Diapause is a genetically programmed developmental or reproductive arrest that helps insects survive adverse seasonal conditions. Our understanding of the molecular regulation of this complex trait has significantly improved through the use of omics technologies over the last few decades. In this review, we have compiled current omics research on insect diapause, spanning transcriptomics, proteomics, metabolomics, lipidomics, epigenomics, genomics, and post-transcriptional regulatory approaches. Transcriptomics has led the field, showing that the diapause program develops gradually through a s

Tools that could be used to subvert the insect diapause response offer potential for insect pest management as well as for the experimental manipulation of insects and the facilitation of mass rearing procedures. In some cases, it is desirable to break diapause on demand and in other cases, it may be attractive to exploit diapause for long-term storage of biocontrol agents or valuable experimental lines. This review highlights some of the diapause disruptors reported in the literature, as well as chemical and physical manipulations that can be used to extend diapause or even induce diapause in

Despite the overwhelming diversity and ecological impact of insects across most ecosystems, their responses to environmental stress remain underexplored. To survive temperate seasonal stressors, particularly winter cold conditions and nutrient and water shortage, insects undergo physiological changes and overwinter in a diapause state. During diapause, cessation of development is coupled with reduced metabolic activity, analogous to hibernation in mammals, in which gut microbiota help manage waste and recycle nitrogen. Similarly, many insects have obligate symbiotic relationships with microbes

Forensic entomology plays an important role in estimating the postmortem interval (PMI). However, during cold seasons, necrophagous insects may enter diapause, resulting in developmental arrest. Currently, effective methods for estimating diapause duration are limited, which may compromise the accuracy of PMI estimation based on insect evidence. Here we show that multi-dimensional data fusion of gene expression and infrared spectral features could improve the accuracy of diapause duration estimation. Using the forensically important flesh fly Sarcophaga peregrina as a model, we performed trans

To survive harsh winter conditions, many insects enter a state of dormancy called diapause which allows them to withstand extreme temperatures and lack of food. When diapause is induced, insects enter a state of arrested development with a low metabolic rate. Because diapause and environmental conditions are closely linked, variation within and between species in diapause induction, depth, and duration is extremely common. Studies investigating the genetic underpinnings of diapause tend to focus on either different populations and/or environmental variation. We use the eastern spruce budworm (

Diapause is a pre-programmed arrest of development allowing insects to survive in unfavorable environments. In adult insects, diapause termination is often followed by a reallocation of macronutrients and a transition to reproduction, and in some insects, this transition can be achieved using narcosis with CO2. However, whether CO2narcosis and diapause act in concert to affect reproduction remains unknown. Here, we investigated the separated and combined effects of diapause and CO2on female reproduction in queens of the common eastern bumble bee Bombus impatiens. Queens were treated with CO2 f

As ectotherms, many insects spend the winter months in a state of suspended animation (i.e., diapause), lowering their metabolic rates to subsist on a limited store of energy reserves. The ability to lower metabolic rate during diapause relies, in part, on cold winter temperatures to intrinsically lower metabolic rate. Winter warming associated with global climate change may pose a challenge to diapausing insects by intrinsically increasing metabolic rate, potentially leading to the exhaustion of energetic reserves. We used stop-flow respirometry to measure oxygen consumption in response to te

Reproductive diapause is an insect survival strategy in which reproduction temporarily halts in response to adverse environmental changes. This process is characterized by arrested ovarian development and lipid accumulation in females. A reduction in juvenile hormone (JH) biosynthesis is known to initiate reproductive diapause, but its regulatory mechanism remains unclear. Seven up (Svp), a transcription factor from the nuclear receptor family, plays a crucial role in various developmental processes in insects. In this study, using the cabbage beetle Colaphellus bowringi as a model, we observe

Adaptive shifts in respiratory patterns during insect diapause represent a key strategy for coping with seasonal adversity, yet the underlying regulatory mechanisms remain poorly understood. Using the swallowtail butterfly Sericinus montelus Gray as a model, this study investigated the regulatory roles of diapause and temperature on respiratory patterns by comparing gas exchange patterns and metabolic characteristics of pupae under different photoperiods and temperature conditions. The results demonstrated that the metabolic rate of diapause-destined pupae was significantly lower than that of

Diapause is a prevalent strategy employed by insects to survive adverse environmental conditions, by halting development and reducing metabolic activity. Although the suppression of DNA replication aligns logically with these reduced developmental and physiological activities, the role of DNA replication in regulating insect diapause remains largely unknown. In this study, we used the cabbage beetle, Colaphellus bowringi, to investigate the role of DNA replication pathway in regulating reproductive diapause. Transcriptome analysis identified DNA replication as a key pathway during diapause pre

