Development of the genitalia in Drosophila melanogaster.

The imaginal discs of Drosophila melanogaster are an excellent material with which to analyze how signaling pathways and Hox genes control growth and pattern formation. The study of one of these discs, the genital disc, offers, in addition, the possibility of integrating the sex determination pathway into this analysis. This disc, whose growth and shape are sexually dimorphic, gives rise to the genitalia and analia, the more posterior structures of the fruit fly. Male genitalia, which develop from the ninth abdominal segment, and female genitalia, which develop mostly from the eighth one, disp

Modeling Neurodegenerative Disorders in Drosophila melanogaster.

Drosophila melanogaster provides a powerful genetic model system in which to investigate the molecular mechanisms underlying neurodegenerative diseases. In this review, we discuss recent progress in Drosophila modeling Alzheimer's Disease, Parkinson's Disease, Amyotrophic Lateral Sclerosis (ALS), Huntington's Disease, Ataxia Telangiectasia, and neurodegeneration related to mitochondrial dysfunction or traumatic brain injury. We close by discussing recent progress using Drosophila models of neural regeneration and how these are likely to provide critical insights into future treatments for neur

Alzheimer's Disease, Drosophila melanogaster and Polyphenols.

Alzheimer's disease (AD) is an insidious neurological disorder that affects memory, one of the human brain's main cognitive functions. Around 5.2 million Americans currently have AD, and the number threatens to climb to 7 million by 2020. Our native country, Colombia, is no exception with an estimated 260,000 individuals to be affected by AD in 2020. A large, genetically-isolated community in Antioquia, Colombia, with early-onset familial Alzheimer's disease due to a presenilin-1 mutation is ideally suited for the study of molecular mechanisms of AD, and hence accelerate the discovery of new o

Endogenously imprinted genes in Drosophila melanogaster.

Genomic imprinting is an epigenetic state that results from differential processing of chromosomes during gametogenesis and which can cause differential expression of genes depending on the sex of the parent transmitting that gene. In Drosophila, many examples of imprinted marker genes have been documented and imprinting of these genes involves highly conserved epigenetic regulators. However, no endogenously imprinted genes have yet been identified. Here we present a phenotypic and transcriptional analysis of parthenogenetic (gynogenetic) and genotypically identical but sexually produced adult

Polycomb Group Proteins in Drosophila melanogaster.

The development of multicellular organisms relies on the precise establishment and stable inheritance of cell type-specific transcriptional programs without changes to the underlying DNA sequence. Polycomb group (PcG) proteins are evolutionarily conserved epigenetic repressors that govern developmental gene expression by assembling into multiprotein complexes to establish robust and spatiotemporally restricted transcriptional silencing. Since their discovery through the classic Polycomb mutant phenotype in Drosophila melanogaster, PcG proteins have become a central paradigm for understanding e

Gut-brain communication in Drosophila melanogaster.

Over the past decades, significant advancements have transformed our understanding of the gut-brain circuits in Drosophila melanogaster. In this review, we explore how mapping these circuits and signaling pathways has deepened our knowledge of the neural and hormonal pathways that regulate nutrient preference, feeding behavior, metabolism, and other homeostatic behaviors in flies. We summarize the recent breakthroughs in gut-brain communication and highlight how these advancements have provided valuable insights into the complex relationship between the gut and the brain. Finally, we emphasize

Drosophila melanogaster Models of Friedreich's Ataxia.

Friedreich's ataxia (FRDA) is a rare inherited recessive disorder affecting the central and peripheral nervous systems and other extraneural organs such as the heart and pancreas. This incapacitating condition usually manifests in childhood or adolescence, exhibits an irreversible progression that confines the patient to a wheelchair, and leads to early death. FRDA is caused by a reduced level of the nuclear-encoded mitochondrial protein frataxin due to an abnormal GAA triplet repeat expansion in the first intron of the human FXN gene. FXN is evolutionarily conserved, with orthologs in essenti

Quantifying Abdominal Pigmentation in Drosophila melanogaster.

Pigmentation is a morphologically simple but highly variable trait that often has adaptive significance. It has served extensively as a model for understanding the development and evolution of morphological phenotypes. Abdominal pigmentation in Drosophila melanogaster has been particularly useful, allowing researchers to identify the loci that underlie inter- and intraspecific variations in morphology. Hitherto, however, D. melanogaster abdominal pigmentation has been largely assayed qualitatively, through scoring, rather than quantitatively, which limits the forms of statistical analysis that

Origin and establishment of the germline in Drosophila melanogaster.

The continuity of a species depends on germ cells. Germ cells are different from all the other cell types of the body (somatic cells) as they are solely destined to develop into gametes (sperm or egg) to create the next generation. In this review, we will touch on 4 areas of embryonic germ cell development in Drosophila melanogaster: the assembly and function of germplasm, which houses the determinants for germ cell specification and fate and the mitochondria of the next generation; the process of pole cell formation, which will give rise to primordial germ cells (PGCs); the specification of p

Mob4 is essential for spermatogenesis in Drosophila melanogaster.

