Detection of specific Treponema species and Dichelobacter nodosus from digital dermatitis (Mortellaro's disease) lesions in Swiss cattle.

The aim of this study was to determine the prevalence of the three Treponema species as well as D. nodosus in Digital dermatitis (DD) and slurry of Swiss cattle using PCR. A total of 86 specimens from 24 farms were enrolled in the study. Slurry samples from 21 DD-affected and one unaffected farm were collected to assess the potential of environmental transmission. Nested and real-time PCR were performed from the specimens to detect Treponema species and D. nodosus, respectively. The DD-stages were positive for at least one or more of the DD-associated Treponema species in 50 of 61 cases (82.0%

The prevalence of Dichelobacter nodosus in clinically footrot-free sheep flocks: a comparative field study on elimination strategies.

BACKGROUND: Ovine footrot caused by Dichelobacter nodosus (D nodosus) is an infectious disease affecting sheep worldwide. Switzerland plans a nationwide footrot eradication program, based on PCR-testing of interdigital swab samples. The aim of this study was to test for the presence of D nodosus in clinically footrot-free sheep flocks which had been subjected to different treatment strategies, to assess whether they were feasible for the eradication process, especially focussing on antimicrobial flock treatments. Clinical scoring and PCR-results were compared. Ten farms had used hoof bathing a

In vitro and ex vivo testing of alternative disinfectants to currently used more harmful substances in footbaths against Dichelobacter nodosus.

A footbath-based control program for ovine footrot, a contagious disease caused by Dichelobacter nodosus, will be implemented in Switzerland. The currently used footbath disinfectants formaldehyde, zinc sulfate and copper sulfate are carcinogenic or environmental pollutants. Hence, the aim of this study was to identify alternative disinfectants, which are highly effective, non-carcinogenic, environmentally acceptable, inexpensive, available as concentrate and suitable for licensing. The antimicrobial effect of a series of potential chemicals such as lactic acid, propionic acid, hydrogen peroxi

Evolution of oxidative/nitrosative stress biomarkers during an open-field vaccination procedure in sheep: effect of melatonin.

Melatonin has been shown to exert immunomodularory properties with broad application in veterinary medicine. In previous work we have described that subcutaneous coadministration of melatonin to seeps vaccinated against two stumps of A1 and C strains of Dichelobacter nodosus enhanced both the antibody titer and serum IgG levels to A1 and C strains of D. nodosus compared to vaccinated animals not treated with melatonin. Following a similar protocol here we have investigated the effect of a higher dose of melatonin (36mg/animal) in the improvement of the immune response and in the possible oxida

Bacterial species associated with interdigital phlegmon outbreaks in Finnish dairy herds.

BACKGROUND: Severe outbreaks of bovine interdigital phlegmon (IP) have occurred recently in several free stall dairy herds in Finland. We studied the aetiology of IP in such herds, and the association of bacterial species with the various stages of IP and herds of various morbidity of IP. Nineteen free stall dairy herds with IP outbreaks and three control herds were visited and bacteriological samples collected from cows suffering from IP (n = 106), other hoof diseases (n = 58), and control cows (n = 64). The herds were divided into high morbidity (morbidity ≥50%) and moderate morbidity groups

Variation in the early-life interdigital skin microbiota of lambs in a commercial sheep flock.

INTRODUCTION: The factors shaping early-life microbial colonisation of the ovine interdigital skin, and their potential relevance to foot health and footrot susceptibility, remain poorly understood. Here, we investigated variation in the interdigital skin microbiota among lambs in different age-location groups. METHODS: Interdigital skin swabs were collected from all four feet of 25 lambs aged 0-15 days on a commercial farm. Bacterial community composition was characterised using 16S rRNA gene amplicon sequencing, and quantitative PCR was used to estimate total bacterial load and detect Diche

Pathology and bacteria related to digital dermatitis in dairy cattle in all year round grazing system in Brazil.

Digital dermatitis (DD) is one of the main causes of lameness in dairy cattle worldwide, and it is frequently reported in high-yielding, free stall dairy herds from regions with a temperate climate. However, DD is also observed with high prevalence in grazing cattle with a low milk yield in tropical regions. To clarify whether these differences have an impact on the etiology of the disease, we studied DD lesions from all year round grazing cattle of mixed breed in Brazil using high-throughput 16S rRNA gene sequencing and fluorescent in situ hybridization. The study included samples from 66 ski

Social Structure of Sheep Flocks at Points of the Production Cycle and Relationship to Disease Spread, Using a Simulated Epidemic of Footrot.

Footrot is one of the top five globally important diseases of sheep and causes lameness, leading to poor welfare and productivity. Transmission of Dichelobacter nodosus, the causative agent, occurs via surfaces such as pasture or bedding and persistence occurs from diseased sheep shedding bacteria into the environment; D. nodosus cannot replicate off host. High resolution proximity sensors were deployed on a flock of Poll Dorset sheep for 10-17 days at several points of the production cycle (teasing, tupping, pregnancy, and lactation (<6-week-old lambs)) between July 2018 and May 2021. Surveying bovine digital dermatitis and non-healing bovine foot lesions for the presence of Fusobacterium necrophorum, Porphyromonas endodontalis and Treponema pallidum.

BACKGROUND: Non-healing bovine foot lesions, including non-healing white line disease, non-healing sole ulcer and toe necrosis, are an increasingly important cause of chronic lameness that are poorly responsive to treatment. Recent studies have demonstrated a high-level association between these non-healing lesions and the Treponema phylogroups implicated in bovine digital dermatitis (BDD). However, a polymicrobial aetiology involving other gram-stain-negative anaerobes is suspected. METHODS: A PCR-based bacteriological survey of uncomplicated BDD lesions (n=10) and non-healing bovine foot le

Plasmid-mediated florfenicol resistance in Mannheimia haemolytica isolated from cattle.

