Dynamic light scattering spectroscopy and the Centiloid scale.

Dynamic light scattering (DLS) spectroscopy measures the Brownian movement of particles. Fifteen patients underwent DLS measurement and positron emission tomography amyloid imaging. The DLS measurement is noninvasive, quantitative, and inexpensive, with immediate results. We report a significant correlation between DLS measurements and Centiloid values.

Earlier diagnosis of Alzheimer's disease by dynamic light scattering spectroscopy.

Dynamic light scattering (DLS) spectroscopy measures changes in the Brownian movement of particles at the molecular level. Since the retina of the eye is a neural tissue and an outgrowth of the brain, a clinical instrument was developed capable of making DLS measurements from the retina in patients with mild cognitive impairment undergoing positron emission tomography nuclide imaging for the presence of cerebral amyloid.

Particle growth of hybrid materials followed by dynamic light scattering.

The hydrolysis of distannylated compounds in which the tin atoms are linked by an organic spacer has been studied under microemulsion conditions using dynamic light scattering and infrared spectroscopy. The experiments provided evidence that the growth of hybrid material particles occurs in the aqueous phase, outside the organic phase of the microemulsion. The growth rates of the particles were found to be strongly dependent on the nature of the spacers, a polymethylene chain inducing the fastest process. This different behavior was explained by a slower condensation process rather than a slow

Dynamic light scattering studies of porcine submaxillary mucin fractions in dilute solution at intermediate scattering vectors.

We report dynamic light scattering measurements over a wide range of scattering vectors for fractionated samples of porcine submaxillary mucin (PSM) glycoproteins in two different solvents: 0.1M NaCl, and 6M GdnHCl. The relaxation spectrum has been successfully resolved into a slow mode corresponding to pure translational diffusion and a fast mode containing information on the relaxation times for intramolecular motion. Analysis of the slow mode permits a light scattering evaluation of the polydispersity of these high molecular weight mucin glycoprotein fractions. Determination of the longest

Dynamic light scattering of the eye in the diagnosis of Alzheimer's disease.

The early detection of Alzheimer's disease, before symptoms have appeared, is integral to the development of effective treatments. Dynamic light scattering spectroscopy measures the Brownian movement of proteins at the molecular level. This technique may facilitate early Alzheimer's disease diagnosis and the discovery of pharmaceuticals that may prevent symptom development.

Resolving measurement artifacts of field-flow fractionation coupled to conventional dynamic light scattering detection by using phase and spatially-resolved dynamic light scattering to improve size accuracy.

The use of dynamic light scattering (DLS) as an online detector in asymmetrical flow field-flow fractionation (AF4 ‒ DLS) for size characterization is known to introduce measurement bias for particles with hydrodynamic size exceeding ∼100 nm. This study demonstrates that the accuracy of hydrodynamic size measurements in AF4 ‒ DLS is significantly improved by applying advanced phase and spatially-resolved DLS (PhaSR-DLS) detection. PhaSR-DLS overcomes two limitations of conventional DLS: (i) overestimation of particle size due to 'number fluctuations' at low particle concentrations, causing 'U-

Zika Virus NS1 Protein Detection Using Gold Nanoparticle-Assisted Dynamic Light Scattering.

The Zika virus (ZIKV) is a global health threat due to its rapid spread and severe health implications, including congenital abnormalities and neurological complications. Differentiating ZIKV from other arboviruses such as dengue virus (DENV) is crucial for effective diagnosis and treatment. This study presents the development of a biosensor for detecting the ZIKV non-structural protein 1 (NS1) using gold nanoparticles (AuNPs) functionalized with monoclonal antibodies employing dynamic light scattering (DLS). The biosensor named ZINS1-mAb-AuNP exhibited specific binding to the ZIKV NS1 protein

Probing intracellular dynamics of prostate cells with coherence-gated dynamic light scattering.

Cells are dynamic biomaterials that continuously modulate their mechanical properties and internal dynamics. This ability is increasingly recognized as a potential biophysical marker for diagnosing and predicting pathological states. We show that the collective intracellular dynamics of prostate cells can be characterized in a passive, non-invasive, contactless manner, using the interferometric amplification of light scattered by individual cells within a localized sensing volume. By employing phenotypically different prostate cell models, namely RWPE 1, C4-2B, and PC-3, we demonstrate that co

Dynamic Light Scattering Microrheology of Phase-Separated Poly(vinyl) Alcohol-Phytagel Blends.

In this investigation, we explored the microrheological characteristics of dilute hydrogels composed exclusively of Poly(vinyl) alcohol (PVA), Phytagel (PHY), and a blend of the two in varying concentrations. Each of these polymers has established applications in the biomedical field, such as drug delivery and lens drops. This study involved varying the sample concentrations from 0.15% to 0.3% (w/w) to assess how the concentration influenced the observed rheological response. Two probe sizes were employed to examine the impact of the size and verify the continuity hypothesis. The use of two po

Postoperative Hemodynamic Index Measurement With Miniaturized Dynamic Light Scattering Predicts Colorectal Anastomotic Healing.

