Echinostoma chankensis nom. nov., other Echinostoma spp. and Isthmiophora hortensis in East Asia: morphology, molecular data and phylogeny within Echinostomatidae.

Life cycles, and morphological and molecular data were obtained for Echinostoma chankensis nom. nov., Echinostoma cinetorchis, Echinostoma miyagawai and Isthmiophora hortensis from East Asia. It was established that, based on both life cycle and morphology data, one of the trematodes is identical to the worms designated as Euparyphium amurensis. Genetic data showed that this trematode belongs to Echinostoma. The complex data on biological, morphological and genetic characterizations establish that the distribution of the morphologically similar species, I. hortensis and Isthmiophora melis, in

Characterization of Echinostoma revolutum and Echinostoma robustum from ducks in Bangladesh based on morphology, nuclear ribosomal ITS2 and mitochondrial nad1 sequences.

Precise discrimination of Echinostoma species within the 'revolutum' group is quite difficult because of their morphological similarities. The objective of this study was to precisely characterize the echinostomes of ducks from Bangladesh based on both morphological and molecular characteristics. Two Echinostoma species were identified: E. revolutum and E. robustum. In the phylogenetic trees (ITS2 and nad1), E. revolutum and E. robustum belonged to their respective Eurasian clade, which is distinct from the American clade. These results suggest that both species have two distinct and geographi

Evaluation of praziquantel effects on Echinostoma paraensei ultrastructure.

Echinostomiasis is a food-borne, intestinal, zoonotic, snail-mediated helminthiasis caused by digenean trematodes of the family Echinostomatidae with seven species of the genus Echinostoma infecting humans or domestic and wildlife animals. Echinostoma paraensei is a peristomic 37-collar-spined echinostome belonging to the "revolutum group". Praziquantel (PZQ) is the drug of choice for treatment and control of human schistosomiasis and food-borne trematodiasis. In the present study we used scanning and transmission electron microscopy to further elucidate the trematocidal effect of PZQ on adult

In vitro and in vivo encystment of the cercariae of Echinostoma caproni.

In vitro and in vivo studies were conducted on the cercariae of Echinostoma caproni. Of the 15 media tried, 2 resulted in effective in vitro encystment in petri dish cultures maintained at 23 +/- 1 C. They were a Locke's--artificial springwater (ASW) (1:1) medium (67% encystment) and a Biomphalaria glabrata embryonic cell line medium (23% encystment). To obtain large numbers of in vitro--formed cysts, finger bowl cultures containing 40 ml of the Locke's-ASW (1:1) medium were used at 23 +/- 1 C. Of 3,000 cercariae tested, 1,890 (63%) were encysted in this medium by 48 hr. Most of these cysts lo

Auto-infection by Echinostoma spp. cercariae in Helisoma anceps.

Auto-infection is a life history strategy used by many parasitic organisms, including digenetic trematodes. The process of autoinfection most frequently involves the transfer of a life cycle stage of the parasite from one site to another inside the same host, usually accompanied by morphological transformation. Moreover, among trematodes, the stage being transferred may also move from one host to another in completing the life cycle, i.e., an indirect cycle. Echinostoma spp. parasites offer the opportunity to study auto-infection because they utilize gastropods as both first and second interme

Ultrastructural observations on the redia of Echinostoma revolutum (Froelich, 1802).

E. revolutum redia digestive and excretory system ultrastructure, as well as body wall tegument were studied in TEM. The body tegument protrudes in short microvilli and contains uni- and multiciliate sensory endings. The anterior part of the digestive system (pharynx, short oesophagus) is lined with body tegument, the caecum being covered with a flat epithelium with sporadic microvilli. Oesophageal gland cells are filled with minute electron-dense granules. The excretory capillaries joined to large flame cells are composed of spirally wrapped cells, in the cytoplasm of which there are clusters

Eating habits associated with Echinostoma malayanum infections in the Philippines.

A survey of 61 residents belonging to 12 pre-selected families (having at least one member positive for echinostomiasis malayanum) from Barangay Malibago, Echague, Isabela (northern Luzon) suggested that infections with Echinostoma malayanum follow a "familial trend". The parasite is endemic because the raw ingestion of Lymnaea (Bullastra) cumingiana, the second intermediate host in the Philippines, is a learned habit passed down from one generation to the next. A questionnaire on eating habits revealed that Lymnaea (Bullastra) cumingiana or "birabid" was usually prepared raw or half-cooked af

A case of Echinostoma cinetorchis (Trematoda: Echinostomatidae) infection diagnosed by colonoscopy.

