Ascorbate-induced lipid peroxidation and the binding of [3H]dihydroalprenolol.

Sodium ascorbate addition caused a modest loss in the specific binding of [3H]dihydroalprenolol ([3H]DHA) in a rat cortical membrane preparation. Sodium ascorbate also caused a modest increase in lipid peroxide formation in the membrane preparation. Both the lipid peroxidation and the loss in [3H]DHA binding were greatly potentiated by the addition of low levels of iron (Fe2+). A kinetic analysis indicated that the iron/ascorbate caused a loss in the number of [3H]DHA binding sites but had no effect on the affinity of [3H]DHA for the binding site. Both the lipid peroxidation and the loss of [3

Effects of age and of chronic antidepressant treatment on [3H]tryptamine and [3H]dihydroalprenolol binding to rat cortical membranes.

1. The effects of age and of chronic antidepressant treatment on [3H]tryptamine and [3H]dihydroalprenolol binding site density were measured in brain cortical membranes from male Sprague-Dawley rats. 2. The density but not the affinity of [3H]tryptamine binding sites was increased in 18-month-old rats relative to 3-month-old rats. Neither the density nor the affinity of [3H]dihydroalprenolol binding sites was affected by age. 3. Chronic administration (28 days s.c. via Alzet osmotic minipumps) of tricyclic antidepressant drugs (daily doses: imipramine.HCl, 30 mg kg-1; desipramine.HCl, 10 mg kg

Autoradiographic localization of peripheral benzodiazepine, dihydroalprenolol and arginine vasopressin binding sites in the pituitaries of control, stalk transected and Brattleboro rats.

The autoradiographic distribution of [3H]arginine vasopressin, [3H]spiperone, [3H]GABA, [3H]dihydroalprenolol and the peripheral-type benzodiazepine ligand [3H]Ro5-4864 were examined in the rat pituitary before and after pituitary stalk transection. Stalk transection produced dramatic changes in the cellular architecture of the pars nervosa. Glial fibrillary acidic protein, an astrocyte marker reported in pituicytes, increased after stalk transection, whereas neurofilament, a marker for neuronal innervation, was lost. These structural changes demonstrated a successful stalk transection, permit

The effect of the beta-adrenoreceptor antagonist Ro03-7894 on rat atrial tension development and (-)-[3H]dihydroalprenolol binding to cardiac and lung membranes.

The ability of 1-(5-chloracetylaminobenzfuran-2-yl)-2-isopropylaminoethanol (Ro03-7894) to irreversibly inactivate beta-adrenoreceptors was studied. In isolated rat atria Ro03-7894 (500 microM) depressed and shifted the tension development curve for isoproterenol to the right. After a 2 hour washout period the dose response curve for isoproterenol was further depressed. At a lower dose of Ro03-7894 (50 microM), the isoproterenol dose response curve was also depressed and shifted to the right although after a 2 hour washout, the sensitivity to isoproterenol was restored but the maximum response

Changes in 3H-spiperone, 3H-WB 4101 and 3H-dihydroalprenolol bindings to brain membranes produced by postnatal pretreatment with chlorpromazine in adult rats.

In order to elucidate possible mechanisms of the learning deficit produced by postnatal pretreatment with chlorpromazine (CPZ), changes in catecholamine receptors in the rat brain were investigated. Male neonates of Wistar strain rats were given s.c. 2 mg/kg/day of CPZ for 7 successive days from days 6 to 12 after birth. Effect of the postnatal pretreatment with CPZ on saturation constants for specific bindings of 3H-spiperone, 3H-WB 4101 and 3H-dihydroalprenolol, respectively, in 8 brain regions was investigated at 60 days after birth. Significant decreases in Bmax values of 3H-WB 4101 bindin

Development of beta-adrenergic receptors in normal and mutant mouse cerebellum.

[3H]-dihydroalprenolol was used to examine the development of beta-adrenoceptors in the cerebellum of weaver, reeler, staggerer, and jimpy neurologically mutant mice and their normal counterparts. In normal animals the greatest increase in [3H] binding occurred during the postnatal interval of 10-15 days, and maximum values were obtained at about 3 weeks. Binding was saturable with an apparent dissociation constant of 0.26 nM, and the affinity of [3]-dihydroalprenolol for its receptor did not change significantly during development. [3H]-dihydroalprenolol binding was significantly reduced in w

Beta-adrenergic receptors and adenylate cyclase in hypertrophic and hyperplastic rat salivary glands.

Isoproterenol induces both the secretion of protein and the stimulation of DNA synthesis and growth in rat salivary glands. The specific binding of the labelled beta-adrenergic antagonist [3H]dihydroalprenolol has been used to measure the number of beta-adrenergic receptors in rat parotid glands during isoproterenol-induced growth. Isoproterenol-enlarged glands display no change in the specific binding capacity per gland for [3H]-dihydroalprenolol compared with normal tissue. Catecholamine sensitive adenylate cyclase activity varies independently of the number of specific [3H]dihydroalprenolol

Human lung adrenoreceptors studied by radioligand binding.

1. Direct radioligand-binding techniques have been used to characterize and quantify adrenoreceptors in human peripheral lung tissue removed at thoracotomy from ten patients, nine of whom had evidence of obstructive airways disease. 2. [3H]Dihydroalprenolol was used to characterize beta-adrenoreceptor sites and [3H]prazosin to identify alpha-adrenoreceptor sites. Binding of both ligands showed saturability, high affinity, rapid kinetics, reversibility and stereospecificity. The rank order of agonists and antagonists inhibiting specific binding correlated well with known physiological potencies

Beta adrenergic receptors in lymphocytes and granulocytes from patients with cystic fibrosis.