Diapause is a critical adaptive strategy in insects to survive unfavourable environmental conditions. The present study investigated molecular mechanisms underlying diapause in Chilo partellus, using high-throughput RNA sequencing and de novo transcriptome assembly. Six transcriptome libraries from diapause and non-diapause larvae yielded over 50 million reads per sample, with >98% mapping efficiency. The Evidential Gene-refined assembly produced 67,116 high-quality transcripts, and BUSCO analysis confirmed its completeness. DEG analysis identified 1204 DEGs, including 660 upregulated and 544

BACKGROUND: Diapause is a vital adaptive strategy enabling insects to endure adverse environmental conditions and significantly extend their lifespan, by suspending development and conserving energy. During diapause, profound physiological and metabolic changes occur. Key genes involved in diapause have long been considered a potential target for pest control.
RESULTS: In this study, we observed elevated glucose levels in the brains of Helicoverpa armigera diapause-destined pupae. Concurrently, the expression of glucose transporter 1 (Glut1), critical for glucose uptake, was upregulated, disp

Transgenerational effects act on a wide range of insects' life-history traits and can be involved in the control of developmental plasticity, such as diapause expression. Decrease in or total loss of winter diapause expression recently observed in some species could arise from inhibiting maternal effects. In this study, we explored transgenerational effects on diapause expression and traits in one commercial and one Canadian field strain of the aphid parasitoid Aphidius ervi. These strains were reared under short photoperiod (8:16 h LD) and low temperature (14 °C) conditions over two generatio

The silkworm Bombyx mori exhibits a photoperiodic response (PR) for embryonic diapause induction. This article provides a comprehensive review of literature on the silkworm PR, starting from early works on population to recent studies uncovering the molecular mechanism. Makita Kogure (1933) conducted extensive research on the PR, presenting a pioneering paper on insect photoperiodism. In the 1970s and 80s, artificial diets were developed, and the influence of nutrition on PR was well documented. The photoperiodic photoreceptor has been investigated from organ to molecular level in the silkworm

Most previous studies on gene expression during insect diapause do not address among-tissue variation in physiological processes. We measured transcriptomic changes during larval diapause in the Asian longhorned beetle, Anoplophora glabripennis (Coleoptera: Cerambycidae). We conducted RNA-seq on fat body, the supraesophageal ganglion, midgut, hindgut, and Malpighian tubules during pre-diapause, diapause maintenance, post-diapause quiescence, and post-diapause development. We observed a small, but consistent, proportion of genes within each gene expression profile that were shared among tissues

Juvenile hormone (JH) has been revealed to be a critical factor in regulating photoperiod reproductive diapause in various insect species, however, little information is known about the detailed mechanisms. In this study, we investigated the roles of JH signaling in photoperiod reproductive diapause in a green lacewing, Chrysoperla nipponensis (Okamoto), which is a potentially important biological control predator. Our results showed that the short-day condition induces a diapause state including JH synthesis suppression, ovarian development arrest, and triglyceride accumulation. The interfere

Ultrastructural examination of diapause and nondiapause larval brains of the European corn borer disclosed anatomical differences that may be related to the insect's "blood-barrier". The perineurial type I cells are quite closely appressed in the diapause brain, but thrown into extensive folds with large intercellular spaces in the nondiapause brain. The perineurial type II cells of diapause and nondiapause larvae are basically similar in general ultrastructure, and most likely form the basis for the "blood-brain barrier." Horseradish peroxidase penetration studies indicated that the outer mar

Diapause is a widespread adaptation of insects that enables them to survive during unfavorable seasons and is characterized by suppressed metabolism and increased lifespan. Previous works have demonstrated that high levels of reactive oxygen species (ROS) and hypoxia-inducible factor-1α (HIF-1α) in the pupal brain of the moth Helicoverpa armigera induce diapause and extend lifespan by downregulating mitochondrial transcription factor A (TFAM). However, the molecular mechanisms of ROS-HIF-1α regulating metabolic activity to extend lifespan are still poorly understood. Here, we show that the mit

BACKGROUND: The brown marmorated stink bug (Halyomorpha halys) (BSMB) is a biosecurity threat globally. Its most likely avenue of entry is via shipping transport of a multitude of commodities. Currently employed biosecurity treatments (methyl bromide, sulfuryl fluoride) pose several concerns including climate change, environmental and occupational health and safety. Ethyl formate with dilution in carbon dioxide has been shown to be an effective biosecurity treatment for controlling BMSB. Carbon dioxide is a known synergist in fumigation and is a greenhouse gas contributing to climate change.

Diapause regulates seasonal insect life cycles and may be highly variable within and among populations due to genetic and environmental variability. Both types of variation may influence how populations respond plastically or evolutionarily to changing climates. We assessed diapause variability in spruce beetle Dendroctonus rufipennis Kirby (Coleoptera: Curculionidae, Scolytinae), a major forest pest whose life cycle timing is regulated by both prepupal and adult diapauses. Using mating studies and ovary dissections, we tested for variability in adult diapause within and between collection sit