Human cases of echinostomiasis have been sporadically diagnosed by extracting worms in the endoscopy in Korea and Japan. Most of these were caused by Echinostoma hortense infection. However, in the present study, we detected 2 live worms of Echinostoma cinetorchis in the ascending colon of a Korean man (68-year old) admitted to the Gyeongsang National University Hospital with complaint of intermittent right lower quadrant abdominal pain for 5 days. Under colonoscopy, 1 worm was found attached on the edematous and hyperemic mucosal surface of the proximal ascending colon and the other was detec

Activity of OZ78 analogues against Fasciola hepatica and Echinostoma caproni.

The rapid spread of triclabendazole resistance in veterinary medicine is an important motivation for the discovery and development of novel fasciocidal drugs. The aim of this study was to characterize the fasciocidal properties of 1,2,4,5-tetraoxane (MT04 and MT14) and 1,2,4-trioxane (ST16 and ST28) analogues of the fasciocidal drug candidate OZ78, a 1,2,4-trioxolane. Dose response relationships were determined against juvenile and adult Fasciola hepatica in rats and Echinostoma caproni in mice. The temporal effects of MT04, MT14, ST16, and ST28 compared to OZ78 on the viability of F. hepatica

Animal Models for Echinostoma malayanum Infection: Worm Recovery and Some Pathology.

Echinostomes are intestinal trematodes that infect a wide range of vertebrate hosts, including humans, in their adult stage and also parasitize numerous invertebrate and cold-blooded vertebrate hosts in their larval stages. The purpose of this study was to compare Echinostoma malayanum parasite growth, including worm recovery, body size of adult worms, eggs per worm, eggs per gram of feces, and pathological changes in the small intestine of experimental animals. In this study, 6-8-week-old male hamsters, rats, mice, and gerbils were infected with echinostome metacercariae and then sacrificed a

Temperature affects the toxicity of pesticides to cercariae of the trematode Echinostoma trivolvis.

Global climate change is predicted to have significant impacts on ecological interactions such as host-parasite relationships. Increased temperatures may also interact with other anthropogenic stressors, such as chemical contaminants, to exacerbate or reduce parasite transmission. However, studies on the effects of pesticides on non-target species are typically conducted at one standard temperature, despite the toxicity of many agrochemicals being temperature-dependent. Furthermore, most studies assessing the effects of temperature on pesticide toxicity have been conducted on host organisms, l

Evaluation of Echinostoma liei worm, metacercaria and redia antigens for schistosomiasis control.

While chemotherepeutic drugs, such as praziquantel, oxamniquine and metrifonate, are currently considered safe and effective drugs for schistosomiasis treatment, reinfection occurs frequently after drug treatment. Thus, a vaccine is sought to provide long-term treatment. Antigens from worm, metacercaria and redia of Echinostoma liei (E. liei) were purified using CNBr-activated Sepharose column, then used for immunization of mice prior to infection with Schistosomiasis mansoni. Worm burden, hepatic and intestinal eggs and oogram count was significantly reduced and that was reflected in normaliz

Echinostoma ilocanum Infection in Two Residents of Savannakhet Province, Lao PDR.

Adult specimens of Echinostoma ilocanum (Garrison, 1908) Odhner, 1911 (Trematoda: Echinostomatidae) were recovered from 2 riparian people who resided along the Mekong River in Savannakhet Province, Lao PDR. In fecal examinations done by the Kato-Katz technique, they revealed echinostome eggs together with eggs of Opisthorchis viverrini (and minute intestinal fluke eggs) and hookworms. To recover the adult flukes, they were treated with praziquantel 30-40 mg/kg in a single dose and purged with magnesium salts. A total of 658 adult fluke specimens were recovered from the 2 people; 456 from case

Infectivity, growth and distribution of preovigerous adults of Echinostoma caproni in ICR mice.

All 23 6-8-week old outbred female ICR mice, each fed 100 metacercarial cysts of Echinostoma caproni, were infected on days 2, 4, 6, and 8 postinfection (PI) with a mean of 45 worms/host. To examine worm distribution in the mouse, the small intestine was divided into five equal sections (segments 1-5 beginning with the pylorus). Worms were found mainly in segments 3 and 4 and were never found in segment 1. Mean body area of worms fixed in 10% neutral buffered formalin (NBF), hot alcohol-formalin-acetic acid (AFA) and aceto-carmine increased slowly until day 4 and then rapidly until day 8 PI. T
