Influence of cAMP on reporter bioassays for dioxin and dioxin-like compounds.

In reporter assays for detection of dioxins, the dioxin-responsive element (DRE) is generally used as a sensor sequence. In several systems, the CYP1A1 promoter containing DREs (DRE(cyp)) is inserted into a part of the long terminal repeat of mouse mammary tumor virus (LTR(MMTV)) to improve sensitivity of assays. We found that DRE(cyp)-LTR(MMTV) responds not only to dioxins and dioxin-like compounds but also to forskolin, a cAMP-elevating agent. This effect was dose-dependent and reproduced by other cAMP-elevating agents including 8-bromo-cAMP and 3-isobutyl-methylxanthine. The cAMP response e

Emissions of dioxins and dioxin-like compounds and incidence of hepatocellular carcinoma in the United States.

Ambient dioxin exposure from industrial sources, excluding exposures from occupations and accidental releases/contamination, may be associated with risk of developing hepatocellular carcinoma (HCC). The objective of this study was to examine the association between county-level ambient dioxin air emissions from industrial sources and HCC risk in the US. We obtained information on 90,359 incident HCC cases diagnosed between 2000 and 2016 from population-based cancer registries across the US in the Surveillance, Epidemiology, and End Results (SEER) database. Dioxin emissions from 1987 to 2007 fr

In utero exposure to dioxins and dioxin-like compounds and anogenital distance in newborns and infants.

BACKGROUND: Anogenital distance in animals is used as a measure of fetal androgen action. Prenatal exposure to dioxins and dioxin-like compounds in rodents causes reproductive changes in male offspring and decreases anogenital distance. OBJECTIVE: We assessed whether in utero exposure to dioxins and dioxin-like compounds adversely influences anogenital distance in newborns and young children (median age, 16 months; range, 1-31 months). METHODS: We measured anogenital distance among participants of the "Rhea" mother-child cohort study in Crete and the Hospital del Mar (HMAR) cohort in Barcelo

Application of a ready-to-use cell sensor for dioxins and dioxin-like compounds screening in foodstuffs.

Dioxins and dioxin-like compounds (DLCs) in foodstuffs are closely related to human health. As China is the largest food-consuming country, there is a potentially large demand for screening bioassays that are rapid, cost-effective and capable of determining dioxins and DLCs in foodstuffs. CBG2.8D is a reporter gene-based recombinant cell sensor that was recently developed for determining dioxin and DLCs in ambient and seafood samples. In this study, we established a bioanalytical method with this ready-to-use cell sensor for the bioanalysis of dioxins and DLCs in different types of meat sample

The burden of cancer attributable to dietary dioxins and dioxin-like compounds exposure in China, 2000-2020.

Dioxin is a typical class of persistent organic pollutants (POPs) that could cause cancer. In China, the contribution of dietary dioxins to the cancer burden remains underexplored. This study evaluates the cancer risk and burden due to dietary dioxins and dioxin-like compounds among Chinese residents from 2000 to 2020. Based on adjustments in China's dioxin policies, the study period was divided into three stages with split years of 2007 and 2014 to estimate the toxic equivalent (TEQ) of dioxins. Participants in dietary surveys conducted in 31 provinces were included. Dietary exposure to dioxi

Biomarkers and bioassays for detecting dioxin-like compounds in the marine environment.

The presence of toxic chemical contaminants in some marine organisms, including those consumed by humans, is well known. Monitoring the levels of such contaminants and their geographic and temporal variability is important for assessing and maintaining the safety of seafood and the health of the marine environment. Chemical analyses are sensitive and specific, but can be expensive and provide little information on the actual or potential biological activity of the contaminants. Biologically-based assays can be used to indicate the presence and potential effects of contaminants in marine animal

Regulation of in vitro human hematopoietic differentiation by dioxin-like compounds.

Certain dioxin-like compounds (DLCs) pose health concerns. However, their impact on human hematopoiesis has not been explored. Role of 2,3,4,7,8-pentachlorodibenzofuran (PeCDF), 3,4,4',5-tetrachlorobiphenyl (PCB81), and 3,3',4,4',5-pentachlorobiphenyl (PCB126) in lineage specification from human cord-blood derived CD34+ hematopoietic stem and progenitor cells (HSPCs) was investigated. We compared these DLCs in relation to 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD). Over a 28-day period, HSPCs were cultured in vitro in the presence of TCDD and DLCs at concentrations ranging from 0.1 to 50 nM. C

Workshop on perinatal exposure to dioxin-like compounds. II. Reproductive effects.

This summary report focuses on current studies on reproductive effects reported at the workshop on Perinatal Exposure to Dioxin-like Compounds and supporting data noted in the discussion. Recent laboratory studies have suggested that altered development (e.g., low birth weight, spontaneous abortion, congenital malformation) and reproductive health (e.g., fertility, sex organ development, reproductive behavior) may be among the most sensitive end points when examining the effects of dioxinlike compounds. Thus, future research should target the reproductive health of both males and females expos

Weight-of-evidence analysis of human exposures to dioxins and dioxin-like compounds and associations with thyroid hormone levels during early development.

