Influence of dietary sucrose on biological aging.

The segment of the population aged > 65 y is the fastest growing age segment in most developed countries. The increasing numbers of individuals living into their eighth and ninth decades of life have shifted the focus of biomedical research from seeking mechanisms for extending life to finding ways to improve health and to reduce age-related morbidity. One area of research that has shown considerable promise for improving the health of elderly people is nutrition. I review recent literature pertinent to the influences of nutrition on biological aging by discussing the effects of dietary sucros

Urinary excretion of sucrose and fructose as a predictor of sucrose intake in dietary intervention studies.

The urinary excretion of sucrose, glucose, and fructose was measured in 9 healthy subjects consuming a common Italian diet and after 3 days of a low sucrose diet, in which the intake of sucrose was restricted but the other main nutrients were unmodified. After the low sucrose diet, we observed a significant drop in the average urinary excretion of sucrose, glucose, and fructose determined at four different times (8:00 and 10:00 a.m.; 3:00 and 10:00 p.m.). The average urinary excretion of fructose in the four urine samples was significantly correlated with dietary sucrose intake. We also found

Metabolic effects of dietary sucrose in type II diabetic subjects.

OBJECTIVE: To assess in diabetic subjects the effects of dietary sucrose on glycemia and lipemia. RESEARCH DESIGN AND METHODS: Twelve type II diabetic subjects consumed, in random order, two isocaloric, 55% carbohydrate study diets for 28 days. In one diet, 19% of energy was derived from sucrose. In the other diet, < 3% of energy was derived from sucrose, and carbohydrate energy came primarily from starch. Both study diets were composed of common foods. All meals were prepared in a metabolic kitchen where foods were weighed during meal preparation. RESULTS: No significant differences were no

Effects of dietary sucrose on factors influencing cholesterol gall stone formation.

Twelve subjects with radiolucent gall stones and bile supersaturated with cholesterol were studied after six weeks each on diets which contained 112 g and 16 g respectively of refined (fibre depleted) sucrose but which allowed free access to other foods. Energy intake was 24.5% higher on the high sugar diet and body weight ended 1.4 kg higher than on the low sugar diet. Biliary secretion rates of cholesterol, phospholipid and bile acid, measured by a perfusion technique, were similar on the two diets, as were the bile acid pool sizes measured by isotope dilution. Cholesterol saturation index o

Effect of variation in dietary sucrose intake on plaque removal by mechanical means.

The influence of dietary sucrose on the rate of plaque formation was investigated for 22 dental personnel. The subjects were instructed to continue their normal oral hygiene procedures throughout the study period. It was found that an increase in the frequency of sucrose intake resulted in an enhancement of plaque deposition and an increase in gingival inflammation. The restriction of dietary sucrose for this group of subjects had a minimal effect on either plaque retention or levels of gingival inflammation; this result might have been caused by the habitual diet of these particular subjects

Source of dietary sucrose influences development of leptin resistance in male and female rats.

Male rats offered 30% sucrose solution in addition to chow develop leptin resistance without an increase in energy intake or body fat. This study tested whether the leptin resistance was dependent on the physical form of the sucrose. Sprague-Dawley rats were offered a sucrose-free (NS) diet, a 66.6% of energy as sucrose (HS) diet, or the NS diet + 30% sucrose solution (LS). Sucrose intake of LS rats equaled that of HS rats, but total carbohydrate intake exceeded that of HS rats. After 33 days, male and female LS rats were resistant to the inhibitory effect of peripherally administered leptin o

Dietary sucrose is essential to the development of liver injury in the methionine-choline-deficient model of steatohepatitis.

Methionine-choline-deficient (MCD) diets cause steatohepatitis in rodents and are used to study the pathophysiology of fatty liver disease in human beings. The most widely used commercial MCD formulas not only lack methionine and choline but also contain excess sucrose and fat. The objective of this study was to determine whether dietary sucrose in the MCD formula plays a role in the pathogenesis of MCD-related liver disease. We prepared two custom MCD formulas, one containing sucrose as the principal carbohydrate and the other substituting sucrose with starch. Mice fed the sucrose-enriched fo

Development of anemia in copper-deficient rats fed high levels of dietary iron and sucrose.

The effects of dietary carbohydrate and iron on the development of copper deficiency were examined. Male Sprague-Dawley rats (n = 48) were limit-fed one of eight diets in a 2 X 2 X 2 factorial design for 19 d. Two levels of copper (0.85 or 8.6 micrograms Cu/g diet) and iron (54 or 226 micrograms Fe/g diet) and two types of carbohydrate (sucrose or cornstarch, 65.3%) were fed. Compared with control rats, copper-deficient rats had lower hematocrits, lower ceruloplasmin levels, lower tissue levels of copper and increased hepatic iron levels. Copper-deficient rats fed sucrose had significantly low

Nucleus accumbens inflammation mediates anxiodepressive behavior and compulsive sucrose seeking elicited by saturated dietary fat.

OBJECTIVE: The incidence of depression is significantly compounded by obesity. Obesity arising from excessive intake of high-fat food provokes anxiodepressive behavior and elicits molecular adaptations in the nucleus accumbens (NAc), a region well-implicated in the hedonic deficits associated with depression and in the control of food-motivated behavior. To determine the etiology of diet-induced depression, we studied the impact of different dietary lipids on anxiodepressive behavior and metabolic and immune outcomes and the contribution of NAc immune activity. METHODS: Adult C57Bl/6 mice wer

The reduction of dietary sucrose improves dyslipidemia, adiposity, and insulin secretion in an insulin-resistant rat model.

