ICTV Virus Taxonomy Profile: Dicistroviridae.

Dicistroviridae is a family of small non-enveloped viruses with monopartite, linear, positive-sense RNA genomes of approximately 8-10 kb. Viruses of all classified species infect arthropod hosts, with some having devastating economic consequences, such as acute bee paralysis virus in domesticated honeybees and taura syndrome virus in shrimp farming. Conversely, the host specificity and other desirable traits exhibited by several members of this group make them potential natural enemies for intentional use against arthropod pests, such as triatoma virus against triatomine bugs that vector Chaga

Identification of Viruses in Molossus Bats from the Brazilian Amazon: A Descriptive Metagenomic Analysis.

Bats are widely distributed in Brazil, including the Amazon region, and their association with viral pathogens is well-known. This work aimed to evaluate the metavirome in samples of Molossus sp. bats captured in the Brazilian Amazon from 2019 to 2021. Lung samples from 58 bats were divided into 13 pools for RNA isolation and sequencing followed by bioinformatic analysis. The Retroviridae family showed the highest abundance of viral reads. Although no complete genome could be recovered, the Paramyxoviridae and Dicistroviridae families showed the formation of contigs with satisfactory identity

Genetic mechanisms underlying the structural elaboration and dissemination of viral internal ribosomal entry sites.

Viral internal ribosomal entry sites (IRESs) are highly structured cis-acting RNAs that mediate end-independent initiation of translation. Their origin remains obscure. The simplest IRESs (type 6) occur in the intergenic region of Dicistroviridae genomes (order Picornavirales), consist of two pseudoknots, and initiate translation by factor-independent binding to ribosomes. Larger variants contain a third pseudoknot that modifies the mechanism of IRES function by engaging with the ribosomal head and promoting binding to the ribosomal aminoacyl site. Metagenomic analyses undertaken to identify s

Mechanical transfer of honey bee (Hymenoptera: Apidae) virus sequences to wax by worker traffic and aerosolization.

Honey bees (Apis mellifera L.) are of undeniable value to agriculture. However, increased mortality of honey bees, mostly due to winter losses associated with parasites and pathogens, have put strain on the apiculture industry. Advancing our knowledge of honey bee viruses and their interactions within the colony environment is vital in mitigating their effect on honey bee health. Our study examined virus sequences detected on beeswax sampled from empty colonies which died during the previous winter. Based on a cage study using virus-containing bees, we confirmed that the introduction of BQCV s

Antiviral activity of curcumin against Israeli acute paralysis virus in Apis mellifera: Screening and mechanistic study.

The Israeli acute paralysis virus (IAPV), widespread in honeybee (Apis mellifera) colonies, causes paralysis, tremors, and eventual mortality, while strongly associated with colony collapse disorder (CCD). Currently, there are no widely approved antiviral therapies for IAPV infection; however, several natural compounds have shown promising antiviral efficacy. In this study, we evaluated the effects of eight natural compounds (myrcene, citral, menthol, chlorogenic acid, resveratrol, curcumin, quercetin, and tea polyphenols) on IAPV-infected honeybees. Bees were artificially or naturally infecte

Nosema ceranae and honey bee-associated virus levels in commercial beekeeping operations across Southwestern Saudi Arabia.

The native honey bee of the Arabian Peninsula, Apis mellifera jemenitica, is well adapted to live in arid environments, but could potentially remain vulnerable to pathogens and parasites. We present a molecular report of Nosema and viral infections from eleven commercial apiaries across Southwestern Saudi Arabia. We confirmed that all Nosema-positive samples belonged to N. ceranae, with infection levels varying by location. The average spore count via microscopy was 3.43 × 107 spores per bee across all locations, with the highest being 3.39 × 108 spores per bee. No spores were detected in thre

Drosophila C virus systemic infection leads to intestinal obstruction.

UNLABELLED: Drosophila C virus (DCV) is a positive-sense RNA virus belonging to the Dicistroviridae family. This natural pathogen of the model organism Drosophila melanogaster is commonly used to investigate antiviral host defense in flies, which involves both RNA interference and inducible responses. Although lethality is used routinely as a readout for the efficiency of the antiviral immune response in these studies, virus-induced pathologies in flies still are poorly understood. Here, we characterize the pathogenesis associated with systemic DCV infection. Comparison of the transcriptome of

Viruses and vectors tied to honey bee colony losses.

