Efferocytosis in atherosclerosis.

Cardiovascular disease is the leading cause of death worldwide, and it commonly results from atherosclerotic plaque progression. One of the increasingly recognized drivers of atherosclerosis is dysfunctional efferocytosis, a homeostatic mechanism responsible for the clearance of dead cells and the resolution of inflammation. In atherosclerosis, the capacity of phagocytes to participate in efferocytosis is hampered, leading to the accumulation of apoptotic and necrotic tissue within the plaque, which results in enlargement of the necrotic core, increased luminal stenosis and plaque inflammation

Macrophage Efferocytosis in Heart.

Efficient clearance of dying cells by efferocytosis is critical for organ development, inflammation resolution, and tissue repair. In the heart, the stakes of efficient clearance are high in that inefficient efferocytosis promotes an inflammatory environment that stresses the health of surrounding nonregenerative cardiomyocytes. Herein, we discuss the fundamental protagonists and therapeutic implications of efferocytosis within the myocardium during cardiac development, myocardial regeneration, and myocardial infarction.

Mechanism of efferocytosis in atherosclerosis.

Atherosclerosis (AS) is a chronic inflammatory vascular disease that occurs in the intima of large and medium-sized arteries with the immune system's involvement. It is a common pathological basis for high morbidity and mortality of cardiovascular diseases. Abnormal proliferation of apoptotic cells and necrotic cells leads to AS plaque expansion, necrotic core formation, and rupture. In the early stage of AS, macrophages exert an efferocytosis effect to engulf and degrade apoptotic, dead, damaged, or senescent cells by efferocytosis, thus enabling the regulation of the organism. In the early s

Interplay between efferocytosis and atherosclerosis.

In an adult human, billions of cells die and turn over daily. During this process, many apoptotic cells are produced and subsequently cleared by phagocytes - a process termed efferocytosis, which plays a critical role in tissue homeostasis. Efferocytosis is an important mechanism in the control of inflammatory processes. Efficient efferocytosis inhibits accumulation of apoptotic cells/debris and maintains homeostasis before the onset of necrosis (secondary necrosis), which promotes inflammation or injury. During efferocytosis, mitochondrial fission and the oxidative stress process are linked t

Efferocytosis is impaired in Gaucher macrophages.

Gaucher disease, the inherited deficiency of lysosomal glucocerebrosidase, is characterized by the presence of glucosylceramide-laden macrophages resulting from impaired digestion of aged erythrocytes or apoptotic leukocytes. Studies of macrophages from patients with type 1 Gaucher disease with genotypes N370S/N370S, N370S/L444P or N370S/c.84dupG revealed that Gaucher macrophages have impaired efferocytosis resulting from reduced levels of p67phox and Rab7. The decreased Rab7 expression leads to impaired fusion of phagosomes with lysosomes. Moreover, there is defective translocation of p67phox

Broad-spectrum antibiotics disrupt homeostatic efferocytosis.

The clearance of apoptotic cells, termed efferocytosis, is essential for tissue homeostasis and prevention of autoimmunity1. Although past studies have elucidated local molecular signals that regulate homeostatic efferocytosis in a tissue2,3, whether signals arising distally also regulate homeostatic efferocytosis remains elusive. Here, we show that large peritoneal macrophage (LPM) display impairs efferocytosis in broad-spectrum antibiotics (ABX)-treated, vancomycin-treated and germ-free mice in vivo, all of which have a depleted gut microbiota. Mechanistically, the microbiota-derived short-c

Measuring efferocytosis by intraperitoneal clearance assay.

Efferocytosis is a critical process by which phagocytes identify, engulf and clear apoptotic cells, thereby preserving immune tolerance and tissue homeostasis. While in vitro assays have advanced our understanding of the molecular mechanisms underlying apoptotic cell clearance, they lack the complexity of the physiological environment. This chapter presents a detailed in vivo approach for assessing efferocytosis by mouse peritoneal macrophages using the intraperitoneal (IP) clearance assay. By administering fluorescently labelled apoptotic Jurkat T cells into the peritoneal cavity, this method

Dual-Fluorescence In Vitro Efferocytosis Assay.

The clearance of dead cells by macrophages, termed "efferocytosis," drives the resolution of inflammation, restricts necrosis, and restores homeostasis. Defects in efferocytosis contribute to many diseases, particularly atherosclerosis. Multiple methods to test efferocytosis by macrophages in vitro exist, but each has distinct disadvantages. This chapter describes an improved method to test apoptotic cell binding and internalization by bone marrow-derived macrophages that takes advantage of the high-affinity between streptavidin and biotin.

