Zoonotic Parasites in Marmosets (Callithrix Spp.) From Southern Brazil: Insights From a One Health Perspective.

BACKGROUND: Currently, six species of Callithrix are frequently observed in anthropized and degraded environments in different regions of Brazil. This occupation favors interactions with humans and increases the risk of infection by parasites with zoonotic potential. This study assessed the occurrence of enteroparasites in free-ranging Callithrix spp. from different municipalities in Paraná state, Brazil, from a One Health approach. METHODS: Faecal samples of 37 specimens of C. penicillata, C. jacchus, and hybrid species were analyzed. RESULTS: The most frequent parasite forms observed inclu

Detection of Spironucleus muris in unpreserved mouse tissue and fecal samples by using a PCR assay.

The purpose of this study was to develop a rapid DNA isolation method and a sensitive and specific PCR assay for detecting Spironucleus muris in mouse tissue and fecal samples. A PCR assay based on the carboxy terminus of the elongation factor 1a gene was developed; the PCR product was confirmed by nucleic acid sequencing and nested PCR. The new PCR assay then was used to test feces from animals that had been screened for S. muris by using direct intestinal examination and histology. The PCR assay was determined to be a more sensitive test than either direct intestinal examination or intestina

Evidence from SSU rRNA phylogeny that Octomitus is a sister lineage to Giardia.

Octomitus intestinalis is a diplomonad flagellate inhabiting the digestive tract of rodents and amphibians. Octomitus is of evolutionary interest because, based on ultrastructural characteristics, it is thought to be closely related to the morphologically derived genus Giardia, and together they have been proposed to make up the Giardiinae. In molecular trees of diplomonads, Giardia is the deepest branching lineage, so identifying a sister group to Giardia that is less derived would be informative. Octomitus is a logical candidate for this position, but unfortunately there are no molecular dat

Comparative biochemistry of Giardia, Hexamita and Spironucleus: Enigmatic diplomonads.

The diplomonad genera are here represented by three highly diverse species, both free-living (Hexamita inflata), and parasitic (Spironucleus vortens and Giardia intestinalis). All three are moderately aerotolerant flagellates, inhabiting environments where O2 tensions are low and fluctuating. Many diplomonads are opportunistic pathogens of avian, terrestrial and aquatic animals. Hexamitids inhabit deep waters and sediments of lakes and marine basins, S. vortens commonly infects the intestinal tract of ornamental fish, particularly of cichlids and cyprinids, and G. intestinalis, the upper intes

Retortamonads from vertebrate hosts share features of anaerobic metabolism and pre-adaptations to parasitism with diplomonads.

Although the mitochondria of extant eukaryotes share a single origin, functionally these organelles diversified to a great extent, reflecting lifestyles of the organisms that host them. In anaerobic protists of the group Metamonada, mitochondria are present in reduced forms (also termed hydrogenosomes or mitosomes) and a complete loss of mitochondrion in Monocercomonoides exilis (Metamonada:Preaxostyla) has also been reported. Within metamonads, retortamonads from the gastrointestinal tract of vertebrates form a sister group to parasitic diplomonads (e.g. Giardia and Spironucleus) and have als

Giardia spp., ten years of parasitological data in the biggest zoo of Latin America.

Giardia spp. (Diplomonadida: Hexamitidae) is an important and widely studied protozoan parasite with worldwide distribution. Nowadays have six described species, and the most important probably is Giardia duodenalis due to the zoonotical potential that some assemblages have. Many studies analysing samples from wild animals have detected Giardia in captive environment, including the zoonotic type. The aim of this study was to determine the prevalence of Giardia sp. in wild captive animals at São Paulo Zoo, using conventional parasitological techniques (direct smear, passive flotation with satur

Interplay between disinfection and the enigmatic diplomonad parasite Spironucleus salmonicida in Atlantic salmon.

The diplomonad parasite Spironucleus salmonicida causes spironucleosis, a systemic infection characterised by internal haemorrhaging, splenomegaly, and granulomatous lesions in multiple organs. Prevention is the primary control strategy, particularly in recirculating aquaculture systems (RAS). This study evaluated the effectiveness of common water-disinfection methods against the parasite. In situ experiments assessed parasite survival following exposure to low- and medium-pressure ultraviolet (UV) radiation at different doses. A subsequent in vivo trial simulated a biosecurity breach in RAS u

Systemic spironucleosis in 2 immunodeficient rhesus macaques (Macaca mulatta).

Spironucleus spp are parasites of fish and terrestrial vertebrates, including mice and turkeys, that rarely cause extraintestinal disease. Two rhesus macaques (Macaca mulatta) were experimentally inoculated with simian immunodeficiency virus mac251. Both progressed to simian acquired immune deficiency syndrome within 1 year of inoculation and developed systemic protozoal infections in addition to common opportunistic infections, including rhesus cytomegalovirus, rhesus lymphocryptovirus, and rhesus adenovirus. In the first case, the protozoa were associated with colitis, multifocal abdominal a

Pathogenesis of mixed infection by Spironucleus sp. and Citrobacter freundii in freshwater angelfish Pterophyllum scalare.

The present study was carried out to identify and describe the pathology of the freshwater angelfish Pterophyllum scalare during chronic mortality in an in-door aquaculture system. Scraping of the integument and gills and the collection of intestinal contents to search for external and internal parasites were performed. Kidneys were collected aseptically for the microbiological analysis and the isolates were subjected to antibiotics to test for susceptibility. Subsequently, necropsy for macroscopic assessment and collection of internal organs for histopathology were performed. The fish exhibit

The redox-active drug metronidazole and thiol-depleting garlic compounds act synergistically in the protist parasite Spironucleus vortens.

Spironucleus vortens is a protozoan parasite associated with significant mortalities in the freshwater angelfish, Pterophyllum scalare. Control of this parasite is especially problematic due to restrictions on the use of the drug of choice, metronidazole (MTZ), on fish farms. Use of garlic (Allium sativum) is undergoing a renaissance following experimental validations of its antimicrobial efficiency. Ajoene ((E,Z)-4,5,9-trithiadodeca-1,6,11-triene 9-oxide), is a stable transformation product of allicin, the primary biologically active component of garlic. In the current study, an ajoene oil cr

Stable transfection of the diplomonad parasite Spironucleus salmonicida.

Eukaryotic microbes are highly diverse, and many lineages remain poorly studied. One such lineage, the diplomonads, a group of binucleate heterotrophic flagellates, has been studied mainly due to the impact of Giardia intestinalis, an intestinal, diarrhea-causing parasite in humans and animals. Here we describe the development of a stable transfection system for use in Spironucleus salmonicida, a diplomonad that causes systemic spironucleosis in salmonid fish. We designed vectors in cassette format carrying epitope tags for localization (3×HA [where HA is hemagglutinin], 2× Escherichia coli Om