Commercial beekeepers in the US reported severe colony losses early in 2025, as colonies were being staged for their critical role in the almond pollination season in California. Average reported losses since the preceding spring exceeded 60%, with substantial variation among operations. Many colonies were still actively collapsing in January 2025, at which time pooled and individual samples were collected and then screened for levels of 13 known honey bee pathogens and parasites. Acute bee paralysis virus and other known viral pathogens were found at high levels in pooled bee samples from all

Honey bee egg composition changes seasonally and after acute maternal virus infection.

Honey bee (Apis mellifera) colonies depend on the reproductive output of their queens, which in turn is contingent on the care they receive from worker bees. Viral infections in queens can compromise their reproductive output, while viral infections in workers can inhibit the successful functioning of the colony and its ability to care for the queen. Transgenerational immune priming (TGIP) occurs when queens transfer immune-related compounds or immune elicitors to their offspring, enhancing the ability of subsequent generations to resist infections. These maternal effects on offspring could po

Seasonal dynamics of seven viruses and Varroa destructor in Apis mellifera colonies: a 4-year survey in Tokyo.

Long-term monitoring of viruses and Varroa destructor within the same honey bee colonies is rare in Japan. Thus, we conducted a 4-year fixed-point survey of three Apis mellifera colonies in Tokyo with monthly screening for seven viruses (using polymerase chain reaction) and Varroa infestation. Deformed wing virus was detected year-round, whereas sacbrood virus and black queen cell virus peaked in spring and summer. Israeli acute paralysis virus appeared sporadically, without seasonal trends. Varroa infestation rates (monitored during the latter 2.5 years) did not correlate with the number of d

Experimental infection of acute bee paralysis virus induces differential responses of selected honey bee immune genes, independent of varroa resistance.

Honeybees (Apis mellifera) have a complex immune system, with both innate and social immunity. The mite-resistant honeybee population from Gotland, Sweden, is well-known for their independent resistance to Varroa destructor mites and tolerance to virus infections, particularly deformed wing virus and acute bee paralysis virus (ABPV). However, the immune mechanisms underlying their virus tolerance remains unclear. Adult bees from this mite-resistant (MR) and a mite-susceptible (MS) control population were orally inoculated with either ABPV or a mock inoculum, in a laboratory time-course experim

Host range of Triatoma virus does not extend to Aedes aegypti and Apis mellifera.

Vector control is the most effective method to prevent transmission of Chagas disease. Control is mostly made through chemical insecticides although they have negative impact on wild pollinators, such as bees. Reducing pesticide use through biological alternatives could minimize the damage to these beneficial insects. Triatoma virus (TrV) is a pathogen able to kill triatomines and thus a valid candidate to be used as biological control agent. In this study we evaluate the capacity of TrV to infect an important beneficial insect (Apis mellifera) as well as a plague insect (Aedes aegypti). Resul

Isolation, characterization, and host specificity of Solenopsis invicta virus 6 (Cripavirus porteri), a dicistrovirus from the red imported fire ant, Solenopsis invicta.

Solenopsis invicta virus 6 (SINV-6; Cripavirus porteri) is a recently discovered dicistrovirus from the red imported fire ant, Solenopsis invicta. SINV-6 was detected in all stages of S. invicta including eggs, pupae, larvae, adult workers, male and female sexual alates, and queens. Electron microscopic examination of negatively stained samples from particles purified from SINV-6-infected fire ant workers revealed isometric particles with a mean diameter of 34.2 ± 2.1 nm. Field surveys of S. invicta nests in Alachua County, Florida, showed that the virus was present year-round. SINV-6 nest pre

Rapid Detection of DNA and RNA Shrimp Viruses Using CRISPR-Based Diagnostics.