INTRODUCTION: Perioperative bowel perfusion (local hemodynamic index [LHI]) was measured with a miniaturized dynamic light scattering (mDLS) device, aiming to determine whether anastomotic perfusion correlates with the anastomotic healing process and whether LHI measurement assists in the detection of anastomotic leakage (AL) in colorectal surgery. METHODS: A partial colectomy was performed in 21 male Wistar rats. Colonic and anastomotic LHIs were recorded during operation. On postoperative day (POD) 3, the rats were examined for AL manifestations. Anastomotic LHI was recorded before determin

Nanoscale and mesoscopic protein assemblies in lysozyme-NaCl solutions probed by dynamic light scattering.

Dynamic light scattering was used to analyze nanoscale and mesoscopic protein-associated objects formed in lysozyme-NaCl solutions under crystallization conditions. The study focuses on the sequence of particle-size populations detected after salt addition and on their relation to the initial state of the protein solution. NaCl reproducibly induced a small population with hydrodynamic diameters of 5.6-6.5 nm, which is consistent with nanoscale oligomeric lysozyme assemblies and with previous SAXS/SANS evidence for dimers and octamers in crystallization solutions. Mesoscopic particles in the te

Antibody orientation drives sensitivity in SARS-CoV-2 detection using dynamic light scattering biosensors.

The performance of nanoparticle-based biosensors strongly depends on the orientation and density of immobilized antibodies, yet these parameters remain underexplored in platforms based on dynamic light scattering (DLS). This study investigates how the length of molecular crosslinkers influences antibody orientation, surface coverage, and biosensor sensitivity for SARS-CoV-2 detection. The impact of crosslinker spacer length on the analytical performance of 53 nm-gold nanoparticles (AuNPs) functionalized with polyclonal antibodies (pAb) was evaluated using both the spike protein (S Ptn) and int

Click-Chemistry-Enhanced Dynamic Light Scattering Immunoassay for Ultrasensitive GFAP Detection in Traumatic Brain Injury.

Traumatic brain injury (TBI) requires a rapid and objective assessment; however, current diagnostic methodologies are constrained by subjectivity, reliance on imaging techniques, and insufficient sensitivity for the early detection of biomarkers. In this study, we present a click-chemistry-enhanced dynamic light scattering (DLS) immunoassay for the ultrasensitive detection of glial fibrillary acidic protein (GFAP), a clinically significant blood biomarker for brain injury. Unlike conventional DLS immunoassays, which rely on weak antigen-antibody interactions to drive DLS probe aggregation, the

Dynamic Light Scattering Analysis for the Determination of the Particle Size of Iron-Carbohydrate Complexes.

Intravenously administered iron-carbohydrate nanoparticle complexes are widely used to treat iron deficiency. This class includes several structurally heterogeneous nanoparticle complexes, which exhibit varying sensitivity to the conditions required for the methodologies available to physicochemically characterize these agents. Currently, the critical quality attributes of iron-carbohydrate complexes have not been fully established. Dynamic light scattering (DLS) has emerged as a fundamental method to determine intact particle size and distribution. However, challenges still remain regarding t

The study of amorphous aggregation of tobacco mosaic virus coat protein by dynamic light scattering.

The kinetics of heat-induced and cetyltrimethylammonium bromide induced amorphous aggregation of tobacco mosaic virus coat protein in Na(+)/Na(+) phosphate buffer, pH 8.0, have been studied using dynamic light scattering. In the case of thermal aggregation (52 degrees C) the character of the dependence of the hydrodynamic radius (R(h)) on time indicates that at certain instant the population of aggregates is split into two components. The size of the aggregates of one kind remains practically constant in time, whereas the size of aggregates of other kind increases monotonously in time reaching

Minimal structural perturbation of Histatin 5 in crowded environments: Insights from small-angle X-ray scattering, dynamic light scattering, and computer simulations.

The structure and dynamics of intrinsically disordered proteins (IDPs) are malleable to solution conditions. Considerable effort has been devoted to understanding the effects of molecular crowding on these properties. Polymer-based crowders, such as polyethylene glycol (PEG) and Ficoll®, are commonly used to simulate the intracellular environment by replicating the high concentration of macromolecules. In this study, we examine the impact of crowding on the IDP Histatin 5 using PEG of various molecular weights and Ficoll® PM 70. Small-angle X-ray scattering (SAXS) reveals minimal effects on th

Idiosyncratic behaviour of pegylated lipid nanoparticles used in RNA delivery: A dynamic light scattering study.

Dynamic light scattering (DLS) is commonly employed to estimate the hydrodynamic radius of particles (RH) and less commonly to assess colloidal stability. Here we suggest DLS can also be used to study the interface between the particle and the surrounding water. DLS RH measurement have two sources of systematic error. Attraction and repulsion between particles alter the apparent RH in concentration dependent manner referred to as the interaction parameter (kD). The second systematic error, which is not concentration dependent, is observed as a change in the calculated hydrodynamic radius at in

Measurement of the Translational Diffusion Coefficient and Hydrodynamic Radius of Proteins by Dynamic Light Scattering.

Diffusion is a fundamental process in biological systems that governs the molecular collisions driving biochemical reactions and membrane and transport. Measurement of the diffusion coefficient and application of the Stokes-Einstein equation produces the hydrodynamic radius, which is a commonly used gauge of particle size. Additionally, measurement of the diffusion coefficient and the sedimentation coefficient, and application of the Svedberg equation, yields the molecular weight, which is particularly useful in the characterization of very large macromolecules. Dynamic light scattering (DLS)