Intact lymphocytes from patients with cystic fibrosis (CF) produce significantly (P less than 0.001) less adenosine 3':5' cyclic monophosphate (cAMP) than normal lymphocytes in response to isoproterenol (10(-8)-10(-4) M), although the basal cAMP content and the response to prostaglandin E1 are normal. Obligate heterozygotes for CF have significantly (P less than 0.005) reduced cAMP response to isoproterenol as well, suggesting a genetic component in the beta adrenergic deficiency in CF. Comparison of beta-adrenoceptors in single smooth muscle cells and in isolated smooth muscle preparations from the guinea-pig taenia caecum.

Single smooth muscle cells were prepared from guinea-pig taenia caecum. To study the beta-adrenoceptors in the single cells, we observed mechanical responses of the single cells to beta-adrenergic drugs and also tested the binding of a [3H]-labelled drug to the single cells. The single cells showed a graded relaxing response to isoprenaline. The concentration-response curve was shifted in a parallel fashion by propranolol, indicating a competitive antagonism. The pA2 value of propranolol was in agreement with that estimated using the intact tissue. The dissociation constant of [3H]-dihydroalpr

Ectopic beta-adrenergic receptor binding sites. possible molecular basis of aberrant catecholamine responsiveness of an adrenocortical tumor adenylate cyclase.

The molecular basis for the aberrant catecholamine responsiveness of the adenylate cyclase of adrenocortical carcinoma 494 was explored. The adenylate cyclase of this corticosteroid-producing, transplanted, adrenal cancer of the rat was stimulated not only by adrenocorticotropic hormone and fluoride, but also by the beta-adrenergic agonist, isoproterenol. The adenylate cyclase of normal adrenal tissue was unresponsive to isoproterenol. Direct binding studies with the specific high affinity B-adrenergic ligand, (-)[3H]dihydroalprenolol, demonstrated the pressure of 0.094 pmol of specific bindin

Modulation of benzodiazepine receptor, adrenoceptor and muscarinic receptor by diazepam in rat parotid gland.

This study investigated the influence of diazepam on the binding characteristics of adrenoceptor, muscarinic and benzodiazepine receptors in rat parotid gland membrane using a radioligand binding assay. At a concentration of >10(-6)M, diazepam competed with [(3)H]dihydroalprenolol for β-adrenoceptor, but not [(3)H]prazosin for α-adrenoceptor or [(3)H]quinuclidinyl benzilate for muscarinic receptor. Irreversible blockade of beta adrenoreceptors and their recovery in the rat heart and lung in vivo.

The interaction of a bromoacetylated derivative of alprenolol (Alm-CO-CH2Br) with cardiac and lung beta adrenoreceptors was partially characterized. After a short incubation period, the concentration of Alm-CO-CH2Br that inhibited specific [3H]dihydroalprenolol binding by 50% in cardiac and lung membranes was 0.5 and 0.11 microM, respectively. The blockade was time-dependent and Scatchard analysis showed no change in the KD value for specific (-)-[3H]dihydroalprenolol binding but a loss of beta adrenoreceptor content after membrane pretreatment with Alm-CO-CH2Br. The blockade was not reversed

Quantitative autoradiography of beta-adrenergic receptors in rabbit eyes.

Beta-adrenergic receptors were localized and quantified with autoradiographic methods in frozen sections of eyes of albino rabbits using [125I]cyanopindolol and [3H]dihydroalprenolol. Quantification in albino rabbits revealed highest beta 2-adrenoceptor densities in the ciliary body, the corneal epithelium and the corneal endothelium. Moreover, the iris showed significant amounts of specific binding, whereas the retina and sclera are nearly free of these receptors.

Glucocorticoid modulation of beta-adrenergic receptors of cultured rat arterial smooth muscle cells.

Since both glucocorticoids and catecholamines are involved in the regulation of normal blood pressure, we investigated the modulation of beta-adrenergic receptors of cultured rat arterial smooth muscle cells by glucocorticoids. Interaction of beta-adrenergic antagonists with local anaesthetic acceptor sites.

H3-dihydroalprenolol (H3-DHA) binding in isolated smooth muscle cells has been studied. Two different binding sites have been detected: a high affinity (highly specific) and a low affinity (nonspecific) binding site. Local anaesthetics are able to displace H3-DHA from nonspecific sites with a rank order of potency that mirrors their local anaesthetic activity. This finding correlates, at the molecular level, with the local anaesthetic properties of beta-blockers, as concluded from classical pharmacological experiments.

Changes in alpha- and beta-receptor densities in rat brain as a result of treatment with monoamine oxidase inhibiting antidepressants.

The effects of chronic administration of the nonselective monoamine oxidase (MAO) inhibitor, phenelzine, and two selective inhibitors, clorgyline, and pargyline on adrenergic receptor binding and MAO activity were studied in the rat brain. Chronic but not acute administration of both phenelzine and the MAO-A inhibitor, clorgyline, resulted in significant decreases in cortical beta-adrenoreceptor binding ([3H]dihydroalprenolol) of 47 and 24% respectively. In contrast pargyline, which only partially inhibited MAO-A, caused only a nonsignificant (7%) change in rat cortical beta-receptor binding.