Timely detection of persistent and emerging pathogens is critical to controlling disease spread, particularly in high-density populations with increased contact between individuals and limited-to-no ability to quarantine. Standard molecular diagnostic tests for surveying pathogenic microbes have provided the sensitivity needed for early detection, but lag in time-to-result leading to delayed action. On-site diagnostics alleviate this lag, but current technologies are less sensitive and adaptable than lab-based molecular methods. Towards the development of improved on-site diagnostics, we demon

Transcriptomic Responses Underlying the High Virulence of Black Queen Cell Virus and Sacbrood Virus following a Change in Their Mode of Transmission in Honey Bees (Apis mellifera).

BACKGROUND: Over the last two decades, honey bees (Apis mellifera) have suffered high rates of colony losses that have been attributed to a variety of factors, chief among which are viral pathogens, such as deformed wing virus (DWV), whose virulence has increased because of vector-based transmission by the invasive, ectoparasitic varroa mite (Varroa destructor). A shift in the experimental mode of transmission of the black queen cell virus (BQCV) and sacbrood virus (SBV) from fecal/food-oral (direct horizontal) to vector-mediated (indirect horizontal) transmission also results in high virulenc

Resident viruses, but not honeybee-associated viruses, impair solitary bee fitness in the field.

Viruses can impact individual host fitness and host population dynamics, especially following host shifts. Thus, the decline of wild solitary bee populations over the last few decades may be linked to viruses or other pathogens. However, evidence for the impact of viruses-transmitted from other genera or resident in solitary bees-on their fitness remains scarce. Here, by assessing solitary bee (Osmia cornuta) foraging, offspring sex ratio, survival and body mass across seven locations in northern Switzerland, we show that resident viruses-but not honeybee-associated viruses-can impact fitness

Seasonal variation of viral infections between the eastern honey bee (Apis cerana) and the western honey bee (Apis mellifera).

It is a widespread practice in China to keep colonies of both the western honey bee, Apis mellifera, and the eastern honey bee, Apis cerana, in close proximity. However, this practice increases opportunities for spillover of parasites and pathogens between the two host bee species, impacting spatial and temporal patterns in the occurrence and prevalence of the viruses that adversely affect bee health. We conducted a 1-year large-scale survey to assess the current status of viral infection in both A. mellifera and A. cerana in China. Our study focused on multiple aspects of viral infections in

Juvenile hormone enhances viral activity in bumble bees.

Bees are susceptible to viral infections that can compromise individual and colony health. As viruses depend on host cellular resources for replication, we investigated the influence of a key insect regulator, Juvenile hormone (JH), which enhances reproduction and metabolic rate on viral activity. We hypothesized that elevated JH levels would promote viral replication in a tissue-specific manner. To test this hypothesis, we used the bumble bee, Bombus terrestris, and the Israeli acute paralysis virus (IAPV). In our first experiment, we compared IAPV replication in pre- and post-diapause queens

Investigation of viral and parasitic pathogens in Bombus terrestris (Hymenoptera: Apidae) populations.

In recent years, with the increase in both abiotic and biotic stress factors, there has been a significant decline in honey bee and bumblebee populations worldwide, which in turn threatens pollination services that are crucial for agricultural production. This study aimed to determine the prevalence and incidence of viral and parasitic pathogens in Bombus terrestris (Linnaeus, 1758) individuals obtained from 23 wild-caught queen bumblebees collected in Ordu Province, Türkiye. For this purpose, analyses conducted on bumblebee samples collected from different businesses revealed that the Acute B

A large-scale, longitudinal study on the epidemiology of Nosema (=Vairimorpha) ceranae and black queen cell virus (BQCV) in Apis mellifera colonies across the Italian agroecosystems.

Honey bee health is increasingly compromised by multiple interacting stressors, including pathogens whose epidemiology is shaped by environmental and temporal variables. Among them, highly prevalent and impactful are the microsporidian Nosema (=Vairimorpha) ceranae and the black queen cell virus (BQCV). This study presents a large-scale, longitudinal assessment of the two pathogens in managed Apis mellifera colonies across all Italian regions, conducted between the years 2021 and 2024 and involving 12028 samples from 398 apiaries. Molecular qPCR diagnostics revealed N. ceranae in 50.9% of the
