currently available methods frequently compromises purity and yield to prioritize speed. Here, we present a next-generation aqueous two-phase system (next-gen ATPS) for the isolation of EVs regardless of scale and volume that is superior to conventional methods such as ultracentrifugation (UC) and commercial kits. This is made possible by the two aqueous phases, one rich in polyethylene glycol (PEG) and the other rich in dextran (DEX), whereby fully encapsulated lipid vesicles preferentially migrate to the DEX-rich phase to achieve a local en

Regulation and extracellular glucosyltransferase production and the relationship between extracellular and cell-associated activities in Streptococcus mutans.

The regulation of extracellular glucosyltransferase production in Streptococcus mutans GS-5 has been studied using a chemically defined medium. Most of the glucosyltransferase activity produced by cells grown in the chemically defined medium was extracellular, in contrast with the distribution between cell-associated and extracellular glucosyltransferase activity when cells were grown in complex medium. The production of extracellular glucosyltransferase activity coincided with the logarithmic growth phase, and further accumulation ceased when glucose was exhausted from the medium. Accumulatio