Gamete formation is essential for sexual reproduction in metazoans. Meiosis in males gives rise to spermatids that must differentiate and individualize into mature sperm. In Drosophila melanogaster, individualization of interconnected spermatids requires the formation of individualization complexes that synchronously move along the sperm bundles. Here, we show that Mob4, a member of the Mps-one binder family, is essential for male fertility but has no detectable role in female fertility. We show that Mob4 is required for proper axonemal structure and its loss leads to male sterility associated

Drosophila melanogaster as a model of systemic dermatophytosis.

BACKGROUND: Dermatophytosis is one of the most common fungal infections worldwide. The distribution of dermatophytes varies across continents, but the genera Trichophyton and Microsporum have emerged as the main isolated agents in humans and animals. OBJECTIVES: To validate Drosophila melanogaster flies as a fast and feasible model to study dermatophytic infections. METHODS: Wild-type (WT) and Toll-deficient D. melanogaster flies were infected by Trichophyton rubrum, T. mentagrophytes, Microsporum canis and Nannizzia gypsea by pricking with a needle previously dipped in inoculum concentratio

Burkholderia thailandensis is virulent in Drosophila melanogaster.

Melioidosis is a serious infectious disease endemic to Southeast Asia and Northern Australia. This disease is caused by the Gram-negative bacterium Burkholderia pseudomallei; Burkholderia thailandensis is a closely-related organism known to be avirulent in humans. B. thailandensis has not previously been used to infect Drosophila melanogaster. We examined the effect of B. thailandensis infection on fly survival, on antimicrobial peptide expression, and on phagocytic cells. In the fruit fly, which possesses only an innate immune system, B. thailandensis is highly virulent, causing rapid death w

Drosophila melanogaster in the study of human neurodegeneration.

Human neurodegenerative diseases are devastating illnesses that predominantly affect elderly people. The majority of the diseases are associated with pathogenic oligomers from misfolded proteins, eventually causing the formation of aggregates and the progressive loss of neurons in the brain and nervous system. Several of these proteinopathies are sporadic and the cause of pathogenesis remains elusive. Heritable forms are associated with genetic defects, suggesting that the affected protein is causally related to disease formation and/or progression. The limitations of human genetics, however,

The transmission of fragmented chromosomes in Drosophila melanogaster.

We investigated the fate of dicentric chromosomes in the mitotic divisions of Drosophila melanogaster. We constructed chromosomes that were not required for viability and that carried P elements with inverted repeats of the target sites (FRTs) for the FLP site-specific recombinase. FLP-mediated unequal sister-chromatid exchange between inverted FRTs produced dicentric chromosomes at a high rate. The fate of the dicentric chromosome was evaluated in the mitotic cells of the male germline. We found that dicentric chromosomes break in mitosis, and the broken fragments can be transmitted. Some of

The protein phosphatases of Drosophila melanogaster and their inhibitors.

Protein phosphatases-1, 2A and 2B have been identified in membrane and soluble fractions of Drosophila melanogaster heads. Similarities between Drosophila and mammalian protein phosphatase-1 included specificity for the beta subunit of phosphorylase kinase, sensitivity to inhibitor-1 and inhibitor-2, inhibition by protamine, retention by heparin-Sepharose and selective interaction with membranes. In addition, an inactive form of protein phosphatase-1, termed protein phosphatase-1I, was detected in the soluble fraction that could be activated by preincubation with MgATP and mammalian glycogen s

Selection for male recombination in Drosophila melanogaster.

Two-way selection for male recombination over seven intervals of the third chromosome in Drosophila melanogaster was practiced for nine generations followed by relaxed selection for five generations. Significant responses in both directions were observed but these mainly occurred in early generations in the low line and in later generations in the high line. Divergence of male recombination frequencies between the two selection lines was not restricted to any specific region but occurred in every measured interval of the chromosome. However, right-arm intervals showed a more pronounced respons

Slmap Is Required For Spermiogenesis in Drosophila melanogaster.

Spermatogenesis is a conserved process across animals, involving the maintenance and differentiation of germ stem cells, haploid spermatid production via meiosis, and morphologically distinct sperm formation during spermiogenesis. We previously demonstrated that Slmap (Sarcolemma-associated protein) knockdown causes male sterility with no sperm in the seminal vesicle. Here, we show that Slmap knockdown in the early Drosophila melanogaster germline led to severe spermiogenesis defects, including aberrant paracrystalline material deposition in both mitochondrial derivatives, fusion of the two de

Regulation of Nucleolar Dominance in Drosophila melanogaster.

In eukaryotic genomes, ribosomal RNA (rRNA) genes exist as tandemly repeated clusters, forming ribosomal DNA (rDNA) loci. Each rDNA locus typically contains hundreds of rRNA genes to meet the high demand of ribosome biogenesis. Nucleolar dominance is a phenomenon whereby individual rDNA loci are entirely silenced or transcribed, and is believed to be a mechanism to control rRNA dosage. Nucleolar dominance was originally noted to occur in interspecies hybrids, and has been shown to occur within a species (i.e, nonhybrid context). However, studying nucleolar dominance within a species has been c
