Reactive vaccination in the presence of disease hotspots.

Reactive vaccination has recently been adopted as an outbreak response tool for cholera and other infectious diseases. Owing to the global shortage of oral cholera vaccine, health officials must quickly decide who and where to distribute limited vaccine. Targeted vaccination in transmission hotspots (i.e. areas with high transmission efficiency) may be a potential approach to efficiently allocate vaccine, however its effectiveness will likely be context-dependent. We compared strategies for allocating vaccine across multiple areas with heterogeneous transmission efficiency. We constructed meta

Genes and environment in chronic kidney disease hotspots.

PURPOSE OF REVIEW: Chronic kidney disease (CKD) can cluster in geographic locations or in people of particular genetic ancestries. We explore APOL1 nephropathy and Balkan nephropathy as examples of CKD clustering that illustrate genetics and environment conspiring to cause high rates of kidney disease. Unexplained hotspots of kidney disease in Asia and Central America are then considered from the perspective of potential gene × environment interactions. RECENT FINDINGS: We report on evidence supporting both genes and environment in these CKD hotspots. Differing genetic susceptibility between

Eating Bushmeat Improves Food Security in a Biodiversity and Infectious Disease "Hotspot".

Hunting and consumption of wild animals, colloquially known as "bushmeat," is associated with health trade-offs. Contact with wildlife increases exposure to wildlife-origin zoonotic diseases yet bushmeat is an important nutritional resource in many rural communities. In this study, we test the hypothesis that bushmeat improves food security in communities that hunt and trade bushmeat regularly. We conducted 478 interviews with men and women in six communities near Cross River National Park in Nigeria. We used interview responses to relate prevalence and diversity of bushmeat consumption to hou

Kidney disease hotspots and water balance in a warming world.

PURPOSE OF REVIEW: Geographically localized areas with a high prevalence of kidney disease exist currently in several regions of the world. Although the exact cause is unclear, environmental exposures accelerated by climate change, particularly heat exposure and ground water contamination, are hypothesized as putative risk factors. Aiming to inform investigations of water-related exposures as risk factors for kidney disease, we excavate the history of major water sources in three regions that are described as hotspots of kidney disease: the low-lying coastal regions in El Salvador and Nicaragu

Genetic and Developmental Factors in Chronic Kidney Disease Hotspots.

Chronic kidney disease increasingly is being recognized as an important global public health problem. Interindividual susceptibility to kidney disease is high and likely is dependent on risk modulation through genetics, fetal and early childhood development, environmental circumstances, and comorbidities. Traditionally, the chronic kidney disease burden has been ascribed largely to hypertension and diabetes. Increasingly, evidence is accumulating that nontraditional risk factors may predominate in some regions and populations, contributing to epidemics of kidney disease. Such nontraditional ri

Epidemic Dynamics of Vibrio parahaemolyticus Illness in a Hotspot of Disease Emergence, Galicia, Spain.

Galicia in northwestern Spain has been considered a hotspot for Vibrio parahaemolyticus infections. Infections abruptly emerged in 1998 and, over the next 15 years, were associated with large outbreaks caused by strains belonging to a single clone. We report a recent transition in the epidemiologic pattern in which cases throughout the region have been linked to different and unrelated strains. Global genome-wide phylogenetic analysis revealed that most of the pathogenic strains isolated from infections were associated with globally diverse isolates, indicating frequent episodic introductions

Hotspots and trends of perianal fistula of Crohn's disease: A bibliometric analysis.

BACKGROUND: Perianal fistula is a frequent and severe complication of Crohn disease (CD). With the rising incidence of perianal fistulas in CD (pfCD), research activity in this field has grown substantially. Although several reviews have addressed CD and pfCD, comprehensive analyses summarizing research trends and hotspots in pfCD remain limited. METHOD: Relevant articles and reviews were retrieved from the Web of Science Core Collection database. Citespace (version 6.3.1), Scimago Graphica (version 1.0.42), and VOSviewer (version 1.6.15) were used to systematically analyze data on publicatio

Frontiers and hotspots evolution in anti-inflammatory studies for Alzheimer's disease.