The aim of this study was to analyse a florfenicol-resistant Mannheimia haemolytica isolated from a calf to determine the genetic basis of its florfenicol-resistance. The antimicrobial susceptibility and plasmid content of the isolate were determined. A florfenicol resistant plasmid carrying the floR gene was identified by PCR and transformed into Escherichia coli JM109 and HB101 strains. The plasmid was then mapped and sequenced completely. The isolate was resistant to chloramphenicol, florfenicol, oxytetracycline, kanamycin, dihydrostreptomycin, nalidixic acid, ampicillin, and amoxicillin; i

The gene for toxic shock toxin is carried by a family of mobile pathogenicity islands in Staphylococcus aureus.

Tst, the gene for toxic shock syndrome toxin-1 (TSST-1), is part of a 15.2 kb genetic element in Staphylococcus aureus that is absent in TSST-1-negative strains. The prototype, in RN4282, is flanked by a 17 nucleotide direct repeat and contains genes for a second possible superantigen toxin, a Dichelobacter nodosus VapE homologue and a putative integrase. It is readily transferred to a recA recipient, and it always inserts into a unique chromosomal copy of the 17 nucleotide sequence in the same orientation. It is excised and circularized by staphylococcal phages phi13 and 80alpha and replicate

Characterization and expression of a novel Porphyromonas gingivalis outer membrane protein, Omp28.

We report the characterization of a Porphyromonas gingivalis gene, designated omp28, encoding a protein that we have previously purified and characterized as a 28-kDa outer membrane protein. The deduced amino acid sequence of the omp28 open reading frame displayed an outer membrane leader sequence and lipoprotein attachment site but did not exhibit any significant overall sequence identity with protein sequences in the databases. A small stretch of amino acids (19 residues) exhibits 50% sequence identity with a segment of a fimbrial protein from Dichelobacter nodosus involved in adhesion, sugg

In vitro susceptibility of bovine digital dermatitis treponemes to conventional and novel antimicrobial agents, and evidence of heavy metal resistance.

OBJECTIVES: Bovine digital dermatitis (BDD) is a globally endemic, painful infectious foot disease of cattle; however, gaps in knowledge regarding treatment efficacy limit our ability to control it. Our objectives were to collect data on minimum inhibitory concentrations and minimum bactericidal concentrations (MICs and MBCs) for conventional and naturally derived therapeutic agents against specific Treponema strains important in BDD pathogenesis in the context of the need for responsible use of antimicrobials. Since antibacterial metals are widely used to control infectious lameness in rumina

Wild UK deer as carriers of livestock lameness associated pathogens.

Lameness is one of the major diseases on UK farms and worldwide and can be caused by environmental damage and/or infection. The pathogens associated with infectious lameness, namely Fusobacterium necrophorum, Dichelobacter nodosus and pathogenic Treponema species are responsible for substantial economic losses. The lack of knowledge on reservoirs of these pathogens is concerning and wild deer may pose a potential risk for livestock infection. This study investigated the prevalence of lameness associated bacteria on the feet of culled wild deer using molecular methods and subsequent isolation t

Detection of treponemes in digital dermatitis lesions of captive European bison (Bison bonasus).

A newly-discovered foot disease of unknown origin in captive European Bison (Bison bonasus) was recently detected at Berne Animal Park. Dermatitis of the interdigital cleft of varying degrees of severity was diagnosed in all animals (n = 10). The aim of this study was to describe the gross and histological lesions of the interdigital cleft found in 10 captive European bison and to identify involved potential pathogens in affected feet using molecular-based methods for Treponema spp., Dichelobacter nodosus and Fusobacterium necrophorum. Lesions were scored according to the degree of gross patho

Histopathological and radiographic characterization of the lesions of pododermatitis in sheep: support for the establishment of the foot injuries degree and its prognosis.

Foot diseases in small ruminants cause locomotor disorders, leading to significant economic, productive, and health concerns in sheep and goat farming worldwide. The diagnosis and classification of lesions caused by footpad dermatitis are complex and based only on clinical observations in the field. In this context, this study assessed the histopathological and radiographic characteristics of lesions caused by pododermatitis in sheep to improve and deepen the classification of lesions and optimize diagnosis and prognosis. In this study 1.701 lame sheep were included and were distributed across

Novel ulcerative leg lesions in yearling lambs: Clinical features, microbiology and histopathology.

Here we report an outbreak of an atypical, ulcerative dermatitis in North Country mule lambs, located in South Gloucestershire, UK. The lesions, which appeared to be contagious, occured between the coronary band and the carpal joint as a focal, well demarcated, circular, ulcerative dermatitis. Histopathological examination of the lesion biopsies revealed areas of ulceration, epidermal hyperplasia, suppurative dermatitis and granulation tissue. Clumped keratohyalin granules and intracellular keratinocyte oedema (ballooning degeneration) were evident within lesion biopsies, consistent with an un

Digital dermatitis in dairy cattle in Egypt: Herd-level risk factors and Treponema spp. prevalence across lesion M-scores.

Digital dermatitis (DD) is an infectious disease of the digital skin of dairy cows that is associated with compromised animal welfare and significant economic losses. The hind feet of 16,098 dairy cows from 55 herds were examined in the milking parlor, and DD lesions identified were classified using the M-score system and swabbed for PCR testing. Swabs were also collected from hind feet with normal digital skin for comparison. Data on farm construction and lameness prevention measures were collected through direct observation and in-person interviews with farm managers. Herd-level risk factors
