Infections with Babesia canis and Ehrlichia canis in dogs in Zimbabwe.

Of 105 dogs examined at a veterinary hospital in Harare, Zimbabwe, 52 per cent had antibodies reactive with Ehrlichia canis in indirect fluorescent antibody tests, 26 per cent had Babesia canis parasites in peripheral blood smears and 17 per cent had both infections. None of the dogs with serological evidence of ehrlichiosis had typical E canis morulae detectable in blood smears. The infections were regarded as incidental findings not readily related to the reasons for examination in 46 per cent of the dogs with antibodies to E canis and 17 per cent of the dogs with both infections. The most c

Mitochondrial Dysfunction in Ehrlichia canis Infection.

Transmitted by tick vectors, Ehrlichia canis (E. canis) is a Gram-negative, obligate intracellular bacterium of zoonotic concern that infects both canine and human hosts. Its pathogenesis centers on the targeting of mononuclear phagocytes, where it establishes an intracellular niche by suppressing phagolysosomal fusion and evading immune detection, thereby facilitating its replication. E. canis infection also compromises mitochondrial integrity, notably by disrupting the mitochondrial membrane potential (MMP, or ΔΨm), which triggers cellular stress and perturbs critical processes such as autop

Sequential evaluation of dogs naturally infected with Ehrlichia canis, Ehrlichia chaffeensis, Ehrlichia equi, Ehrlichia ewingii, or Bartonella vinsonii.

Historically, disease manifestations in dogs seroreactive to Ehrlichia canis antigens by indirect immunofluorescent antibody testing have been attributed to infection with either E. canis or Ehrlichia ewingii. A 1996 study by Dawson and colleagues provided PCR evidence that healthy dogs from southeastern Virginia could be naturally infected with Ehrlichia chaffeensis. This observation stimulated us to determine which Ehrlichia spp. infected sick dogs that were referred to our hospital from the same region. Based upon PCR amplification with species-specific primers, sick dogs seroreactive to E.

Disseminated protothecosis in a dog coinfected with Hepatozoon canis and Ehrlichia canis.

Protothecosis is a severe, emerging opportunistic infection caused by the saprophytic, achlorophyllous microalgae of the genus Prototheca. Though uncommon, human and animal cases are increasing worldwide, making awareness of this fungal-like pathogen important in both human and veterinary medicine. We report a fatal case of disseminated protothecosis caused by P. wickherhamii in a 10-year-old, spayed, female, mixed-breed collie dog. Clinical signs included uveitis, ulcerated cutaneous nodules on the limbs and face, pododermatitis with nail loss and, lymphadenomegaly in the prescapular and popl

Ehrlichia canis: Is It a Pathogen for Humans and Other Primates?

Ehrlichioses and anaplasmosis are among the most commonly reported tick-borne diseases in humans and some animal species. Ehrlichia canis is the causative agent of canine monocytic ehrlichiosis; it primarily affects dogs and is usually transmitted by Rhipicephalus sanguineus ticks. Some reports suggest that this bacterium is a zoonotic pathogen capable of causing clinical symptoms consistent with human monocytotropic ehrlichiosis caused by Ehrlichia chaffeensis. Non-human primates seem to be susceptible to E. canis-infection, although it is not clear whether the bacterium can cause disease in

Ehrlichia spp. close to Ehrlichia ruminantium, Ehrlichia canis, and "Candidatus Ehrlichia regneryi" linked to heartwater-like disease in Kenyan camels (Camelus dromedarius).

We present findings from an outbreak of a heartwater-like disease in camels that killed at least 2000 adult animals in Kenya in 2016. Clinical signs included excitability, head pressing, aimless wandering, recumbency, and fast breathing followed by death after about 4 days. The observed morbidity in one herd was 40% with an average mortality of 7.5% in animals that received early antibiotic treatments. In untreated adults, the case fatality rate reached 100%. Gross pathology showed pulmonary edema, pleural exudate, hydrothorax, hydropericardium, ascites, enlarged "cooked" liver, nephrosis, and

Multiplex real-time qPCR for the detection of Ehrlichia canis and Babesia canis vogeli.

Ehrlichia canis and Babesia canis vogeli are two tick-borne canine pathogens with a worldwide importance. Both pathogens are transmitted by Rhipicephalus sanguineus, the brown dog tick, which has an increasing global distribution. A multiplex quantitative real-time PCR (qPCR) assay for the simultaneous detection of the tick-borne pathogens E. canis and B. canis vogeli was developed using dual-labeled probes. The target genes were the 16S rRNA of E. canis and the heat shock protein 70 (hsp70) of B. canis vogeli. The canine beta actin (ACTB) gene was used as an internal control gene. The assay w

Two cases of Ehrlichia canis acquired locally in Victoria.