BACKGROUND: Alzheimer's disease (AD) is a neurodegenerative disorder that seriously affects the quality of the elderly's lives worldwide. The main pathological features of AD are amyloid plaques formed by β-amyloid (Aβ) and neuronal fibrillary tangls (NFTs) formed by hyperphosphorylated Tau protein. The formation process of these pathological features is closely related to inflammatory response, so anti-inflammatory treatment has become a potential treatment for AD. In recent years, more and more research has shown that the anti-inflammatory therapy can relieve the symptoms of AD and improve c

Heterogeneity in hotspots: spatio-temporal patterns in neglected parasitic diseases.

Cryptosporidiosis and giardiasis have been recognized by the WHO as 'Neglected Diseases'. Minimal attention has been paid to the spatial and temporal distribution of disease incidence patterns. Using disease notification data, we detected spatio-temporal clusters of cryptosporidiosis and giardiasis across three time periods: (i) 1997-2000, (ii) 2001-2004, (iii) 2005-2008. There was substantial variation in the geographical location and timing of recurrent cryptosporidiosis and giardiasis clusters. Statistically significant (P < 0·05) giardiasis clusters tended to occur in predominantly urban a

Genotype-Phenotype Correlation of GNAS Gene: Review and Disease Management of a Hotspot Mutation.

Defects of the GNAS gene have been mainly associated with pseudohypoparathyroidism Ia. To date, pathogenic missense, frameshift, non-sense and splicing variants have been described in all the 13 exons of the GNAS gene. Of them, a specific mutation, namely the 4 bp deletion c.565_568delGACT, is currently considered a mutation hotspot. Recent articles performed genotype-phenotype correlations in patients with GNAS-related pseudohypoparathyroidism Ia (PHP1a) but a specific focus on this hotspot is still lacking. We reported two cases, from our department, of PHP1a associated with c.565_568delGACT

Plaque vicinity as a hotspot of microglial turnover in a mouse model of Alzheimer's disease.

Microglia, the major immune cells of the brain, are functionally heterogeneous but in vivo functional properties of these cells are rarely studied at single-cell resolution. By using microRNA-9 regulated viral vectors for multicolor labeling and longitudinal in vivo monitoring of individual microglia, we followed their fate in the cortex of healthy adult mice and at the onset of amyloidosis in a mouse model of Alzheimer's disease. In wild-type mice, microglia were rather mobile (16% of the cells migrated at least once in 10-20 days) but had a low turnover as documented by low division and deat

Molecular surveillance reveals a potential hotspot of tick-borne disease in Yakeshi City, Inner Mongolia.

A molecular surveillance of tick-borne diseases was performed in Hulunbuir City, Inner Mongolia. A total of 149 ticks including three species (Ixodes persulcatus, Haemaphysalis concinna, and Dermacentor silvarum) were collected. As many as 11 tick-borne bacterial pathogens were identified in them. Some of them have high positive rates. For example, Candidatus Rickettsia tarasevichiae was detected with a high prevalence of 72.48%, while Candidatus Lariskella sp. was detected in 31.54% of ticks. For both Rickettsia raoultii and Anaplasma phagocytophilum, two distinct genotypes were identified ba

Bibliometric analysis of clinical treatment in Hirschsprung disease: trends and emerging hotspots.

BACKGROUND: Hirschsprung disease (HSCR) is a congenital gastrointestinal malformation defined by the absence of enteric ganglion cells in the distal bowel. Although surgical resection remains the primary treatment, the field has witnessed rapid evolution toward precision minimally invasive strategies, including laparoscopic/robotic-assisted procedures and standardized perioperative protocols. The expanding literature volume necessitates a systematic synthesis of the research landscape. METHODS: We retrieved HSCR clinical treatment publications from Web of Science Core Collection and Scopus (2

Visualizing synaptic disruptions in the release and regulation of dopamine hotspots in Huntington's Disease.

Dopamine neuromodulation is a critical process that facilitates learning, motivation, and motor control. Disruption of these processes has been implicated in several neurodegenerative disorders including Huntington's Disease (HD). While dopaminergic signaling is a therapeutic target for treating physical and psychiatric HD symptoms, the mechanism by which dopaminergic dysfunction occurs during HD is unknown. New tools for the visualization of dopamine dynamics at the spatiotemporal resolution of neuromodulator release (ms) and dopaminergic boutons (µm) provide a richer understanding of how dop
