Essential functions of mosquito ecdysone importers in development and reproduction.

The primary insect steroid hormone ecdysone requires a membrane transporter to enter its target cells. Although an organic anion-transporting polypeptide (OATP) named Ecdysone Importer (EcI) serves this role in the fruit fly Drosophila melanogaster and most likely in other arthropod species, this highly conserved transporter is apparently missing in mosquitoes. Here we report three additional OATPs that facilitate cellular incorporation of ecdysone in Drosophila and the yellow fever mosquito Aedes aegypti. These additional ecdysone importers (EcI-2, -3, and -4) are dispensable for development

Stage- and cell-specific expression of ecdysone receptors and ecdysone-induced transcription factors during midgut remodeling in the yellow fever mosquito, Aedes aegypti.

In insects, especially in mosquitoes that are adult blood feeders, midgut remodeling is an important event during metamorphosis. It involves two processes viz., programmed cell death (PCD) of larval cells, and proliferation and differentiation of imaginal cells to form pupal/adult midgut. These processes are regulated by 20-hydroxyecdysone (20E) and juvenile hormone (JH), but the signaling mechanisms, which trigger specific changes remain poorly understood. Here, we report stage- and cell-specific expression of ecydone receptor (EcR), ultraspiracle (USP), broad (Br), E75B and hormone receptor

Effects of the Ecdysone Receptor on the Regulation of Reproduction in Coccinella septempunctata.

The effects of the gene encoding the ecdysone receptor (EcR) on the reproduction of the ladybug Coccinella septempunctata was evaluated. EcR transcription was measured by quantitative real-time PCR in ladybug adults reared on artificial diets with and without 20-hydroxyecdysone (20E). EcR expression levels in 5 d old male and female ladybugs supplied with the 20E-amended artificial diet were lower than expression levels in ladybugs reared on an artificial diet lacking 20E. However, EcR expression levels in 10 d old ladybugs supplied with the 20E artificial diet were higher than those lacking 2

Genetic and Molecular Regulation of Ecdysone Biosynthesis During Insect Metamorphosis.

Steroid hormones govern diverse physiological processes during juvenile-to-adult development across vertebrates and invertebrates, ranging from pubertal initiation in mammals to metamorphosis in insects. Ecdysteroids, the collective name for many moulting hormones in insects, act as master effectors driving metamorphic development. Their biosynthesis begins with dietary cholesterol as the direct precursor (phytosterols are converted as needed) and is driven by a series of enzymatic reactions catalyzed by the evolutionarily conserved Halloween genes to produce ecdysone, which is ultimately conv

polished rice mediates ecdysone-dependent control of Drosophila embryonic organogenesis.

In many animals, progression of developmental stages is temporally controlled by steroid hormones. In Drosophila, the level of ecdysone titer oscillates and developmental stage transitions, such as larval molting and metamorphosis, are induced at each of ecdysone peaks. Ecdysone titer also peaks at the stage of mid-embryogenesis and the embryonic ecdysone is necessary for morphogenesis of several organs, although the regulatory mechanisms of embryonic organogenesis dependent on ecdysone signaling are still open questions. In this study, we find that absence or interruption of embryonic ecdyson

Ecdysone Elicits Chronic Renal Impairment via Mineralocorticoid-Like Pathogenic Activities.

BACKGROUND/AIMS: Ecdysteroids are steroidal insect molting hormones that also exist in herbs. Ecdysteroid-containing adaptogens have been popularly used to improve well-being and by bodybuilders for muscle growth. However, the use of ecdysone in mammals is also associated with kidney growth and enlargement, indications of disturbed kidney homeostasis. The underlying pathogenic mechanism remains to be clarified. METHODS: Virtual screening tools were employed to identify compounds that are homologous to ecdysone and to predict putative ecdysone-interacting proteins. The kidney effect of ecdyson

Molecular evidence for a functional ecdysone signaling system in Brugia malayi.