A 10-year-old, female desexed, Yorkshire Terrier (case 1) and a 2-year-old, female entire, Great Dane (case 2) were referred for incidentally detected anaemia, thrombocytopenia, hyperglobulinaemia, pyrexia and neutropenia, respectively. Both dogs lived in central Victoria and had no history of travel outside of the state. Both cases were presumptively diagnosed with immune-mediated disease affecting the bone marrow and treated with immunosuppressive agents (prednisolone, cyclosporine and mycophenolate, respectively). Failure of response to therapy prompted further investigation which confirmed

A serological survey of Ehrlichia canis, Ehrlichia equi, Rickettsia rickettsii, and Borrelia burgdorferi in dogs in Oklahoma.

Serum samples from 259 dogs were tested for antibodies to Ehrlichia canis, Ehrlichia equi, Rickettsia rickettsii, and Borrelia burgdorferi using the indirect fluorescent antibody test. The sera were obtained from submissions to the Oklahoma Animal Disease Diagnostic Laboratory during a 14-month period from June 1986 through July 1987. The rate for positive antibody titers to E. canis was 53%, to E. equi was 33%, to R. rickettsii was 38%, and to B. burgdorferi was 18%. Higher percentages of sera serologically positive to E. canis were found in the spring through the fall months, but there were

Canine vector-borne co-infections: Ehrlichia canis and Hepatozoon canis in the same host monocytes.

The protozoon Hepatozoon canis and the rickettsia Ehrlichia canis are tick-borne pathogens, transmitted by Rhipicephalus sanguineus, which cause canine hepatozoonosis and canine monocytic ehrlichiosis, respectively. Co-infection of the same host monocytes with H. canis and E. canis confirmed by molecular characterization of the infecting agents and quantitative assessment of co-infected cells is described for the first time in three naturally-infected dogs. Blood smear evaluation indicated that at least 50% of the leukocytes infected with H. canis gamonts contained E. canis morulae. Co-infecti

Ehrlichia canis in Rhipicephalus sanguineus ticks in the Ivory Coast.

Canine monocytic ehrlichiosis caused by Ehrlichia canis is distributed globally, but its prevalence in Africa is poorly known. In the study reported herein, 27% of Rhipicephalus sanguineus ticks collected from watchdogs in Abidjan, Ivory Coast, were positive for E. canis using quantitative real-time PCR. A new molecular strategy is proposed that can be used not only for epidemiological study, but also for the diagnosis of canine monocytic ehrlichiosis. Our findings show for the first time the presence of E. canis using molecular tools in the Ivory Coast, providing direct evidence for the prese

Molecular detection and genetic characterization of Ehrlichia canis and Ehrlichia sp. in neotropical primates from Brazil.

The Anaplasmataceae family includes obligate, arthropod-transmitted intracellular bacteria that can be zoonotic and potentially fatal. Studies focusing on the interaction between neotropical primates and the agents of this family are scarce. The present study aimed to identify agents of the Anaplasmataceae family in the whole blood of free-living and captive neotropical primates in the State of Mato Grosso, Central-West Brazil. Thirty-eight samples of six nonhuman primate (NHP) species were collected in seven municipalities and analysed through polymerase chain reaction (PCR), nucleotide seque

Evaluation of antibiotic susceptibilities of Ehrlichia canis, Ehrlichia chaffeensis, and Anaplasma phagocytophilum by real-time PCR.

We determined MICs of antibiotics against Anaplasma phagocytophilum, Ehrlichia chaffeensis, and Ehrlichia canis by real-time quantitative PCR. The doubling times of the organisms were established: 19 h for E. chaffeensis, 26 h for A. phagocytophilum, and 28 h for E. canis. In comparison to the reference method for determining sensitivities, which uses Diff-Quick staining, our PCR assay was very sensitive and specific. We confirmed that doxycycline and rifampin are highly active against these bacteria and found variable susceptibilities to fluoroquinolones; A. phagocytophilum was susceptible, b

Genetic variants of Ehrlichia canis detected in two dogs in Japan.

Ehrlichia canis, the causative agent of canine monocytic ehrlichiosis, infects monocytes and macrophages in dogs and is transmitted mainly by hard ticks Rhipicephalus sanguineus sensu lato. Although E. canis has been reported worldwide, autochthonous E. canis infection in Japan was not confirmed before 2020. In this study, molecular characterization of E. canis detected in two dogs in Tokyo in 2020 and 2023 was conducted. DNA extracted from spleen and blood samples was subjected to polymerase chain reaction (PCR) targeting the 16S ribosomal RNA (rRNA), groEL (heat shock protein), p28 (major ou