Thyroid hormones play a critical role in the proper development of brain function and cell growth. Several epidemiological studies have been conducted to assess potential associations between pre- and post-natal exposure to dioxins or dioxin-like compounds (DLCs) and the levels of circulating thyroid hormones during early development. Dioxins and DLCs include chlorinated dibenzo-p-dioxins, chlorinated dibenzofurans, and mono- and non-ortho polychlorinated biphenyls (PCBs). We identified a total of 23 relevant epidemiological studies (21 cohort studies and 1 case-control study) that measured ex

Polycyclic aromatic hydrocarbons in oysters and sediments from the Yatsushiro Sea, Japan: comparison of potential risks among PAHs, dioxins and dioxin-like compounds in benthic organisms.

Polycyclic aromatic hydrocarbons (PAHs) were analyzed in oysters collected from 18 stations in the Yatsushiro Sea, western Japan. PAHs were detected in all samples analyzed, and the highest concentration (mean 230 ng/g wet weight) was found in oysters from Tanoura Bay. The high molecular weight PAHs to low molecular weight PAHs ratios in oysters from Tanoura Bay were higher than at other stations. Sediment samples collected from 42 stations in Tanoura Bay were analyzed for PAHs to understand their concentrations and distribution. Higher concentrations were found in sediment samples at two stat

Plant lignan secoisolariciresinol suppresses pericardial edema caused by dioxin-like compounds in developing zebrafish: Implications for suppression of morphological abnormalities.

Dioxins and dioxin-like compounds (DLCs) enter the body mainly through diet and cause various toxicological effects through activation of the aryl hydrocarbon receptor (AhR), a ligand activated transcription factor. Some plant extracts and phytochemicals are reported to suppress this transformation. However, most of these reports have been from in vitro experiments and few reports have been from in vivo experiments. In addition, there has been no report of foodstuffs that effectively prevent AhR-associated morphological abnormalities such as deformities caused by dioxins and DLCs in vivo. In t

A20/AN1 zinc-finger proteins positively regulate major latex-like proteins, transporting factors toward dioxin-like compounds in Cucurbita pepo.

The Cucurbitaceae family accumulates dioxin-like compounds in its fruits. We previously showed that A20/AN1 zinc finger protein (ZFP) genes were highly expressed in the zucchini (Cucurbita pepo) subspecies pepo, which accumulates dioxin-like compounds at high concentrations. Transgenic tobacco (Nicotiana tabacum) plants overexpressing A20/AN1 ZFP genes show accumulation of dioxin-like compounds in their upper parts. However, the mechanisms underlying the accumulation of dioxin-like compounds regulated by the A20/AN1 ZFPs remain unclear. Here, we show that A20/AN1 ZFPs positively regulate the e

A rapid and reagent-free bioassay for the detection of dioxin-like compounds and other aryl hydrocarbon receptor (AhR) agonists using autobioluminescent yeast.

An autonomously bioluminescent Saccharomyces cerevisiae BLYAhS bioreporter was developed in this study for the simple and rapid detection of dioxin-like compounds (DLCs) and aryl hydrocarbon receptor (AhR) agonists. This recombinant yeast reporter was based on a synthetic bacterial luciferase reporter gene cassette (lux) that can produce the luciferase as well as the enzymes capable of self-synthesizing the requisite substrates for bioluminescent production from endogenous cellular metabolites. As a result, bioluminescent signal production is generated continuously and autonomously without cel

The Role of Aryl Hydrocarbon Receptor-Dependent Signaling Pathway in the Profibrotic Effects of Dioxin-like Compounds in Human Renal Tubular Epithelial Cells.

The aryl hydrocarbon receptor (AhR) signaling pathway mediates nephrotoxic compound effects on the kidney, although its mechanisms are incompletely understood. Given that renal tubulointerstitial fibrosis is a central pathological feature of progressive kidney diseases, we investigated AhR-induced profibrotic events at the molecular and cellular levels in human renal tubular epithelial cells (HKC). We found that the AhR activation by the potent agonist 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD) promoted epithelial-mesenchymal transition (EMT) and inflammatory responses. This was evidenced by a

Association of prenatal exposure to dioxin-like compounds, polychlorinated biphenyl, and methylmercury with event-related brain potentials in school-aged children: The Hokkaido study.

Previous studies have indicated that prenatal exposure to dioxin-like compounds (DLC) or polychlorinated biphenyl (PCB) has a negative association with neurodevelopment in school-aged children. Event-related brain potentials (ERP) can reveal subtle and specific differences in the modulation of cognitive processes that are assumed when they are associated with lower levels of prenatal exposure to DLC or PCBs. This prospective birth cohort study was conducted to examine the association between prenatal exposure to relatively low levels of DLC, PCB or methylmercury (MeHg), and ERP. A total of 55