Efferocytosis: molecular mechanisms and pathophysiological perspectives.

During the life of a human being, several tons of apoptotic cells and debris are produced. These apoptotic particles should be cleared quickly and accurately from the body, as they may lose their membrane integrity with the probability of leakage of cytotoxic materials and other intracellular antigens into the environment. The action of removing apoptotic particles occurs by a process called efferocytosis. Efferocytosis is a highly regulated balance among a set of find-me, eat-me and don't-eat-me signals. Efferocytosis is accompanied by a suppression of the immune system that can explain its n

Efferocytosis of vascular cells in cardiovascular disease.

Cell death and the clearance of apoptotic cells are tightly regulated by various signaling molecules in order to maintain physiological tissue function and homeostasis. The phagocytic removal of apoptotic cells is known as the process of efferocytosis, and abnormal efferocytosis is linked to various health complications and diseases, such as cardiovascular disease, inflammatory diseases, and autoimmune diseases. During efferocytosis, phagocytic cells and/or apoptotic cells release signals, such as "find me" and "eat me" signals, to stimulate the phagocytic engulfment of apoptotic cells. Primar

Insights of efferocytosis in normal and pathological pregnancy.

Efferocytosis, which is known as the phagocytic clearance of dying cells by professional as well as non-professional phagocytes, including a great number of intracellular/extracellular factors and signals, is interrelated with the immune system, contributing to local and systemic homeostasis, especially in tissues with high constitutive rates of apoptosis. Accumulating studies have indicated that immune dysregulation is associated with the pathogenesis of the female reproductive system, which causes preeclampsia (PE), recurrent spontaneous abortion (RSA), ruptured ectopic pregnancy, and so on.

Macrophage efferocytosis in pregnancy and pregnancy complications.

ABSTRACT: Efferocytosis is a vital process during early placental development that removes apoptotic cells generated through decidualisation and controls inflammation. As pregnancy progresses and levels of apoptotic placental cells increase, macrophage efferocytosis remains essential to maintain a healthy pregnancy. There is mounting evidence that dysfunctional macrophage efferocytosis is implicated in the pathogenesis of multiple pregnancy complications. This includes disorders of malplacentation, such as preeclampsia and fetal growth restriction, wherein impaired spiral artery remodelling an

Efferocytosis and Its Role in Rheumatic Diseases.

Efferocytosis, the process through which phagocytes clear apoptotic cells (ACs), has rapidly emerged as an expanding field of research. This multistep process involves recognizing, binding, internalizing, and degrading ACs. Efferocytosis clears ACs, preventing their secondary necrosis, generating anti-inflammatory mediators, and promoting the resolution of inflammation. Appropriate efferocytosis is essential for preserving immune tolerance and preventing exposure to self-antigens, whereas defective efferocytosis is linked to the onset of various rheumatic diseases. Here, we review the process

Assessing Efferocytosis in Atherosclerotic Lesions In Situ.

Whereas most atherosclerotic lesions are relatively benign, some plaques with large necrotic cores and thin fibrous caps are vulnerable to rupture, which results in many cardiovascular events and sudden death. Defects in the clearance of apoptotic cells, termed "efferocytosis," is the leading cause of necrotic core expansion. This chapter describes a method that identifies macrophage-associated terminal deoxynucleotidyl transferase (TUNEL)-positive events (i.e., efferocytosis events) and TUNEL events free from association with macrophages (i.e., uncleared apoptotic events) in atherosclerotic l

The role of efferocytosis in neuro-degenerative diseases.

Efferocytosis has a critical role in maintaining tissues and organs' homeostasis by removing apoptotic cells. It is essential for human health, and disturbances in efferocytosis may result indifferent illnesses. In case of inadequate clearance of the dead cells, the content in the cells would be released. In fact, it induces some damages to the tissue and leads to the prolonged inflammation, so unsuitable phagocytosis of the apoptotic cells is involved in occurrence as well as expansion of numerous human chronic inflammatory diseases. Studies have shown age dependence of the neuro-degenerative

Drugging the efferocytosis process: concepts and opportunities.

The daily removal of billions of apoptotic cells in the human body via the process of efferocytosis is essential for homeostasis. To allow for this continuous efferocytosis, rapid phenotypic changes occur in the phagocytes enabling them to engulf and digest the apoptotic cargo. In addition, efferocytosis is actively anti-inflammatory and promotes resolution. Owing to its ubiquitous nature and the sheer volume of cell turnover, efferocytosis is a point of vulnerability. Aberrations in efferocytosis are associated with numerous inflammatory pathologies, including atherosclerosis, cancer and infe