OBJECTIVE: The purpose of the present work was to investigate whether changes in the type of carbohydrate in the diet are able to improve and/or reverse hyperlipemia, impaired glucose homeostasis, and insulin secretion from beta-cells induced in rats by chronically feeding a high sucrose intake. METHODS: For 30 wk male Wistar rats received a sucrose-rich diet (63% w/w) or a control diet in which sucrose was replaced by starch. After this period, the sucrose-fed animals were randomly divided into two groups: the first group continued with this diet up to 42 wk and the other received the same d

Impact of dietary sucralose and sucrose-sweetened water intake on lipid and glucose metabolism in male mice.

AIMS: Overconsumption of sugar-sweetened beverages (SSBs) is associated with an increased risk of metabolic disorders, including obesity and diabetes. However, accumulating evidence also suggests the potential negative impact of consuming nonnutritive sweeteners (NNSs) on weight and glycaemic control. The metabolic effects of sucralose, the most widely used NNS, remain controversial. This study aimed to compare the impact of intake of dietary sucralose (acceptable daily intake dose, ADI dose) and sucrose-sweetened water (at the same sweetness level) on lipid and glucose metabolism in male mice

Effect of dietary sucrose on the SHR/N-corpulent rat: a new model for insulin-independent diabetes.

A new congenic strain of genetically obese rat, SHR/N-corpulent (cp), was studied. Young male corpulent (cp/cp) and lean (cp/+ or +/+) rats approximately 5 wk of age were fed a diet containing 54% carbohydrate as either sucrose or cooked cornstarch for 9 wk. A phenotype effect was observed with body weight, fasting levels of serum insulin, triglyceride and total cholesterol, levels of serum insulin and glucose after an oral glucose load, and level of urine glucose (corpulent greater than lean), and with systolic blood pressure (corpulent less than lean). Only lean rats were hypertensive. Corpu

High-Sucrose Diet-Induced Reprogramming of Plasma Oxylipin Profiles Reflects Dietary Risk for Gut Barrier Dysfunction.

High-sucrose diet (HSD)-induced alterations in colonic oxylipins have been documented as a key contributor to gut barrier dysfunction. However, the impact of HSD on the systemic oxylipin profiles and their potential link to gut barrier impairment remain largely unknown. Here, we report HSD reprograms plasma oxylipin profiles, with implications for monitoring the risk of HSD-induced gut barrier impairment. In a mouse model, HSD altered plasma oxylipin metabolism, preferentially affecting metabolites enriched in the CYP epoxygenase pathway. Among these, 8(9)-epoxyeicosatrienoic acid [8(9)-EET] w

Endothelial mechanotransduction proteins and vascular function are altered by dietary sucrose supplementation in healthy young male subjects.

KEY POINTS: Mechanotransduction in endothelial cells is a central mechanism in the regulation of vascular tone and vascular remodelling Mechanotransduction and vascular function may be affected by high sugar levels in plasma because of a resulting increase in oxidative stress and increased levels of advanced glycation end-products (AGE). In healthy young subjects, 2 weeks of daily supplementation with 3 × 75 g of sucrose was found to reduce blood flow in response to passive lower leg movement and in response to 12 W of knee extensor exercise. This vascular impairment was paralleled by up-regul

Multi-tissue network analysis reveals the effect of JNK inhibition on dietary sucrose-induced metabolic dysfunction in rats.

Excessive consumption of sucrose, in the form of sugar-sweetened beverages, has been implicated in the pathogenesis of metabolic dysfunction-associated fatty liver disease (MAFLD) and other related metabolic syndromes. The c-Jun N-terminal kinase (JNK) pathway plays a crucial role in response to dietary stressors, and it was demonstrated that the inhibition of the JNK pathway could potentially be used in the treatment of MAFLD. However, the intricate mechanisms underlying these interventions remain incompletely understood given their multifaceted effects across multiple tissues. In this study,

Gene expression and metabolic activity of Streptococcus mutans during exposure to dietary carbohydrates glucose, sucrose, lactose, and xylitol.

Recent RNA sequencing studies have given us a deeper insight into the cariogenic impact of carbohydrate sources in the bacterium Streptococcus mutans, the principal microbial agent in dental caries etiopathogenesis. The process of dental caries development is facilitated by the ability of this bacterium to ferment some carbohydrates into organic acids contributing to a pH decrease in the oral cavity and the demineralization of the hard tissues of the tooth. Furthermore, in dental caries progression, biofilm formation, which starts and ends with free planktonic cells, plays an important role an

High dietary fat and sucrose results in an extensive and time-dependent deterioration in health of multiple physiological systems in mice.

Obesity is associated with metabolic dysfunction, including insulin resistance and hyperinsulinemia, and with disorders such as cardiovascular disease, osteoporosis, and neurodegeneration. Typically, these pathologies are examined in discrete model systems and with limited temporal resolution, and whether these disorders co-occur is therefore unclear. To address this question, here we examined multiple physiological systems in male C57BL/6J mice following prolonged exposure to a high-fat/high-sucrose diet (HFHSD). HFHSD-fed mice rapidly exhibited metabolic alterations, including obesity, hyper