BACKGROUND: Filarial nematodes, including Brugia malayi, the causative agent of lymphatic filariasis, undergo molting in both arthropod and mammalian hosts to complete their life cycles. An understanding of how these parasites cross developmental checkpoints may reveal potential targets for intervention. Pharmacological evidence suggests that ecdysteroids play a role in parasitic nematode molting and fertility although their specific function remains unknown. In insects, ecdysone triggers molting through the activation of the ecdysone receptor: a heterodimer of EcR (ecdysone receptor) and USP

On the control of ecdysone biosynthesis by the central nervous system of blowfly larvae.

Ecdysone was found to be the major secreted steroid of ring glands dissected from blowfly larvae and incubated in vitro. Other secretory products such as 3-dehydroecdysone and 20-deoxy-makisterone A could not be detected when the glands were labelled with tritiated cholesterol. Ecdysone synthesis and secretion were found to be tightly coupled. The highest rate of secretion was observed a few hours before pupariation. In vitro, the rate of ecdysone secretion by ring glands was affected significantly by coincubation with the central nervous system (CNS). Modulating effects from the CNS to the gl

Ectopic expression of ecdysone oxidase impairs tissue degeneration in Bombyx mori.

Metamorphosis in insects includes a series of programmed tissue histolysis and remolding processes that are controlled by two major classes of hormones, juvenile hormones and ecdysteroids. Precise pulses of ecdysteroids (the most active ecdysteroid is 20-hydroxyecdysone, 20E), are regulated by both biosynthesis and metabolism. In this study, we show that ecdysone oxidase (EO), a 20E inactivation enzyme, expresses predominantly in the midgut during the early pupal stage in the lepidopteran model insect, Bombyx mori. Depletion of BmEO using the transgenic CRISPR/Cas9 (clustered regularly intersp

Ecdysone signaling and transcript signature in Drosophila cells resistant against methoxyfenozide.

Methoxyfenozide (RH-2485) is a non-steroidal ecdysteroid agonist with a dibenzoylhydrazine structure, representing a group used as novel biorational insecticides in the control of insect pests. Here we report on the selection of Drosophila melanogaster S2 cells for resistance to inhibition of cell proliferation by methoxyfenozide by ∼ 1000-fold over 4 months. Cells were exposed to gradually increasing concentrations of methoxyfenozide and selected out based on the ecdysteroid-sensitive response for cell proliferation. In the resistant cells, the ecdysteroid receptor (EcR/USP) complex was no lo

Induction of breast and lung neoplastic lesions in mice by alpha-ecdysone.

Alpha ecdysone is naturally present in the leaves of some plants, including the carcinogenic plant bracken fern (Pteris aquilina). Feeding mice with alpha-ecdysone at a dose level of 3 ppm/25 g body weight, 2 times per week for 22 months induced a significant increase in the body weight of the animals. Breast and lung tumors developed in the experimental mice. Other pathological changes such as fibrocystic disease of the breast and bronchocentric granulomas appeared in these animals. The results of this study proved that alpha-ecdysone may be one of the active carcinogens in bracken fern plant

Ecdysone receptor autonomously controls germ cell differentiation in the Drosophila ovary.

BACKGROUND: Brucellosis is a zoonotic disease that causes substantial public health problems and endangers the development of animal husbandry in endemic areas. Early diagnosis of infected animals and humans is a crucial step in reducing the incidence of brucellosis. In this study, we designed different combinations of Brucella major outer membrane proteins (omps) including omp10, omp16, omp19, omp25, omp31 and BP26 as antigens and evaluated their efficiency in serodiagnosis for brucellosis. The efficiency assay was conducted using the method of indirect enzyme-linked immunosorbent assay (iELI

Ecdysone Receptor autonomously controls germ cell differentiation in the Drosophila ovary.

In the Drosophila ovary, the steroid hormone ecdysone controls germline stem cell (GSC) maintenance and germ cell differentiation. Prior studies demonstrated that ecdysone regulates germ cell differentiation non-autonomously via the nuclear receptor Ecdysone Receptor (EcR) in ovarian somatic cells. Although EcR is also expressed in GSCs and their differentiating daughters, potential direct roles for EcR in GSCs independent of the soma have not been examined. Here, we demonstrate that EcR functions autonomously in GSCs and cystoblasts to control germline differentiation. While depletion of EcR

The ecdysone receptor regulates several key physiological factors in Anopheles funestus.

BACKGROUND: Malaria is a devastating disease, transmitted by female Anopheles mosquitoes infected with Plasmodium parasites. Current insecticide-based strategies exist to control the spread of malaria by targeting vectors. However, the increase in insecticide resistance in vector populations hinder the efficacy of these methods. It is, therefore, essential to develop novel vector control methods that efficiently target transmission reducing factors such as vector density and competence. A possible vector control candidate gene, the ecdysone receptor, regulates longevity, reproduction, immunity

β-ecdysone promotes osteogenic differentiation of bone marrow mesenchymal stem cells.

BACKGROUND: β-ecdysone (βEcd) has numerous pharmacological effects, although its role in the osteogenic differentiation of bone marrow mesenchymal stem cells (BMSCs) has not yet been explored. METHODS: In cell experiments, BMSCs were induced to differentiate by osteogenic induction medium (OIM) or βEcd. In animal experiments, an osteonecrosis of the femoral head (ONFH) rat model was established using lipopolysaccharide plus methylprednisolone and treating the rats with βEcd. The osteogenic differentiation capacity of human BMSCs (hBMSCs) was analyzed by alkaline phosphatase and alizarin red S

Mutations of a Drosophila NPC1 gene confer sterol and ecdysone metabolic defects.

The molecular mechanisms by which dietary cholesterol is trafficked within cells are poorly understood. Previous work indicates that the NPC1 family of proteins plays an important role in this process, although the precise functions performed by this protein family remain elusive. We have taken a genetic approach to further explore the NPC1 family in the fruit fly Drosophila melanogaster. The Drosophila genome encodes two NPC1 homologs, designated NPC1a and NPC1b, that exhibit 42% and 35% identity to the human NPC1 protein, respectively. Here we describe the results of mutational analysis of t

SOX12 and DELTA mediate ecdysone-driven adult neural cell proliferation during metamorphosis.

During metamorphic development from larva to adult, insect brains exhibit cell proliferation; however, the molecular mechanisms governing this process remain poorly understood. Employing the lepidopteran insect Helicoverpa armigera (cotton bollworm), an agricultural pest, as a model system, this study demonstrates that the steroid hormone 20-hydroxyecdysone (20E) regulates neural cell proliferation during imaginal brain development by upregulating the expression of SOX12 and DELTA via nuclear receptor EcRA. SOX12 exhibits significantly elevated expression and is localized in the brain during m

Ecdysone regulates phagocytic cell fate of epithelial cells in developing Drosophila eggs.

Acquisition of nonprofessional phagocytic cell fate plays an important role in sculpting functional metazoan organs and maintaining overall tissue homeostasis. Though physiologically highly relevant, how the normal epithelial cells acquire phagocytic fate is still mostly unclear. We have employed the Drosophila ovary model to demonstrate that the classical ecdysone signaling in the somatic epithelial follicle cells (AFCs) aids the removal of germline nurse cells (NCs) in late oogenesis. Our live-cell imaging data reveal a novel phenomenon wherein collective behavior of 4-5 AFCs is required for

Systemic positive feedback regulation of ecdysone biosynthesis via sugar metabolism in insects.

Insects are recognized as the most diverse species in the world. They are classified into three separate groups based on their developmental characteristics: ametabola, hemimetabola, and holometabola. Holometabolous insects have attracted the attention of many developmental biologists because of their dramatic metamorphosis as a key model for studying regulatory mechanisms of development. Central to this process is ecdysone, a steroid hormone that governs insect growth and maturation. Extensive research has clarified ecdysone's structure, functions, and signaling pathways. In particular, the r

NlCYP417A1 is a critical regulator of molting by governing ecdysone homeostasis in the brown planthopper.

BACKGROUND: The brown planthopper (BPH), Nilaparvata lugens, is a destructive rice pest across Asia. Suppressing insect development is enduring goals in insect pest management, underpinning the action of existing insecticides and guiding the design of new ones. Insect molting, which is tightly regulated by ecdysteroid signaling, is essential for insect growth and development. Cytochrome P450 (CYP) enzymes play critical roles in ecdysteroid biosynthesis, and identifying P450 genes involved in this process may provide new insights into insect developmental regulation and potential targets for pe
